AIM:To describe the prevalence and demographic characteristics of corneal blindness in an urban and rural region of Ningxia,located in the northwest part of China.METHODS:A stratified,randomized sampling procedure was...AIM:To describe the prevalence and demographic characteristics of corneal blindness in an urban and rural region of Ningxia,located in the northwest part of China.METHODS:A stratified,randomized sampling procedure was employed in the study,including urban and rural area of all age group.Visual acuity,anterior segment and ocular fundus were checked.Related factor of corneal disease,including age,gender,education status,ethnic group,location and occupation,were identified according to uniform customized protocol.An eye was defined to be corneal blindness if the visual acuity was【20/400 due to a corneal disease.RESULTS:Three thousand individuals(1290 from urban area and 1710 from rural area)participated in the investigation,with a response rate of 80.380%.The prevalence of corneal blindness was 0.023%in both eyes and 0.733%in at least one eye.The blindness in at least one eye with varied causes was present in 106participants(3.533%)and in bilateral eyes in 34participants(1.133%).The corneal diseases accounted for 20.754%of blindness in at least one eye and 20.588%of bilateral blindness.The prevalence of corneal disease was higher in older and Han ethnic group,especially those who occupied in agriculture and outdoor work.People with corneal blindness were more likely to be older and lower education.Rural population were more likely to suffer from bilateral corneal blindness than the urban population in≥59-year group(χ2=6.716,P=0.019).Infectious,trauma and immune corneal disease were the three leading causes of corneal disease.Trauma cornealdisease was more likely leading to blindness in one eye.However,infectious and immune corneal diseases make more contribution to the bilateral corneal blindness.CONCLUSION:Corneal blindness is a significant burden of in Ningxia population,encompassing a variety of corneal infections and trauma;the majority of those were avoidable.Health promotion strategies and good hygienic conditions have to be developed.展开更多
To study the effects of the deformation degree and cooling rate on the microstructure and phase transformation temperature for the B 1500HS steel, the samples were heated at 900 ℃ for 5 min, compressed by 10, 20, 30 ...To study the effects of the deformation degree and cooling rate on the microstructure and phase transformation temperature for the B 1500HS steel, the samples were heated at 900 ℃ for 5 min, compressed by 10, 20, 30 and 40% at the strain rate of 0.1 s^-1, and then cooled down at the rates of 50, 40, 25, 20 and 15 ℃/s by the thermo-mechanical simulator, respectively. The start and finish temperatures of the phase transformation were determined by the tangent method, and the volume fraction of the phase transformation was ascertained by the level principle according to the dilatometric curves. The volume fraction of the retained austenite was determined by X-ray diffraction. The results show that the volume fraction of the bainite rises with an increase in the deformation degree as the cooling rate is lower than the critical rate. At the same cooling rate, the phase transformation temperature rises with an increase in the deformation degree, and the sizes of both the martensite and bainite phases reduce due to the austenite grain refinement induced by the deformation. The volume fraction of the retained austenite reduces as the deformation degree increases. The critical cooling rate of the undeformed samples is approximately 25 ℃/s and the critical cooling rate rises as the deformation degree increases.展开更多

METHODS: Patients received the comprehensive ophthalmic evaluation. Genomic DNA was extracted from peripheral blood and subjected to Sure Select Human All Exon 6+ UTR exon capture kit. The exons were sequenced as 100 base paired reads on Illumina HiS eq2500 system. Only mutations that resulted in a change in amino acid sequence were selected. A pattern of inheritance of the RP family was aligned to identified causal mutation.RESULTS: We analysed the data of WES information from XLRP family. The analysis revealed a hemizygous large genomic deletion of RPGR c.29113 del was responsible for this XLRP. The gross deletion lead to a frame-shift mutation and generate stop codon at 7 animo acid behind Asp(D10 Afs*7), which would serious truncate RPGR protein. The novel frame-shift mutation was found to segregate with retinitis pigmentosa(RP) phenotype in this family. Bilateral myopia was present on the male patients, but carrier female showed unilateral myopia without RP.CONCLUSION: Our study identifies a novel frame-shift mutation of RPGR in a Chinese family, which would expand the spectrum of RPGR mutations. The geno-phenotypic analysis reveals a correlation between RP and myopia. Although exact mechanism of RP related myopia is still unknown, but the novel frame-shift mutation will give our hit on studying the molecular pathogenesis of RP and myopia.展开更多

To lessen myocardial ischemia-reperfusion injury after cardiac arrest and CPR,it is important to reduce energy consumption and to increase energy supply in the myocardium.This study aimed to observe changes of cell apoptosis and expression of Bcl-2 and Bax proteinon the myocardium after CPR in rats, and the protective effects of different doses of exogenousphosphocreatine (creatine phosphate, CP) on them.METHODS: A total of 32 male adult Sprague-Dawley rats were randomly divided into 4 groups:control group (group A), CPR group (group B), low-dose CP group (group C, CP 0.5 g/kg at beginning ofCPR and 1.0 g/kg at 2 hours after CPR) and high-dose CP group (group D, CP 1.0 g/kg at beginning ofCPR and 2.0 g/kg at 2 hours after CPR). Cardiac arrest was induced by asphyxiation and CPR startedat 7 minutes after asphyxiation in groups B, C and D. Myocardium samples were taken at 24 hoursafter CPR. Cardiomycytic apoptosis was detected by the TdT-mediated dUTP-biotin nick end labeling(TUNEL) method. The expression of Bcl-2 and Bax protein was measured by immunohistochemistry.RESULTS: Cardiomyocytic apoptosis index (AI) and expression of Bcl-2 and Bax proteinincreased more significantly in groups B, C and D than in group A (P〈0.01), but Bcl-2/Bax ratiosignificantly decreased (P〈0.01). Cardiomyocytic AI and expression of Bcl-2 and Bax proteindecreased more significantly in groups C and D than in group B (P〈0.01), but Bcl-2/Bax ratioincreased more significantly (P〈0.01). AIM:To screen mutations in the retinitis pigmentosa 1(RP1) gene and the rhodopsin(RHO) gene in Chinese patients with retinitis pigmentosa sine pigmento(RPSP)and describe the genotype-phenotype relationship of the muta...AIM:To screen mutations in the retinitis pigmentosa 1(RP1) gene and the rhodopsin(RHO) gene in Chinese patients with retinitis pigmentosa sine pigmento(RPSP)and describe the genotype-phenotype relationship of the mutations.·METHODS:Twenty affected,unrelated Chinese individuals with RPSP(4 autosomal dominant RPSP,12autosomal recessive RPSP and 4 unknown inheritance pattern) were recruited between 2009 and 2012.The clinical features were determined by complete ophthalmologic examinations.Polymerase chain reaction(PCR) and direct DNA sequencing were used to screen the entire coding region and splice junctions of the RP1gene and the RHO gene.The cosegregation analysis and population frequency studies were performed for patients with identified mutations.·RESULTS:Five variants in the RP1 gene and one in the RHO gene were detected in 20 probands.Four missense changes(rs444772,rs446227,rs414352,rs441800) and one non-coding variant(rs56340615) were common SNPs and none of them showed a significant relationship with RPSP.A missense mutation p.R1443W was identified in the RP1 gene in three affected individuals from a family with autosomal dominant RPSP and was found to cosegregate with the phenotype in this family,suggestive of pathogenic.In addition,population frequency analysis showed the p.R1443W mutation was absent in 300 healthy controls.·CONCLUSION:The identification of p.R1443W mutationcosegregating in a family with autosomal dominant RPSP highlights an atypical phenotype of the RP1 gene mutation,while RHO gene is not associated with the pathogenesis of RPSP in this study.To our knowledge,this is the fist mutation identified to associate with RPSP.展开更多

Neuroglobin (NGB) is an oxygen-binding heme protein found in the brain with a protection role against ischemic-hypoxic brain injury. Hemin is an effective activator of neuroglobin. This study was undertaken to assess the effect of hemin on expression of neuroglobin (NGB) in the cerebral cortex, neuro-defi cit score (NDS) and pathological changes after cardiopulmonary resuscitation (CPR) in rats.METHODS: A total of 120 male Sprague-Dawley (SD) rats were randomly divided into a control group (A), a CPR group (B) and a Hemin group (C). The animal model of cardiac arrest (CA) induced by asphyxia and CPR was established. NGB expression in the cerebral cortex with immunohistochemistry, NDS and pathological changes in the cerebral cortex were examined at 3, 6, 12, 24 hours after recovery of spontaneous circulation (ROSC) in each group. Experimental data were treated as one-factor analysis of variance and the Tukey test.RESULTS: In comparison with group A, NGB expression was increased signifi cantly at 12 and 24 hours after ROSC (P〈0.05 or P〈0.01), NDS was decreased signifi cantly at each time point after ROSC (P〈0.01), and pathological changes were severe at each time point after ROSC in group B. In comparison with group A, NGB expression was increased signifi cantly at 6, 12, 24 hours after ROSC (P〈0.05 or P〈0.01), NDS was decreased signifi cantly at 3, 6, 12 hours after ROSC (P〈0.01) in group C. In comparison with group B, NGB expression was increased signifi cantly at 12 and 24 hours after ROSC, NDS was increased signifi cantly at 12 and 24 hours after ROSC, and pathological changes were milder in group C.CONCLUSION: There were increased NGB expression in the cerebral cortex, decreased NDS, and severe pathological changes after CPR in rats. Hemin treatment up-regulated expression of NGB, improved NDS, mitigated pathological changes, and alleviated cerebral injury after CPR.展开更多
BACKGROUND:The outcome of cardiopulmonary resuscitation(CPR) may depend on a variety of factors related to patient status or resuscitation management.To evaluate the factors influencing the outcome of CPR after cardia...BACKGROUND:The outcome of cardiopulmonary resuscitation(CPR) may depend on a variety of factors related to patient status or resuscitation management.To evaluate the factors influencing the outcome of CPR after cardiac arrest(CA) will be conducive to improve the effectiveness of resuscitation.Therefore,a study was designed to assess these factors in the emergency department(ED) of a city hospital.METHODS:A CPR registry conforming to the Utstein-style template was conducted in the ED of the First Affiliated Hospital of Wenzhou Medical College from January 2005 to December 2011.The outcomes of CPR were compared in various factors groups.The primary outcomes were rated to return of spontaneous circulation(ROSC),24-hour survival,survival to discharge and discharge with favorable neurological outcomes.Univariate analysis and multivariable logistic regression analysis were performed to evaluate factors associated with survival.RESULTS:A total of 725 patients were analyzed in the study.Of these patients,187(25.8%) had ROSC,100(13.8%) survived for 24 hours,48(6.6%) survived to discharge,and 23(3.2%) survived to discharge with favorable neurologic outcomes.A logistic regression analysis demonstrated that the independent predictors of ROSC included traumatic etiology,first monitored rhythms,CPR duration,and total adrenaline dose.The independent predictors of 24-hour survival included traumatic etiology,cardiac etiology,first monitored rhythm and CPR duration.Previous status,cardiac etiology,first monitored rhythms and CPR duration were included in independent predictors of survival to discharge and neurologically favorable survival to discharge.CONCLUSIONS:Shockable rhythms,CPR duration ≤15 minutes and total adrenaline dose≤5 mg were favorable predictors of ROSC,whereas traumatic etiology was unfavorable.Cardiac etiology,shockable rhythms and CPR duration ?15 minutes were favorable predictors of 24-hour survival,whereas traumatic etiology was unfavorable.Cardiac etiology,shockable rhythms,CPR duration ≤15 minutes were favorable predictors of survival to discharge and neurologically favorable survival to discharge,but previous terminal illness or multiple organ failure(MOF) was unfavorable.展开更多
BACKGROUND:Spontaneous isolated superior mesenteric artery(SMA)dissection(SISMAD)is a rare cause of abdominal pain.The aim of the study is to investigate the role of a new parameter,the ratio of the SMA diameter to th...BACKGROUND:Spontaneous isolated superior mesenteric artery(SMA)dissection(SISMAD)is a rare cause of abdominal pain.The aim of the study is to investigate the role of a new parameter,the ratio of the SMA diameter to the superior mesenteric vein(SMV)diameter(SMA/SMV)based on non-enhanced computed tomography(CT),in the early diagnosis of SISMAD.METHODS:In a registry study from December 2013 to June 2021,97 abdominal pain SISMAD patients(SISMAD group)admitted to our hospital were enrolled.Meanwhile,the matched sex and age abdominal pain non-SISMAD patients at 1:2 were collected in reverse chronological order as the control group.Student’s t-test,Wilcoxon rank-sum test,and Chi-square test were used to compare differences between the SISMAD and control groups.Med Calc was used to generate receiver operating characteristic(ROC)curve.RESULTS:A total of 291 abdominal pain patients,including 97 SISMAD patients and 194 nonSISMAD patients,were included in the current study.The maximum SMA diameter,perivascular exudation,and SMA/SMV based on non-enhanced CT were significant between the two groups(all P<0.05).ROC curves showed that for the maximum SMA diameter,the area under the curve(AUC),cut-off,sensitivity,and specificity were 0.926,9.80,93.8%,and 79.4%,respectively.For SMA/SMV,its AUC,cut-off,sensitivity,and specificity were 0.956,0.83,88.7%,and 92.3%,respectively.The diagnostic efficiency of SMA/SMV was better than that of the maximum SMA diameter(P<0.05).The combined parameters of SMA/SMV and maximum SMA diameter had the best diagnostic efficiency(AUC=0.970).CONCLUSION:SMA/SMV may be a potential marker for SISMAD.展开更多
Pulmonary fibrosis(PF)is a chronic and progressively fatal disease,but clinically available therapeutic drugs are limited due to efficacy and side effects.The possible mechanism of pulmonary fibrosis includes the dama...Pulmonary fibrosis(PF)is a chronic and progressively fatal disease,but clinically available therapeutic drugs are limited due to efficacy and side effects.The possible mechanism of pulmonary fibrosis includes the damage of alveolar epithelial cells II(AEC2),and activation of immune cells such as macrophages.The ions released from bioceramics have shown the activity in stimulating soft tissue derived cells such as fibroblasts,endothelia cells and epithelia cells,and regulating macrophage polarization.Therefore,this study proposes an“ion therapy”approach based on the active ions of bioceramic materials,and investigates the therapeutic effect of bioactive ions derived from calcium silicate(CS)bioceramics on mouse models of pulmonary fibrosis.We demonstrate that silicate ions significantly reduce pulmonary fibrosis by simultaneously regulating the functions of AEC2 and macrophages.This result suggests potential clinical applications of ion therapy for lung fibrosis.展开更多

Cardiomyocytic AI and expression of Bcl-2 and Bax proteindecreased more signifi cantly in group D than in group C (P〈0.05), but Bcl-2/Bax ratio increased moresignifi cantly (P〈0.05).CONCLUSION: Exogenous phosphocreatine, especially at a large dose, could inhibitcardiomyocytic apoptosis and alleviate myocardial injury after CPR in rats.

文摘AIM:To screen mutations in the retinitis pigmentosa 1(RP1) gene and the rhodopsin(RHO) gene in Chinese patients with retinitis pigmentosa sine pigmento(RPSP)and describe the genotype-phenotype relationship of the mutations.·METHODS:Twenty affected,unrelated Chinese individuals with RPSP(4 autosomal dominant RPSP,12autosomal recessive RPSP and 4 unknown inheritance pattern) were recruited between 2009 and 2012.The clinical features were determined by complete ophthalmologic examinations.Polymerase chain reaction(PCR) and direct DNA sequencing were used to screen the entire coding region and splice junctions of the RP1gene and the RHO gene.The cosegregation analysis and population frequency studies were performed for patients with identified mutations.·RESULTS:Five variants in the RP1 gene and one in the RHO gene were detected in 20 probands.Four missense changes(rs444772,rs446227,rs414352,rs441800) and one non-coding variant(rs56340615) were common SNPs and none of them showed a significant relationship with RPSP.A missense mutation p.R1443W was identified in the RP1 gene in three affected individuals from a family with autosomal dominant RPSP and was found to cosegregate with the phenotype in this family,suggestive of pathogenic.In addition,population frequency analysis showed the p.R1443W mutation was absent in 300 healthy controls.·CONCLUSION:The identification of p.R1443W mutationcosegregating in a family with autosomal dominant RPSP highlights an atypical phenotype of the RP1 gene mutation,while RHO gene is not associated with the pathogenesis of RPSP in this study.To our knowledge,this is the fist mutation identified to associate with RPSP.
文摘BACKGROUND: Despite a large amount of resuscitation research, the survival rate after cardiac arrest remains low, and brain injury is the key issue. Neuroglobin (NGB) is an oxygen-binding heme protein found in the brain with a protection role against ischemic-hypoxic brain injury. Hemin is an effective activator of neuroglobin. This study was undertaken to assess the effect of hemin on expression of neuroglobin (NGB) in the cerebral cortex, neuro-defi cit score (NDS) and pathological changes after cardiopulmonary resuscitation (CPR) in rats.METHODS: A total of 120 male Sprague-Dawley (SD) rats were randomly divided into a control group (A), a CPR group (B) and a Hemin group (C). The animal model of cardiac arrest (CA) induced by asphyxia and CPR was established. NGB expression in the cerebral cortex with immunohistochemistry, NDS and pathological changes in the cerebral cortex were examined at 3, 6, 12, 24 hours after recovery of spontaneous circulation (ROSC) in each group. Experimental data were treated as one-factor analysis of variance and the Tukey test.RESULTS: In comparison with group A, NGB expression was increased signifi cantly at 12 and 24 hours after ROSC (P〈0.05 or P〈0.01), NDS was decreased signifi cantly at each time point after ROSC (P〈0.01), and pathological changes were severe at each time point after ROSC in group B. In comparison with group A, NGB expression was increased signifi cantly at 6, 12, 24 hours after ROSC (P〈0.05 or P〈0.01), NDS was decreased signifi cantly at 3, 6, 12 hours after ROSC (P〈0.01) in group C. In comparison with group B, NGB expression was increased signifi cantly at 12 and 24 hours after ROSC, NDS was increased signifi cantly at 12 and 24 hours after ROSC, and pathological changes were milder in group C.CONCLUSION: There were increased NGB expression in the cerebral cortex, decreased NDS, and severe pathological changes after CPR in rats. Hemin treatment up-regulated expression of NGB, improved NDS, mitigated pathological changes, and alleviated cerebral injury after CPR.

文摘BACKGROUND:The outcome of cardiopulmonary resuscitation(CPR) may depend on a variety of factors related to patient status or resuscitation management.To evaluate the factors influencing the outcome of CPR after cardiac arrest(CA) will be conducive to improve the effectiveness of resuscitation.Therefore,a study was designed to assess these factors in the emergency department(ED) of a city hospital.METHODS:A CPR registry conforming to the Utstein-style template was conducted in the ED of the First Affiliated Hospital of Wenzhou Medical College from January 2005 to December 2011.The outcomes of CPR were compared in various factors groups.The primary outcomes were rated to return of spontaneous circulation(ROSC),24-hour survival,survival to discharge and discharge with favorable neurological outcomes.Univariate analysis and multivariable logistic regression analysis were performed to evaluate factors associated with survival.RESULTS:A total of 725 patients were analyzed in the study.Of these patients,187(25.8%) had ROSC,100(13.8%) survived for 24 hours,48(6.6%) survived to discharge,and 23(3.2%) survived to discharge with favorable neurologic outcomes.A logistic regression analysis demonstrated that the independent predictors of ROSC included traumatic etiology,first monitored rhythms,CPR duration,and total adrenaline dose.The independent predictors of 24-hour survival included traumatic etiology,cardiac etiology,first monitored rhythm and CPR duration.Previous status,cardiac etiology,first monitored rhythms and CPR duration were included in independent predictors of survival to discharge and neurologically favorable survival to discharge.CONCLUSIONS:Shockable rhythms,CPR duration ≤15 minutes and total adrenaline dose≤5 mg were favorable predictors of ROSC,whereas traumatic etiology was unfavorable.Cardiac etiology,shockable rhythms and CPR duration ?15 minutes were favorable predictors of 24-hour survival,whereas traumatic etiology was unfavorable.Cardiac etiology,shockable rhythms,CPR duration ≤15 minutes were favorable predictors of survival to discharge and neurologically favorable survival to discharge,but previous terminal illness or multiple organ failure(MOF) was unfavorable.

文摘BACKGROUND:Spontaneous isolated superior mesenteric artery(SMA)dissection(SISMAD)is a rare cause of abdominal pain.The aim of the study is to investigate the role of a new parameter,the ratio of the SMA diameter to the superior mesenteric vein(SMV)diameter(SMA/SMV)based on non-enhanced computed tomography(CT),in the early diagnosis of SISMAD.METHODS:In a registry study from December 2013 to June 2021,97 abdominal pain SISMAD patients(SISMAD group)admitted to our hospital were enrolled.Meanwhile,the matched sex and age abdominal pain non-SISMAD patients at 1:2 were collected in reverse chronological order as the control group.Student’s t-test,Wilcoxon rank-sum test,and Chi-square test were used to compare differences between the SISMAD and control groups.Med Calc was used to generate receiver operating characteristic(ROC)curve.RESULTS:A total of 291 abdominal pain patients,including 97 SISMAD patients and 194 nonSISMAD patients,were included in the current study.The maximum SMA diameter,perivascular exudation,and SMA/SMV based on non-enhanced CT were significant between the two groups(all P<0.05).ROC curves showed that for the maximum SMA diameter,the area under the curve(AUC),cut-off,sensitivity,and specificity were 0.926,9.80,93.8%,and 79.4%,respectively.For SMA/SMV,its AUC,cut-off,sensitivity,and specificity were 0.956,0.83,88.7%,and 92.3%,respectively.The diagnostic efficiency of SMA/SMV was better than that of the maximum SMA diameter(P<0.05).The combined parameters of SMA/SMV and maximum SMA diameter had the best diagnostic efficiency(AUC=0.970).CONCLUSION:SMA/SMV may be a potential marker for SISMAD.

文摘Pulmonary fibrosis(PF)is a chronic and progressively fatal disease,but clinically available therapeutic drugs are limited due to efficacy and side effects.The possible mechanism of pulmonary fibrosis includes the damage of alveolar epithelial cells II(AEC2),and activation of immune cells such as macrophages.The ions released from bioceramics have shown the activity in stimulating soft tissue derived cells such as fibroblasts,endothelia cells and epithelia cells,and regulating macrophage polarization.Therefore,this study proposes an“ion therapy”approach based on the active ions of bioceramic materials,and investigates the therapeutic effect of bioactive ions derived from calcium silicate(CS)bioceramics on mouse models of pulmonary fibrosis.We demonstrate that silicate ions significantly reduce pulmonary fibrosis by simultaneously regulating the functions of AEC2 and macrophages.This result suggests potential clinical applications of ion therapy for lung fibrosis.

Since the pioneer description in 1980, only a few cases of GSD-associated PH have been reported. Here, we reported a Chinese case with informed consent by the patient and the guardian.
