Simulated Asymmetric Connection of SST in the Tasman Sea with Respect to the Opposite Phases of ENSO in Austral Summer in CMIP6 Models

A prior observational study indicated an asymmetric link between sea surface temperature(SST)in the Tasman Sea and ENSO during austral summer.Specifically,El Niño is associated with a dipolar SST anomaly pattern,featuring warming in the northwest and cooling in the southeast,whereas La Niña corresponds to basin-scale warming.This study employs the experiments of coupled models from the sixth phase of the Coupled Model Intercomparison Project(CMIP6)to assess ENSO’s impact on Tasman Sea SST.While all 15 models capture the observed dipolar SST anomalies(SSTAs)in the Tasman Sea during El Niño years,only 7 models capture the basin-scale warmth in the Tasman Sea during La Niña years.Consequently,the models are bifurcated into two groups:group-one models yield one physically reasonable asymmetric connection as observed,including the asymmetry of oceanic heat transport,especially the Ekman meridional transport anomalies induced by zonal wind stress driven by the asymmetric atmospheric circulation over the Tasman Sea.However,due to abnormal responses to ENSO and systematic biases in model simulations,including jet and storm tracks,oceanic heat fluxes,ocean currents,and SST,the group-two models fail to reproduce the asymmetric connection between the Tasman Sea and ENSO.This study not only validates the observational asymmetric connection of SSTAs in the Tasman Sea with respect to the two opposite ENSO phases,but also provides evidence and clues to reduce the bias in group-two models. 展开更多

A Oilfield is the most representative mid to deep oil field in the Bohai Sea, with the main oil bearing intervals being the upper and middle Shahejie-3 sections of the Paleogene Shahejie Formation. By combining well seismic analysis, the middle section of Shahejie-3 is divided into high-level system tract and forced lake retreat system tract, corresponding to the II oil formation and I oil formation, respectively. Using sequence stratigraphy methods, based on seismic profiles and drilling lithological cycles, the high stand system tract is divided into 5 stages of delta progradation. The first and second stages are high angle S-type progradation with large sedimentary thickness, the third stage is oblique progradation, and the fourth and fifth stages are S-oblique composite progradation;By combining seismic data, we characterized the large-scale (8 small-scale) progradation bodies of 5 periods, clarified the distribution characteristics of reservoir planes, and laid the foundation for the later exploration of oilfield potential. 展开更多

BACKGROUND Gastric cancer has a high incidence and fatality rate,and surgery is the preferred course of treatment.Nonetheless,patient survival rates are still low,and the incidence of major postoperative complications cannot be disregarded.The systemic inflammatory response,nutritional level,and coagulation status are key factors affecting the postoperative recovery and prognosis of gastric cancer patients.The systemic inflammatory response index(SIRI)and the albumin fibrinogen ratio(AFR)are two valuable comprehensive indicators of the severity and prognosis of systemic inflammation in various medical conditions.AIM To assess the clinical importance and prognostic significance of the SIRI scores and the AFR on early postoperative outcomes in patients undergoing radical gastric cancer surgery.METHODS We conducted a retrospective analysis of the clinicopathological characteristics and relevant laboratory indices of 568 gastric cancer patients from January 2018 to December 2019.We calculated and compared two indicators of inflammation and then examined the diagnostic ability of combined SIRI and AFR values for serious early postoperative complications.We scored the patients and categorized them into three groups based on their SIRI and AFR levels.COX analysis was used to compare the three groups of patients the prognostic value of various preoperative SIRI-AFR scores for 5-year overall survival(OS)and disease-free survival(DFS).RESULTS SIRI-AFR scores were an independent risk factor for prognosis[OS:P=0.004;hazards ratio(HR)=3.134;DFS:P<0.001;HR=3.543]and had the highest diagnostic power(area under the curve:0.779;95%confidence interval:0.737-0.820)for early serious complications in patients with gastric cancer.The tumor-node-metastasis stage(P=0.001),perioperative transfusion(P=0.044),positive carcinoembryonic antigen(P=0.014)findings,and major postoperative complications(P=0.011)were factors associated with prognosis.CONCLUSION Preoperative SIRI and AFR values were significantly associated with early postoperative survival and the occurrence of severe complications in gastric cancer patients. 展开更多

Comprehensive analysis of the potential pathogenesis of COVID-19 infection and liver cancer

BACKGROUND A growing number of clinical examples suggest that coronavirus disease 2019(COVID-19)appears to have an impact on the treatment of patients with liver cancer compared to the normal population,and the prevalence of COVID-19 is significantly higher in patients with liver cancer.However,this mechanism of action has not been clarified.Gene sets for COVID-19(GSE180226)and liver cancer(GSE87630)were obtained from the Gene Expression Omnibus database.After identifying the common differentially expressed genes(DEGs)of COVID-19 and liver cancer,functional enrichment analysis,protein-protein interaction network construction and scree-ning and analysis of hub genes were performed.Subsequently,the validation of the differential expression of hub genes in the disease was performed and the regulatory network of transcription factors and hub genes was constructed.RESULTS Of 518 common DEGs were obtained by screening for functional analysis.Fifteen hub genes including aurora kinase B,cyclin B2,cell division cycle 20,cell division cycle associated 8,nucleolar and spindle associated protein 1,etc.,were further identified from DEGs using the“cytoHubba”plugin.Functional enrichment analysis of hub genes showed that these hub genes are associated with P53 signalling pathway regulation,cell cycle and other functions,and they may serve as potential molecular markers for COVID-19 and liver cancer.Finally,we selected 10 of the hub genes for in vitro expression validation in liver cancer cells.CONCLUSION Our study reveals a common pathogenesis of liver cancer and COVID-19.These common pathways and key genes may provide new ideas for further mechanistic studies. 展开更多

BACKGROUND The pyruvate dehydrogenase E1 subunitβ(PDHB)gene which regulates energy metabolism is located in mitochondria.However,few studies have elucidated the role and mechanism of PDHB in different cancers.AIM To comprehensive pan-cancer analysis of PDHB was performed based on bioinformatics approaches to explore its tumor diagnostic and prognostic value and tumor immune relevance in cancer.In vitro experiments were performed to examine the biological regulation of PDHB in liver cancer.METHODS Pan-cancer data related to PDHB were obtained from the Cancer Genome Atlas(TCGA)database.Analysis of the gene expression profiles of PDHB was based on TCGA and Genotype Tissue Expression Dataset databases.Cox regression analysis and Kaplan-Meier methods were used to assess the correlation between PDHB expression and survival prognosis in cancer patients.The correlation between PDHB and receiver operating characteristic diagnostic curve,clinicopathological staging,somatic mutation,tumor mutation burden(TMB),microsatellite instability(MSI),DNA methylation,and drug susceptibility in pan-cancer was also analyzed.Various algorithms were used to analyze the correlation between PDHB and immune cell infiltration and tumor chemotaxis environment,as well as the co-expression analysis of PDHB and immune checkpoint(ICP)genes.The expression and functional phenotype of PDHB in single tumor cells were studied by single-cell sequencing,and the functional enrichment analysis of PDHB-related genes was performed.The study also validated the level of mRNA or protein expression of PDHB in several cancers.Finally,in vitro experiments verified the regulatory effect of PDHB on the proliferation,migration,and invasion of liver cancer.RESULTS PDHB was significantly and differently expressed in most cancers.PDHB was significantly associated with prognosis in patients with a wide range of cancers,including kidney renal clear cell carcinoma,kidney renal papillary cell carcinoma,breast invasive carcinoma,and brain lower grade glioma.In some cancers,PDHB expression was clearly associated with gene mutations,clinicopathological stages,and expression of TMB,MSI,and ICP genes.The expression of PDHB was closely related to the infiltration of multiple immune cells in the immune microenvironment and the regulation of tumor chemotaxis environment.In addition,single-cell sequencing results showed that PDHB correlated with different biological phenotypes of multiple cancer single cells.This study further demonstrated that down-regulation of PDHB expression inhibited the proliferation,migration,and invasion functions of hepatoma cells.CONCLUSION As a member of pan-cancer,PDHB may be a novel cancer marker with potential value in diagnosing cancer,predicting prognosis,and in targeted therapy. 展开更多

BACKGROUND The systemic inflammatory response index(SIRI)has been demonstrated to make a significant difference in assessing the prognosis of patients with different solid neoplasms.However,research is needed to ascertain the accuracy and reliability of applying the SIRI to patients who undergo robotic radical gastric cancer sur-gery.AIM To validate the applicability of the SIRI in assessing the survival of gastric cancer patients and evaluate the clinical contribution of preoperative SIRI levels to predicting long-term tumor outcomes in patients,who received robotic radical gastric cancer surgery.METHODS Initially,an exhaustive retrieval was performed in the PubMed,the Cochrane Library,EMBASE,Web of Science,and Scopus databases to identify relevant studies.Subsequently,a meta-analysis was executed on 6 cohort studies iden-tifying the value of the SIRI in assessing the survival of gastric cancer patients.Additionally,the clinical data of 161 patients undergoing robotic radical gastric cancer surgery were retrospectively analyzed to evaluate their clinicopathological characteristics and relevant laboratory indicators.The association between preoperative SIRI levels and 5-year overall survival(OS)and disease-free survival(DFS)was assessed.RESULTS The findings demonstrated an extensive connection between SIRI values and the outcome of patients with gastric cancer.Preoperative SIRI levels were identified as an independent hazard feature for both OS and DFS among those who received robotic surgery for gastric cancer.SIRI levels in gastric cancer patients were observed to be associated with the presence of comorbidities,T-stage,carcinoembryonic antigen levels,the development of early serious postoperative complications,and the rate of lymph node metastasis.CONCLUSION SIRI values are correlated with adverse in the gastric cancer population and have the potential to be utilized in predicting long-term oncological survival in patients who undergo robotic radical gastric cancer surgery. 展开更多

目的:介绍程序化达芬奇机器人“3+2”模式远端胃癌根治手术清扫“七步法”。方法:回顾性分析2017年6月至2020年6月于甘肃省人民医院完成全腔镜下达芬奇机器人远端胃癌根治术的患者100例。手术分七步:1)建立戳孔,腹腔探查,悬调肝脏,摆体... 目的:介绍程序化达芬奇机器人“3+2”模式远端胃癌根治手术清扫“七步法”。方法:回顾性分析2017年6月至2020年6月于甘肃省人民医院完成全腔镜下达芬奇机器人远端胃癌根治术的患者100例。手术分七步:1)建立戳孔,腹腔探查,悬调肝脏,摆体位,连接机器;2)右侧大弯侧区域及幽门下区域(4d,6);3)幽门上区域,离断十二指肠,清扫肝十二指肠韧带(5,12a);4)胰腺前方区域(7,8a,8p,9,11p);5)小弯侧区域(1,3);6)左侧胃大弯区域(4sb);7)离断胃标本,近端胃和空肠吻合。结果:全部患者无中转开腹,术中无输血。手术时间(175.0±16)min,出血量(99±36.4)mL,淋巴结清扫数目(30.3±2.5)枚,机器人相关费用(22794±3660)元。并发症发生5例,其中肠梗阻2例,腹腔内出血1例、伤口感染1例以及胃排空延迟1例,均采用保守方法治愈。未出现吻合口狭窄及吻合口瘘。结论:本程序化方法安全可行,重复性好,手术时间相对较短,费用相对较低。该程序化方法有利于新开展机器人手术的机构培养手术团队、缩短学习曲线、降低手术难度,从而更好地服务于临床教学工作。 展开更多

目的:明确miR-519d-3p对高糖诱导的人视网膜微血管内皮细胞(HRMEC)功能障碍与血管生成的影响,并阐明其对低氧诱导因子-1α(HIF-1α)的调控机制。方法:通过5、30mmol/L葡萄糖分别诱导HRMEC建立正常(NG)和高糖(HG)细胞模型。将HRMEC分为... 目的:明确miR-519d-3p对高糖诱导的人视网膜微血管内皮细胞(HRMEC)功能障碍与血管生成的影响,并阐明其对低氧诱导因子-1α(HIF-1α)的调控机制。方法:通过5、30mmol/L葡萄糖分别诱导HRMEC建立正常(NG)和高糖(HG)细胞模型。将HRMEC分为对照组(HG细胞模型转染阴性对照模拟物)、甘露醇组(对照组加入25mmol/L甘露醇)、miR-519d-3p过表达组(HG细胞模型转染miR-519d-3p模拟物)、miR-519d-3p联合HIF-1α过表达组(HG细胞模型共转染miR-519d-3p模拟物和HIF-1α过表达载体)。实时荧光定量PCR法检测各组miR-519d-3p的表达情况。Western blotting法检测各组HIF-1α蛋白的表达情况。荧光素酶报告基因实验检测miR-519d-3p和HIF-1α的结合位点情况。CCK-8法检测各组细胞增殖情况。Hoechst 33342染色法检测各组细胞凋亡情况。ELISA法检测各组细胞外液炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6蛋白的表达情况。小管形成实验检测各组新生毛细血管管腔样结构形成情况。结果:与NG相比,HG细胞模型中miR-519d-3p表达显著减少,而HIF-1α蛋白表达显著增加(均P<0.01)。与对照组比较,miR-519d-3p过表达组中HIF-1α蛋白表达显著降低(P<0.01)。miR-519d-3p中“CGUGAAA”序列可以与HIF-1α3'-非编码区(3'-UTR)中“GCACUUU”序列特异性结合。与对照组比较,miR-519d-3p过表达组细胞24、48、72h吸光度值均显著增加,细胞凋亡率显著减少,细胞外液TNF-α、IL-1β、IL-6浓度均显著减少,新生毛细血管管腔样结构数量显著减少(均P<0.01)。与miR-519d-3p过表达组比较,miR-519d-3p联合HIF-1α过表达组细胞24、48、72h吸光度值均显著减少,细胞凋亡率显著增加,细胞外液TNF-α、IL-1β、IL-6浓度均显著增加,新生毛细血管管腔样结构数量显著增加(均P<0.01)。对照组和甘露醇组中上述各指标比较无差异(均P>0.05)。结论:高糖诱导HRMEC模型中miR-519d-3p表达下调,而HIF-1α蛋白表达上调。HIF-1α是miR-519d-3p的靶基因,miR-519d-3p靶向HIF-1α增加细胞增殖并降低细胞凋亡和炎症反应,从而减轻高糖诱导的HRMEC功能障碍并抑制血管生成。 展开更多

Optimization of Injection Parameters for Profile Control and Flooding in an Oilfield during High Water Cut Period

In order to improve the effect of water control and oil stabilization during high water cut period, a mathematical model of five point method well group was established with the high water cut well group of an Oilfield as the target area, the variation law of water cut and recovery factor of different injection parameters was analyzed, and the optimization research of injection parameters of polymer enhanced foam flooding was carried out. The results show that the higher the injection rate, the lower the water content curve, and the higher the oil recovery rate. As the foam defoamed when encountering oil, when the injection time was earlier than 80% of water cut, the later the injection time was, the better the oil displacement effect would be. When the injection time was later than 80% of water cut, the later the injection time was, the worse the oil displacement effect would be. The larger the injection volume, the lower the water content curve and the higher the recovery rate. After the injection volume exceeded 0.2 PV, the amplitude of changes in water content and recovery rate slowed down. The optimal injection parameters of profile control agent for high water content well group in Oilfield A were: injection rate of 15 m<sup>3</sup>/d, injection timing of 80% water content, and injection volume of 0.2 PV. 展开更多

Normal spermatogenic processes require the scrotal temperature to be lower than that of the body as excessive heat affects spermatogenesis in the testes,reduces sperm quality and quantity,and even causes infertility.Endoplasmic reticulum stress(ERS)is a crucial factor in many pathologies.Although several studies have linked ERS to heat stress,researchers have not yet determined which ERS signaling pathways contribute to heat-induced testicular damage.Melatonin activates antioxidant enzymes,scavenges free radicals,and protects the testes from inflammation;however,few studies have reported on the influence of melatonin on heat-induced testicular damage.Using a murine model of testicular hyperthermia,we observed that heat stress causes both ERS and apoptosis in the testes,especially in the spermatocytes.These observations were confirmed using the mouse spermatocyte cell line GC2,where the Atf6 and Perk signaling pathways were activated during heat stress.Knockout of the above genes effectively reduced spermatocyte damage caused by heat stress.Pretreatment with melatonin alleviated heat-induced apoptosis by inhibiting the Atf6 and Perk signaling pathways.This mitigation was dependent on the melatonin receptors.In vivo experiments verified that melatonin treatment relieved heat-induced testicular damage.In conclusion,our results demonstrated that ATF6 and PERK are important mediators for heat-induced apoptosis,which can be prevented by melatonin treatment.Thus,our study highlights melatonin as a potential therapeutic agent in mammals for subfertility/infertility induced by testicular hyperthermia. 展开更多

Postmenopausal osteoporosis is a kind of degenerative disease,also described as“invisible killer.”Estrogen is generally considered as the key hormone for women to maintain bone mineral content during their lives.Iron accumulation refers to a state of human serum ferritin that is higher than the normal value but less than 1000μg/L.It has been found that iron accumulation and osteoporosis could occur simultaneously with the decrease in estrogen level after menopause.In recent years,many studies indicated that iron accumulation plays a vital role in postmenopausal osteoporosis,and a significant correlation has been found between iron accumulation and fragility fractures.In this review,we summarize and analyze the relevant literature including randomized controlled trials,systematic reviews,and meta-analyses between January 1996 and July 2022.We investigate the mechanism of the effect of iron accumulation on bone metabolism and discuss the relationship of iron accumulation,osteoporosis,and postmenopausal fragility fractures,as well as the main clinical treatment strategies.We conclude that it is necessary to pay attention to the phenomenon of iron accumulation in postmenopausal women with osteoporosis and explore the in-depth mechanism of abnormal bone metabolism caused by iron accumulation,in order to facilitate the discovery of effective therapeutic targets for postmenopausal osteoporosis. 展开更多

BACKGROUND Conventional Billroth Ⅱ(BⅡ) anastomosis after laparoscopic distal gastrectomy(LDG) for gastric cancer(GC) is associated with bile reflux gastritis, and Roux-enY anastomosis is associated with Roux-Y stasis syndrome(RSS). The uncut Rouxen-Y(URY) gastrojejunostomy reduces these complications by blocking the entry of bile and pancreatic juice into the residual stomach and preserving the impulse originating from the duodenum, while BⅡ with Braun(BB) anastomosis reduces the postoperative biliary reflux without RSS. Therefore, the purpose of this study was to compare the efficacy and safety of laparoscopic URY with BB anastomosis in patients with GC who underwent radical distal gastrectomy.AIM To evaluate the value of URY in patients with GC.METHODS PubMed, Embase, Web of Science, Cochrane Library, Chinese National Knowledge Infrastructure, Wanfang, Chinese Biomedical Database, and VIP Database for Chinese Technical Periodicals(VIP) were used to search relevant studies published from January 1994 to August 18, 2021. The following databases were also used in our search: Clinicaltrials.gov, Data Archiving and Networked Services, the World Health Organization International Clinical Trials Registry Platform Search Portal(https://www.who.int/clinical-trials-registry-platform/the-ictrp-search-portal), the reference lists of articles and relevant conference proceedings in August 2021. In addition, we conducted a relevant search by Reference Citation Analysis(RCA)(https://www.referencecitationanalysis.com). We cited highquality references using its results analysis functionality. The methodological quality of the eligible randomized clinical trials(RCTs) was evaluated using the Cochrane Risk of Bias Tool, and the non-RCTs were evaluated using the Newcastle-Ottawa scale. Statistical analyses were performed using Review Manager(Version 5.4).RESULTS Eight studies involving 704 patients were included in this meta-analysis. The incidence of reflux gastritis [odds ratio = 0.07, 95% confidence interval(CI): 0.03-0.19, P < 0.00001] was significantly lower in the URY group than in the BB group. The pH of the postoperative gastric fluid was lower in the URY group than in the BB group at 1 d [mean difference(MD) =-2.03, 95%CI:(-2.73)-(-1.32),P < 0.00001] and 3 d [MD =-2.03, 95%CI:(-2.57)-(-2.03), P < 0.00001] after the operation. However,no significant difference in all the intraoperative outcomes was found between the two groups.CONCLUSION This work suggests that URY is superior to BB in gastrointestinal reconstruction after LDG when considering postoperative outcomes. 展开更多

Background:Acupuncture has been widely used to relieve migraine-related symptoms.However,the findings of previous systematic reviews(SRs)and meta-analyses(MAs)are still not completely consistent.Their quality is also unknown,so a comprehensive study is needed.Objective:To evaluate the reporting and methodological quality of these MAs concerning acupuncture for migraine,and summarize evidence about the efficacy and safety of acupuncture for migraine.Search strategy:Pub Med,Embase,Cochrane Library,China National Knowledge Infrastructure,Chinese Biomedical Databases,Wanfang Data,and VIP databases were searched from inception to September 2020,with a comprehensive search strategy.Inclusion criteria:The pairwise MAs of randomized controlled trials(RCTs)concerning migraine treated by acupuncture or acupuncture-based therapies,with a control group that received sham acupuncture,medication,no treatment,or acupuncture at different acupoints were included.Data extraction and analysis:Two independent investigators screened studies,extracted relevant data,and assessed reporting and methodological quality using Preferred Reporting Items for Systematic Reviews and Meta-Analyses(PRISMA)2009 and A Measurement Tool to Assess Systematic Reviews 2(AMSTAR 2),then all results were cross-checked.Spearman correlation test was used to evaluate the correlation between reporting and methodological quality scores.Results:A total of 20 MAs were included in this study.The included MAs indicated that acupuncture was efficacious and safe in preventing and treating migraine when compared with control intervention.There was a high correlation between reporting and methodological quality scores(rs=0.87,P<0.001).The quality of the included SRs needs to be improved mainly with regard to protocol and prospective registration,using a comprehensive search strategy,summarizing the strength of evidence body for key outcomes,a full list of excluded studies with reasons for exclusion,reporting of RCTs’funding sources,and assessing the potential impact of risk of bias in RCTs on MA results.Conclusion:Acupuncture is an effective and safe intervention for preventing and treating migraine,and could be considered as a good option for patients with migraine.However,the reporting and methodological quality of MAs included in this overview is suboptimal.In the future,AMSTAR 2 and PRISMA tools should be followed when making and reporting an SR with MA. 展开更多

Gastric cancer(GC) is the most prevalent malignancy in the world,especially in China.GC has been postulated to spread via several different routes,including through hematogenous channels,lymphatic vessels,the seeding of peritoneal surfaces,direct extension through the gastric wall,and retrograde extension through the vas deferens or lymphatics.Testicular metastasis is rare.We show here a 53-year-old patient with GC who underwent a radical total gastrectomy approximately 22 mo ago after he presented with a sensation of heaviness and swelling of the right hemiscrotum.The diagnosis of metastatic adenocarcinoma was made after a rightside orchiectomy.We report the first case of testicular metastasis from gastric adenocarcinoma in China's Mainland and summarize the clinicopathologic features of the disease based on previously published papers. 展开更多

A recent genome-wide association study identified a new susceptibility locus for breast cancer, rs2046210, which is a single nucleotide polymorphism (SNP) located upstream of the estrogen receptor α (ESR1) gene on chromosome 6q25.1. Given that endometrial cancer shares many risk factors with breast cancer and both are related to estrogen exposure and that rs2046210 is in close proximity to the ESR1 gene, we evaluated the association of SNP rs2046210 with endometrial cancer risk among 953 cases and 947 controls in a population-based, case-control study conducted in Shanghai, China. Logistic regression models were used to derive odds ratios (ORs) and 95% confidence intervals (95% CIs) after adjusting for potential confounders. We found that the A allele of rs2046210, linked to an increased risk of breast cancer, was associated with increased but not statistically significant risk of endometrial cancer (OR = 1.16, 95% CI = 0.96-1.41 for the GA and AA genotypes compared with the GG genotype); the association was stronger among post-menopausal women (OR = 1.28, 95% CI = 1.00-1.65). The association tended to be stronger among women with higher or longer estrogen exposure than among women with relatively lower or shorter exposure to estrogen. Our study suggests that rs2046210 may play a role in the etiology of endometrial cancer. Additional studies are needed to confirm our findings. 展开更多
