BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS)is an effective intervention for managing complications of portal hypertension,particularly acute variceal bleeding(AVB).While effective in reducing portal...BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS)is an effective intervention for managing complications of portal hypertension,particularly acute variceal bleeding(AVB).While effective in reducing portal pressure and preventing rebleeding,TIPS is associated with a considerable risk of overt hepatic encephalopathy(OHE),a complication that significantly elevates mortality rates.AIM To develop a machine learning(ML)model to predict OHE occurrence post-TIPS in patients with AVB using a 5-year dataset.METHODS This retrospective single-center study included 218 patients with AVB who underwent TIPS.The dataset was divided into training(70%)and testing(30%)sets.Critical features were identified using embedded methods and recursive feature elimination.Three ML algorithms-random forest,extreme gradient boosting,and logistic regression-were validated via 10-fold cross-validation.SHapley Additive exPlanations analysis was employed to interpret the model’s predictions.Survival analysis was conducted using Kaplan-Meier curves and stepwise Cox regression analysis to compare overall survival(OS)between patients with and without OHE.RESULTS The median OS of the study cohort was 47.83±22.95 months.Among the models evaluated,logistic regression demonstrated the highest performance with an area under the curve(AUC)of 0.825.Key predictors identified were Child-Pugh score,age,and portal vein thrombosis.Kaplan-Meier analysis revealed that patients without OHE had a significantly longer OS(P=0.005).The 5-year survival rate was 78.4%,with an OHE incidence of 15.1%.Both actual OHE status and predicted OHE value were significant predictors in each Cox model,with model-predicted OHE achieving an AUC of 88.1 in survival prediction.CONCLUSION The ML model accurately predicts post-TIPS OHE and outperforms traditional models,supporting its use in improving outcomes in patients with AVB.展开更多
The Görtler vortex is a characteristic flow feature observed in the boundary layer on compression ramp in hypersonic flow.In the context of high-enthalpy plasma flows during aerospace re-entry processes,there is ...The Görtler vortex is a characteristic flow feature observed in the boundary layer on compression ramp in hypersonic flow.In the context of high-enthalpy plasma flows during aerospace re-entry processes,there is currently a lack of effective means to visualize the boundary layer.In this study,the Nitric Oxide Planar Laser-Induced Fluorescence(NO-PLIF)technique was employed to visualize the boundary layer of a compression ramp in a 50-MW arc-heated plasma wind tunnel.Görtler-like vortex structures were observed in the boundary layer of the ramp.This is the first time that Görtler vortices have been clearly observed in a high-enthalpy plasma flow.By varying the flow conditions,the Görtler vortices persisted in the boundary layer of the ramp when the total enthalpy of the arc-heated wind tunnel exceeded 12.3 MJ/kg.Several image processing techniques were applied to extract the structure of high-speed Görtler streaks,and the position of the high-speed streaks was found to be non-fixed,whereas the average Görtler wavelength remained at approximately 30 mm at a 10°ramp and showed limited variation with the total enthalpy.Additionally,a sheet-forming optics system with an adjustable angle and height was designed to enable visualization of the Görtler vortices in the boundary layer of the ramp at different angles and heights.The vortices on the low-angle ramp exhibited better stability and shorter wavelengths.Visualization results at different heights confirmed that the Görtler vortex wavelength was approximately twice the boundary layer thickness.This study demonstrates the feasibility and potential of the PLIF technique for the visualization of the boundary layer in plasma flows,especially with regard to Görtler vortices.展开更多
With the development of urban infrastructure,it is inevitable that shield tunnels will undercross intercity railways.However,the safe operation of intercity railways requires strict subgrade deformation.On the basis o...With the development of urban infrastructure,it is inevitable that shield tunnels will undercross intercity railways.However,the safe operation of intercity railways requires strict subgrade deformation.On the basis of the engineering background of the Lianghu Tunnel in Wuhan,the three-dimensional centrifuge test and numerical back analysis were used to study the development of subgrade surface settlement during shield tunneling.A three-dimensional numerical model with the same size as the prototype was subsequently established to further study the settlement development and torsion behavior of the subgrade during tunnel excavation.The results show that the maximum settlement point of the transverse settlement trough gradually moves to the tunnel axis during tunnel excavation and that the entire subgrade experiences torsional deformation.Moreover,the effect of the intersection angle between the axes of the tunnel and the subgrade on the surface settlement of the subgrade was further studied.The results show that the intersection angle has no effect on the maximum settlement,but the width of the settlement trough increases gradually with increasing angle.Finally,on the basis of the soil arching effect caused by tunnel excavation,the subgrade settlement during tunnel excavation is reduced by reinforcing the soil in different zones of soil arching.The results show that the settlement of the subgrade caused by the shield tunnel can be effectively controlled by adding reinforcement directly to the top of the tunnel,and the maximum settlement of the subgrade surface is reduced from 24.41 mm to 9.47 mm,a reduction of approximately 61.2%.展开更多
In clinical settings,regenerating critical-sized calvarial bone defects presents substantial problems owing to the intricacy of surgical methods,restricted bone growth medications,and a scarcity of commercial bone gra...In clinical settings,regenerating critical-sized calvarial bone defects presents substantial problems owing to the intricacy of surgical methods,restricted bone growth medications,and a scarcity of commercial bone grafts.To treat this life-threatening issue,improved biofunctional grafts capable of properly healing critical-sized bone defects are required.In this study,we effectively created anti-fracture hydrogel systems using spongy-like metal-organic(magnesium-phosphate)coordinated chitosan-modified injectable hydrogels(CPMg)loaded with a bioinspired neobavaisoflavone(NBF)component.The CPMg-NBF hydrogels showed outstanding anti-fracture capabilities during compression testing and retained exceptional mechanical stability even after 28 d of immersion in phosphatebuffered saline.They also demonstrated prolonged and stable release profiles of Mg^(2+)and NBF.Importantly,CPMg-NBF hydrogels revealed robust biphasic mineralization and were non-toxic to MC3T3-E1 cells.To better understand the underlying mechanism of Mg^(2+)and NBF component,as well as their synergistic effect on osteogenesis,we investigated the expression of key osteogenic proteins in the p38 MAPK and NOTCH pathways.Our results showed that CPMg-NBF hydrogels greatly increased the expression of osteogenic proteins(Runx2,OCN,OPN,BMPS and ALP).In vivo experiments showed that the implantation of CPMg-NBF hydrogels resulted in a significant increase in new bone growth within critical-sized calvarial defects.Based on these findings,we expect that the CPMg-NBF supramolecular hydrogel has tremendous promise for use as a therapeutic biomaterial for treating critical-sized calvarial defects.展开更多

Methods: The proliferation of GA-treated JeKo-1 cells was measured by CCK-8 assay and Ki-67 immunocytochemical detection. Apopt0sis, cell cycle and mitochondrial membrane potential were measured by flow cytometric analysis. Caspase-3, -8 and -9 were detected by colorimetric assay. Bcl-2 and Bax were analyzed by Western blotting. Results: GA inhibited cell growth in a time- and dose- dependent manner. GA induces apoptosis in JeKo- 1 cells but not in normal bone marrow cells, which was involved in reducing the membrane potential of mitochondria, activating caspases-3, -8 and -9 and decreasing the ratio of Bd-2 and Bax without cell cycle arresting. Conclusions: GA induced apoptosis in human MCL JeKo-1 cells by regulating Bcl-2/Bax and activating caspase-3, -8 and -9 via mitochondrial pathway without affecting cell cycle.展开更多
Triptolide,a component of the Chinese herb Tripterygium wilfordii Hook F,has been proved to be effective in the treatment of rheumatoid arthritis(RA).However,its underlying mechanisms on RA have not yet been well esta...Triptolide,a component of the Chinese herb Tripterygium wilfordii Hook F,has been proved to be effective in the treatment of rheumatoid arthritis(RA).However,its underlying mechanisms on RA have not yet been well established.We observed the inhibitory effect of triptolide on the expression of inflammatory cytokines and proliferation of fibroblast-like synoviocytes(FLS)induced by the complex of interleukin-6(IL-6)and the soluble form of the IL-6 receptor(sIL-6R).Furthermore,to clarify the underlying mechanisms,we treated FLS with the Janus-activated kinase 2(JAK2)inhibitor/signal transducer and activator of transcription 3(STAT3)activation blocker AZD1480.In this study,immunohistochemical staining was used to identify vimentin(+)and CD68(−)in FLS.The FLS proliferation was measured by cell proliferation assay,and the cell cycles were analyzed by flow cytometry.Furthermore,ELISA was used to detect the expression of the inflammatory factors in culture solution.The expression levels of p-JAK2,JAK2,p-STAT3 and STAT3 were investigated through Western blotting analysis.The results showed that IL-6/sIL-6R significantly increased the cell proliferation and expression of inflammatory cytokines,including IL-6,interleukin-1β(IL-1β)and vascular endothelial growth factor(VEGF).Triptolide or AZD1480 inhibited the cell proliferation and inflammatory cytokine expression in IL-6/sIL-6R-stimulated FLS by suppressing JAK2/STAT3.The study suggested that the physiological effects of triptolide on RA were due to its contribution to the inhibition of the inflammatory cytokine expression and FLS proliferation by suppressing the JAK2/STAT3 signaling pathway.It may provide an innovative insight into the effect of triptolide in preventing RA pathogenesis.展开更多
Due to their capability of fabricating geometrically complex structures,additive manufacturing(AM)techniques have provided unprecedented opportunities to produce biodegradable metallic implants—especially using Mg al...Due to their capability of fabricating geometrically complex structures,additive manufacturing(AM)techniques have provided unprecedented opportunities to produce biodegradable metallic implants—especially using Mg alloys,which exhibit appropriate mechanical properties and outstanding biocompatibility.However,many challenges hinder the fabrication of AM-processed biodegradable Mg-based implants,such as the difficulty of Mg powder preparation,powder splash,and crack formation during the AM process.In the present work,the challenges of AM-processed Mg components are analyzed and solutions to these challenges are proposed.A novel Mg-based alloy(Mg-Nd-Zn-Zr alloy,JDBM)powder with a smooth surface and good roundness was first synthesized successfully,and the AM parameters for Mg-based alloys were optimized.Based on the optimized parameters,porous JDBM scaffolds with three different architectures(biomimetic,diamond,and gyroid)were then fabricated by selective laser melting(SLM),and their mechanical properties and degradation behavior were evaluated.Finally,the gyroid scaffolds with the best performance were selected for dicalcium phosphate dihydrate(DCPD)coating treatment,which greatly suppressed the degradation rate and increased the cytocompatibility,indicating a promising prospect for clinical application as bone tissue engineering scaffolds.展开更多
The current COVID-19 pandemic urges the extremely sensitive and prompt detection of SARS-CoV-2 virus.Here,we present a Human Angiotensin-converting-enzyme 2(ACE2)-functionalized gold“virus traps”nanostructure as an ...The current COVID-19 pandemic urges the extremely sensitive and prompt detection of SARS-CoV-2 virus.Here,we present a Human Angiotensin-converting-enzyme 2(ACE2)-functionalized gold“virus traps”nanostructure as an extremely sensitive SERS biosensor,to selectively capture and rapidly detect S-protein expressed coronavirus,such as the current SARS-CoV-2 in the contaminated water,down to the single-virus level.Such a SERS sensor features extraordinary 106-fold virus enrichment originating from high-affinity of ACE2 with S protein as well as“virus-traps”composed of oblique gold nanoneedles,and 109-fold enhancement of Raman signals originating from multi-component SERS effects.Furthermore,the identification standard of virus signals is established by machine-learning and identification techniques,resulting in an especially low detection limit of 80 copies mL^(−1) for the simulated contaminated water by SARS-CoV-2 virus with complex circumstance as short as 5 min,which is of great significance for achieving real-time monitoring and early warning of coronavirus.Moreover,here-developed method can be used to establish the identification standard for future unknown coronavirus,and immediately enable extremely sensitive and rapid detection of novel virus.展开更多

Methods The skeleta...Objective To distinguish the edema, injury, or rupture in the traumatic skeletal muscle fiber in vivo using diffusion tensor imaging (DTI) and tractography on magnetic resonance imaging (MRI). Methods The skeletal muscle trauma models were made in 4 rabbits ( eight hindlimbs) by iron discus ( weight 1. 0 kg, diameter 6 cm) falling down vertically from 45 cm height to rabbits' thighs. Conventional sequences and two-dimensional (2D) diffusion-weighted (DW) spin-echo (SE) echo planar imaging (EPI) sequence with fat suppression ( b = 600 s/mm^2 ) were performed on 1.5T MRI scanner. The grading of edema, injury, and fiber rupture in the damaged muscle were made according to their histopathological views, which was consistent with the images. The mean apparent diffusion coefficient (ADC) values and fractional anisotropy (FA) values were measured from the region of interests (ROIs) of all groups on 2D DW images used for tractography. Analysis of variance test was performed to analyze all data. Results ADC values of the areas in normal muscle, edema muscle, injury muscle, and ruptured muscle were (6. 12±1.34 ) × 10^-3, ( 6.38±1.30 ) × 10^-3, ( 8.06±0. 97 ) × 10^-3, and ( 9. 57±0. 93 ) × 10^-3 mm^2/s, respectively. There was significant difference among groups (P 〈0. 001 ), but no difference between edema muscle and normal muscle group (P 〉0. 05). The FA values of normal muscle, edema muscle, injury muscle, and ruptured muscle were 0. 42±0. 12, 0. 36±0. 12, 0. 26±0. 09, 0. 12±0. 08, respectively, with a significant difference among groups ( P 〈 0. 001 ). In the edema muscle, the tracking cross-fiber could be seen but it decreased slightly. In the injury muscle, the tracking fiber decreased markedly. In the ruptured muscle, the transverse-orientation tracking fiber vanished, yet some interrupted longitudinal-orientation tracking fiber could be found. Conclsion The edema, injury, and rupture of muscle fiber in rabbit damaged skeletal muscle can be verified according to the ADC and the FA on DTI and tractography.展开更多

Ltd. were studied. It is found that C, Zn, K, Na, etc. exist in the fabric filter dust, off gas (OG) sludge, fine ash in converter, and electrical field dust in sinter. Among these, OG sludge gives the finest particle, more than 90% of which is less than 2.51 mm. The dust can lead to a serious negative influence on the production of sintering and blast furnaces (BF) if it is recycled in sintering. The briquette and reduction experimental results showed that the qualified strength could be obtained in the case of 8wt% molasses or 4wt% QT-10 added as binders. Also, more than 75% of metallization ratio, more than 95% of dezincing ratio, as well as more than 80% of K and Na removal rates were achieved for the briquettes kept at 1250℃ for 15 min during the direct reduction process. SEM observation indicated that the rates of indirect reduction and carbonization became dominating when the bri-quettes were kept at 1250℃ for 6 min.展开更多
Mg−Nd−Zn−Zr magnesium alloy(JDBM)has been studied widely as biodegradable medical material.To process high quality JDBM wires,effects of annealing on the mechanical properties and degradation behavior after drawing we...Mg−Nd−Zn−Zr magnesium alloy(JDBM)has been studied widely as biodegradable medical material.To process high quality JDBM wires,effects of annealing on the mechanical properties and degradation behavior after drawing were studied by microscopic observations,tensile and immersion tests.The as-extruded wires with a diameter of 3 mm could be drawn up to 9 passes without annealing until 125%cumulative drawing deformation.Complete recrystallization occurred after annealing at 325℃ for 30 min,350℃ for 5 min or 450℃for 3 min,respectively.Room temperature tensile tests and simulated body fluid immersion tests showed that annealing at slightly elevated temperature for short time could obtain better properties due to the finer grain size and more dispersive distribution of precipitates.For this study,annealing at 350℃ for 5 min is the best parameters which can be utilized to further fabricate fine wires.展开更多
Background: The LiBackpack is a recently developed backpack light detection and ranging(LiDAR) system that combines the flexibility of human walking with the nearby measurement in all directions to provide a novel and...Background: The LiBackpack is a recently developed backpack light detection and ranging(LiDAR) system that combines the flexibility of human walking with the nearby measurement in all directions to provide a novel and efficient approach to LiDAR remote sensing, especially useful for forest structure inventory. However, the measurement accuracy and error sources have not been systematically explored for this system.Method: In this study, we used the LiBackpack D-50 system to measure the diameter at breast height(DBH) for a Pinus sylvestris tree population in the Saihanba National Forest Park of China, and estimated the accuracy of LiBackpack measurements of DBH based on comparisons with manually measured DBH values in the field. We determined the optimal vertical slice thickness of the point cloud sample for achieving the most stable and accurate LiBackpack measurements of DBH for this tree species, and explored the effects of different factors on the measurement error.Result: 1) A vertical thickness of 30 cm for the point cloud sample slice provided the highest fitting accuracy(adjusted R2= 0.89, Root Mean Squared Error(RMSE) = 20.85 mm);2) the point cloud density had a significant negative, logarithmic relationship with measurement error of DBH and it explained 35.1% of the measurement error;3) the LiBackpack measurements of DBH were generally smaller than the manually measured values, and the corresponding measurement errors increased for larger trees;and 4) by considering the effect of the point cloud density correction, a transitional model can be fitted to approximate field measured DBH using LiBackpackscanned value with satisfactory accuracy(adjusted R2= 0.920;RMSE = 14.77 mm), and decrease the predicting error by 29.2%. Our study confirmed the reliability of the novel LiBackpack system in accurate forestry inventory, set up a useful transitional model between scanning data and the traditional manual-measured data specifically for P.sylvestris, and implied the applicable substitution of this new approach for more species, with necessary parameter calibration.展开更多
Interconnectivity is the key characteristic of bone tissue engineering scaffold modulating cell migration,blood vessels invasion and transport of nutrient and waste.However,efforts and understanding of the interconnec...Interconnectivity is the key characteristic of bone tissue engineering scaffold modulating cell migration,blood vessels invasion and transport of nutrient and waste.However,efforts and understanding of the interconnectivity of porous Mg is limited due to the diverse architectures of pore struts and pore size distribution of Mg scaffold systems.In this work,biomimetic hierarchical porous Mg scaffolds with tailored interconnectivity as well as pore size distribution were prepared by template replication of infiltration casting.Mg scaffold with better interconnectivity showed lower mechanical strength.Enlarging interconnected pores would enhance the interconnectivity of the whole scaffold and reduce the change of ion concentration,pH value and osmolality of the degradation microenvironment due to the lower specific surface area.Nevertheless,the degradation rates of five tested Mg scaffolds were no different because of the same geometry of strut unit.Direct cell culture and evaluation of cell density at both sides of four typical Mg scaffolds indicated that cell migration through hierarchical porous Mg scaffolds could be enhanced by not only bigger interconnected pore size but also larger main pore size.In summary,design of interconnectivity in terms of pore size distribution could regulate mechanical strength,microenvironment in cell culture condition and cell migration potential,and beyond that it shows great potential for personalized therapy which could facilitate the regeneration process.展开更多

文摘With the development of urban infrastructure,it is inevitable that shield tunnels will undercross intercity railways.However,the safe operation of intercity railways requires strict subgrade deformation.On the basis of the engineering background of the Lianghu Tunnel in Wuhan,the three-dimensional centrifuge test and numerical back analysis were used to study the development of subgrade surface settlement during shield tunneling.A three-dimensional numerical model with the same size as the prototype was subsequently established to further study the settlement development and torsion behavior of the subgrade during tunnel excavation.The results show that the maximum settlement point of the transverse settlement trough gradually moves to the tunnel axis during tunnel excavation and that the entire subgrade experiences torsional deformation.Moreover,the effect of the intersection angle between the axes of the tunnel and the subgrade on the surface settlement of the subgrade was further studied.The results show that the intersection angle has no effect on the maximum settlement,but the width of the settlement trough increases gradually with increasing angle.Finally,on the basis of the soil arching effect caused by tunnel excavation,the subgrade settlement during tunnel excavation is reduced by reinforcing the soil in different zones of soil arching.The results show that the settlement of the subgrade caused by the shield tunnel can be effectively controlled by adding reinforcement directly to the top of the tunnel,and the maximum settlement of the subgrade surface is reduced from 24.41 mm to 9.47 mm,a reduction of approximately 61.2%.

文摘In clinical settings,regenerating critical-sized calvarial bone defects presents substantial problems owing to the intricacy of surgical methods,restricted bone growth medications,and a scarcity of commercial bone grafts.To treat this life-threatening issue,improved biofunctional grafts capable of properly healing critical-sized bone defects are required.In this study,we effectively created anti-fracture hydrogel systems using spongy-like metal-organic(magnesium-phosphate)coordinated chitosan-modified injectable hydrogels(CPMg)loaded with a bioinspired neobavaisoflavone(NBF)component.The CPMg-NBF hydrogels showed outstanding anti-fracture capabilities during compression testing and retained exceptional mechanical stability even after 28 d of immersion in phosphatebuffered saline.They also demonstrated prolonged and stable release profiles of Mg^(2+)and NBF.Importantly,CPMg-NBF hydrogels revealed robust biphasic mineralization and were non-toxic to MC3T3-E1 cells.To better understand the underlying mechanism of Mg^(2+)and NBF component,as well as their synergistic effect on osteogenesis,we investigated the expression of key osteogenic proteins in the p38 MAPK and NOTCH pathways.Our results showed that CPMg-NBF hydrogels greatly increased the expression of osteogenic proteins(Runx2,OCN,OPN,BMPS and ALP).In vivo experiments showed that the implantation of CPMg-NBF hydrogels resulted in a significant increase in new bone growth within critical-sized calvarial defects.Based on these findings,we expect that the CPMg-NBF supramolecular hydrogel has tremendous promise for use as a therapeutic biomaterial for treating critical-sized calvarial defects.

文摘In this work,the phase structure of a holographic s+d model with quartic potential terms from 4D Einstein–Gauss–Bonnet gravity is studied in the probe limit.We first show the qd-μphase diagram with a very small value of the Gauss–Bonnet coefficientα=1×10-7and in the absence of the quartic terms to locate the suitable choice of the value of qd,where the system admits coexistent s+d solutions.Then we consider the various values of the Gauss–Bonnet coefficientαand present theα-μphase diagram to show the influence of the Gauss–Bonnet term on the phase structure.We also give an example of the re-entrant phase transition which is also realized in the holographic s+s and s+p models.After that we confirm the universality of the influence of the quartic term with coefficientλdon the d-wave solutions,which is similar to the case of s-wave and p-wave solutions previously studied in the s+p model.Finally we give the dependence of the special values of the quartic term coefficientλdon the Gauss–Bonnet coefficientα,below which the d-wave condensate grows to an opposite direction at the(quasi-)critical point,which is useful in realizing first order phase transitions in further studies of the holographic d-wave superfluids.

文摘Objective: To study the mechanisms in gambogic acid (GA) -induced JeKo-1 human Mantle Cell Lymphoma cell apoptosis in vitro. Methods: The proliferation of GA-treated JeKo-1 cells was measured by CCK-8 assay and Ki-67 immunocytochemical detection. Apopt0sis, cell cycle and mitochondrial membrane potential were measured by flow cytometric analysis. Caspase-3, -8 and -9 were detected by colorimetric assay. Bcl-2 and Bax were analyzed by Western blotting. Results: GA inhibited cell growth in a time- and dose- dependent manner. GA induces apoptosis in JeKo- 1 cells but not in normal bone marrow cells, which was involved in reducing the membrane potential of mitochondria, activating caspases-3, -8 and -9 and decreasing the ratio of Bd-2 and Bax without cell cycle arresting. Conclusions: GA induced apoptosis in human MCL JeKo-1 cells by regulating Bcl-2/Bax and activating caspase-3, -8 and -9 via mitochondrial pathway without affecting cell cycle.

文摘Triptolide,a component of the Chinese herb Tripterygium wilfordii Hook F,has been proved to be effective in the treatment of rheumatoid arthritis(RA).However,its underlying mechanisms on RA have not yet been well established.We observed the inhibitory effect of triptolide on the expression of inflammatory cytokines and proliferation of fibroblast-like synoviocytes(FLS)induced by the complex of interleukin-6(IL-6)and the soluble form of the IL-6 receptor(sIL-6R).Furthermore,to clarify the underlying mechanisms,we treated FLS with the Janus-activated kinase 2(JAK2)inhibitor/signal transducer and activator of transcription 3(STAT3)activation blocker AZD1480.In this study,immunohistochemical staining was used to identify vimentin(+)and CD68(−)in FLS.The FLS proliferation was measured by cell proliferation assay,and the cell cycles were analyzed by flow cytometry.Furthermore,ELISA was used to detect the expression of the inflammatory factors in culture solution.The expression levels of p-JAK2,JAK2,p-STAT3 and STAT3 were investigated through Western blotting analysis.The results showed that IL-6/sIL-6R significantly increased the cell proliferation and expression of inflammatory cytokines,including IL-6,interleukin-1β(IL-1β)and vascular endothelial growth factor(VEGF).Triptolide or AZD1480 inhibited the cell proliferation and inflammatory cytokine expression in IL-6/sIL-6R-stimulated FLS by suppressing JAK2/STAT3.The study suggested that the physiological effects of triptolide on RA were due to its contribution to the inhibition of the inflammatory cytokine expression and FLS proliferation by suppressing the JAK2/STAT3 signaling pathway.It may provide an innovative insight into the effect of triptolide in preventing RA pathogenesis.

文摘Due to their capability of fabricating geometrically complex structures,additive manufacturing(AM)techniques have provided unprecedented opportunities to produce biodegradable metallic implants—especially using Mg alloys,which exhibit appropriate mechanical properties and outstanding biocompatibility.However,many challenges hinder the fabrication of AM-processed biodegradable Mg-based implants,such as the difficulty of Mg powder preparation,powder splash,and crack formation during the AM process.In the present work,the challenges of AM-processed Mg components are analyzed and solutions to these challenges are proposed.A novel Mg-based alloy(Mg-Nd-Zn-Zr alloy,JDBM)powder with a smooth surface and good roundness was first synthesized successfully,and the AM parameters for Mg-based alloys were optimized.Based on the optimized parameters,porous JDBM scaffolds with three different architectures(biomimetic,diamond,and gyroid)were then fabricated by selective laser melting(SLM),and their mechanical properties and degradation behavior were evaluated.Finally,the gyroid scaffolds with the best performance were selected for dicalcium phosphate dihydrate(DCPD)coating treatment,which greatly suppressed the degradation rate and increased the cytocompatibility,indicating a promising prospect for clinical application as bone tissue engineering scaffolds.

文摘The current COVID-19 pandemic urges the extremely sensitive and prompt detection of SARS-CoV-2 virus.Here,we present a Human Angiotensin-converting-enzyme 2(ACE2)-functionalized gold“virus traps”nanostructure as an extremely sensitive SERS biosensor,to selectively capture and rapidly detect S-protein expressed coronavirus,such as the current SARS-CoV-2 in the contaminated water,down to the single-virus level.Such a SERS sensor features extraordinary 106-fold virus enrichment originating from high-affinity of ACE2 with S protein as well as“virus-traps”composed of oblique gold nanoneedles,and 109-fold enhancement of Raman signals originating from multi-component SERS effects.Furthermore,the identification standard of virus signals is established by machine-learning and identification techniques,resulting in an especially low detection limit of 80 copies mL^(−1) for the simulated contaminated water by SARS-CoV-2 virus with complex circumstance as short as 5 min,which is of great significance for achieving real-time monitoring and early warning of coronavirus.Moreover,here-developed method can be used to establish the identification standard for future unknown coronavirus,and immediately enable extremely sensitive and rapid detection of novel virus.
文摘Objective To distinguish the edema, injury, or rupture in the traumatic skeletal muscle fiber in vivo using diffusion tensor imaging (DTI) and tractography on magnetic resonance imaging (MRI). Methods The skeletal muscle trauma models were made in 4 rabbits ( eight hindlimbs) by iron discus ( weight 1. 0 kg, diameter 6 cm) falling down vertically from 45 cm height to rabbits' thighs. Conventional sequences and two-dimensional (2D) diffusion-weighted (DW) spin-echo (SE) echo planar imaging (EPI) sequence with fat suppression ( b = 600 s/mm^2 ) were performed on 1.5T MRI scanner. The grading of edema, injury, and fiber rupture in the damaged muscle were made according to their histopathological views, which was consistent with the images. The mean apparent diffusion coefficient (ADC) values and fractional anisotropy (FA) values were measured from the region of interests (ROIs) of all groups on 2D DW images used for tractography. Analysis of variance test was performed to analyze all data. Results ADC values of the areas in normal muscle, edema muscle, injury muscle, and ruptured muscle were (6. 12±1.34 ) × 10^-3, ( 6.38±1.30 ) × 10^-3, ( 8.06±0. 97 ) × 10^-3, and ( 9. 57±0. 93 ) × 10^-3 mm^2/s, respectively. There was significant difference among groups (P 〈0. 001 ), but no difference between edema muscle and normal muscle group (P 〉0. 05). The FA values of normal muscle, edema muscle, injury muscle, and ruptured muscle were 0. 42±0. 12, 0. 36±0. 12, 0. 26±0. 09, 0. 12±0. 08, respectively, with a significant difference among groups ( P 〈 0. 001 ). In the edema muscle, the tracking cross-fiber could be seen but it decreased slightly. In the injury muscle, the tracking fiber decreased markedly. In the ruptured muscle, the transverse-orientation tracking fiber vanished, yet some interrupted longitudinal-orientation tracking fiber could be found. Conclsion The edema, injury, and rupture of muscle fiber in rabbit damaged skeletal muscle can be verified according to the ADC and the FA on DTI and tractography.
文摘Basic physicochemical properties of the dust from Laiwu Iron and Steel Co. Ltd. were studied. It is found that C, Zn, K, Na, etc. exist in the fabric filter dust, off gas (OG) sludge, fine ash in converter, and electrical field dust in sinter. Among these, OG sludge gives the finest particle, more than 90% of which is less than 2.51 mm. The dust can lead to a serious negative influence on the production of sintering and blast furnaces (BF) if it is recycled in sintering. The briquette and reduction experimental results showed that the qualified strength could be obtained in the case of 8wt% molasses or 4wt% QT-10 added as binders. Also, more than 75% of metallization ratio, more than 95% of dezincing ratio, as well as more than 80% of K and Na removal rates were achieved for the briquettes kept at 1250℃ for 15 min during the direct reduction process. SEM observation indicated that the rates of indirect reduction and carbonization became dominating when the bri-quettes were kept at 1250℃ for 6 min.

文摘Mg−Nd−Zn−Zr magnesium alloy(JDBM)has been studied widely as biodegradable medical material.To process high quality JDBM wires,effects of annealing on the mechanical properties and degradation behavior after drawing were studied by microscopic observations,tensile and immersion tests.The as-extruded wires with a diameter of 3 mm could be drawn up to 9 passes without annealing until 125%cumulative drawing deformation.Complete recrystallization occurred after annealing at 325℃ for 30 min,350℃ for 5 min or 450℃for 3 min,respectively.Room temperature tensile tests and simulated body fluid immersion tests showed that annealing at slightly elevated temperature for short time could obtain better properties due to the finer grain size and more dispersive distribution of precipitates.For this study,annealing at 350℃ for 5 min is the best parameters which can be utilized to further fabricate fine wires.

文摘Background: The LiBackpack is a recently developed backpack light detection and ranging(LiDAR) system that combines the flexibility of human walking with the nearby measurement in all directions to provide a novel and efficient approach to LiDAR remote sensing, especially useful for forest structure inventory. However, the measurement accuracy and error sources have not been systematically explored for this system.Method: In this study, we used the LiBackpack D-50 system to measure the diameter at breast height(DBH) for a Pinus sylvestris tree population in the Saihanba National Forest Park of China, and estimated the accuracy of LiBackpack measurements of DBH based on comparisons with manually measured DBH values in the field. We determined the optimal vertical slice thickness of the point cloud sample for achieving the most stable and accurate LiBackpack measurements of DBH for this tree species, and explored the effects of different factors on the measurement error.Result: 1) A vertical thickness of 30 cm for the point cloud sample slice provided the highest fitting accuracy(adjusted R2= 0.89, Root Mean Squared Error(RMSE) = 20.85 mm);2) the point cloud density had a significant negative, logarithmic relationship with measurement error of DBH and it explained 35.1% of the measurement error;3) the LiBackpack measurements of DBH were generally smaller than the manually measured values, and the corresponding measurement errors increased for larger trees;and 4) by considering the effect of the point cloud density correction, a transitional model can be fitted to approximate field measured DBH using LiBackpackscanned value with satisfactory accuracy(adjusted R2= 0.920;RMSE = 14.77 mm), and decrease the predicting error by 29.2%. Our study confirmed the reliability of the novel LiBackpack system in accurate forestry inventory, set up a useful transitional model between scanning data and the traditional manual-measured data specifically for P.sylvestris, and implied the applicable substitution of this new approach for more species, with necessary parameter calibration.

文摘Interconnectivity is the key characteristic of bone tissue engineering scaffold modulating cell migration,blood vessels invasion and transport of nutrient and waste.However,efforts and understanding of the interconnectivity of porous Mg is limited due to the diverse architectures of pore struts and pore size distribution of Mg scaffold systems.In this work,biomimetic hierarchical porous Mg scaffolds with tailored interconnectivity as well as pore size distribution were prepared by template replication of infiltration casting.Mg scaffold with better interconnectivity showed lower mechanical strength.Enlarging interconnected pores would enhance the interconnectivity of the whole scaffold and reduce the change of ion concentration,pH value and osmolality of the degradation microenvironment due to the lower specific surface area.Nevertheless,the degradation rates of five tested Mg scaffolds were no different because of the same geometry of strut unit.Direct cell culture and evaluation of cell density at both sides of four typical Mg scaffolds indicated that cell migration through hierarchical porous Mg scaffolds could be enhanced by not only bigger interconnected pore size but also larger main pore size.In summary,design of interconnectivity in terms of pore size distribution could regulate mechanical strength,microenvironment in cell culture condition and cell migration potential,and beyond that it shows great potential for personalized therapy which could facilitate the regeneration process.
