MM: To develop a protocol for direct hepatic lineage differentiation from early developmental progenitors to a population of mature hepatocytes, METHODS: Hepatic progenitor cells and then mature hepatocytes from mou...MM: To develop a protocol for direct hepatic lineage differentiation from early developmental progenitors to a population of mature hepatocytes, METHODS: Hepatic progenitor cells and then mature hepatocytes from mouse embryonic stem (ES) cells were obtained in a sequential manner, induced by valproic acid (VPA) and cytokines (hepatocyte growth factor, epidermal growth factor and insulin), Morphological changes of the differentiated cells were examined by phase-contrast microscopy and electron microscopy, Reverse transcription polymerase chain reaction and immunocytochemical analyses were used to evaluate the gene expression profiles of the VPA-induced hepatic progenitors and the hepatic progenitor-derived hepa- tocytes, Glycogen storage, cytochrome P450 activity, transplantation assay, differentiation of bile duct-like structures and tumorigenic analyses were performed for the functional identification of the differentiated cells, Furthermore, FACS and electron microscopy were used for the analyses of cell cycle profile and apoptosis in VPA-induced hepatic differentiated cells.RESULTS: Based on the combination of VPA and cytokines, mouse ES cells differentiated into a uniform and homogeneous cell population of hepatic progenitor cells and then matured into functional hepatocytes. The progenitor population shared several characteristics with ES cells and hepatic stem/progenitor cells, and represented a novel progenitor cell between ES and hepatic oval cells in embryonic development. The dif- ferentiated hepatocytes from progenitor cells shared typical characteristics with mature hepatocytes, including the patterns of gene expression, immunological markers,in vitro hepatocyte functions and in vivo capacity to restore acute-damaged liver function. In addition, the differentiation of hepatic progenitor cells from ES cells was accompanied by significant cell cycle arrest and selective survival of differentiating cells to-wards hepatic lineages. CONCLUSION: Hepatic cells of different developmental stages from early progenitors to matured hepatocytes can be acquired in the appropriate order based on sequential induction with VPA and cytokines.展开更多
BACKGROUND Depression is the most common mental illness in postpartum mothers,and the etiology of postpartum depression remains poorly understood.Over the past several decades,studies have reported that postpartum dep...BACKGROUND Depression is the most common mental illness in postpartum mothers,and the etiology of postpartum depression remains poorly understood.Over the past several decades,studies have reported that postpartum depression is caused by multiple factors,such as genetic,psychological,pregnancy,and environmental factors,with the family environment being an important environmental factor.The theory of family cohesion and adaptability put forward by Olson is a classic model that describes the level of family function.However,to date,this model has not been examined regarding its applicability to patients with postpartum depression.AIM To investigate the relationship between family cohesion and adaptability and the risk of postpartum depressive symptoms.METHODS We retrospectively analyzed 1446 patients admitted to the postpartum healthcare clinic of the Affiliated Foshan Maternity and Child Healthcare Hospital from April 2021 to December 2021.Patients were grouped according to whether postpartum depression symptoms were reported(symptoms,n=454;no symptoms,n=992).All patients completed the Edinburgh Postpartum Depression Scale and the Chinese version of the Family Cohesion and Adaptability Assessment Scale II.Baseline and clinical data were compared between groups.Univariate regression analysis was used to investigate the association between different types of family cohesion and postpartum depressive symptoms and the association between different family adaptability types and postpartum depressive symptoms.RESULTS After adjusting for age,education,occupation,gravidity,parity,and mode of delivery,disengaged[adjusted odds ratio(AOR)=3.36,95%CI:1.91-5.91],and separated(AOR=1.97,95%CI:1.34-2.90)family cohesion types showed a higher risk of postpartum depression than the connection type,whereas the enmeshed type(AOR=0.38,95%CI:0.28-0.51)protected against postpartum depressive symptoms.Rigid(AOR=4.41,95%CI:3.02-6.43)and structured families(AOR=1.88,95%CI:1.34-2.63)had a higher risk of postpartum depressive symptoms than flexible families,whereas chaotic families(AOR=0.35,95%CI:0.24-0.51)protected against postpartum depressive symptoms.CONCLUSION Family cohesion and adaptability are influencing factors for postpartum depressive symptoms,with higher family cohesion and adaptability being associated with a lower risk of postpartum depressive symptoms.展开更多
BACKGROUND Guillain-Barrésyndrome(GBS)is an autoimmune-mediated peripheral neuropathy characterized by symmetric weakness.Asymmetric weakness in GBS is uncommon and may be easily confused with other differential ...BACKGROUND Guillain-Barrésyndrome(GBS)is an autoimmune-mediated peripheral neuropathy characterized by symmetric weakness.Asymmetric weakness in GBS is uncommon and may be easily confused with other differential diagnoses.We herein present three cases of asymmetric GBS and review the literature on this atypical subtype of GBS in order to describe the characteristics of asymmetric GBS and to provide experience for clinicians.CASE SUMMARY Different from patients in the previous reports,our patients showed persistent asymmetric limb weakness from the onset to recovery phase.All three patients were serologically positive for antecedent infections.Two of the three cases had IgG antibodies against ganglioside GM1.Two patients received immunotherapy including intravenous immunoglobulin and plasma exchange,while one patient received only supportive treatment.Autoantibodies against gangliosides,asymmetry of congenital development of blood-nerve barrier and limb use may contribute to the development of asymmetric limb weakness in GBS.CONCLUSION Asymmetric GBS may be a rare clinical variant and should be considered when a patient develops acute and progressive asymmetric limb weakness.The differences in clinical features and prognosis between asymmetric GBS and classic GBS deserve further investigation in a large study.展开更多
A design of ultrathin crystalline silicon solar cell with Si_(3)N_(4)circular truncated cone holes(CTCs)arrays on the top is proposed.In this article,we perform an optical simulation of the structure.The finite-differ...A design of ultrathin crystalline silicon solar cell with Si_(3)N_(4)circular truncated cone holes(CTCs)arrays on the top is proposed.In this article,we perform an optical simulation of the structure.The finite-difference time-domain method is used to calculate the optical absorption of different periods,radius of top and bottom circles and depth of Si_(3)N_(4)CTCs.The short-circuit current density generated by the optimized cells(30.17 mA/cm^2)is 32.44%more than the value gained by control group(with flat Si_(3)N_(4)).Then adding a layer of back silver to allow us to better analyze optical absorption.Later,we simulate the optimization of the same configuration of different silicon thicknesses andfind that our structure does enhance the light absorption.This work uses a combined path towards achieving higher photocurrent ultrathin crystalline silicon solar cells by constructing the texture of anti-reflection coating.展开更多
To realize the application of the floating offshore wind turbine(FOWT)from deep to relatively shallow waters,a new concept of multi-column floating wind turbine platform with low center of gravity(CG)is designed and v...To realize the application of the floating offshore wind turbine(FOWT)from deep to relatively shallow waters,a new concept of multi-column floating wind turbine platform with low center of gravity(CG)is designed and validated.The multi-column low CG platform is designed to support a 6MW wind turbine class and operated at a water depth of 50m in the South China Sea.The frequency domain software WADAM and time domain software NREL-FAST are used to simulate coupled dynamic responses of the floating wind turbine system with second-order wave loads considering.The dynamic behaviors of multi-column low CG FOWT system under normal operation and parked conditions are presented.The influence of second-order wave force on the motion responses of the multi-column platform,fore-aft force and moment of the tower base and mooring force are researched respectively.The results demonstrate that the coupled dynamic responses at rated operating condition and extreme condition meet the normal operating requirements and extreme survival requirements of FOWT system in the shallow water(50m)of South China Sea.In addition,it is found that,the wave frequency response gradually replaces the second-order low frequency response as the main influencing factor of the coupled dynamic response of the FOWT system with the increasing severity of the sea states.However,in general,the magnitude of second-order low frequency response increases with the increasing severity of the design load case.Thus,in the subsequent design of the shallow water FOWT system,the second-order effects should be paid enough attention.展开更多
