目的:建立一种简便、快速和灵敏的皮肤组织原位活力检测方法。方法:采用SYTO 13和溴化乙啶(EB)两种核酸染色剂对皮肤组织进行双重染色,荧光显微镜下通过颜色不同判断细胞的活力状态并计算出活力百分比。本研究利用SYTO/EB法和一种快速... 目的:建立一种简便、快速和灵敏的皮肤组织原位活力检测方法。方法:采用SYTO 13和溴化乙啶(EB)两种核酸染色剂对皮肤组织进行双重染色,荧光显微镜下通过颜色不同判断细胞的活力状态并计算出活力百分比。本研究利用SYTO/EB法和一种快速、灵敏的间接活力测定方法WST-1法同时对4℃保存不同时间的皮肤进行活力的检测,并将结果进行比较。结果:两种方法测定的结果均显示皮肤在一定的保存液中4℃保存后两周时活力为新鲜皮肤的30%左右,两个结果在各时相点均未有明显差异。结论:SYTO/EB测定皮肤的活力快速、灵敏且简便,并可原位观察皮肤内各种细胞的活力,不失为一种可取的皮肤活力检测方法。 展开更多

目的:了解在无保护剂条件下细胞连接在组织冷冻损伤中的作用。方法:采用细胞单层中可形成细胞连接的MDCK细胞和不形成细胞连接的V79细胞,分为细胞悬液组(Suspension)、贴壁组(Single Tough)和单层细胞组(Monolayer)分别进行0-40℃之间... 目的:了解在无保护剂条件下细胞连接在组织冷冻损伤中的作用。方法:采用细胞单层中可形成细胞连接的MDCK细胞和不形成细胞连接的V79细胞,分为细胞悬液组(Suspension)、贴壁组(Single Tough)和单层细胞组(Monolayer)分别进行0-40℃之间的梯度冷冻实验,然后采用WST-1和MTT两种方法测定细胞的活力并进行比较。结果:两种方法测定的结果一致,均显示两种细胞的细胞悬液冷冻后活力高于贴壁组和单层细胞组;MDCK细胞贴壁组活力高于单层细胞组,V79细胞则无明显差异;MDCK单层细胞组活力却明显低于V79单层细胞组。结论:组织细胞间的连接以及细胞与细胞基质的连接对组织冷冻的损伤有重要的作用,它们的存在直接影响组织细胞的活力,其原因可能与构成细胞连接的细胞骨架系统有关。 展开更多

Precipitation strengthening provides one of the most widely-used mechanisms for strengthen-ing multi-principal-element alloys(MPEAs).Here,we report dual-morphology B2 precipitates in Co_(36)Cr_(15)Fe_(18)Ni_(18)Al_(8)Ti_(4)Mo_(1)MPEA obtained by thermo-mechanical processing.Electron microscopy charac-terization reveals that the dual-morphology B2 precipitates are either recrystallized B2 particles formed at the grain boundaries or triple junctions with recrystallization process,or rod-like within the non-recrystallized FCC matrix.The dual-morphology B2 precipitates enhance the yield strength and ultimate tensile strength up to 1120 MPa and 1480 MPa,respectively.This work suggests the mechanical proper-ties of the alloy can be optimized by B2 precipitation strengthening to meet the needs of engineering applications. 展开更多

Tungsten powder was fabricated from the system CaWO4-Mg by self-propagating high-temperature synthesis (SHS) with a magnesium thermit stage. The physic-chemical change during heating and the effects of pressure of sample and diluents (W powder) on product have been studied. The experimental results show that the porosity of combustion product and the particle size of final tungsten powder decrease with increasing pressure of sample. Addition of diluents could increase the particle size of final tungsten powder. The purity of tungsten is improved by leaching in NaOH solution. The results of spectral analysis and particle size distribution of final tungsten powder show that the final Tungsten powder has a median diameter of 0.87μm,specific surface area of 1.09m2/g and purity of above 99.0%. 展开更多

We report a detailed study of polarization characteristics and subpulse drifting in PSR J1110-5637 with the observations of the Parkes 64 m radio telescope at 1369 MHz.The observations revealed that the trailing component of the pulse profile has obvious subpulse drifting,while the leading component has no subpulse drifting.Using the two-dimensional fluctuation spectrum(2DFS),we detected three distinct emission modes in the trailing component(modes A,B and C).The emission in mode A is chaotic and indistinguishable,while modes B and C have obvious subpulse drifting.The vertical modulation periods P3of modes B and C are around the mean values of 12 P and 8 P,respectively.The subpulse drifting of PSR J1110-5637 will expand the pulsar sample with multiple subpulse drifting rates,and this will help future systematic studies on the physical origin of the subpulse drifting phenomenon. 展开更多
