Traumatic brain injury involves complex pathophysiological mechanisms,among which oxidative stress significantly contributes to the occurrence of secondary injury.In this study,we evaluated hypidone hydrochloride(YL-0919),a self-developed antidepressant with selective sigma-1 receptor agonist properties,and its associated mechanisms and targets in traumatic brain injury.Behavioral experiments to assess functional deficits were followed by assessment of neuronal damage through histological analyses and examination of blood-brain barrier permeability and brain edema.Next,we investigated the antioxidative effects of YL-0919 by assessing the levels of traditional markers of oxidative stress in vivo in mice and in vitro in HT22 cells.Finally,the targeted action of YL-0919 was verified by employing a sigma-1 receptor antagonist(BD-1047).Our findings demonstrated that YL-0919 markedly improved deficits in motor function and spatial cognition on day 3 post traumatic brain injury,while also decreasing neuronal mortality and reversing blood-brain barrier disruption and brain edema.Furthermore,YL-0919 effectively combated oxidative stress both in vivo and in vitro.The protective effects of YL-0919 were partially inhibited by BD-1047.These results indicated that YL-0919 relieved impairments in motor and spatial cognition by restraining oxidative stress,a neuroprotective effect that was partially reversed by the sigma-1 receptor antagonist BD-1047.YL-0919 may have potential as a new treatment for traumatic brain injury. 展开更多

Male infertility can result from impaired sperm motility caused by multiple morphological abnormalities of the flagella(MMAF).Distinct projections encircling the central microtubules of the spermatozoal axoneme play pivotal roles in flagellar bending and spermatozoal movement.Mammalian sperm-associated antigen 17(SPAG17)encodes a conserved axonemal protein of cilia and flagella,forming part of the C1a projection of the central apparatus,with functions related to ciliary/flagellar motility,skeletal growth,and male fertility.This study investigated two novel homozygous SPAG17 mutations(M1:NM_206996.2,c.829+1G>T,p.Asp212_Glu276del;and M2:c.2120del,p.Leu707*)identified in four infertile patients from two consanguineous Pakistani families.These patients displayed the MMAF phenotype confirmed by Papanicolaou staining and scanning electron microscopy assays of spermatozoa.Quantitative real-time polymerase chain reaction(PCR)of patients’spermatozoa also revealed a significant decrease in SPAG17 mRNA expression,and immunofluorescence staining showed the absence of SPAG17 protein signals along the flagella.However,no apparent ciliary-related symptoms or skeletal malformations were observed in the chest X-rays of any of the patients.Transmission electron microscopy of axoneme cross-sections from the patients showed incomplete C1a projection and a higher frequency of missing microtubule doublets 1 and 9 compared with those from fertile controls.Immunofluorescence staining and Western blot analyses of spermatogenesis-associated protein 17(SPATA17),a component of the C1a projection,and sperm-associated antigen 6(SPAG6),a marker of the spring layer,revealed disrupted expression of both proteins in the patients’spermatozoa.Altogether,these findings demonstrated that SPAG17 maintains the integrity of spermatozoal flagellar axoneme,expanding the phenotypic spectrum of SPAG17 mutations in humans. 展开更多

Asan emerging poserful tool to provide structural informstion af tissue specimens label-freely,Mueller matrix(MM)polarimetry has garnered extensive attention in biomedical studies and pathological diagnois.However,for the commonly used constant-step rotating MM polarimetricsystem,beam drift induæd by the rotation of polarization eements can lead to distortions in messurement results,severely affect ing MM imaging accuracy.Here,based on our previous study,we prоровe an optimizad self-registration method to mitigate the psæudo-depolarization effects introduced by image artifacts in constant-step rotatin g MM polarimetry.By addresing the prevalent issue of beam drift and image distortions in such polarimetric imaging systems,the effectivenes of the proposed method is experimentally validated using tissue samples.The result.s demonstrate a significant enhanæment in the accuIrsсy of depolarization parameter estimation after applying the optimized self-registration method.Furthermore,the method enhances the coarseness and contrsst of MM-derived parameters images,thereby bolstering their capacity to characterize tissuestructures.The optimized self-registration method proposed in this study can provide an innovstive spproach for quantitative tissue polarimetry bssæd on constant-step ro tating MM messurement,and contribute to the advanæment of polarimetric imaging technology in biomedical applications. 展开更多

Recently,Mueller matrix(MM)polarimetric imaging-assisted pathology detection methods are showing great potential in clinical diagnosis.However,since our human eyes cannot observe polarized light directly,it raises a notable challenge for interpreting the measurement results by pathologists who have limited familiarity with polarization images.One feasible approach is to combine MM polarimetric imaging with virtual staining techniques to generate standardized stained images,inheriting the advantages of information-abundant MM polarimetric imaging.In this study,we develop a model using unpaired MM polarimetric images and bright-field images for generating standard hematoxylin and eosin(H&E)stained tissue images.Compared with the existing polarization virtual staining techniques primarily based on the model training with paired images,the proposed Cycle-Consistent Generative Adversarial Networks(CycleGAN)-based model simplifies data acquisition and data preprocessing to a great extent.The outcomes demonstrate the feasibility of training CycleGAN with unpaired polarization images and their corresponding bright-field images as a viable approach,which provides an intuitive manner for pathologists for future polarization-assisted digital pathology. 展开更多

As a common fault of the aero-engine,the blade-casing rubbing(BCR)has the potential to cause catastrophic accidents.In this paper,to investigate the dynamic responses and wear characteristics of the system,the laminated shell element is used to establish the finite element model(FEM)of a flexibly coated casing system.Using the shell element,the blade is modeled,and the surface stress of the blade is calculated.The stress-solving method of the blade is validated through comparisons with the measured time-domain waveform of the stress.Then,a dynamic model of a blade-flexibly coated casing system with rubbing is proposed,accounting for the time-varying mass and stiffness of the casing caused by coating wear.The effects of the proposed flexible casing model are compared with those of a rigid casing model,and the stress changes induced by rubbing are investigated.The results show that the natural characteristics of the coated casing decrease due to the coating wear.The flexibly coated casing model is found to be more suitable for studying casing vibration.Additionally,the stress changes caused by rubbing are slight,and the change in the stress maximum is approximately 5%under the influence of the abrasive coating. 展开更多

Background Higher embryonic mortality,especially in aged breeding hens,is associated with insufficient hepatic functionality in maintaining redox homeostasis.Our previous study demonstrated that egg exosome-derived miRNAs may play a key role in modulating embryonic oxidation-reduction process,whereas the exact function and mechanism were still poorly understood.The present study aimed to investigate the roles of egg exosome miRNAs in maintaining dynamic equilibrium of free radicals and peroxide agents in embryonic liver,as well as demonstrate the specific mechanism using oxidative stress-challenged hepatocytes.Results Compared to 36-week-old breeding hens,decreased hatchability and increased embryonic mortality were observed in 65-week-old breeding hens.Meanwhile,the older group showed the increased MDA levels and decreased SOD and GSH-Px activities in embryonic liver,muscle and serum.Embryonic mortality was significantly positively correlated with MDA level and negatively correlated with GSH-Px activity in embryonic liver.In addition,363 differentially expressed genes(DEGs)were identified in embryonic liver,13 differentially expressed miRNAs(DE-miRNAs)were identified in egg exosomes.These DEGs and DE-miRNAs were involved in oxidoreductase activity,glutathione metabolic process,MAPK signaling pathway,apoptosis and autophagy.miRNA-mRNA network analysis further found that DEGs targeted by DE-miRNAs were mainly enriched in programmed cell death,such as apoptosis and autophagy.Wherein,MAPK10 with highest MCC and AUC values was significantly related to GSH-Px activity and MDA level,and served as the target gene of miR-145-5p based on dual luciferase reporter experiment and correlation analysis.Bioinformatics analysis found that miR-145-5p/MAPK10 axis might alleviate peroxide generation and apoptosis.In primary hepatocytes of chick embryos,miR-145-5p transfection significantly reversed H_(2)O_(2)-induced mitochondrial ROS increase,MAPK10,BAX and CASP3 overexpression and excessive apoptosis.Conclusion Exosome miR-145-5p in eggs could target MAPK10 and decrease mitochondrial ROS,attenuating oxidative damage and apoptosis in hepatocytes of chick embryos.These findings may provide new theoretical basis for the improvement of maternal physiological status to maintain embryonic redox homeostasis by nutritional or genetic modifications. 展开更多

Sexual maturation heterosis has been widely exploited in animal crossbreeding.However,the underlying mechanism has been rarely explored in chicken.In the present study,we performed the reciprocal crossing between White Leghorn and Beijing You chicken to evaluate the phenotypes related to sexual maturation,and profiled the ovary circRNAs of purebreds(WW,YY)and crossbreds(WY,YW)to elucidate the molecular mechanism underlying heterosis for sexual maturation.Pubic space and oviduct length exhibited positive heterosis,and age at first egg(AFE)exhibited negative heterosis in the crossbreds.We identified 3,025 known circRNAs and 624 putative circRNAs,which were mainly derived from the exons.Among these circRNAs,141 and 178circRNAs were specially expressed in WY and YW,respectively.There were 52.38 and 64.63%of total circRNAs in WY and YW exhibited non-additive expression pattern,respectively.GO enrichment and KEGG pathway analysis showed that the host genes of non-additive circRNAs were mainly involved in TGF-beta signaling pathway,oocyte development,ATPase activator activity,oocyte meiosis,progesterone-mediated oocyte maturation and GnRH signaling pathway.Weighted gene co-expression network analysis identified that 4 modules were significantly(P<0.05)correlated with oviduct length and pubic space.The host genes of non-additive circRNAs harbored in the 4 modules were associated with MAPK signaling pathway and Wnt signaling pathway.Furthermore,competing endogenous RNAs(ceRNA)network analysis characterized non-additive circRNAs gal-FGFR2_0005 and galMAPKAP1_0004 could interact with gga-miR-1612 and gga-miR-12235-5p to regulate CNOT6,COL8A1,and FHL2,which were essential for ovary development,indicating that the non-additive circRNAs involved in the formation of sexual maturation heterosis through regulating genes related to the reproductive and developmental process.The findings would provide a deeper understanding of the molecular mechanism underlying sexual maturation heterosis from a novel perspective. 展开更多

Multiple morphological abnormalities of the flagella(MMAF)represent a severe form of sperm defects leading to asthenozoospermia and male infertility.In this study,we identified a novel homozygous splicing mutation(c.871-4 ACA>A)in the adenylate kinase 7(AK7)gene by whole-exome sequencing in infertile individuals.Spermatozoa from affected individuals exhibited typical MMAF characteristics,including coiled,bent,short,absent,and irregular flagella.Transmission electron microscopy analysis showed disorganized axonemal structure and abnormal mitochondrial sheets in sperm flagella.Immunofluorescence staining confirmed the absence of AK7 protein from the patients’spermatozoa,validating the pathogenic nature of the mutation.This study provides direct evidence linking the AK7 gene to MMAF-associated asthenozoospermia in humans,expanding the mutational spectrum of AK7 and enhancing our understanding of the genetic basis of male infertility. 展开更多

Microbial fuel cells(MFCs)have become more prevalent in groundwater remediation due to their capacity for power generation,removal of pollution,ease of assembly,and low secondary contamination.It is currently being evaluated for practical application in an effort to eliminate groundwater pollution.However,a considerable majority of research was conducted in laboratories.But the operational circumstances including anaerobic characteristics,pH,and temperature vary at different sites.In addition,the complexity of contaminants and the positioning of MFCs significantly affect remediation performance.Taking the aforementioned factors into consideration,this reviewsummarizes a bibliography on the application of MFCs for the remediation of groundwater contamination during the last ten decades and assesses the impact of environmental conditions on the treatment performance.The design of the reactor,including configuration,dimensions,electrodes,membranes,separators,and target contaminants are discussed.This review aims to provide practical guidance for the future application of MFCs in groundwater remediation. 展开更多

Infertility represents a significant health concern,with sperm quantity and quality being crucial determinants of male fertility.Oligoasthenoteratozoospermia(OAT)is characterized by reduced sperm motility,lower sperm concentration,and morphological abnormalities in sperm heads and flagella.Although variants in several genes have been implicated in OAT,its genetic etiologies and pathogenetic mechanisms remain inadequately understood.In this study,we identified a homozygous nonsense mutation(c.916C>T,p.Arg306*)in the coiled-coil domain containing 146(CCDC146)gene in an infertile male patient with OAT.This mutation resulted in the production of a truncated CCDC146 protein(amino acids 1-305),retaining only two out of five coiled-coil domains.To validate the pathogenicity of the CCDC146 mutation,we generated a mouse model(Ccdc146^(mut/mut))with a similar mutation to that of the patient.Consistently,the Ccdc146mut/mut mice exhibited infertility,characterized by significantly reduced sperm counts,diminished motility,and multiple defects in sperm heads and flagella.Furthermore,the levels of axonemal proteins,including DNAH17,DNAH1,and SPAG6,were significantly reduced in the sperm of Ccdc146^(mut/mut) mice.Additionally,both human and mouse CCDC146 interacted with intraflagellar transport protein 20(IFT20),but this interaction was lost in the mutated versions,leading to the degradation of IFT20.This study identified a novel deleterious homozygous nonsense mutation in CCDC146 that causes male infertility,potentially by disrupting axonemal protein transportation.These findings offer valuable insights for genetic counseling and understanding the mechanisms underlying CCDC146 mutant-associated infertility in human males. 展开更多

Crossed beak is a complex mode of inheritance with prevalence ranging from 0.2 to 7.4% in at least 12 chicken strains worldwide.To reveal the intrinsic factors causing crossed beaks,genes expression patterns in bilateral mandibular condyle between affected and normal birds were characterized by RNA sequencing analysis in the present studies.Crossed beak was induced by short length of unilateral mandibular ramus,and a total of 110differentially expressed genes were up-or down-regulated in the affected(short)mandibular condyle side as compared to the normal side.Carbonic anhydrase 2(CA2)and Carbonic anhydrase 13(CA13)were enriched in the carbonate dehydratase activity,and high-expressed in mandibular condyle and osteoblasts(P<0.05).However,both were low-expressed in short mandibular condyle side of affected birds(P<0.05).The carbonate dehydratase inhibitor experiments confirmed that there is positive association between the calcification and carbonic anhydrase isoenzymes.Quantitative analysis with cetylpyridinium chloride showed a decrease in calcification when the cells were transfected with an anti-CA13 shRNA.Our research suggested that CA2 and CA13 are down-calcified in shortside mandibular condyle,and caused mandibular ramus to grow slowly.CA2 and CA13 have the critical role in crossed beaks by regulating calcification of mandibular condyle. 展开更多

Mechanically cleaved two-dimensional materials are random in size and thickness.Recognizing atomically thin flakes by human experts is inefficient and unsuitable for scalable production.Deep learning algorithms have been adopted as an alternative,nevertheless a major challenge is a lack of sufficient actual training images.Here we report the generation of synthetic two-dimensional materials images using StyleGAN3 to complement the dataset.DeepLabv3Plus network is trained with the synthetic images which reduces overfitting and improves recognition accuracy to over 90%.A semi-supervisory technique for labeling images is introduced to reduce manual efforts.The sharper edges recognized by this method facilitate material stacking with precise edge alignment,which benefits exploring novel properties of layered-material devices that crucially depend on the interlayer twist-angle.This feasible and efficient method allows for the rapid and high-quality manufacturing of atomically thin materials and devices. 展开更多

To better understand the challenges faced by public hospitals in implementing charitable medical assistance,this study examines Nanjing R Hospital as a case study.By combining literature review and field research,the current status and fundamental methods of charitable medical assistance at R Hospital are systematically analyzed.The findings indicate that medical social workers play a multifaceted role in the hospital’s charitable assistance efforts.They contribute significantly to building assistance networks and addressing patients’multi-dimensional needs.However,challenges persist in the assistance process,including both external and internal obstacles.This study explores ways to overcome these barriers to enhance the efficient utilization of healthcare resources.Through empirical investigation,the study identifies the types of charitable aid resources available and highlights the practical difficulties in their application within public hospitals.These findings provide a valuable reference for optimizing resource allocation,improving the effectiveness of assistance programs,and fostering institutional collaboration. 展开更多

Background Crossbreeding is widely promoted as an efficient strategy to improve the productivity in agriculture.The molecular mechanism underlying heterosis for egg production is always intriguing in chicken.The transcriptional dynamic changes play a crucial role in the formation of heterosis,but little is known for the egg production traits.Results In present study,we measured the continuous manifestation of heterosis ranging from 2.67%to 10.24%for egg number in the crossbreds generated by reciprocal crossing White Leghorn and Beijing You chicken.The high-quality transcriptomes of ovary for purebreds(WW and YY)and crossbreds(WY and YW)in 5 laying stages were sequenced and integrated to identify regulatory networks relevant to the heterosis.We found highly conserved transcriptional features among 4 genetic groups.By using weighted gene co-expression network analysis(WGCNA),we obtained multiple gene co-expression modules that were significantly correlated with egg number for each group.The common KEGG pathways including apelin signaling pathway,cell cycle,ribosome,spliceosome and oxidative phosphorylation,were screened for the 2 crossbreds.Then,we identified consensus co-expression modules(CMs)that showed divergent expression pattern among crossbred(WY or YW)and purebreds(WW and YY).The hub genes of CMs were again overrepresented in the cell cycle pathway,and the crossbreds exhibited temporally complementary dominance of hub genes in the 5 laying stages.These results suggested that the crossbreds inherited from both parents to maintain the ovary function by cell cycle-related genes,contributing to the persistent heterosis for egg production.Furthermore,the dominant genes including MAD2L1,CHEK2 and E2F1 were demonstrated to function in ovarian follicle development and maturation and could be the candidate genes for egg production heterosis.Conclusion Our study characterized the dynamic profile of genome-wide gene expression in ovary and highlighted the role of dominant expression of cell cycle pathway genes in heterosis.These findings provided new insights for the molecular mechanism of egg production heterosis,which would facilitate the rational choice of suitable parents for producing crossbred chickens with higher egg production. 展开更多

Histone H2B ubiquitination(H2Bub)is positively linked to transcriptional activation,but the genetic programs affected by H2Bub to enhance drought tolerance remain largely unknown.Here,we show that OsbZIP27 interacts directly with OsHUB1/2 to regulate drought tolerance in rice by binding to the promoters of OsHAK1 and OsGLN1 to achieve H2Bub and transcriptional activation.Consistently,mutations in OsbZIP27 reduce transcription of OsHAK1 and OsGLN1,resulting in increased sensitivity to drought stress.Moreover,loss of OsHUB1 and OsHUB2 function causes hypersensitivity to drought stress,whereas OsHUB2 overexpression enhances drought tolerance.Together,our results indicate that OsbZIP27 coordinates with OsHUB1/2 to enhance rice drought tolerance by increasing H2Bub and expression of OsHAK1 and OsGLN1. 展开更多

Degradation of nitrobenzene(NB)via Fenton-like reaction is considered as an efficient approach for contaminated groundwater remediation.However,the poor stability of H2O2limits the application of traditional Fenton reactions in soil and groundwater due to the transportation risks of H_(2)O_(2).In this study,we synthesized a controlled release nano calcium peroxide(n CP)by coating it with polydopamine(PDA)as a solid H2O2to construct a Fe(Ⅱ)/PDA@n CP Fenton-like system for contaminants degradation.The phenolquinone transformations of catechol groups on the PDA surface facilitated the Fe(Ⅱ)/Fe(Ⅲ)cycle,resulting in enhanced generation of hydroxyl radicals(HO·)and effective long-term degradation of NB.Moreover,the PDA shell modulated the n CP decomposition rate and inhibited sharp p H fluctuations,and the NB removal efficiency was achieved up to 96.8%at p H ranging from 3.0 to 9.0.This study demonstrated the promising application potential of PDA@n CP as a solid-controlled release H2O2source in Fenton-like system for groundwater contamination remediation. 展开更多
