Background Maternal nutrition significantly influences offspring development.This study investigated the effects of maternal or post-weaning cinnamaldehyde(CA)supplementation in sows and their offspring on reproductive performance and health.Sixty sows,selected based on body condition score and parity,were randomly allocated to control or CA(500 mg/kg)diets from d 107 of gestation to d 24 of lactation.At weaning,128 piglets were assigned to four groups(n=8)based on weight and source litter for a 21-d experiment.The four groups were CON-CON(both sow and piglet on CON),CON-CA(sow on CON,piglet on CA),CA-CON(sow on CA,piglet on CON),and CA-CA(both sow and piglet on CA).Results Maternal CA supplementation tended to improve body weight(+15%,P=0.09)and average daily gain(+21%,P=0.07)of suckling piglets,along with increased levels of milk IgG(P=0.01)and IgM(P=0.02),colostrum crude fat(P=0.01),and plasma glutathione peroxidase(GSH-Px)activity(P=0.02)at farrowing.Moreover,maternal CA supplementation significantly improved plasma antioxidant capacity,expressions of intestinal barrier and antiinflammatory genes,and gut microbiota structure of piglets at the end of suckling.Additionally,maternal CA supplementation increased the apparent total tract digestibility(ATTD)of crude protein(P<0.01),gross energy(GE;P=0.03),and dry matter(P=0.01),improved jejunal sucrase activity(P<0.01),villus height(P=0.03),the ratio of villi height to crypt depth(P=0.02),and the expressions of intestinal barrier and anti-inflammatory genes in post-weaning piglets.Furthermore,post-weaning CA supplementation tended to decrease diarrhea scores of piglets during d 14–21 and increased the ATTD of GE(P=0.02),activities of jejunal sucrase(P=0.02),plasma catalase(P=0.01),and total superoxide dismutase(P<0.01)in piglets.Conclusion Maternal CA supplementation tended to increase the growth rate and weaning weight of suckling piglets,associated with improved antioxidant capacity and milk composition.Moreover,maternal CA supplementation or post-weaning CA supplementation improved nutrient digestibility,redox status,and intestinal function-related parameters of weaned piglets. 展开更多

Background While maternal proline(Pro)supplementation has demonstrated efficacy in enhancing placental angiogenesis and farrowing efficiency in swine,its regulatory role in fetal skeletal muscle ontogeny remains undefined.This study systematically evaluated the temporal-specific impacts of dietary Pro supplementation during critical phases of fetal myogenesis(encompassing primary myofiber formation and secondary myofiber hyperplasia)on offspring muscle development.A total of 120 sows with similar farrowing schedules were assigned to three groups:CON(basal diet),ST-Pro(0.5%Pro supplementation during secondary myofiber formation period,from d 60 gestation to farrowing),LT-Pro(0.5%Pro supplementation spanning primary and secondary myofiber formation period:from d 20 gestation to farrowing).Results LT-Pro group significantly increased the longissimus dorsi(LD)muscle mass per unit body weight in newborn piglets compared to CON group(P<0.05),while no such effect was observed in the ST-Pro group.Metabolomic profiling revealed elevated Pro,lysine,and tryptophan levels in the LD muscle of LT-Pro group piglets,accompanied by reduced branched-chain amino acids(BCAAs;leucine,isoleucine,and valine)in both serum and muscle(P<0.05).Histological analysis demonstrated a 45.74%increase in myofiber cross-sectional area in the LT-Pro group(P<0.05).At the molecular level,LT-Pro group piglets exhibited upregulated mRNA expression levels of myogenic regulatory genes(MYOD1,MYF6)and the cell cycle accelerator CCND1(P<0.05),coupled with activation of the STAT3 signaling pathway(phosphorylated STAT3 protein increased by 2.53-fold,P<0.01).Furthermore,Pro supplementation enhanced oxidative metabolism,evidenced by elevated mitochondrial biogenesis markers(the mRNA expression levels of PPARGC1A,OPA1,and SQSTM1)and a 61.58%increase in succinate dehydrogenase activity(P<0.05).Notably,LT-Pro group piglets showed a selective shift toward slow-twitch oxidative fibers,with both MyHC1 mRNA and protein expression levels significantly upregulated(P<0.05),while the mRNA expression levels of MyHCIIb showed no significant change.Conclusions This study identified the primary fiber formation period as a critical window.Supplementation with Pro during G20–114 reprogrammed offspring skeletal muscle development through STAT3-CCND1-mediated myoblast proliferation,enhanced mitochondrial bioenergetics,and oxidative fiber specification.However,no such effects were observed during G60–114.These findings propose maternal Pro intervention as a novel strategy to enhance muscle yield and metabolic efficiency in swine production,with potential applications for improving meat quality traits linked to oxidative muscle phenotypes. 展开更多

Background Boars fed a mixed form of inorganic and organic iron in excess of the NRC recommended levels still develop anemia,which suggested that the current level and form of iron supplementation in boar diets may be inappropriate.Therefore,56 healthy Topeka E line boars aged 15–21 months were randomly divided into 5 groups:basal diet supplemented with 96 mg/kg ferrous sulfate(FeSO_(4))and 54 mg/kg glycine chelated iron(Gly-Fe,control);80 mg/kg or 115 mg/kg Gly-Fe;80 mg/kg or 115 mg/kg methionine hydroxyl analogue chelated iron(MHA-Fe,from CalimetFe)for 16 weeks.The effects of dietary iron supplementation with different sources and levels on semen quality in boars were investigated.Results 1)Serum Fe and hemoglobin concentrations were not affected by reduced dietary iron levels in the 80 mg/kg or 115 mg/kg Gly-Fe and MHA-Fe groups compared with the control group(P>0.05).2)Serum interleukin-6(IL-6)and sperm malondialdehyde(MDA)levels in the 80 mg/kg or 115 mg/kg MHA-Fe groups were lower than those in the control group(P<0.05),and higher serum superoxide dismutase levels and lower MDA levels in the 115 mg/kg MHA-Fe group(P<0.05).3)Boars in the 80 mg/kg or 115 mg/kg Gly-Fe and MHA-Fe groups had lower serum hepcidin(P<0.01),ferritin(P<0.05),and transferrin receptor(P<0.01)concentrations,and boars in the 115 mg/kg MHA-Fe group had higher seminal plasma Fe concentrations compared with the control group.4)Boars in the 80 mg/kg and 115 mg/kg MHA-Fe groups had lower abnormal sperm rate and in situ oscillating sperm ratio compared to the control group at weeks 12 and/or 16 of the trial.However,the effect of Gly-Fe on improving semen quality in boars was not evident.5)Serum IL-6 level was positively correlated with hepcidin concentration(P<0.05),which in turn was significantly positively correlated with abnormal sperm rate(P<0.05).Furthermore,significant correlations were also found between indicators of iron status and oxidative stress and semen quality parameters.Conclusions Dietary supplementation with 80 mg/kg or 115 mg/kg MHA-Fe did not induce iron deficiency,but rather reduced serum inflammatory levels and hepcidin concentration,alleviated oxidative stress,increased body iron utilization,and improved semen quality in adult boars. 展开更多

Background:This study aimed to investigate the hydration properties of different-source fibrous materials by com-paring their water-binding capacity(WBC),water swelling capacity(WSC),viscosity,and in vivo effects of selected samples on growth performance,nutrient digestibility,diarrhea,and intestinal health in weaned piglets.Methods:A total of 13 commercially available fibrous materials were first compared in chemical composition and in vitro hydration property.Subsequently,40 weaned piglets were randomized to five experimental dietary groups(8 piglets per group):control diet(a basal diet without dietary fiber,CON),basal diet supplemented with 5%microcrys-talline cellulose(MCC),5%wheat bran(WB),5%Moringa oleifera leaf powder(MOLP),or 5%sugar beet pulp(SBP),fol-lowed by analyzing their growth performance and diarrhea rate in a 28-d experiment.After the feeding experiment,anaesthetized piglets were killed,and their intestinal and colon content or plasma samples were analyzed in nutrient digestibility,intestinal morphology,intestinal barrier,short-chain fatty acids(SCFAs),and bacterial population.Results:In vitro studies showed low hydration properties for WB and MCC,while medium hydration properties for MOLP and SBP.In vivo studies indicated that compared with medium hydration property groups,low hydration prop-erty groups showed(1)exacerbated diarrhea,impaired intestinal health,and reduced apparent fecal digestibility of dry matter,gross energy,acid detergent fiber,and neutral detergent fiber;(2)decreased SCFAs concentration and rela-tive levels of Lactobacillus and Bifidobacterium,but increased levels of Escherichia coli and Brachyspira hyodysenteriae in colon contents.Additionally,SBP showed optimal performance in reducing diarrhea and increasing SCFAs produc-tion.Correlation analysis revealed a positive correlation of fiber hydration properties with in vitro SCFAs production,and diarrhea index and nutrient digestibility were negatively and positively correlated with SCFAs levels in the colon contents of weaned piglets,respectively.Conclusions:Different-source dietary fibers varied in their hydration properties and impacts on diarrhea,microbial composition and SCFAs production in weaned piglets.WB and MCC could exacerbate diarrhea and impair nutrient digestibility,probably because their low hydration properties were detrimental to gut microbial homeostasis and fermentation.Our findings provide new ideas for rational use of fiber resources in weaned piglets. 展开更多

Maternal proline(Pro)supplementation enhances fetal survival and placental development in mice.However,the effect of Pro on fetal and placental development in gilts remains to be investigated,particularly in the context of obesity-induced impaired pregnancy.Here,we investigated the effect of dietary Pro on fetal and placental development in obese gilts.Exp.1:On day 60 of gestation,48 gilts with similar delivery times were selected and followed up until delivery to determine the relationship between maternal obesity,litter performance,and Pro abundance in term placentae.The results showed that impaired reproductive performance was associated with body condition parameters and inadequate placental Pro availability of gilts.Exp.2:A total of 114 gilts were then used in a 2×3 factorial design to investigate the interaction between body condition(factor I:normal or obese gilts)and dietary Pro levels(factor II:low[0.89%,L-Pro],medium[1.39%,M-Pro],and high[1.89%,H-Pro])on farrowing performance and placental angiogenesis.This resulted in six treatment combinations:normal-L-Pro,obese-L-Pro,normal-M-Pro,obese-M-Pro,normal-H-Pro,and obese-H-Pro.The effective number of replicates per group was 17,21,19,21,18,and 18,respectively(1 gilt per replicate).The results showed that increasing Pro intake increased piglet birth weight(P=0.001),litter weight(P<0.001),placental efficiency(P=0.036)and placental vascular density(P<0.001),and decreased the number of mummified fetuses(P=0.001),the rate of low-birth-weight piglets(P=0.005),and the rate of invalid piglets(P=0.029).Interaction effects were observed between body condition and dietary Pro levels on piglet birth weight(P=0.046),within-litter birth weight variation(P=0.012),and placental vascular density(P=0.007).Moreover,the beneficial effect of Pro on farrowing performance may be related to the improvement of sirtuin 1-superoxide dismutase 2-mitochondrial reactive oxygen species axis homeostasis and angiogenesis in the placenta.Our results suggest that gestation diets need to provide adequate Pro to meet the needs of fetal and placental development,particularly in obese gilts. 展开更多

Abnormal placental angiogenesis is associated with the occurrence of intrauterine growth restriction(IUGR)in piglets,and effective treatment strategies against this occurrence remain to be explored.Adenosine has been reported to play an important role in angiogenesis,but its role in placental angio-genesis is still unknown.Here,we investigated the effect of dietary adenosine supplementation on IUGR occurrence in piglets by analyzing the role of adenosine in placental angiogenesis for Normal and IUGR piglets.Specifically,88 sows were allotted to 2 treatments(n=44)and fed a basal diet supplemented with 0%or 0.1%of adenosine from day 65 of gestation until farrowing,followed by collecting the placental samples of Normal and IUGR piglets,and recording their characteristics.The results showed that adenosine supplementation increased the mean birth weight of piglets(P<0.05)and placental efficiency(P<0.05),while decreasing the IUGR piglet rate(P<0.05).Expectedly,the placenta for IUGR neonates showed a down-regulated vascular density(P<0.05)and angiogenesis as evidenced by the expression level of vascular cell adhesion molecule-1(VCAM1)(P<0.05).Notably,dietary adenosine supplementation promoted angiogenesis(P<0.05)both in the Normal and IUGR placenta.More importantly,the expression level of adenosine A2a receptor(ADORA2A)was lower(P<0.05)in the IUGR placenta than in Normal placenta,whereas adenosine treatment could significantly increase ADORA2A expression,and also had an interaction effect between factors IUGR and Ado.Collectively,placentae for IUGR piglets showed impaired angiogenesis and down-regulated expression level of ADORA2A,while dietary adenosine supplementation could activate ADORA2A expression,improve the placental angio-genesis,and ultimately decrease the occurrence of IUGR in piglets. 展开更多
