Stem cells play a crucial role in maintaining tissue regenerative capacity and homeostasis.However,mechanisms associated with stem cell senescence require further investigation.In this study,we conducted a proteomic a...Stem cells play a crucial role in maintaining tissue regenerative capacity and homeostasis.However,mechanisms associated with stem cell senescence require further investigation.In this study,we conducted a proteomic analysis of human dental pulp stem cells(HDPSCs)obtained from individuals of various ages.Our findings showed that the expression of NUP62 was decreased in aged HDPSCs.We discovered that NUP62 alleviated senescence-associated phenotypes and enhanced differentiation potential both in vitro and in vivo.Conversely,the knocking down of NUP62 expression aggravated the senescence-associated phenotypes and impaired the proliferation and migration capacity of HDPSCs.Through RNA-sequence and decoding the epigenomic landscapes remodeled induced by NUP62 overexpression,we found that NUP62 helps alleviate senescence in HDPSCs by enhancing the nuclear transport of the transcription factor E2F1.This,in turn,stimulates the transcription of the epigenetic enzyme NSD2.Finally,the overexpression of NUP62 influences the H3K36me2 and H3K36me3 modifications of anti-aging genes(HMGA1,HMGA2,and SIRT6).Our results demonstrated that NUP62 regulates the fate of HDPSCs via NSD2-dependent epigenetic reprogramming.展开更多

Photoisomerization-induced phase change are important for co-harvesting the latent heat and isomerization energy of azobenzene molecules.Chemically optimizing heat output and energy delivery at alternating temperature...Photoisomerization-induced phase change are important for co-harvesting the latent heat and isomerization energy of azobenzene molecules.Chemically optimizing heat output and energy delivery at alternating temperatures are challenging because of the differences in crystallizability and isomerization.This article reports two series of asymmetrically alkyl-grafted azobenzene(Azo-g),with and without a methyl group,that have an optically triggered phase change.Three exothermic modes were designed to utilize crystallization enthalpy(△H_(c))and photothermal(isomerization)energy(△H_(p))at different temperatures determined by the crystallization.Azo-g has high heat output(275-303 J g^(-1))by synchronously releasing△H_(c)and△H_(p)over a wide temperature range(-79℃to 25℃).We fabricated a new distributed energy utilization and delivery system to realize a temperature increase of 6.6℃at a temperature of-8℃.The findings offer insight into selective utilization of latent heat and isomerization energy by molecular optimization of crystallization and isomerization processes.展开更多
With the aid of non-contact measurements of vibrating surfaces through laser scanning,operating deflection shapes(ODSs)with high spatial resolutions can be used to graphically characterize damage in plane structures.A...With the aid of non-contact measurements of vibrating surfaces through laser scanning,operating deflection shapes(ODSs)with high spatial resolutions can be used to graphically characterize damage in plane structures.Although numerous damage identification approaches relying on laser-measured ODSs have been developed for plate-type structures,they cannot be directly applied to circular cylinders due to the gap between equations of motions of plates and circular cylinders.To fill this gap,a novel approach is proposed in this study for damage identification of circular cylinders.Damage-induced discontinuities of the derivatives of ODSs can be used to gra-phically manifest the occurrence of the damage,and characterize the location and size of the damage.The approach is experimentally validated on a specimen of the circular cylinder component,whose out-of-plane ODSs in an inspection region are acquired through laser scanning using a scanning laser vibrometer.The results suggest that the occurrence,location,and size of the internal damage of the circular cylinder can be identified.展开更多

Sixty adult mice were randomly divided into four groups (mild, moderate and severe salt deficiency groups and control) during ...Low salt intake is associated with depression, but the experimental evidence is not clear. Sixty adult mice were randomly divided into four groups (mild, moderate and severe salt deficiency groups and control) during the seasonal splitting from winter to spring. The mice in the control group were fed with standard ordinary diet (salt content 0.26%), while in the mild, moderate and severe groups, the mice were fed with 10%, 30% and 50% salt deficient feedstuff, lasting for approximately 3 months. The results showed that the salt content of feed was negatively correlated with the food intake and body weight of mice. The sucrose preference test found that only the mild salt deficiency group had no differ-ence between the beginning and the end of the experiment, and the other three groups in-cluding the control, showed a significant decrease. These results suggest that dietary salt content has an impact on the food intake and body weight of mice and is associated with the emergence of depressive-like behavior. Furthermore, the seasonal splitting from winter to spring may also have a differential synergistic effect on the change of depression-like behavior associated with low salt intake in mice.展开更多
Self-regulating heating and self-powered flexibility are pivotal for future wearable devices.However,the low energy-conversion rate of wearable devices at low temperatures limits their application in plateaus and othe...Self-regulating heating and self-powered flexibility are pivotal for future wearable devices.However,the low energy-conversion rate of wearable devices at low temperatures limits their application in plateaus and other environments.This study introduces an azopolymer with remarkable semicrystallinity and reversible photoinduced solid-liquid transition ability that is obtained through copolymerization of azoben-zene(Azo)monomers and styrene.A composite of one such copolymer with an Azo:styrene molar ratio of 9:1(copolymer is denoted as PAzo9:1-co-polystyrene(PS))and nylon fabrics(NFs)is prepared(composite is denoted as PAzo9:1-co-PS@NF).PAzo9:1-co-PS@NF exhibits hydrophobicity and high wear resistance.Moreover,it shows good responsiveness(0.624 s^(−1))during isomerization under solid ultraviolet(UV)light(365 nm)with an energy density of 70.6 kJ kg^(−1).In addition,the open-circuit voltage,short-circuit current and quantity values of PAzo9:1-co-PS@NF exhibit small variations in a temperature range of−20℃ to 25℃ and remain at 170 V,5 μA,and 62 nC,respectively.Notably,the involved NFs were cut and sewn into gloves to be worn on a human hand model.When the model was exposed to both UV radiation and friction,the temperature of the finger coated with PAzo9:1-co-PS was approximately 6.0°C higher than that of the other parts.Therefore,developing triboelectric nanogenerators based on the in situ photothermal cycles of Azo in wearable devices is important to develop low-temperature self-regulating heating and self-powered flexible devices for extreme environments.展开更多

Cell growth was investigated after enhancing expression of SATB1. Wound-healing assay and Transwell assay were used to investigate the impact of SATB1 on migratory and invasive abilities of SW480 cells in vitro . Nude mice that received subcutaneous implantation or lateral tail vein were used to study the effects of SATB1 on tumor growth or metastasis in vivo . RESULTS: SATB1 was over-expressed in CRC tissues and CRC cell lines. SATB1 promotes cell proliferation and cell cycle progression in CRC SW480 cells. SATB1 over-expression could promote cell growth in vivo . In addition, SATB1 could significantly raise the ability of cell migration and invasion in vitro and promote the ability of tumor metastasis in vivo . SATB1 could up-regulate matrix metalloproteases 2, 9, cyclin D1 and vimentin, meanwhile SATB1 could down-regulate E-cadherin in CRC. CONCLUSION: SATB1 acts as a potential growth and metastasis promoter in CRC. SATB1 may be useful as a therapeutic target for CRC.展开更多

To establish a hyperhomocysteinemic pregnant rat model, 2 mL of DL-homocysteine was administered daily by intraperitoneal injection at a dose of 200 mg/kg from day 10 to day 19 of gestation. Folic acid was administered by intragastric administration at a dose of 20 mg/kg during the period of preeclampsia induction. Results showed that systolic blood pressure, proteinuria/creatinine ratio, and plasma homocysteine levels in the hyperhomocysteinemic pregnant rats increased significantly, and that body weight and brain weight of rat pups significantly decreased. Folic acid supplementation markedly reversed the above-mentioned abnormal changes of hyperhomocysteinemic pregnant rats and rat pups. These findings suggest that folic acid can alleviate the symptoms of hyperhomocysteinemia- induced preeclampsia in pregnant rats without influencing brain development of rat pups.展开更多
The establishment of the upper-level South Asian high (SAH) over the Indo-China Peninsula (ICP) during late boreal spring and its possible causes are investigated using long-term NCEP-NCAR and ERA-40 reanalysis an...The establishment of the upper-level South Asian high (SAH) over the Indo-China Peninsula (ICP) during late boreal spring and its possible causes are investigated using long-term NCEP-NCAR and ERA-40 reanalysis and satellite-observed OLR data. Results show that, from early March to mid-April, deep convection stays south of -6°N over the northern Sumatran islands. As the maximum solar radiation moves over the latitudes of the ICP (10-20°N) in late April, the air over the ICP becomes unstable. It ascends over the ICP and descends over the adjacent waters to the east and west. This triggers deep convection over the ICP that induces large latent heating and strong updrafts and upper-level divergence, leading to the formation of an upper-level anticyclonic circulation and the SAH over the ICE During early to mid-May, deep convection over the ICP intensifies and extends northwards to the adjacent waters. Strong latent heating from deep convection enhances and maintains the strong updrafts and upper-level divergence, and the SAH is fully established by mid-May. Thus, the seasonal maximum solar heating and the land-sea contrast around the ICP provide the basic conditions for deep convection to occur preferentially over the ICP, which leads to the formation of the SAH over the ICP from late April to mid-May. Simulations using RegCM4 also indicate that the diabatic heating over the ICP is conducive to the generation and development of upper-level anticyclonic circulation, which leads to an earlier establishment of the SAH.展开更多

This study established a pregnant rat model of hyperhomocysteinemia, in which blood plasma homocysteine concentrations were twice or three times greater than that of normal pregnant rats. TUNEL revealed an increase in the number of apoptotic cells in the frontal cortex of pregnant rats with hyperhomocysteinemia. In addition, immunohistochemical staining detected activated nuclear factor-KB-positve cells in the frontal cortex. Reverse transcription-PCR detected that mRNA expression of the anti-apoptotic gene bcl-2 diminished in the frontal cortex. In situ hybridization and western blotting revealed that N-methyi-D- aspartate receptor 1 mRNA and protein expression was upregulated in the frontal cortex and hippocampus. These results indicate that hyperhomocysteinemia can induce brain cell apoptosis, increase nerve excitability, and promote the occurrence of preeclampsia in pregnant rats.展开更多

Thus, these pregnant rats experienced preeclampsia-like syndromes such as hyper-tension and proteinuria. Simultaneously, metabotropic glutamate receptor 1 mRNA and protein ex-pressions were upregulated in the rat hippocampus. These findings indicate that increased expres-sion of metabotropic glutamate receptor 1 promotes the occurrence of high-fat diet-induced pree-clampsia in pregnant rats.展开更多
Purpose-It is quite universal for high-speed turnouts to be exposed to the wear of the stock rail of the switch rail during the service process.The wear will cause the change of railhead profile and the relative posit...Purpose-It is quite universal for high-speed turnouts to be exposed to the wear of the stock rail of the switch rail during the service process.The wear will cause the change of railhead profile and the relative positions of the switch rail and the stock rail,which will directly affect the wheel-rail contact state and wheel load transition when a train passes the turnout and will further impose serious impacts on the safety and stability of train operation.The purpose of this paper is to provide suggestions for wear management of high-speed turnout.Design/methodology/approach-The actual wear characteristics of switch rails of high-speed turnouts in different guiding directions were studied based on the monitoring results on site;the authorized wear limits for the switch rails of high-speed turnout were studied through derailment risk analysis and switch rail strength analysis.Findings-The results show that:the major factor for the service life of a curved switch rail is the lateral wear.The wear characteristics of the curved switch rail of a facing turnout are significantly different from those of a trailing turnout.To be specific,the lateral wear of the curved switch rail mainly occurs in the narrower section at its front end for a trailing turnout,but in the wider section at its rear end when for a facing turnout.The maximum lateral wear of a dismounted switch rail from a trailing turnout is found on the 15-mm wide section and is 3.9 mm,which does not reach the specified limit of 6 mm.For comparison,the lateral wear of a dismounted switch rail from a facing turnout is found from the 35-mm wide section to the full-width section and is greater than 7.5 mm,which exceeds the specified limit.Based on this,in addition to meeting the requirements of maintenance rules,the allowed wear of switch rails of high-speed turnout shall be so that the dangerous area with a tangent angle of wheel profile smaller than 43.68 will not contact the switch rail when the wheel is lifted by 2 mm.Accordingly,the lateral wear limit at the 5-mm wide section of the curved switch rail shall be reduced from 6 mm(as specified)to 3.5 mm.Originality/value-The work in this paper is of reference significance to the research on the development law of rail wear in high-speed turnout area and the formulation of relevant standards.展开更多

Meth...Objective This study aimed to investigate the expression and significance of PIN2/TERF1 interacting, telomerase inhibitor 1(Pin X1) and human telomerase reverse transcriptase(h TERT) in basal cell carcinoma(BCC). Methods Real-time polymerase chain reaction and immunohistochemistry were performed to quantify the m RNA expressions and integrated optical density(IOD), respectively, of Pin X1 and h TERT in BCC specimens(n = 30), as well as in normal skin specimens(n = 15). Results The m RNA expression level and IOD of Pin X1 in the BCC samples were both significantly lower than those in the control specimens(P < 0.05). Conversely, the m RNA expression level and IOD of h TERT in BCC were both significantly higher than that in the control samples(P < 0.05). The correlation between the expression levels of Pin X1 and h TERT showed no statistical significance(P > 0.05). Conclusion Downregulation of Pin X1 and upregulation of h TERT expression may be associated with the activation and maintenance of telomerases in the induction of BCC.展开更多

文摘Self-regulating heating and self-powered flexibility are pivotal for future wearable devices.However,the low energy-conversion rate of wearable devices at low temperatures limits their application in plateaus and other environments.This study introduces an azopolymer with remarkable semicrystallinity and reversible photoinduced solid-liquid transition ability that is obtained through copolymerization of azoben-zene(Azo)monomers and styrene.A composite of one such copolymer with an Azo:styrene molar ratio of 9:1(copolymer is denoted as PAzo9:1-co-polystyrene(PS))and nylon fabrics(NFs)is prepared(composite is denoted as PAzo9:1-co-PS@NF).PAzo9:1-co-PS@NF exhibits hydrophobicity and high wear resistance.Moreover,it shows good responsiveness(0.624 s^(−1))during isomerization under solid ultraviolet(UV)light(365 nm)with an energy density of 70.6 kJ kg^(−1).In addition,the open-circuit voltage,short-circuit current and quantity values of PAzo9:1-co-PS@NF exhibit small variations in a temperature range of−20℃ to 25℃ and remain at 170 V,5 μA,and 62 nC,respectively.Notably,the involved NFs were cut and sewn into gloves to be worn on a human hand model.When the model was exposed to both UV radiation and friction,the temperature of the finger coated with PAzo9:1-co-PS was approximately 6.0°C higher than that of the other parts.Therefore,developing triboelectric nanogenerators based on the in situ photothermal cycles of Azo in wearable devices is important to develop low-temperature self-regulating heating and self-powered flexible devices for extreme environments.

文摘AIM: To evaluate the expression of special AT-rich sequence-binding protein 1 (SATB1 ) gene in colorectal cancer and its role in colorectal cancer cell proliferation and invasion.METHODS: Immunohistochemistry was used to detect the protein expression of SATB1 in 30 colorectal cancer (CRC) tissue samples and pair-matched adjacent nontumor samples. Cell growth was investigated after enhancing expression of SATB1. Wound-healing assay and Transwell assay were used to investigate the impact of SATB1 on migratory and invasive abilities of SW480 cells in vitro . Nude mice that received subcutaneous implantation or lateral tail vein were used to study the effects of SATB1 on tumor growth or metastasis in vivo . RESULTS: SATB1 was over-expressed in CRC tissues and CRC cell lines. SATB1 promotes cell proliferation and cell cycle progression in CRC SW480 cells. SATB1 over-expression could promote cell growth in vivo . In addition, SATB1 could significantly raise the ability of cell migration and invasion in vitro and promote the ability of tumor metastasis in vivo . SATB1 could up-regulate matrix metalloproteases 2, 9, cyclin D1 and vimentin, meanwhile SATB1 could down-regulate E-cadherin in CRC. CONCLUSION: SATB1 acts as a potential growth and metastasis promoter in CRC. SATB1 may be useful as a therapeutic target for CRC.

文摘Folic acid participates in the metabolism of homocysteine and lowers plasma homocysteine levels directly or indirectly. To establish a hyperhomocysteinemic pregnant rat model, 2 mL of DL-homocysteine was administered daily by intraperitoneal injection at a dose of 200 mg/kg from day 10 to day 19 of gestation. Folic acid was administered by intragastric administration at a dose of 20 mg/kg during the period of preeclampsia induction. Results showed that systolic blood pressure, proteinuria/creatinine ratio, and plasma homocysteine levels in the hyperhomocysteinemic pregnant rats increased significantly, and that body weight and brain weight of rat pups significantly decreased. Folic acid supplementation markedly reversed the above-mentioned abnormal changes of hyperhomocysteinemic pregnant rats and rat pups. These findings suggest that folic acid can alleviate the symptoms of hyperhomocysteinemia- induced preeclampsia in pregnant rats without influencing brain development of rat pups.

文摘The establishment of the upper-level South Asian high (SAH) over the Indo-China Peninsula (ICP) during late boreal spring and its possible causes are investigated using long-term NCEP-NCAR and ERA-40 reanalysis and satellite-observed OLR data. Results show that, from early March to mid-April, deep convection stays south of -6°N over the northern Sumatran islands. As the maximum solar radiation moves over the latitudes of the ICP (10-20°N) in late April, the air over the ICP becomes unstable. It ascends over the ICP and descends over the adjacent waters to the east and west. This triggers deep convection over the ICP that induces large latent heating and strong updrafts and upper-level divergence, leading to the formation of an upper-level anticyclonic circulation and the SAH over the ICE During early to mid-May, deep convection over the ICP intensifies and extends northwards to the adjacent waters. Strong latent heating from deep convection enhances and maintains the strong updrafts and upper-level divergence, and the SAH is fully established by mid-May. Thus, the seasonal maximum solar heating and the land-sea contrast around the ICP provide the basic conditions for deep convection to occur preferentially over the ICP, which leads to the formation of the SAH over the ICP from late April to mid-May. Simulations using RegCM4 also indicate that the diabatic heating over the ICP is conducive to the generation and development of upper-level anticyclonic circulation, which leads to an earlier establishment of the SAH.

文摘Hyperhomocysteinemia is an important risk factor for preeclampsia-eclampsia. This study established a pregnant rat model of hyperhomocysteinemia, in which blood plasma homocysteine concentrations were twice or three times greater than that of normal pregnant rats. TUNEL revealed an increase in the number of apoptotic cells in the frontal cortex of pregnant rats with hyperhomocysteinemia. In addition, immunohistochemical staining detected activated nuclear factor-KB-positve cells in the frontal cortex. Reverse transcription-PCR detected that mRNA expression of the anti-apoptotic gene bcl-2 diminished in the frontal cortex. In situ hybridization and western blotting revealed that N-methyi-D- aspartate receptor 1 mRNA and protein expression was upregulated in the frontal cortex and hippocampus. These results indicate that hyperhomocysteinemia can induce brain cell apoptosis, increase nerve excitability, and promote the occurrence of preeclampsia in pregnant rats.

文摘Changes in neurotransmitter levels in the brain play an important role in epilepsy-like attacks after pregnancy-induced preeclampsia-eclampsia. Metabotropic glutamate receptor 1 participates in the onset of lipid metabolism disorder-induced preeclampsia. Pregnant rats were fed with a high-fat diet for 20 days. Thus, these pregnant rats experienced preeclampsia-like syndromes such as hyper-tension and proteinuria. Simultaneously, metabotropic glutamate receptor 1 mRNA and protein ex-pressions were upregulated in the rat hippocampus. These findings indicate that increased expres-sion of metabotropic glutamate receptor 1 promotes the occurrence of high-fat diet-induced pree-clampsia in pregnant rats.

文摘Purpose-It is quite universal for high-speed turnouts to be exposed to the wear of the stock rail of the switch rail during the service process.The wear will cause the change of railhead profile and the relative positions of the switch rail and the stock rail,which will directly affect the wheel-rail contact state and wheel load transition when a train passes the turnout and will further impose serious impacts on the safety and stability of train operation.The purpose of this paper is to provide suggestions for wear management of high-speed turnout.Design/methodology/approach-The actual wear characteristics of switch rails of high-speed turnouts in different guiding directions were studied based on the monitoring results on site;the authorized wear limits for the switch rails of high-speed turnout were studied through derailment risk analysis and switch rail strength analysis.Findings-The results show that:the major factor for the service life of a curved switch rail is the lateral wear.The wear characteristics of the curved switch rail of a facing turnout are significantly different from those of a trailing turnout.To be specific,the lateral wear of the curved switch rail mainly occurs in the narrower section at its front end for a trailing turnout,but in the wider section at its rear end when for a facing turnout.The maximum lateral wear of a dismounted switch rail from a trailing turnout is found on the 15-mm wide section and is 3.9 mm,which does not reach the specified limit of 6 mm.For comparison,the lateral wear of a dismounted switch rail from a facing turnout is found from the 35-mm wide section to the full-width section and is greater than 7.5 mm,which exceeds the specified limit.Based on this,in addition to meeting the requirements of maintenance rules,the allowed wear of switch rails of high-speed turnout shall be so that the dangerous area with a tangent angle of wheel profile smaller than 43.68 will not contact the switch rail when the wheel is lifted by 2 mm.Accordingly,the lateral wear limit at the 5-mm wide section of the curved switch rail shall be reduced from 6 mm(as specified)to 3.5 mm.Originality/value-The work in this paper is of reference significance to the research on the development law of rail wear in high-speed turnout area and the formulation of relevant standards.
文摘Objective This study aimed to investigate the expression and significance of PIN2/TERF1 interacting, telomerase inhibitor 1(Pin X1) and human telomerase reverse transcriptase(h TERT) in basal cell carcinoma(BCC). Methods Real-time polymerase chain reaction and immunohistochemistry were performed to quantify the m RNA expressions and integrated optical density(IOD), respectively, of Pin X1 and h TERT in BCC specimens(n = 30), as well as in normal skin specimens(n = 15). Results The m RNA expression level and IOD of Pin X1 in the BCC samples were both significantly lower than those in the control specimens(P < 0.05). Conversely, the m RNA expression level and IOD of h TERT in BCC were both significantly higher than that in the control samples(P < 0.05). The correlation between the expression levels of Pin X1 and h TERT showed no statistical significance(P > 0.05). Conclusion Downregulation of Pin X1 and upregulation of h TERT expression may be associated with the activation and maintenance of telomerases in the induction of BCC.
