During obesity treatment,clinicians have been burdened by the ever-increasing number of patients and the side effects of drug treatment.Finding new plant-derived active ingredients as an intervention for obesity has b...During obesity treatment,clinicians have been burdened by the ever-increasing number of patients and the side effects of drug treatment.Finding new plant-derived active ingredients as an intervention for obesity has become a research focus.In this study,Plackett-Burman and Central Composite Design experiments,plant-wide target metabolomics technology,simulated human colon fermentation test,and cell experiment in vitro were successively used to investigate the effects of fermented and unfermented polyphenols(FP and UP,respectively)of blue honeysuckle on obesity.After the optimization of the response surface method,polyphenol content((1878.08±14.53)mg/100 g)increased by 2.63 times after Lactobacillus rhamnosus 6224 and Saccharomyces cerevisiae W5 mixed fermentation.There were 23 significantly different metabolites were observed in FP and UP.Of them,7 different metabolites were significantly and positively correlated with short-chain fatty acids(SCFAs),whereas negatively correlated with lipopolysaccharides(LPS).The contents of these 7 metabolites were significantly increased in FP.Furthermore,the effects of FP and UP on the SCFA biosynthesis pathway were different.Cell experiments revealed that polyphenols can significantly promote the secretion of gastrointestinal endogenous hormones and insulin and inhibit interleukin-6 levels.Linear regression analysis revealed SCFAs had a positive regulatory effect on glycolipid metabolism,whereas LPS had a negative effect.Overall,these crucial findings provide an economical and simple method for obtaining blue honeysuckle polyphenols and laying a theoretical foundation for its application in the therapeutic intervention against obesity.展开更多

Bacillus megaterium HDYM-09 produced pectin lyase that exhibited the highest activity of 2116.71 ± 11.55 U/mL. Bacillus licheniformis HDYM-03 produced pectate lyase that exhibited the highest activity of 611.21 ± 14.54 U/mL. Based on these fi ndings, we constructed four retting systems to degrade the pectin substance. The results showed that the content of galacturonic acid in the mixed system was 529.21 μg/mL, the content of reducing sugar was 98.14 mg/mL, and the weight loss ratio of cells reached 19.49%, which were signifi cantly higher than those in other systems. The mixed system has more advantages, and the utilization rate of degumming was higher, which further ensured that the degumming can be carried out effi ciently and quickly. The mixed system exhibits feasible applications in the fi ber and textile industry.展开更多
In order to improve the yield of β-mannase and to investigate the rules of fermentation production, a high-yield β-mannase producing strain, Bacillus licheniformis HDYM-04, was used to investigate the kinetics model...In order to improve the yield of β-mannase and to investigate the rules of fermentation production, a high-yield β-mannase producing strain, Bacillus licheniformis HDYM-04, was used to investigate the kinetics models based on the optimal fermentation conditions: HDYM-04 strain was fermented at 37℃ for 30 h with agitation speed at 300 r/min and aeration rate at 3 L/min in a 5 L fermenter, the initial addition amount of konjac flour was 2%(w/v), the initial pH of medium was 8.0, and the inoculum concentration was 6.7%(v/v). Three batch fermentation kinetic models were established (cell growth kinetic model, substrate consumption kinetic model, product formation kinetic model) bases on Logistic and Luedeking-Piret equations. To be specific, cell growth kinetic model was dX/dt =0.431X (1- X/ 15.522 ), substrate consumption kinetic model was -ds/dt =1.11 dX/dt +0.000 2 dP/dt +0.000 8X, and product formation kinetic model was dP/dt=133.1 dX +222.87X. The correlation coefficients R^2 of the three equations were 0.990 21, 0.989 08 and 0.988 12, respectively, which indicated a good correlation between experimental values and models. Therefore, the three equations could be used to describe the processes of cell growth, enzyme synthesis and substrate consumption during batch fermentation using B. licheniformis strain HDYM-04. The establishment of batch fermentation kinetic models (cell growth kinetic model, substrate depletion kinetic model, product formation kinetic model) could lay the theoretical foundation and provide practical reference for the applica- tion of HDYM-04 in fermentation industry.展开更多

Methods: Fresh tissues were obtained from patients with BTC (9 cases; 3 cases with grade Ⅰ, 3 cases with grade Ⅱ, 3 cases with grade III) and those with normal bladder mucosa (3 cases) and stored in liquid nitrogen. Total RNA was extracted using TRizol reagent and RNA was quantified and quality control was performed, miRNA probes were labeled with Hy3TM fluorescence, then hybridized with a miRCUR^M array labeling kit. miRNAarrays were scanned and analyzed and the scanned result was validated using reverse transcription-polymerase chain reaction (RT-PCR). Results: In four groups of differentially expressed genes obtained from grade Ⅰ, grade Ⅱ, grade Ⅲ, and grade Ⅰ + grade Ⅱ + grade Ⅲ BTC tissues compared with normal bladder mucosa, hsa-miR-29b-1* was upregulated, and hsa-miR-923 and hsa-miR-300 were downregulated. The hsa-miR-29b-1*, hsa-miR-300, and hsa-miR-923 findings were confirmed by real-time RT-PCR. The first patient was incidentally discovered by health examination, with lower nuc...We reported the clinical and pathologic features of two different types of renal mucinous tubular and spindle cell carcinoma (MTSCC). The first patient was incidentally discovered by health examination, with lower nuclear grade, no part and distant metastasis. The second patient presented with persistence hyperpyrexia, part and distant metastasis, and high nuclear grade. Surgery were both performed successfully. The first patient had no recurrences and no distant metastases. The second patient died of multiple organ failure 3 months postoperatively. Although MTSCC is usually a low potential malignancy carcinoma, high malignancy may occur and lead to a fatal course. So it needs a proper management and prognostication.展开更多
