烯丙孕素纳米混悬递送系统的制备、表征及体外释放特性

烯丙孕素(ALT)作为一种动物用口服孕激素,其水溶性极低(约10μg/mL),且在体内代谢快,这严重制约了其临床疗效的发挥。纳米混悬递送系统(NS)可以将药物颗粒减小至纳米尺寸,有效提升难溶性药物的溶出能力和生物活性。本研究采用反溶剂法... 烯丙孕素(ALT)作为一种动物用口服孕激素,其水溶性极低(约10μg/mL),且在体内代谢快,这严重制约了其临床疗效的发挥。纳米混悬递送系统(NS)可以将药物颗粒减小至纳米尺寸,有效提升难溶性药物的溶出能力和生物活性。本研究采用反溶剂法制备了烯丙孕素纳米混悬剂(ALT-NS),通过单因素实验及星点设计-效应面法(CCD-RSM)对制备工艺和处方组成展开优化,并深入分析了ALT-NS的形成机理及制备影响因素。结果显示,ALT-NS最优处方和工艺参数为:烯丙孕素含量0.4wt%、十二烷基硫酸钠(SDS)浓度25 mg/mL、羟丙基甲基纤维素(HPMC)浓度4 mg/mL、搅拌速度1000 r/min、搅拌时间10 min。所制备的ALT-NS为淡黄色澄清液体,扫描电镜下呈类球形颗粒,平均粒径为104.87±1.10 nm、粒径分布指数(PDI)为0.19±0.01、Zeta电位为-37.87±0.81 mV。分析表明,在ALT-NS制备过程中,ALT由结晶状转变为无定形态。体外释放度研究表明,ALT-NS在72 h的体外累计释放度为98.4%,约是原药组的1.54倍,展现出高效、快速的释药特性。综上,本研究制备的ALT-NS能够显著减小ALT的粒径,有效提高ALT的溶解和释放效率,这对于ALT在临床应用中的疗效具有重要意义。 展开更多

Titanium(Ti)and its alloys are frequently utilized as critical components in a variety of engineering ap-plications because of their high specific strength and excellent corrosion resistance.Compared to conven-tional surface strengthening technologies,laser shock peening(LSP)has increasingly attracted attention from researchers and industries,since it significantly improves the surface strength,biocompatibility,fa-tigue resistance,and anti-corrosion ability of Ti and its alloys.Despite numerous studies that have been carried out to elucidate the effects of LSP on microstructural evolution and mechanical properties of Ti and its alloys in recent years,a comprehensive review of recent advancements in the field of Ti and its alloys subjected to LSP is still lacking.In this review,the standard LSP and the novel process designs of LSP assisted by thermal,cryogenic,electropulsing and magnetic fields are discussed and compared.Microstructural evolution,with focuses on the dislocation dynamics,deformation twinning,grain refine-ment and surface amorphization,during LSP processing of Ti alloys is reviewed.Furthermore,the en-hanced engineering performance of the L SP-processed(L SPed)Ti alloys,including surface hardness,wear resistance,fatigue life and corrosion resistance are summarized.Finally,this review concludes by present-ing an overview of the current challenges encountered in this field and offering insights into anticipated future trends. 展开更多

Location estimation of underwater sensor networks(USNs)has become a critical technology,due to its fundamental role in the sensing,communication and control of ocean volume.However,the asynchronous clock,security attack and mobility characteristics of underwater environment make localization much more challenging as compared with terrestrial sensor networks.This paper is concerned with a privacy-preserving asynchronous localization issue for USNs.Particularly,a hybrid network architecture that includes surface buoys,anchor nodes,active sensor nodes and ordinary sensor nodes is constructed.Then,an asynchronous localization protocol is provided,through which two privacy-preserving localization algorithms are designed to estimate the locations of active and ordinary sensor nodes.It is worth mentioning that,the proposed localization algorithms reveal disguised positions to the network,while they do not adopt any homomorphic encryption technique.More importantly,they can eliminate the effect of asynchronous clock,i.e.,clock skew and offset.The performance analyses for the privacy-preserving asynchronous localization algorithms are also presented.Finally,simulation and experiment results reveal that the proposed localization approach can avoid the leakage of position information,while the location accuracy can be significantly enhanced as compared with the other works. 展开更多

Gut microbiota and circulating metabolite dysbiosis predate important pathological changes in glucose metabolic disorders;however,comprehensive studies on impaired glucose tolerance(IGT),a diabetes mellitus(DM)precursor,are lacking.Here,we perform metagenomic sequencing and metabolomics on 47 pairs of individuals with IGT and newly diagnosed DM and 46 controls with normal glucose tolerance(NGT);patients with IGT are followed up after 4 years for progression to DM.Analysis of baseline data reveals significant differences in gut microbiota and serum metabolites among the IGT,DM,and NGT groups.In addition,13 types of gut microbiota and 17 types of circulating metabolites showed significant differences at baseline before IGT progressed to DM,including higher levels of Eggerthella unclassified,Coprobacillus unclassified,Clostridium ramosum,L-valine,L-norleucine,and L-isoleucine,and lower levels of Eubacterium eligens,Bacteroides faecis,Lachnospiraceae bacterium 3_1_46FAA,Alistipes senegalensis,Megaspaera elsdenii,Clostridium perfringens,α-linolenic acid,10E,12Z-octadecadienoic acid,and dodecanoic acid.A random forest model based on differential intestinal microbiota and circulating metabolites can predict the progression from IGT to DM(AUC=0.87).These results suggest that microbiome and metabolome dysbiosis occur in individuals with IGT and have important predictive values and potential for intervention in preventing IGT from progressing to DM. 展开更多

A simple and sensitive high performance liquid chromatography with fluorescence detection (HPLC-FD) has been developed for simultaneous quantification of doxorubicin (DOX) and its dipeptide conjugate prodrug (PDOX) in mice plasma. The chromatographic separation was carried out on an Amethyst C18-H column with gradient mobile phase of 0.1% formic acid and 0.1% formic acid in acetonitrile at a flow rate of 1.0 mL/min. The excitation and emission wavelengths were set at 490 and 550 nm, respectively. The method was comprehensively validated. The limits of detection were low up to 5.0 ng/mL for DOX and 25.0 ng/mL for PDOX. And the limits of quantification were low up to 12.5 ng/mL for DOX and 50 ng/mL for PDOX, which were lower than those for most of the current methods. The calibration curves showed good linearity (R2 〉 0.999) over the concentration ranges. The extraction recoveries ranged from 84.0% to 88.2% for DOX and from 85.4% to 89.2% for PDOX. Satisfactory intra-day and inter-day precisions were achieved with RSDs less than 9.1%. The results show that the developed HPLC-FD method is accurate, reliable and will be helpful for preclinical pharmacokinetic study of DOX and PDOX. 展开更多

The electromigration force and Joule heating took on the main driving forces for Bi diffusion and migration. In addition, two appearance types of Bi layers, planar-type and groove-type, were found during current stressing. In order to effectively achieve torque demand in electric vehicles (EVs), this paper presents a torque control strategy based on model predictive control (MPC) for permanent magnet synchronous motor (PMSM) driven by a two-level three-phase inverter. A centralized control strategy is established in the MPC framework to track the torque demand and reduce energy loss, by directly optimizing the switch states of inverter. To fast determine the optimal control sequence in predictive process, a searching tree is built to look for optimal inputs by dynamic programming (DP) algorithm on the basis of the principle of optimality. Then we design a pruning method to check the candidate inputs that can enter the next predictive loop in order to decrease the computational burden of evaluation of input sequences. Finally, the simulation results on different conditions indicate that the proposed strategy can achieve a tradeoff between control performance and computational efficiency. 展开更多

Underwater data collection is an importance part in the process of network monitoring,network management and intrusion detection.However,the limited-energy of nodes is a major challenge to collect underwater data.The solution of this problem are not only in the hands of network topology but in the hands of path of autonomous underwater vehicle(AUV).With the problem in hand,an energy-efficient data collection scheme is designed for mobile underwater network.Especially,the data collection scheme is divided into two phases,i.e.,routing algorithm design for sensor nodes and path planing for AUV.With consideration of limited-energy and network robustness,Q-learning based dynamic routing algorithm is designed in the first phase to optimize the routing selection of nodes,through which a potential-game based optimal rigid graph method is proposed to balance the trade-off between the energy consumption and the network robustness.With the collected data,Q-learning based path planning strategy is proposed for AUV in the second phase to find the desired path to gather the data from data collector,then a mode-free tracking controller is developed to track the desired path accurately.Finally,the performance analysis and simulation results reveal that the proposed approach can guarantee energy-efficient and improve network stability. 展开更多

The Chinese medicine Extractum trametes robiniophila murr(ETRM)is the extract of a type of fungus.Recent studies have suggested that ETRM efficiently improves the effacement of podocyte foot processes in adriamycin(ADR)-induced nephrotic rats.In the present study,we aimed to assess whether ETRM modulated the actin rearrangements of podocytes and involved signaling molecules,includingα-actinin-4 and nephrin.Podocytes were treated with ADR(0.5μmol/L),ADR(0.5μmol/L)+dexamethasone(Dex)(1μmol/L),ADR(0.5μmol/L)+ETRM(10 mg/mL).The F-actin in the podocytes was stained by fluorescent phallotoxins and observed by confocal microscopy.The expression levels ofα-actinin-4 and nephrin were tested by real-time PCR and Western blotting analysis.The administration of ETRM could significantly prevent ADR-treated podocytes from actin rearrangement.Both ETRM and Dex could stabilize podocyte actin cytoskeletons.Moreover,α-actinin-4 might act as a potential target for ETRM functionality in podocyte actin rearrangements.However,pretreatment with ETRM could not inhibit the up-regulation of nephrin as a result of ADR treatment.ETRM could improve the cytoskeleton stability in ADR-induced actin rearrangement of podocytes via regulatingα-actinin-4 expression. 展开更多

ASIC2 Synergizes with TRPV1 in the Mechano-Electrical Transduction of Arterial Baroreceptors

Mechanosensitive ion channels(MSCs)are key molecules in the mechano-electrical transduction of arterial baroreceptors.Among them,acid-sensing ion channel 2(ASIC2)and transient receptor potential vanilloid subfamily member 1(TRPV1)have been studied extensively and documented to play important roles.In this study,experiments using aortic arch-aortic nerve preparations isolated from rats revealed that both ASIC2 and TRPV1 are functionally necessary,as blocking either abrogated nearly all pressure-dependent neural discharge.However,whether ASIC2 and TRPV1 work in coordination remained unclear.So we carried out cell-attached patch-clamp recordings in HEK293T cells co-expressing ASIC2 and TRPV1 and found that inhibition of ASIC2 completely blocked stretch-activated currents while inhibition of TRPV 1 only partially blocked these currents.Immunofluorescence staining of aortic arch-aortic adventitia from rats showed that ASIC2 and TRPV1 are co-localized in the aortic nerve endings,and co-immunoprecipitation assays confirmed that the two proteins form a compact complex in HEK293T cells and in baroreceptors.Moreover,protein modeling analysis,exogenous co-immunoprecipitation assays,and biotin pull-down assays indicated that ASIC2 and TRPV1 interact directly.In summary,our research suggests that ASIC2 and TRPV1 form a compact complex and function synergisti-cally in the mechano-electrical transduction of arterial baroreceptors.The model of synergism between MSCs may have important biological significance beyond ASIC2 and TRPV 1. 展开更多

Objective:In this research,we tried to explore how short-term mindfulness(STM)intervention affects adoles-cents’anxiety,depression,and negative and positive emotion during the COVID-19 pandemic.Design:10 classes were divided into experiment groups(5 classes;n=238)and control(5 classes;n=244)randomly.Hospital Anxi-ety and Depression Scale(HADS)and Positive and Negative Affect Schedule(PANAS)were used to measure par-ticipants’dependent variables.In the experiment group,we conducted STM practice interventions every morning in theirfirst class from March to November 2020.No interventions were conducted in the control group.Methods:Paired-sample t-tests were used to identify if a significant difference exists between every time point of the experimental and control groups.Repeated ANOVA and Growth Mixture Model(GMM)were used to analyze the tendency of positive and negative emotions,anxiety,and depression in the experimental group.Results and Conclusions:(1)With the intervention of STM,there was a significant decrease in negative emotions and an increase in positive emotions in the experimental group,whereas there were non-significant differences in the control group.(2)To explore the heterogeneity trajectories of dependent variables,we built a GMM and found there were two latent growth classes in the trajectories.(3)The results of the models showed their trajec-tories were downward,which meant that the levels of anxiety,depression,and negative emotions of participants decreased during the STM training period.Nonetheless,the score of positive affect showed upward in three loops of intervention,which indicated that the level of the participants’positive affect increased through the STM inter-vention.(4)This research indicated that STM should be given increasing consideration to enhance mental health during the worldwide outbreak of COVID-19. 展开更多
