Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,th...Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans.展开更多

OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carci...OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. CONCLUSION: The mechanism accounting for the prolonged survival of HCC-bearing mice treated with JPJDF might be related to the reduction in weight loss and the benign regulation of PTEN, PI3K, and FAK.展开更多

However, in...Soil contamination in agroecosystems remains a global environmental problem. Biochar has been suggested as an organic amendment to alleviate soil pollution, sequester carbon(C), and improve soil fertility. However, information on how bacterial and fungal communities in acidic bulk and rhizosphere soils respond to swine manure and its biochar is still lacking. In this study, biochar and swine manure were applied at two rates of 1.5 and 3 t ha-1in a rice-wheat rotation field to assess how soil characteristics, especially p H and chemical element availability, correlate to compositional variations of bacteria and fungi in bulk and rhizosphere soils. Our results showed that high rates of biochar and manure promoted the bacterial richness in bulk and rhizosphere soils by increasing soil pH and reducing soil arsenic(As) and copper(Cu) availability. Compared with soil As and Cu availability, soil p H had opposite effects on beta diversity of both the bacterial and fungal communities. Specifically, biochar and swine manure applications stimulated the bacterial classes Gemmatimonadetes, Deltaproteobacteria, and Gammaproteobacteria by increasing soil pH and decreasing soil available chemical elements. Opposite trends were observed in fungal communities responding to biochar and manure. For example, biochar restrained the fungal class Eurotiomycetes by decreasing soil As and Cu availability, but manure inhibited Leotiomycetes mainly because of an increase in soil pH and a decrease in soil dissolved organic C. These suggest that both bacterial and fungal communities respond significantly to biochar and manure amendments in both bulk and rhizosphere soils, possibly because of their sensitive adaptation to variations in soil environmental factors, such as pH level and chemical element availability.展开更多

Our patient presented with bilateral l...Angiolipoma of the spine is a benign neoplasm consisting of both mature fatty tissue and abnormal vascular elements, and usually presents with a slow progressive clinical course. Our patient presented with bilateral lower extremity weakness and chest-back numbness. Physical examination revealed adipose elements superficial hypesthesia below the T5 level and analgesia below the T6 level. Magnetic resonance imaging (MRI) scan showed an avidly and heterogeneously enhancing mass which was located in the posterior epidural space. Compression of the thoracic cord by the fusiform mass was seen between T3-T4. During the operation, a flesh pink vascular mass (4.7 cm × 1.0 cm × 1.0 cm) with obscure margin and strong but pliable texture was found in the posterior epidural space extending from T3 to T4. There was no infiltration of the dura or the adjacent bony spine. Histopathological study of the surgical specimen showed a typical angiolipoma. We review the previously documented cases of spinal extradural angiolipomas performed with MRI.展开更多

METHODS: Thirty-two SD rats were divided ran- domly into control group A (given regular feed), model group B (food high in sugar and fat), inter- vention group C (food high in sugar and fat as well as regular doses of Danqidihuang Granules), and in- tervention group D (food high in sugar and fat as well as double doses of Danqidihuang Granules). The interventions were for 8 weeks. Motion, change in color, body weight, and food intake, as well as plasma lipids (including low-density lipopro- tein-cholesterol (LDL-C), high-density lipopro- tein-cholesterol (HDL-C), total cholesterol (TC) and triglyceride (TG), fasting blood glucose (FBG), fast-ing insulin (FINs) levels, insulin sensitivity index (ISI), and insulin resistance index (HOMO-IR) were observed. RESULTS: At the end of the second week of the ex- periment, the appetite and activities of rats in groups B, C and D decreased significantly com- pared with group A. The fur of the rats in those three groups was curly. After the fourth week, the activities, food intake and color of rats in group B were worse than those in groups C and D, but there were no significant differences in weight (P〉0.05). Compared with group A, LDL-C, TC, FBG and HO- MO-IR in model group 13 were increased significant- ly (P〈0.05), whereas the FINs and ISI increased obvi- ously (P〈0.05). The levels of LDL-C and TC in group D was decreased obviously compared with those in group C, and HOMO-IR in group D was less than that in group B CP〈0.05). CONCLUSION: Danqidihuang Granules helped to prevent and improved the insulin resistance of rats.展开更多
Phosphorus(P)is an essential element for plant growth but is often limiting in ecosystems;therefore,improving the P fertilizer use efficiency is important.Biochar and arbuscular mycorrhizal fungi(AMF)may enhance P cyc...Phosphorus(P)is an essential element for plant growth but is often limiting in ecosystems;therefore,improving the P fertilizer use efficiency is important.Biochar and arbuscular mycorrhizal fungi(AMF)may enhance P cycling in paddy soils that contain high content of total P but low content of available P(AP).In this study,the effects of biochar addition and Rhizophagus irregularis inoculation on the organic and inorganic P contents and phosphatase activities in paddy soils,rice seedling growth,and AMF colonization were investigated.Compared with no biochar addition,biochar addition enhanced the percentage of spore germination at day 7,hyphal length,most probable number,and mycorrhizal colonization rate of R.irregularis by 32%,662%,70%,and 28%on average,respectively.Biochar and R.irregularis altered soil P cycling and availability.Biochar and R.irregularis,either individually or in combination,increased soil AP content by 2%–48%.Rice seedlings treated with biochar and R.irregularis produced greater biomass,improved root morphology,and increased nutrient uptake compared with those of the control without biochar and R.irregularis.The results suggest that combined application of biochar and R.irregularis is beneficial to rice cultivation in paddy soils with high content of total P but low content of AP.展开更多

We constructed a time-based gene regulatory network of zebrafish cardiogenesis on the basis of a spatio-temporal neighborhood method. Through the network, specific regulatory pathways and order of gene expression during zebrafish cardiogenesis were obtained. By comparing the order with locations of these genes on chromosomes, we discovered that there exists a reversal phenomenon between the order and order of gene locations. The discovery provides an inherent rule to instruct exploration of gene regulatory relationships. Specifically, the discovery can help to predict if regulatory relationships between genes exist and contribute to evaluating the correctness of discovered gene regulatory relationships.展开更多

Our model achieves high recognition performance,with accuracy rates of 89.4% for the training dataset and 86.4% for the validation dataset.Through statistical analysis of the weight parameter distribution and activation output in the CNN, we find that most of the convolution kernels and hidden layer neurons remain inactive,while only two convolution kernels and two hidden layer neurons play active roles. By examining the weight parameters of connections between the active convolution kernels and the active hidden neurons, we can automatically differentiate various types of El Nino and La Nina,thereby identifying the specific functions of each part of the CNN. We anticipate that this progress will be helpful for future studies on both climate prediction and a deeper understanding of artificial neural networks.展开更多

Applying the kinetic theory of rarefied gases, we construct a two-group kinetic model for the evolution of wealth distribution. Under the continuous trading limit, the Fokker–Planck equation is derived and its steady-state solution is obtained. For the non-Maxwellian collision kernel, we find a suitable redistribution operator to match the taxation. Arbuscular mycorrhi...Tomato(Solanum lycopersicum), an economically important vegetable crop cultivated worldwide, often suffers massive financial losses due to Phytophthora infestans(P. infestans) spread and breakouts. Arbuscular mycorrhiza(AM) fungi mediated biocontrol has demonstrated great potential in plant resistance. However, little information is available on the regulation of mycorrhizal tomato resistance against P. infestans.Therefore, microRNAs(miRNAs) sequencing technology was used to analyse miRNA and their targets in the mycorrhizal tomato after P.infestans infection. Our study showed a lower severity of necrotic lesions in mycorrhizal tomato than in nonmycorrhizal controls. We investigated 35 miRNAs that showed the opposite expression tendency in mycorrhizal and nonmycorrhizal tomato after P. infestans infection when compared with uninfected P. infestans. Among them, miR319c was upregulated in mycorrhizal tomato leaves after pathogen infection. Overexpression of miR319c or silencing of its target gene(TCP1) increased tomato resistance to P. infestans, implying that miR319c acts as a positive regulator in tomato after pathogen infection. Additionally, we examined the induced expression patterns of miR319c and TCP1 in tomato plants exposed to salicylic acid(SA) treatment, and SA content and the expression levels of SA-related genes were also measured in overexpressing transgenic plants. The result revealed that miR319c can not only participates in tomato resistance to P. infestans by regulating SA content, but also indirectly regulates the expression levels of key genes in the SA pathway by regulating TCP1. In this study, we propose a novel mechanism in which the miR319c in mycorrhizal tomato increases resistance to P. infestans.展开更多

Magnetic resonance imaging (MRI) scan showed an avidly and heterogeneously enhancing mass which was located in the posterior epidural space. Compression of the thoracic cord by the fusiform mass was seen between T3-T4. During the operation, a flesh pink vascular mass (4.7 cm × 1.0 cm × 1.0 cm) with obscure margin and strong but pliable texture was found in the posterior epidural space extending from T3 to T4. There was no infiltration of the dura or the adjacent bony spine. Histopathological study of the surgical specimen showed a typical angiolipoma. We review the previously documented cases of spinal extradural angiolipomas performed with MRI.
文摘As prostate cancer is a biologically heterogeneous disease for which a variety of treatment options are available, the major objective of prostate cancer imaging is to achieve more precise disease characterization. In clinical practice, magnetic resonance imaging (MRI) is one of the imaging tools for the evaluation of prostate cancer, the fusion of MRI or dynamic contrast-enhanced MRI (DCE-MRI) with magnetic resonance spectroscopic imaging (MRSI) is improving the evaluation of cancer locafon, size, and extent, while providing an indication of tumor aggressiveness. This review summarizes the role of MRI in the application of prostate cancer and describes molecular MRI techniques (including MRSI and DCE-MRI) for aiding prostate cancer management.
文摘OBJECTIVE: To explore whether the insulin resis- tance (IR) model could be established through feed- ing Sprague-Dawley (SD) rats high-sugar and high-fat diets and to further observe the preven- tive and treatment effects of different doses of Dan- qidihuang Granules in rats. METHODS: Thirty-two SD rats were divided ran- domly into control group A (given regular feed), model group B (food high in sugar and fat), inter- vention group C (food high in sugar and fat as well as regular doses of Danqidihuang Granules), and in- tervention group D (food high in sugar and fat as well as double doses of Danqidihuang Granules). The interventions were for 8 weeks. Motion, change in color, body weight, and food intake, as well as plasma lipids (including low-density lipopro- tein-cholesterol (LDL-C), high-density lipopro- tein-cholesterol (HDL-C), total cholesterol (TC) and triglyceride (TG), fasting blood glucose (FBG), fast-ing insulin (FINs) levels, insulin sensitivity index (ISI), and insulin resistance index (HOMO-IR) were observed. RESULTS: At the end of the second week of the ex- periment, the appetite and activities of rats in groups B, C and D decreased significantly com- pared with group A. The fur of the rats in those three groups was curly. After the fourth week, the activities, food intake and color of rats in group B were worse than those in groups C and D, but there were no significant differences in weight (P〉0.05). Compared with group A, LDL-C, TC, FBG and HO- MO-IR in model group 13 were increased significant- ly (P〈0.05), whereas the FINs and ISI increased obvi- ously (P〈0.05). The levels of LDL-C and TC in group D was decreased obviously compared with those in group C, and HOMO-IR in group D was less than that in group B CP〈0.05). CONCLUSION: Danqidihuang Granules helped to prevent and improved the insulin resistance of rats.

文摘Phosphorus(P)is an essential element for plant growth but is often limiting in ecosystems;therefore,improving the P fertilizer use efficiency is important.Biochar and arbuscular mycorrhizal fungi(AMF)may enhance P cycling in paddy soils that contain high content of total P but low content of available P(AP).In this study,the effects of biochar addition and Rhizophagus irregularis inoculation on the organic and inorganic P contents and phosphatase activities in paddy soils,rice seedling growth,and AMF colonization were investigated.Compared with no biochar addition,biochar addition enhanced the percentage of spore germination at day 7,hyphal length,most probable number,and mycorrhizal colonization rate of R.irregularis by 32%,662%,70%,and 28%on average,respectively.Biochar and R.irregularis altered soil P cycling and availability.Biochar and R.irregularis,either individually or in combination,increased soil AP content by 2%–48%.Rice seedlings treated with biochar and R.irregularis produced greater biomass,improved root morphology,and increased nutrient uptake compared with those of the control without biochar and R.irregularis.The results suggest that combined application of biochar and R.irregularis is beneficial to rice cultivation in paddy soils with high content of total P but low content of AP.

文摘We propose that locations of genes on chromosomes can contribute to the prediction of gene regulatory relationships. We constructed a time-based gene regulatory network of zebrafish cardiogenesis on the basis of a spatio-temporal neighborhood method. Through the network, specific regulatory pathways and order of gene expression during zebrafish cardiogenesis were obtained. By comparing the order with locations of these genes on chromosomes, we discovered that there exists a reversal phenomenon between the order and order of gene locations. The discovery provides an inherent rule to instruct exploration of gene regulatory relationships. Specifically, the discovery can help to predict if regulatory relationships between genes exist and contribute to evaluating the correctness of discovered gene regulatory relationships.

文摘By training a convolutional neural network(CNN) model, we successfully recognize different phases of the El Nino-Southern oscillation. Our model achieves high recognition performance,with accuracy rates of 89.4% for the training dataset and 86.4% for the validation dataset.Through statistical analysis of the weight parameter distribution and activation output in the CNN, we find that most of the convolution kernels and hidden layer neurons remain inactive,while only two convolution kernels and two hidden layer neurons play active roles. By examining the weight parameters of connections between the active convolution kernels and the active hidden neurons, we can automatically differentiate various types of El Nino and La Nina,thereby identifying the specific functions of each part of the CNN. We anticipate that this progress will be helpful for future studies on both climate prediction and a deeper understanding of artificial neural networks.

文摘A non-Maxwellian collision kernel is employed to study the evolution of wealth distribution in a multi-agent society.The collision kernel divides agents into two different groups under certain conditions. Applying the kinetic theory of rarefied gases, we construct a two-group kinetic model for the evolution of wealth distribution. Under the continuous trading limit, the Fokker–Planck equation is derived and its steady-state solution is obtained. For the non-Maxwellian collision kernel, we find a suitable redistribution operator to match the taxation. Our results illustrate that taxation and redistribution have the property to change the Pareto index.

文摘Tomato(Solanum lycopersicum), an economically important vegetable crop cultivated worldwide, often suffers massive financial losses due to Phytophthora infestans(P. infestans) spread and breakouts. Arbuscular mycorrhiza(AM) fungi mediated biocontrol has demonstrated great potential in plant resistance. However, little information is available on the regulation of mycorrhizal tomato resistance against P. infestans.Therefore, microRNAs(miRNAs) sequencing technology was used to analyse miRNA and their targets in the mycorrhizal tomato after P.infestans infection. Our study showed a lower severity of necrotic lesions in mycorrhizal tomato than in nonmycorrhizal controls. We investigated 35 miRNAs that showed the opposite expression tendency in mycorrhizal and nonmycorrhizal tomato after P. infestans infection when compared with uninfected P. infestans. Among them, miR319c was upregulated in mycorrhizal tomato leaves after pathogen infection. Overexpression of miR319c or silencing of its target gene(TCP1) increased tomato resistance to P. infestans, implying that miR319c acts as a positive regulator in tomato after pathogen infection. Additionally, we examined the induced expression patterns of miR319c and TCP1 in tomato plants exposed to salicylic acid(SA) treatment, and SA content and the expression levels of SA-related genes were also measured in overexpressing transgenic plants. The result revealed that miR319c can not only participates in tomato resistance to P. infestans by regulating SA content, but also indirectly regulates the expression levels of key genes in the SA pathway by regulating TCP1. In this study, we propose a novel mechanism in which the miR319c in mycorrhizal tomato increases resistance to P. infestans.
