文中基于同轴电场驱动喷印技术制备了聚合物有机半导体微结构,并将其用于可弯曲有机场效应晶体管(OFET)器件制备。通过实验探究了外液黏度、喷印速度、工作电压和喷印高度4个工艺参数对聚-3已基噻吩(P3HT)结构线宽的影响,实现了P3HT亚... 文中基于同轴电场驱动喷印技术制备了聚合物有机半导体微结构,并将其用于可弯曲有机场效应晶体管(OFET)器件制备。通过实验探究了外液黏度、喷印速度、工作电压和喷印高度4个工艺参数对聚-3已基噻吩(P3HT)结构线宽的影响,实现了P3HT亚微米阵列线的可靠制备,平均直径150 nm。通过调整喷印工艺参数,完成了聚合物有机半导体材料P3HT/IDT-BT/PDVT-10的亚微米阵列线制备。基于引达省二噻吩-苯并噻二唑(IDT-BT)亚微米阵线列制备了可弯曲OFET,器件载流子迁移率可达0.98 cm^(2)/(V·s),开关比可达1.21×10^(5),阈值电压为-12 V,在弯曲半径5 mm下,仍具有良好的电学性能,展示了同轴电场驱动喷印技术在柔性电子器件制造中的巨大应用潜力。 展开更多

N6-methyladenosine(m^(6)A)modification of mRNA is a critical post-transcriptional regulatory mechanism that modulates mRNA metabolism and neuronal function.The m^(6)A reader YTHDF1 has been shown to enhance the translational efficiency of m^(6)A-modified mRNAs in the brain and is essential for learning and memory.However,its role in the mature retina remains unclear.Herein,we report a novel role of Ythdf1 in the maintenance of retinal function using a genetic knockout model.Loss of Ythdf1 resulted in impaired scotopic electroretinogram(ERG)responses and progressive retinal degeneration.Detailed analyses of rod photoreceptors confirmed substantial degenerative changes in the absence of ciliary defects.Single-cell RNA sequencing revealed comprehensive molecular alterations across all retinal cell types in Ythdf1-deficient retinas.Integrative analysis of methylated RNA immunoprecipitation(MeRIP)sequencing and RIP sequencing identified Tulp1 and Dhx38,two inheritable retinal degeneration disease-associated gene homologs,as direct targets of YTHDF1 in the retina.Specifically,YTHDF1 recognized and bound m^(6)A-modified Tulp1 and Dhx38 mRNA at the coding sequence(CDS),enhancing their translational efficiency without altering mRNA levels.Collectively,these findings highlight the essential role of YTHDF1 in preserving visual function and reveal a novel regulatory mechanism of m^(6)A reader proteins in retinal degeneration,identifying potential therapeutic targets for severe retinopathies. 展开更多

为探讨不同温度下干旱与盐胁迫对3种大黄种子萌发和幼苗生长的影响,以聚乙二醇6000和NaCl分别模拟不同强度的干旱和盐胁迫,纯水处理为对照组(CK),分别置于3种温度(10、20、30℃)的人工气候箱内培养。结果表明:20℃各处理下3种大黄的最... 为探讨不同温度下干旱与盐胁迫对3种大黄种子萌发和幼苗生长的影响,以聚乙二醇6000和NaCl分别模拟不同强度的干旱和盐胁迫,纯水处理为对照组(CK),分别置于3种温度(10、20、30℃)的人工气候箱内培养。结果表明:20℃各处理下3种大黄的最终发芽率和发芽势均较高,30℃与干旱或盐的交互胁迫会抑制大黄种子萌发且不成苗;10℃时,随盐胁迫的加剧掌叶大黄地上和地下部分的脱落酸(ABA)含量呈升高趋势;20℃时,随干旱、盐胁迫的加重掌叶大黄地上和地下部分赤霉素(GA)、生长素(IAA)含量均呈降低趋势,且随干旱加剧3种大黄地上/地下非结构性碳水化合物(NSC)呈先降低后升高的趋势;10℃和20℃下,随盐胁迫加剧药用大黄地上GA/地下GA呈先升后降的趋势,地上IAA/地下IAA为先降后升的趋势,而随干旱加剧掌叶大黄和唐古特大黄的地上和地下部分NSC含量呈先增后减的趋势;不同处理下药用大黄的萌发指标低于其他2种大黄,表现为更易受到温度和干旱胁迫的影响;20℃为3种大黄萌发适宜温度,高温会加剧渗透胁迫对大黄种子萌发和幼苗生长的危害而适宜的温度能对渗透胁迫起到一定的缓解作用;大黄通过调节不同部位的可溶性糖和淀粉含量及其比例以保持细胞渗透平衡,通过调整激素含量与分配模式维持生理与代谢活动以缓解逆境危害。 展开更多

Obesity caused by poor eating habits has become a global public health issue and harms human health.Rosmarinic acid(RA)is a natural active substance with many biological functions like antioxidant.However,the function of RA's lipid-lowering effect and mechanism are not entirely clear.This study aimed to explore the effect of RA on obesity and its action mechanism in Caenorhabditis elegans.RA not only alleviated intestinal inflammation and change the composition of unsaturated fatty acid,but also decreased fat storage and adjusted the size and quantity of fat droplets without threatening development and reproduction or affecting energy intake.In addition,RA improved the integrity of intestinal barrier function in C.elegans.RNA sequencing and quantitative reverse transcription-polymerase chain reaction(qRT-PCR)comprehensively revealed that the lipid-lowering mechanism mediated by RA might involve lipid metabolism and anti-inflammatory pathways.Among them,RA regulates lipid metabolism by reducing fat synthesis through SBP-1 and increasing fatty acidβ-oxidation via NHR-49.Furthermore,the anti-inflammatory pathway was associated with RA's enhancement of the ratio of unsaturated fatty acid and alleviation of inflammatory reactions caused by Pseudomonas aeruginosa(PAO1).Therefore,RA regulated lipid metabolism and relieved inflammation,which had the potential for dietary supplements or functional foods. 展开更多

AIM:To evaluate the short-term efficacy and safety of ultrasound cycloplasty(UCP)procedure in Chinese patients with angle-closure glaucoma(ACG),and the mechanisms of its intraocular pressure(IOP)lowering effects.METHODS:Fifty-six patients(56 eyes)diagnosed with primary and/or secondary ACG were enrolled in this prospective study.Visual acuity,IOP,slit-lamp examinations,structural parameters of anterior segments including anterior chamber depth(ACD),pupillary diameter(PD),anterior chamber area(ACAr),anterior chamber angle(ACAn),and side effects were evaluated.Seven rhesus macaques(Macaca Mulatta)were used for the analyses of IOP lowering effects,including inflammatory reactions,pathohistological evaluation,scanning electron microscopy(SEM),and aqueous outflow pathway via 1%basic fuchsin perfusion.RESULTS:Fifty six eyes of 24 male and 32 female patients with an average age of 58.93±15.97 years old were recruited in primary and secondary ACG affected 31 and 25 eyes,respectively.Clinically,the mean IOP was 17.89±7.93 mm Hg(n=53,53.63%reduction,P<0.001)at 1wk and 22.47±12.00 mm Hg at 12mo(n=16,18.67%reduction,P<0.01)after UCP,compared to the baseline of 39.08±14.75 mm Hg(n=56).Postoperative tolerance was favorable in 94.64%of cases,with visual acuity unchanged or improved in 96.43%of patients.Mild and transient inflammatory reactions were observed post UCP.UBM analysis revealed statistically significant changes in anterior chamber parameters:increased ACD(n=22,P<0.05),enlarged PD(n=22,P<0.05),expanded ACAr(n=16,P<0.01),and widened ACAn(n=19,P<0.05)compared to preoperative measurements.As in normal monkey eyes,IOP was also reduced upon UCP.Mild inflammatory reactions were seen 1-7d post treatment.Hematoxylin and Eosin(H&E)staining showed enlarged spaces among ciliary muscle bundles.Ciliary process stromal edema was evident,but significant loss of two layers of ciliary epitheliums was not seen.SEM confirmed ciliary muscle fibers concentrated.Fuchsin anterior chamber perfusion showed the whole ciliary body staining in UCP group,but only surface staining in controls.CONCLUSION:UCP is efficient and safe to lower the IOP of patients with ACG.Changed anterior chamber structure and increased aqueous humour outflow via uveoscleral pathway may significantly attribute to IOP lowering effects of UCP. 展开更多

目前我国蜂蜜市场掺假现象严重,研究一种快速、准确的方法用于市场流通领域掺假蜂蜜的鉴别具有重要的现实意义。采用近红外光谱(NIR)结合化学计量学方法对常见的天然蜂蜜以及掺假(掺杂常见糖浆)蜂蜜进行建模识别,并比较偏最小二乘-判别... 目前我国蜂蜜市场掺假现象严重,研究一种快速、准确的方法用于市场流通领域掺假蜂蜜的鉴别具有重要的现实意义。采用近红外光谱(NIR)结合化学计量学方法对常见的天然蜂蜜以及掺假(掺杂常见糖浆)蜂蜜进行建模识别,并比较偏最小二乘-判别分析(PLS-DA)及支持向量机(SVM)对糖浆掺假蜂蜜鉴别模型的影响。首先,采集来自中国10个省份、20种常见蜂蜜的112个天然纯蜂蜜样品,以及6种常见糖浆样品按不同糖浆含量(10%,20%,30%,40%,50%,60%)配制的112个掺假蜂蜜样品,共计224个样品;通过近红外光仪器扫描获得所有样品的近红外光谱数据(波长范围400~2 500nm);然后,分别采用一阶导数(FD)、二阶导数(SD)、多元散射校正(MSC)、标准正态变化(SNVT)四种方式对原始光谱进行预处理;再结合PLS-DA和SVM建立天然蜂蜜和糖浆掺假蜂蜜的鉴别模型,比较不同预处理方法对两种不同建模算法建立的蜂蜜掺假鉴别模型效果。其中SVM算法的惩罚参数c和核函数参数g通过网格搜索法(GS)、遗传算法(GA)、粒子群算法(PSO)三种寻优算法进行优化。分析结果表明:光谱数据进行预处理后所建立的模型准确率均有明显提升,而对于SVM模型,惩罚参数c和核函数参数g对模型准确率的提升效果要比光谱预处理带来的提升效果更明显。在PLS-DA算法中,经FD光谱预处理后建立的模型效果最好,最佳PLS-DA模型准确率为87.50%;在SVM算法中,经MSC预处理后,再通过GS寻优,获得惩罚参数c为3.0314,核函数参数g为0.3298的条件下所建立的模型效果最好,最佳SVM模型准确率为94.64%。由此可见,非线性的SVM算法结合NIR光谱数据所建立的天然蜂蜜与糖浆掺假蜂蜜鉴别模型要优于线性的PLS-DA模型,同时表明NIR光谱结合化学计量学方法对常见糖浆掺杂的中国蜂蜜鉴别是可行的。 展开更多

选取可可托海3号脉石英-锂辉石带、卡鲁安806号脉、库卡拉盖650脉,开展石英、锂辉石中流体包裹体显微测温以及石英中捕获的单个流体包裹体组成的LA-ICP-MS原位分析。结果表明,锂矿化伟晶岩晚期出溶流体相为H_(2)O-NaCl-CO_(2)体系,石英... 选取可可托海3号脉石英-锂辉石带、卡鲁安806号脉、库卡拉盖650脉,开展石英、锂辉石中流体包裹体显微测温以及石英中捕获的单个流体包裹体组成的LA-ICP-MS原位分析。结果表明,锂矿化伟晶岩晚期出溶流体相为H_(2)O-NaCl-CO_(2)体系,石英中流体包裹体均一温度为233~407℃,盐度为5.77%~20.15%;w(Li)为1527×10^(-6)~17767×10^(-6)、w(Cs)为32×10^(-6)~4563×10^(-6)、w(Rb)为10×10^(-6)~100×10^(-6)。Li与Cs、Rb与Cs之间存在正相关系,并且Li/Rb与Li、Cs/Rb与Cs显示正相关关系,表明Li、Rb、Cs强烈分配进入流体相。流体包裹体中K/Na<0.4,指示锂矿化伟晶岩晚期分异出的流体显示富钠性质。26个流体包裹体LA-ICP-MS分析结果显示锂矿化伟晶岩晚期流体相中含有0.33%~3.82%Li_(2)O(w(Li_(2)O)平均值为1.10%),考虑到Li、Cs、Rb在流体/熔体间的分配系数>1,由此提出伟晶岩型锂床具有岩浆热液成因的可能性。 展开更多

In this study,28,691 genome sequences and16,566 expressed sequence tags(ESTs) of Eucalyptus were derived from the Gen Bank database.A total of 2292 SSR loci were sought out from 1785 effective sequences.Through analyses of SSR loci information,the SSR motif length was negatively correlated with the abundance of the SSRs.In the EST sequences of Eucalyptus,triplet repeat motifs were the most abundant,and dinucleotide repeats motifs had the highest frequencies.Subsequently,395 pairs of primers were designed based on the SSR loci.Using optimized SSR-PCR conditions,340 pairs of primers were successfully screened,with a success rate of 86.1%.By construction of a maximum likelihood phylogenetic tree of six eucalypt species,represented by five species of the genus Eucalyptus and one of the genus Corymbia,the genetic relationships of Eucalyptus urophylla and E.camaldulensis suggested by this tree was found to differ from that suggested by traditional morphological taxonomy.The results provide insights for evaluating geneticdiversity of Eucalyptus and analysis of Eucalyptus phylogenetics using SSR markers. 展开更多

The shuttle effect of soluble lithium polysulfides(LiPSs)between electrodes and slow reaction kinetics lead to extreme inefficiency and poor high current cycling stability,which limits the commercial application of Li-S batteries.Herein,the multi-dimensional composite frame has been proposed as the modified separator(MCCoS/PP)of Li-S battery,which is composed of CoS_(2) nanoparticles on alkali-treated MXene nanosheets and carbon nanotubes.Both experiments and theoretical calculations show that bifunctional catalytic activity can be achieved on the MCCoS/PP separator.It can not only promote the liquid-solid conversion in the reduction process,but also accelerate the decomposition of insoluble Li_(2)S in the oxidation process.In addition,LiPSs shuttle effect has been inhibited without a decrease in lithium-ion transference numbers.Simultaneously,the MCCoS/PP separator with good LiPSs adsorption capability arouses redistribution and fixing of active substances,which is also beneficial to the rate performance and cycling stability.The Li-S batteries with the MCCoS/PP separator have a specific capacity of 368.6 mAh g^(−1) at 20C,and the capacity decay per cycle is only 0.033%in 1000 cycles at 7C.Also,high area capacity(6.34 mAh cm^(−2))with a high sulfur loading(7.7 mg cm^(−2))and a low electrolyte/sulfur ratio(7.5μL mg^(−1))is achieved. 展开更多

Micro-supercapacitors(MSCs)are attractive electrochemical energy storage devices owing to their high power density and extended cycling stability.However,relatively low areal energy density still hinders their practical applications.Here,an asymmetric Mg ion MSC with promising high energy density is fabricated.Firstly,indium tin oxide(ITO)NWs were synthesized by chemical vapor deposition as the excellent current collector.Subsequently,nanostructured Mn_(3)O_(4)and Ppy@FeOOH were deposited on the laser-engraved interdigital structure ITO NWs electrodes as the positive and negative electrodes,respectively.Beneficial from the hierarchical micro-nano structures of active materials,high conductive electron transport pathways,and charge-balanced asymmetric electrodes,the obtained MSC possesses a high potential window of 2.2 V and a high areal capacitance of 107.3 mF cm^(-2)at 0.2 mA cm^(-2).The insitu XRD,VSM,and ex-situ XPS results reveal that the primary energy storage mechanism of Mg ions in negative FeOOH electrode is Mg ions de-/intercalation and phase transition reaction of FeOOH.Furthermore,the MSC exhibits a high specific energy density of 71.18μWh cm^(-2)at a power density of 0.22 mWh cm^(-2)and capacitance retention of 85%after 5000 cycles with unvaried Coulombic efficiency.These results suggest promising applications of our MSC in miniaturized energy storage devices. 展开更多

AIM:To evaluate the potential of two trabecular meshwork(TM)-specific promoters,Chitinase 3-like 1(Ch3L1)and matrix gla protein(MGP),for improving specificity and safety in glaucoma gene therapy based on self-complementary AAV2(scAAV2)vector technologies.METHODS:An scAAV2 vector with C3 transferase(C3)as the reporter gene(scAAV2-C3)was selected.The scAAV2-C3 vectors were driven by Ch3L1(scAAV2-Ch3L1-C3),MGP(scAAV2-MGP-C3),enhanced MGP(scAAV2-eMGP-C3)and cytomegalovirus(scAAV2-CMV-C3),respectively.The cultured primary human TM cells were treated with each vector at different multiplicities of infections.Changes in cell morphology were observed by phase contrast microscopy.Actin stress fibers and Rho GTPases/Rho-associated protein kinase pathway-related molecules were assessed by immunofluorescence staining,real-time quantitative polymerase chain reaction and Western blot.Each vector was injected intracamerally into the one eye of each rat at low and high doses respectively.In vivo green fluorescence was visualized by a Micron III Retinal Imaging Microscope.Intraocular pressure(IOP)was monitored using a rebound tonometer.Ocular responses were evaluated by slit-lamp microscopy.Ocular histopathology analysis was examined by hematoxylin and eosin staining.RESULTS:In TM cell culture studies,the vectormediated C3 expression induced morphologic changes,disruption of actin cytoskeleton and reduction of fibronectin expression in TM cells by inhibiting the Rho GTPases/Rhoassociated protein kinase signaling pathway.At the same dose,these changes were significant in TM cells treated with scAAV2-CMV-C3 or scAAV2-Ch3L1-C3,but not in cells treated with scAAV2-eMGP-C3 or scAAV2-MGP-C3.At lowinjected dose,the IOP was significantly decreased in the scAAV2-Ch3L1-C3-injected eyes but not in scAAV2-MGPC3-injected and scAAV2-eMGP-C3-injected eyes.At highinjected dose,significant IOP reduction was observed in the scAAV2-eMGP-C3-injected eyes but not in scAAV2-MGP-C3-injected eyes.Similar to scAAV2-CMV-C3,scAAV2-Ch3L1-C3 vector showed efficient transduction both in the TM and corneal endothelium.In anterior segment tissues of scAAV2-eMGP-C3-injected eyes,no obvious morphological changes were found except for the TM.Inflammation was absent.CONCLUSION:In scAAV2-transduced TM cells,the promoter-driven efficiency of Ch3L1 is close to that of cytomegalovirus,but obviously higher than that of MGP.In the anterior chamber of rat eye,the transgene expression pattern of scAAV2 vector is presumably affected by MGP promoter,but not by Ch3L1 promoter.These findings would provide a useful reference for improvement of specificity and safety in glaucoma gene therapy using scAAV2 vector. 展开更多
