针对倾斜煤层露天煤矿内排土场与端帮之间搭桥位置和搭桥高度问题开展研究。在搭桥位置选取时,分别分析了反向排土、正向排土和双向排土的优缺点,选取正向由南向北的排土方式,确定搭桥的排土位置;在西帮建立倾斜煤层压帮二次剥离量与剥... 针对倾斜煤层露天煤矿内排土场与端帮之间搭桥位置和搭桥高度问题开展研究。在搭桥位置选取时,分别分析了反向排土、正向排土和双向排土的优缺点,选取正向由南向北的排土方式,确定搭桥的排土位置;在西帮建立倾斜煤层压帮二次剥离量与剥采比的分段函数,通过剥采比分析了压帮高度和开采长度的关系,确定了二次剥离阶段剥采比变化规律;由排土场基底承载力计算了内排土场的单台阶排土高度;根据南北向的排土空间参数、排土作业参数、生产接续规划,设计了最终的搭桥方案。结果表明:选取西帮靠南部区域作为搭桥位置;从搭桥高度195 m处排土至+665 m,设计桥面的南坡运输道路宽度为30 m,北坡排土平盘宽度80 m;距离南帮深部采煤作业区50 m;现场有效排土长度790 m,初始压帮高度到+575 m。 展开更多

目的探讨双开门椎管扩大成形术对脊髓型颈椎病合并高血压患者血压的影响。方法2018年6月至2020年9月共42例于中山大学附属第七医院骨科行双开门椎管扩大成形术的脊髓型颈椎病合并高血压患者;其中男24例,女18例;年龄41~86(56.3±9.4... 目的探讨双开门椎管扩大成形术对脊髓型颈椎病合并高血压患者血压的影响。方法2018年6月至2020年9月共42例于中山大学附属第七医院骨科行双开门椎管扩大成形术的脊髓型颈椎病合并高血压患者;其中男24例,女18例;年龄41~86(56.3±9.4)岁。监测术前术后血压及降压药物使用情况。结果术后1周、术后1个月的收缩压、舒张压均较术前下降(t=2.41、2.65、3.58、3.12,P<0.05)。术后高血压各分级所占比例均较术前降低(χ^(2)=4.27、4.92、4.16,P<0.05)。全部42病例降压药物未见增药;31%病例术后减药或停药。结论颈椎后路双开门椎管扩大成形术可降低脊髓型颈椎病合并高血压患者的血压水平。 展开更多

Salinity is one of the most severe environmental factors that may impair crop productivity.A proteomic study based on two-dimensional gel electrophoresis is performed in order to analyze the long-term salinity stress response of Thellungiella halophila,an Arabidopsis-related halophyte.Four-week-old seedlings are exposed to long-term salinity treatment.The total crude proteins are extracted from leaf blades,separated by 2-DE,stained with Coomassie Brilliant Blue,and differentially displayed spots are identified by MALDI-TOF MS or QTOF MS/MS.Among 900 protein spots reproducibly detected on each gel,30 spots exhibit significant change and some of them are identified.The identified proteins include not only some previously characterized stress-responsive proteins such as TIR-NBS-LRR class disease resistance protein,ferritin-1,and pathogenesis-related protein 5,but also some proteins related to energy pathway,metabolism,RNA processing and protein degradation,as well as proteins with unknown functions.The possible functions of these proteins in salinity tolerance of T.halophila are discussed and it is suggested that the long-term salinity tolerance of T.halophila is achieved,at least partly,by enhancing defense system,adjusting energy and metabolic pathway and maintaining RNA structure. 展开更多

CENP-K and CENP-H may form coiled-coils in the kinetochores

Kinetochores are large proteinaceous structure on the surface of chromosomes’primary constriction during mitosis.They link chromosomes to spindle microtubules and also regulate the spindle assem-bly checkpoint,which is crucial for correct chromosome segregation in all eukaryotes.The better known core networks of kinetochores include the KMN network(K,KNL1;M,Mis12 complex;N,Ndc80 complex)and CCAN(constitutive centromere-associated network).However,the detailed molecular mechanism of the kinetochore protein network remains unclear.This study demonstrates that CENP-H and CENP-K form quite stable subcomplex by TAP(tandem affinity purification)with HEK 293 cells which express TAP-CENP-K,with the ratio of purified CENP-H and CENP-K being close to 1︰1 even with high salt.Bioinformatic analysis suggests that CENP-H and CENP-K are enriched with coiled-coil regions.This implies that CENP-H and CENP-K form heterodimeric coiled-coils.Furthermore,the func-tional regions which form the complex are respectively located on their N-and C-terminals,but the association between the C-terminals is more complex.It is possible that this is the first identified het-erodimeric coiled-coils within the inner kinetochore,which is directly involved in the attachment be-tween kinetochores and the spindle microtubules. 展开更多

Two hundred and eighteen serum samples from 175 lung cancer patients and 43 healthy individuals were analyzed by using Surface Enhaced Laser Desorption/Ionization Time of Flight Mass Spectrome-try(SELDI-TOF-MS).The data analyzed by both Biomarker Wizard?and Biomarker Patterns?software showed that a protein peak with the molecular weight of 11.6 kDa significantly increased in lung cancer.Meanwhile,the level of this biomarker was progressively increased with the clinical stages of lung cancer.The candidate biomarker was then obtained from tricine one-dimensional sodium dodecyl sul-fate-polyacrylamide gel electrophoresis by matching the molecular weight with peaks on WCX2 chips and was identified as Serum Amyloid A protein(SAA)by MALDI/MS-MS and database searching.It was further validated in the same serum samples by immunoprecipitation with commercial SAA antibody.To confirm the SAA differential expression in lung cancer patients,the same set of serum samples was measured by ELISA assay.The result showed that at the cutoff point 0.446(OD value)on the Receiver Operating Characteristic(ROC)curve,SAA could better discriminate lung cancer from healthy indi-viduals with sensitivity of 84.1%and specificity of 80%.These findings demonstrated that SAA could be characterized as a biomarker related to pathological stages of lung cancer. 展开更多

Compared to other subcellular organelles,centrosome proteome can hardly be studied,due to the dif-ficulties in separation and purification of centrosome.Auto-antisera from 6 autoimmune patients,which recognized centrosome specifically in immunofluorescence,were used to identify the corresponding centrosomal proteins.The sera were first tested by Western blot on whole cell lysate,and all bound antibodies were then eluted from each single band in Western blot membrane to assure which antibody was responsible for the centrosome specific immunofluorescence staining.The corresponding pro-teins were obtained by immunoprecipitation and identified by mass spectrometry.Six centrosomal proteins,including 2 known centrosomal proteins and 4 proteins with unknown localization or report-edly non-centrosomal localization,were identified.These proteins apparently involve in cell cycle regulation,signal transduction pathways,molecular chaperons,and metabolism enzymes,which may reflect the expected functional diversity of centrosome. 展开更多

There are multiple reports of autoimmune response in patients with lung cancer.To investigate whether a novel autoantibody is present in patients with lung cancer and evaluate its clinical diagnostic and prognostic value,sera from 10 patients with lung cancer and 10 normal individuals were analyzed using immunofluorescence and Western blotting.It was found that one serum sample from the patients with squamous carcinoma gave a fine speckled pattern staining in nucleus and had a high titer antinuclear autoantibody which could recognize 31 kD of nuclear protein isolated from both cancer cells and normal cells.The same patient’s serum was further used to immunoprecipitate the target antigen.The protein bands were excised from the SDS-PAGE gels and were analyzed with a Qstar Pulser I Quadrupole time-flight mass spec-trometer,and the 31 kD target antigen was identified as U1-A snRNP.To test the prevalence of anti-U1-A snRNP antibody,sera from 93 patients including 36 squmaous carcinomas(SCC),26 adenocarcinomas(Ad),and 31 small cell carcinomas(SCLC)were screened by Western blotting.The results demonstrated that anti-U1-A snRNP antibody was present in 50%of SCC sera,26.9%of Ad sera and 54.8%of SCLC sera.In this paper,we report for the first time that anti-U1-A snRNP antibody could be detected in the patients with lung cancer. 展开更多
