混菌发酵制备柏树籽多肽饲料添加剂及其抗氧化作用研究

目的:探讨绝经后女性宫腔积液分流管理中血清人附睾蛋白4(HE4)、糖类抗原125(CA125)检测价值。方法:选取2019年1月—2019年12月门诊确诊的绝经后宫腔积液300例,均经阴道超声、宫腔镜检查及诊断性刮宫术确诊。测定血清HE4、CA125水平及... 目的:探讨绝经后女性宫腔积液分流管理中血清人附睾蛋白4(HE4)、糖类抗原125(CA125)检测价值。方法:选取2019年1月—2019年12月门诊确诊的绝经后宫腔积液300例,均经阴道超声、宫腔镜检查及诊断性刮宫术确诊。测定血清HE4、CA125水平及阳性率,评估HE4、CA125鉴别诊断宫颈积液者良恶性疾病的效能。结果:300例中原发子宫内膜癌33例,良性子宫内膜疾病267例;子宫内膜癌者HE4(126.93±42.79 pmol/L)、CA125(41.49±17.27 U/ml)水平均高于良性疾病者(73.98±26.75 pmol/L、22.22±10.64 U/ml),HE4阳性率(33.3%)、CA125阳性率(63.6%)均高于良性疾病者(0,12.0%)(均P<0.05);阴道出血发生率在子宫良恶性疾病者间未见差异(P>0.05)。ROC曲线显示,HE4联合CA125诊断子宫内膜癌的AUC=0.948,HE4敏感性最高为81.8%,CA125特异性最高为91.8%。结论:血清HE4联合CA125鉴别诊断绝经后宫腔积液患者良恶性病变效能较高,有利于其分流管理。 展开更多

基于Kano模型的训练伤康复护理需求分析与干预策略优化

目的了解训练伤患者康复护理需求的属性并进行优化排序,探究影响因素,为制定针对性的优化策略提供理论依据。方法采用便利抽样法抽取333名训练伤康复患者,基于Kano模型设计问卷并进行调查,分析训练伤康复患者护理需求的Kano属性,通过重... 目的了解训练伤患者康复护理需求的属性并进行优化排序,探究影响因素,为制定针对性的优化策略提供理论依据。方法采用便利抽样法抽取333名训练伤康复患者,基于Kano模型设计问卷并进行调查,分析训练伤康复患者护理需求的Kano属性,通过重要性和满意度调查,进行重要性-满意度(IPA)矩阵分析,确定患者需求改进的优先序。结果单因素分析显示,训练伤患者的性别、年龄、文化程度、病程、家庭收入对满意度的影响有高度统计学意义(P<0.01)。根据Kano属性分类,10个条目中9个属于期望型,1个属于魅力型;各需求满意度均分4.024~4.625,重要性均分4.393~4.679;IPA矩阵图显示,5项服务位于第1象限,3项服务位于第2象限,2项服务位于第3象限。分析发现家人/朋友参与和疼痛控制的重要性被训练伤患者相对低估,下一步应重点关注。结论Kano-IPA方法能够有效分析训练伤患者的需求属性,并确定优先改进顺序,为资源优化配置提供方向,有助于提升康复护理质量和训练伤患者满意度。 展开更多

目的研究慢性肾病(chronic kidney disease,CKD)患者血清Klotho,人附睾蛋白4(HE4)和尿调节素(UMOD)表达水平在评估疾病预后中的参考价值。方法对120例CKD患者及100例健康体检对照者血清同时应用ELISA检测Klotho和HE4,电化学发光法检测U... 目的研究慢性肾病(chronic kidney disease,CKD)患者血清Klotho,人附睾蛋白4(HE4)和尿调节素(UMOD)表达水平在评估疾病预后中的参考价值。方法对120例CKD患者及100例健康体检对照者血清同时应用ELISA检测Klotho和HE4,电化学发光法检测UMOD表达水平,分析各因子与肾小球滤过率(GFR)之间的关系。患者随访12个月后,根据GFR下降幅度分为恶化组和稳定组,比较两组之间血清因子变化差异,采用Kaplan-Meier生存曲线分析血清因子与预后的关系。结果CKD患者血清Klotho,UMOD,GFR和HE4表达水平与健康对照组相比,差异均有统计学意义(t=22.492,58.235,23.668和33.341,均P<0.05)。Klotho,UMOD与GFR呈现正相关性(r=0.682,0.582,P<0.001),HE4与GFR呈负相关性(r=-0.627,P<0.001)。随访结束时血清Klotho,UMOD水平明显低于随访前,HE4水平明显高于随访前,恶性组患者的变化幅度与稳定组相比差异有统计学意义(t=13.263,18.600和16.645,均P<0.0015)。随访12个月后患者死亡45例(37.50%),高Klotho,UMOD水平组不良预后风险低于低Klotho,UMOD水平组,高HE4水平组不良预后风险高于低HE4水平组,差异均有统计学意义(χ^(2)=4.511,3.960,7.484;P=0.034,0.047,0.006)。结论慢性肾病患者血清中Klotho,HE4,UMOD均存在异常表达,其中血清Klotho,UMOD水平降低以及HE4水平升高可增加肾功能恶化风险,三种血清因子联合可提高对预后评估的参考价值。 展开更多

鹿茸菇多糖的提取工艺优化及其体外抗氧化作用研究

为实现鹿茸菇多糖的高效制备,本研究系统比较了水提法、酶解辅助水提醇沉法、微波水提法、超声水提法及冻融水提法对鹿茸菇多糖提取率与多糖含量的影响。在单因素试验基础上,通过正交试验法优化鹿茸菇多糖提取工艺,并通过DPPH·和AB... 为实现鹿茸菇多糖的高效制备,本研究系统比较了水提法、酶解辅助水提醇沉法、微波水提法、超声水提法及冻融水提法对鹿茸菇多糖提取率与多糖含量的影响。在单因素试验基础上,通过正交试验法优化鹿茸菇多糖提取工艺,并通过DPPH·和ABTS^(+)·清除试验探究该多糖的抗氧化活性。结果显示,水提法、酶解辅助水提醇沉法、微波水提法、超声水提法、冻融水提法多糖提取率分别为(4.63±0.40)%、(19.38±1.90)%、(7.32±0.47)%、(8.57±0.48)%、(11.46±0.50)%,多糖含量分别为(41.87±1.39)%、(77.03±1.76)%、(44.78±0.76%)、(42.57±1.36)%、(54.59±2.22)%,其中酶解辅助水提醇沉法效果最佳,故选用酶解辅助水提醇沉法提取鹿茸菇多糖。在此基础上,通过单因素与正交试验对酶解时间、酶浓度、固液比及提取次数4个关键参数进行优化,确定最佳工艺条件为提取3次,酶解时间150 min,酶浓度2.5%,固液比1∶20(g∶mL)。在此工艺下进行3次重复验证试验,鹿茸菇多糖含量分别为83.19%、82.08%和81.96%,多糖提取率分别为20.18%、21.34%和20.96%,工艺稳定可靠。体外抗氧化活性检测发现,由此制备的鹿茸菇多糖对DPPH·和ABTS^(+)·均表现出良好的自由基清除活性,且清除效果呈现剂量依赖关系,质量浓度为4.0 mg/mL的鹿茸菇多糖对两种自由基的清除率分别高达80.17%和88.42%。综上所述,本研究建立了一种高效的鹿茸菇多糖提取工艺,并证实该多糖具有显著的抗氧化潜力,为鹿茸菇在功能性食品中的开发与应用提供了重要依据。 展开更多

背景:相较于2D培养技术,3D培养技术能够更准确地模拟细胞自然生长的体内环境,成为基础科学研究和转化医学领域的关键焦点。目的:通过3D培养技术构建预血管化的成骨类器官。方法:从同一人体脂肪组织中分别分离提取血管基质成分、脂肪间... 背景:相较于2D培养技术,3D培养技术能够更准确地模拟细胞自然生长的体内环境,成为基础科学研究和转化医学领域的关键焦点。目的:通过3D培养技术构建预血管化的成骨类器官。方法:从同一人体脂肪组织中分别分离提取血管基质成分、脂肪间充质干细胞,通过流式细胞学鉴定细胞组分。将血管基质成分、脂肪间充质干细胞分别接种于96孔U型板内,培养3 d后血管基质成分自组装成型、脂肪间充质干细胞成球,培养第14天,通过苏木精-伊红染色与细胞骨架染色观察球体内部结构和细胞形态;培养0,7,14,21 d,Presto Blue实验检测细胞活性;培养3 d后分别进行成骨、成软骨与成脂诱导分化,诱导14 d后,茜素红染色观察成骨分化,阿利新蓝染色观察成软骨分化,油红O染色观察成脂分化,免疫荧光染色共定位成骨诱导后CD31(内皮细胞标志物)、RUNX2(早期成骨标志物)、CD44(间充质干细胞标志物)、Ⅰ型胶原(晚期成骨标志物)、过氧化物酶体增殖物激活受体γ(成脂标志物)的表达,qRT-PCR检测成骨诱导后RUNX2、骨桥蛋白、Ⅰ型胶原、Sp7转录因子、CD31和血管内皮生长因子mRNA表达。结果与结论:①流式细胞分析表明,血管基质成分包含脂肪间充质干细胞和内皮细胞等多种细胞类型,脂肪间充质干细胞球主要由脂肪间充质干细胞构成;②苏木精-伊红染色及细胞骨架染色显示,相对于脂肪间充质干细胞球,血管基质成分类器官结构更贴近天然组织,细胞排列有序,细胞外基质更丰富;细胞活性测试显示,脂肪间充质干细胞球培养7 d的细胞活性高于血管基质成分类器官(P<0.05),培养21 d的细胞活性低于血管基质成分类器官(P<0.05),说明血管基质成分类器官的细胞活性持续时间更长;③相较于脂肪间充质干细胞球,血管基质成分类器官具有更优的骨、软骨和脂肪形成能力;多色免疫荧光共定位显示,血管基质成分类器官中血管网络和成骨标志物表达丰富,细胞间和细胞-基质相互作用复杂,脂肪间充质干细胞球结构单一且缺乏内皮细胞基因表达;qRT-PCR检测结果显示,血管基质成分类器官中RUNX2、骨桥蛋白、Ⅰ型胶原、Sp7转录因子、CD31和血管内皮生长因子mRNA表达均高于脂肪间充质干细胞球(P<0.05);④结果表明,血管基质成分类器官具备复杂的生理结构和更强的血管形成和骨形成能力,以及更持久的细胞活性。 展开更多

基于熵权TOPSIS-RSR模型的临床专科能力评价研究

目的综合评价某三级甲等综合医院临床专科竞争力,为医院临床重点专科的申报工作提供科学依据。方法基于某三级甲等综合医院拟申报的15个临床专科2021年—2023年DRG数据,从医疗服务能力、服务效率与质量安全3个维度筛选DRG指标,采用熵权T... 目的综合评价某三级甲等综合医院临床专科竞争力,为医院临床重点专科的申报工作提供科学依据。方法基于某三级甲等综合医院拟申报的15个临床专科2021年—2023年DRG数据,从医疗服务能力、服务效率与质量安全3个维度筛选DRG指标,采用熵权TOPSIS-RSR模型进行综合评价。结果熵权TOPSIS法结果显示,服务能力维度指标权重最高,外科总权重为20.62%,内科总权重为28.56%;而质量安全维度权重显著偏低,其中中低风险组死亡率权重为1.88%。RSR分档结果显示,推荐专科4个:胸外科、神经外科、心血管内科和消化内科;培养专科9个;孵化专科2个。表明专科发展水平存在明显差异。结论DRG相关指标结合熵权TOPSIS-RSR综合评价法可有效克服传统申报策略对经验的过度依赖,为医院临床重点专科的推荐申报工作提供优化策略,并为提升专科医疗质量提供依据。 展开更多

Objective Tick-borne encephalitis virus(TBEV) is an emerging pathogen in Europe and North Asia that causes tick-borne encephalitis(TBE). A simple, rapid method for detecting TBEV RNA is needed to control this disease. Methods A reverse-transcription recombinase-aided amplification(RT-RAA) assay was developed. This assay can be completed in one closed tube at 39℃ within 30 minutes. The sensitivity and specificity of RT-RAA were validated using non-infectious synthetic RNA representing a fragment of the NS5 region of the wild-type(WT) TBEV genome and the Senzhang strain. Additionally, 10 batches of tick samples were used to evaluate the performance of the RT-RAA assay. Results The analytical limit of detection of the assay was 20 copies per reaction of the TBEV synthetic transcript and 3 plaque-forming units(pfu) per reaction of TBEV titers. With the specific assay, no signal due to other arboviruses was observed. Of the 10 batches of tick samples obtained from the Changbai Mountains of China, three were TBEV-positive, which was consistent with the results of the quantitative real-time PCR assay. Conclusion A rapid, highly sensitive, specific, and easy-to-use method was developed for the detection of the TBEV Far-Eastern subtype. 展开更多

A real-time RT-PCR (RT-qPCR) assay for the detection of Tahyna virus was developed to monitor Tahyna virus infection in field-collected vector mosquito samples. The targets selected for the assay were S segment sequences encoding the nucleocapsid protein from the Tahyna virus. Primers and probes were selected in conserved regions by aligning genetic sequences from various Tahyna virus strains available from GenBank. The sensitivity of the RT-qPCR approach was compared to that of a standard plaque assay in BHK cells. RT-qPCR assay can detect 4.8 PFU of titrated Tahyna virus. Assay specificities were determined by testing a battery of arboviruses, including representative strains of Tahyna virus and other arthropod-borne viruses from China. Seven strains of Tahyna virus were confirmed as positive; the other seven species of arboviruses could not be detected by RT-qPCR. Additionally, the assay was used to detect Tahyna viral RNA in pooled mosquito samples. The RT-qPCR assay detected Tahyna virus in a sensitive, specific, and rapid manner; these findings support the use of the assay in viral surveillance. 展开更多

Rhododendron is a well-known genus consisting of commercially valuable ornamental woody plant species.Heat stress is a major environmental factor that affects rhododendron growth.Melatonin was recently reported to alleviate the effects of abiotic stress on plants.However,the role of melatonin in rhododendron plants is unknown.In this study,the effect of melatonin on rhododendron plants exposed to heat stress and the potential underlying mechanism were investigated.Analyses of morphological characteristics and chlorophyll a fluorescence indicated 200μmol L–1 was the optimal melatonin concentration for protecting rhododendron plants from heat stress.To elucidate how melatonin limits the adverse effects of high temperatures,melatonin contents,photosynthetic indices,Rubisco activity,and adenosine triphosphate(ATP)contents were analyzed at 25,35,and 40℃,respectively.Compared with the control,exogenous application of melatonin improved the melatonin contents,electron transport rate,photosystem II and I activities,Rubisco activity,and ATP contents under heat stress.The transcriptome analysis revealed many of the heat-induced differentially expressed genes were associated with the photosynthetic pathway;the expression of most of these genes was down-regulated by heat stress more in the melatonin-free plants than in the melatonin-treated plants.We identified Rh PGR5A,Rh ATPB,Rh LHCB3,and Rh Rbs A as key genes.Thus,we speculate that melatonin promotes photosynthetic electron transport,improves Calvin cycle enzyme activities,and increases ATP production.These changes lead to increased photosynthetic efficiency and CO_(2) assimilation under heat stress conditions via the regulated expression of specific genes,including Rh Rbs A.Therefore,the application of exogenous melatonin may increase the tolerance of rhododendron to heat stress. 展开更多

Based on the Culex flavivirus (CxFV) E gene sequences in GenBank, CxFV-specific primers and probes were designed for real-time reverse transcription-polymerase chain reaction (RT-qPCR). The specificity test revealed that CxFV could be detected using RT-qPCR with the specific CxFV primers and probes; other species of arboviruses were not detected. The stability test demonstrated a coefficient of variation of <1.5%. A quantitative standard curve for CxFV RT-qPCR was established. Quantitative standard curve analysis revealed that the lower detection limit of the RT-qPCR system is 100 copies/mu L. Moreover, RT-qPCR was used to detect CxFV viral RNA in mosquito pool samples. In conclusion, we established a real-time RT-PCR assay for CxFV detection, and this assay is more sensitive and efficient than general RT-PCR. This technology may be used to monitor changes in the environmental virus levels. 展开更多

Objective This study aimed to compare the current Essen rabies post-exposure immunization schedule(0-3-7-14-28)in China and the simple 4-dose schedule(0-3-7-14)newly recommended by the World Health Organization in terms of their safety,efficacy,and protection.Methods Mice were vaccinated according to different immunization schedules,and blood was collected for detection of rabies virus neutralizing antibodies(RVNAs)on days 14,21,28,35,and 120after the first immunization.Additionally,different groups of mice were injected with lethal doses of the CVS-11 virus on day 0,subjected to different rabies immunization schedules,and assessed for morbidity and death status.In a clinical trial,185 rabies-exposed individuals were selected for post-exposure vaccination according to the Essen schedule,and blood was collected for RVNAs detection on days 28and 42 after the first immunization.Results A statistically significant difference in RVNAs between mice in the Essen and 0-3-7-14 schedule groups was observed on the 35th day(P<0.05).The groups 0-3-7-14,0-3-7-21,and 0-3-7-28 showed no statistically significant difference(P>0.05)in RVNAs levels at any time point.The post-exposure immune protective test showed that the survival rate of mice in the control group was 20%,whereas that in the immunization groups was 40%.In the clinical trial,the RVNAs positive conversion rates on days 28(14 days after 4 doses)and 42(14 days after 5 doses)were both 100%,and no significant difference in RVNAs levels was observed(P>0.05).Conclusion The simple 4-dose schedule can produce sufficient RVNAs levels,with no significant effect of a delayed fourth vaccine dose(14–28 d)on the immunization potential. 展开更多

Detection of pigs epidemic diarrhea virus (PEDV) was conducted on 331 piglets diarrhea fecal samples collected in Nanning, Yulin and other 12 areas of Guangxi Province from January of 2011 to April of 2014 by the method of reverse transcription-polymerase chain reaction (RT-PCR). The results showed that the positive samples of PEDV were 210 and the positive rate was 63.44%. The clone and sequencing of M gene was carried out on 25 positive samples. PEDV reference strains were selected from GeneBank to conduct the sequence homology alignment analysis and the phylogenetic tree of M gene. The M gene homology and amino acid sequence identity between 25 isolated strains and 51 reference strains were 96.0% - 99.6% and 94.3% - 99.6%, respectively. The genetic variation anal- ysis of M gene showed that the genetic relationship of PEDV prevalent strains in Guangxi Province from 2013 to 2014 was close to that of the prevalent strains in Bei- jing, Anhui, Wuhan, Hebei and Guangdong from 2010 to 2013, and which were far from that of the Chinese early isolates CH/S (GenBank number: JN547228 ), vaccine strain CV777 (GenBank number: AF353511 ) and Attenuated DR13 (GenBank number: JQ023162). Indicating that the PEDV strains prevalent in Guan- gxi in recent years showed significant variation with the early isolates. 展开更多
