However, little is known about mGluR5 in the prenatal human brain. Here, we aimed to explore the expression pattern and cellular distribution of mGluR5 in human fetal hippocampus. Methods Thirty-four human fetuses were divided into four groups according to gestational age: 9-11, 14-16, 22-24 and 32-36 weeks. The hippocampus was dissected out and prepared. The protein and mRNA expression of mGluR5 were evaluated by Western blot and immunohistochemistry or real-time PCR. The cellular distribution of mGluR5 was observed with double-labeling immunofluorescence. Results Both mGluR5 mRNA and protein were detected in the prenatal human hippocampus by real-time PCR and immunoblotting, and the expression levels increased gradually over time. The immunohistochemistry results were consistent with immunoblotting and showed that mGluR5 immunoreactivity was mainly present in the inner marginal zone (IMZ), hippocampal plate (HP) and ventricular zone (VZ). The double-labeling immunoftuorescence showed that mGluR5 was present in neural stem cells (nestin-positive), neuroblasts (DCX-positive) and mature neurons (NeuN-positive), but not in typical astrocytes (GFAP- positive). The cells co-expressing mGluR5 and nestin were mainly located in the IMZ, HP and subplate at 11 weeks, all layers at 16 weeks, and CA 1 at 24 weeks. As development proceeded, the number of mGluR5/nestin double-positive cells decreased gradually so that there were only a handful of double-labeled cells at 32 weeks. However, mGluR5/DCX double-positive cells were only found in the HP, IZ and IMZ at 11 weeks. Conclusion The pattern ofmGluR5 expression by neural stern/progenitor cells, neuroblasts and neurons provides important anatomical evidence for the role of mGluR5 in the regulation of human hippocampal development.展开更多

Objective:To review trials on mindfulness intervention for chronic pain in primary care to clarify the evidence base and establish whether mindfulness is an important intervention for relieving pain and improving psyc...Objective:To review trials on mindfulness intervention for chronic pain in primary care to clarify the evidence base and establish whether mindfulness is an important intervention for relieving pain and improving psychological comorbidity.Methods:We performed a literature search using PubMed,the Cochrane Database,EBSCOhost,Elsevier,Wiley,Springer,and the references of retrieved articles.We included articles written in English and that were published up to January 2012.We found 428 empirical studies,but only eight were included as randomized controlled trials of mindfulness intervention for chronic pain in our meta-analysis.After extracting and synthesizing data from these eight trials,we analyzed the data extracted and synthesized from these eight trials.Results:Compared with control intervention,mindfulness intervention had no specific effect on reducing pain intensity(weighted mean difference 3.24,95%confidence interval[CI]:8.92 to 2.45).Mindfulness intervention led to greater improvement in psychological comorbidity with chronic pain,such as depression(weighted mean difference3.91,95%CI5.94 to2.32)and trait anxiety(weighted mean difference4.07,95%CI4.48 to3.65).Conclusion:There is insufficient evidence that mindfulness intervention relieves pain intensity.However,it improves depression and trait anxiety in patients with chronic pain.Further research in larger,properly powered,and better-designed studies is warranted.展开更多

Until recently, n...Neural regeneration was once considered to be impossible, especially in the central nervous system where neural regeneration comprise the generation of new neurons, glia, axons, myelin, and synapses. Until recently, neural stem cells/neural progenitor cells (NSCs/NPCs) were identified from various areas of brain and brought hopes to the neural repair and regeneration. Tissue engineering has revolutionized the current neural regeneration technology and it has become a pioneering interdisciplinary field in the areas of biomedical research. Polyhydoxyalkanoate (PHA) as one of biodegradable material has been successfully used as tissue engineering materials. It has also been applied in nerve tissue engineering due to the high biocompatibility and low cytotoxicity. Over the past 10 years, different kinds of modification strategies have been undertaken to improve the properties of PHA to fit the requirements from various fields. Several members of PHA family have been attempted for neural regeneration. This article reviewed the recent modification strategies for improving the properties of PHA and highlighted the pioneer applications in neural regeneration.展开更多

[Objectives]To compare the effects of traditional processing and fresh processing on the quality of Polygonatum odoratum decoction piece.[Methods]The effects of fresh processing and traditional processing on the quali...[Objectives]To compare the effects of traditional processing and fresh processing on the quality of Polygonatum odoratum decoction piece.[Methods]The effects of fresh processing and traditional processing on the quality of P.odoratum decoction piece were compared and analyzed with appearance characteristics,total ash content,extract content,total polysaccharides content,and total flavonoids content as the evaluation indexes.[Results]Fresh processing method in different production areas has different effects on P.odoratum decoction piece.P.odoratum was dried in oven of 50℃.When moisture content was 41.44%-59.67%,it was cut.After complete drying at 50℃,the moisture content of dried P.odoratum was 8.94%-9.60%,and ethanol-soluble extract content was 77.29%-78.20%,and water-soluble extract was 77.7%-78.14%.At this time,the appearance characteristics of section of P.odoratum decoction piece were better than that of traditional processing,which was yellowish white.The total polysaccharide content was higher than that of traditional processing,and the content of total flavonoids was statistically significant different from that of traditional processing.[Conclusions]The quality of P.odoratum decoction piece by fresh processing is better than that of the traditional processing,and it is feasible to use fresh processing.展开更多
Background:Countries that aimed for eliminating the cases of COVID-19 with test-trace-isolate policy are found to have lower infections,deaths,and better economic performance,compared with those that opted for other m...Background:Countries that aimed for eliminating the cases of COVID-19 with test-trace-isolate policy are found to have lower infections,deaths,and better economic performance,compared with those that opted for other mitigation strategies.However,the continuous evolution of new strains has raised the question of whether COVID-19 eradication is still possible given the limited public health response capacity and fatigue of the epidemic.We aim to investigate the mechanism of the Zero-COVID policy on outbreak containment,and to explore the possibility of eradication of Omicron transmission using the citywide test-trace-isolate(CTTI)strategy.Methods:We develop a compartmental model incorporating the CTTI Zero-COVID policy to understand how it contributes to the SARS-CoV-2 elimination.We employ our model to mimic the Delta outbreak in Fujian Province,China,from September 10 to October 9,2021,and the Omicron outbreak in Jilin Province,China for the period from March 1 to April 1,2022.Projections and sensitivity analyses were conducted using dynamical system and Latin Hypercube Sampling/Partial Rank Correlation Coefcient(PRCC).Results:Calibration results of the model estimate the Fujian Delta outbreak can end in 30(95%confdence interval CI:28–33)days,after 10(95%CI:9–11)rounds of citywide testing.The emerging Jilin Omicron outbreak may achieve zero COVID cases in 50(95%CI:41–57)days if supported with sufcient public health resources and population compliance,which shows the efectiveness of the CTTI Zero-COVID policy.Conclusions:The CTTI policy shows the capacity for the eradication of the Delta outbreaks and also the Omicron outbreaks.Nonetheless,the implementation of radical CTTI is challenging,which requires routine monitoring for early detection,adequate testing capacity,efcient contact tracing,and high isolation compliance,which constrain its benefts in regions with limited resources.Moreover,these challenges become even more acute in the face of more contagious variants with a high proportion of asymptomatic cases.Hence,in regions where CTTI is not possible,personal protection,public health control measures,and vaccination are indispensable for mitigating and exiting the COVID-19 pandemic.展开更多
