Salinity is one of the most severe environmental factors that may impair crop productivity.A proteomic study based on two-dimensional gel electrophoresis is performed in order to analyze the long-term salinity stress ...Salinity is one of the most severe environmental factors that may impair crop productivity.A proteomic study based on two-dimensional gel electrophoresis is performed in order to analyze the long-term salinity stress response of Thellungiella halophila,an Arabidopsis-related halophyte.Four-week-old seedlings are exposed to long-term salinity treatment.The total crude proteins are extracted from leaf blades,separated by 2-DE,stained with Coomassie Brilliant Blue,and differentially displayed spots are identified by MALDI-TOF MS or QTOF MS/MS.Among 900 protein spots reproducibly detected on each gel,30 spots exhibit significant change and some of them are identified.The identified proteins include not only some previously characterized stress-responsive proteins such as TIR-NBS-LRR class disease resistance protein,ferritin-1,and pathogenesis-related protein 5,but also some proteins related to energy pathway,metabolism,RNA processing and protein degradation,as well as proteins with unknown functions.The possible functions of these proteins in salinity tolerance of T.halophila are discussed and it is suggested that the long-term salinity tolerance of T.halophila is achieved,at least partly,by enhancing defense system,adjusting energy and metabolic pathway and maintaining RNA structure.展开更多
Kinetochores are large proteinaceous structure on the surface of chromosomes’primary constriction during mitosis.They link chromosomes to spindle microtubules and also regulate the spindle assem-bly checkpoint,which ...Kinetochores are large proteinaceous structure on the surface of chromosomes’primary constriction during mitosis.They link chromosomes to spindle microtubules and also regulate the spindle assem-bly checkpoint,which is crucial for correct chromosome segregation in all eukaryotes.The better known core networks of kinetochores include the KMN network(K,KNL1;M,Mis12 complex;N,Ndc80 complex)and CCAN(constitutive centromere-associated network).However,the detailed molecular mechanism of the kinetochore protein network remains unclear.This study demonstrates that CENP-H and CENP-K form quite stable subcomplex by TAP(tandem affinity purification)with HEK 293 cells which express TAP-CENP-K,with the ratio of purified CENP-H and CENP-K being close to 1︰1 even with high salt.Bioinformatic analysis suggests that CENP-H and CENP-K are enriched with coiled-coil regions.This implies that CENP-H and CENP-K form heterodimeric coiled-coils.Furthermore,the func-tional regions which form the complex are respectively located on their N-and C-terminals,but the association between the C-terminals is more complex.It is possible that this is the first identified het-erodimeric coiled-coils within the inner kinetochore,which is directly involved in the attachment be-tween kinetochores and the spindle microtubules.展开更多
Two hundred and eighteen serum samples from 175 lung cancer patients and 43 healthy individuals were analyzed by using Surface Enhaced Laser Desorption/Ionization Time of Flight Mass Spectrome-try(SELDI-TOF-MS).The da...Two hundred and eighteen serum samples from 175 lung cancer patients and 43 healthy individuals were analyzed by using Surface Enhaced Laser Desorption/Ionization Time of Flight Mass Spectrome-try(SELDI-TOF-MS).The data analyzed by both Biomarker Wizard?and Biomarker Patterns?software showed that a protein peak with the molecular weight of 11.6 kDa significantly increased in lung cancer.Meanwhile,the level of this biomarker was progressively increased with the clinical stages of lung cancer.The candidate biomarker was then obtained from tricine one-dimensional sodium dodecyl sul-fate-polyacrylamide gel electrophoresis by matching the molecular weight with peaks on WCX2 chips and was identified as Serum Amyloid A protein(SAA)by MALDI/MS-MS and database searching.It was further validated in the same serum samples by immunoprecipitation with commercial SAA antibody.To confirm the SAA differential expression in lung cancer patients,the same set of serum samples was measured by ELISA assay.The result showed that at the cutoff point 0.446(OD value)on the Receiver Operating Characteristic(ROC)curve,SAA could better discriminate lung cancer from healthy indi-viduals with sensitivity of 84.1%and specificity of 80%.These findings demonstrated that SAA could be characterized as a biomarker related to pathological stages of lung cancer.展开更多
