龟龄集通过调节血清葡萄糖-6-磷酸水平改善小鼠少弱精子症的机制研究

目的探索龟龄集(GLJ)改善少弱精子症的相关机制。方法将72只8周龄C57BL/6J雄性小鼠随机分为对照组(CTL组)、奥硝唑模型组(Orn组)、不同浓度GLJ组(Orn+GLJ组)和不同浓度葡萄糖-6-磷酸(G6P)组(Orn+G6P组),检测各组小鼠附睾尾精子浓度和精... 目的探索龟龄集(GLJ)改善少弱精子症的相关机制。方法将72只8周龄C57BL/6J雄性小鼠随机分为对照组(CTL组)、奥硝唑模型组(Orn组)、不同浓度GLJ组(Orn+GLJ组)和不同浓度葡萄糖-6-磷酸(G6P)组(Orn+G6P组),检测各组小鼠附睾尾精子浓度和精子活力,ELISA法检测血清卵泡刺激素(FSH)、黄体生成素(LH)、抑制素B(INHB)、睾酮(T)水平,苏木精-伊红(HE)染色观察睾丸和附睾组织结构,免疫荧光分析各级生殖细胞的数量。另将SD大鼠分为空白组(Base组)、GLJ组(灌胃)、GLJ+3-溴丙酮酸(3-BP)组(GLJ+3-BP组,腹腔注射),检测各组大鼠血清中G6P浓度并取各组血清加入培养基,进行小鼠精原细胞系(GC1)、精母细胞系(GC2)和支持细胞系(TM4)培养,观察血清中G6P浓度对睾丸各细胞系的影响。结果与Orn组小鼠比较,Orn+GLJ组和Orn+G6P组小鼠的血清G6P含量均显著增加(P<0.05),血清中FSH、LH、INHB水平均得到显著改善(P<0.05),精子浓度和精子活力均显著升高(P<0.05),睾丸内的生殖细胞数量均显著增加(P<0.05)。细胞培养结果显示,与Base组相比,GLJ组的GC1、GC2、TM4的细胞活力显著上升(P<0.05),且TM4的ATP水平显著增加(P<0.05);与GLJ组比较,GLJ+3-BP组中改善效果被显著抑制(P<0.01)。结论GLJ可能通过升高血清中G6P水平,增强生精微环境中的细胞能量代谢活性,促进各级生殖细胞及支持细胞的活力,最终改善OA模型小鼠的精子发生功能。 展开更多

目的NOD样受体家族蛋白3(NLRP3)在子宫内膜中的生理性功能研究较少。文中旨在研究NLRP3在子宫内膜间质细胞蜕膜化过程中的表达和调控作用。方法选自2020年4月至6月在东部战区总医院生殖中心20例不孕症患者的子宫内膜样本。收集人增殖期(... 目的NOD样受体家族蛋白3(NLRP3)在子宫内膜中的生理性功能研究较少。文中旨在研究NLRP3在子宫内膜间质细胞蜕膜化过程中的表达和调控作用。方法选自2020年4月至6月在东部战区总医院生殖中心20例不孕症患者的子宫内膜样本。收集人增殖期(n=10)及蜕膜期(n=10)的子宫内膜。另选用SPF级30只6~8周龄雌鼠和10只雄鼠,获取小鼠交配后见栓天数(dpc)0.5~7.5天的子宫内膜,采用Western blot、免疫组化检测NLRP3的表达及定位。体外利用对羟基苯甲酸甲酯(MPA)和8-溴-环腺苷酸(8-Br-cAMP)联合诱导人间质细胞系T-HESCs蜕膜化,检测蜕膜化过程中NLRP3的表达情况。干扰NLRP3的表达后,通过实时荧光定量PCR及ELISA分别检测细胞及其培养上清中蜕膜化标志物催乳素(PRL)的表达情况。结果免疫组化结果显示,在人蜕膜期子宫内膜中,可见大量NLRP3阳性细胞,蜕膜期NLRP3的表达量较增殖期显著上升。Western blot结果所示,人蜕膜期子宫内膜较增殖期NLRP3显著高表达。免疫组化结果所示,NLRP3在小鼠妊娠早期上调,dpc 3.5天表达最高,可见大量NLRP3阳性细胞表达,细胞质棕黄色深染,之后下降。Western blot结果所示,在小鼠妊娠早期,随着妊娠时间增加,NLRP3表达量先上升后下降,dpc 3.5天达到峰值,与dpc 0.5天和dpc 7.5天比较,差异具有统计学意义(P<0.01)。Western blot结果所示,随着诱导人间质细胞蜕膜化时间延长,NLRP3及caspase-1表达量显著升高。与12 h NLRP3、caspase-1蛋白表达量(1.76±0.04、1.28±0.02)比较,0 h(1.00±0.01、0.98±0.01)明显降低,24 h(2.26±0.03、1.95±0.05)明显升高(P<0.05)。结论NLRP3可促进间质细胞蜕膜化过程中PRL的表达和分泌,为研究NLRP3在子宫内膜围种植期调控间质细胞蜕膜化奠定基础。 展开更多

In this paper, the application of Chinese calligraphy fonts to the modem design art is discussed from computer font design and the components of Chinese characters, development trend, and modem decoration design. Wound management is a crucial measure for skin wound healing and is significantly important to maintaining the integrity of skins and their functions.Electrical stimulation at the wound site is a compelling strategy for skin wound repair.However,there has been an urgent need for wearable and point-of-care electrical stimulation devices that have self-adhesive and mechanical properties comparable to wound tissue.Herein,we develop a bioinspired hybrid patch with self-adhesive and piezoelectric nanogenerator(HPSP)for promoting skin wound healing,which is composed of a mussel-inspired hydrogel matrix and a piezoelectric nanogenerator based on aligned electrospun poly(vinylidene fluoride)nanofibers.The device with optimized modulus and permeability for skin wear can self-adhere to the wound site and locally produce a dynamic voltage caused by motion.We show that the HPSP not only promotes fibroblast proliferation and migration in vitro,but also effectively facilitates the collagen deposition,angiogenesis,and re-epithelialization in vivo with the increased expressions of crucial growth factors.The HPSP reduces the wound closure time of full-thickness skin defects by about 1/3,greatly accelerating the healing process.This patch can serve as wearable and real-time electrical stimulation devices,potentially useful in clinical applications of skin wound healing. 展开更多

Utilizing photothermal hydrogels as a wound dressing is a promising strategy to accelerate wound healing.Usually,a photothermal hydrogel has a strong light-absorbing capability,and hence its transparency can be largely sacrificed,which is unbeneficial for the visual monitoring of wound states.It remains challenging to balance the trade-off between the photothermal conversion and wound visualization for the photothermal hydrogel dressing.Herein,a composite photothermal hydrogel film with high transparency is presented for the visual monitor of the wound,which is constructed by incorporating CsxW03 nanorods into the networks of polyacrylamide hydrogels.The composite photothermal hydrogel film exhibits high light absorption in the near-infrared region and high transmittance in the visible light region.Under 980 nm laser irradiation,the composite hydrogel can be heated up to 45°C.In vivo animal experiment on mouse skin wound model shows that the composite hydrogel film can locally heat the skin wound to accelerate healing while maintaining more than 70%transparency to realize real-time observation of the wound.This study provides the first attempt to solve the problem of opacity in photothermal hydrogel dressings,promoting the possibility of its clinical applications. 展开更多

目的 探索慢性前列腺炎(Chronic prostatitis,CP)发生与人前列腺组织免疫优势抗原的因果关联,挖掘潜在的CP实验室诊断标志物。方法 本课题组前期的一项研究发现了16种人前列腺组织免疫优势抗原。本研究利用大规模人群全基因组关联研究(G... 目的 探索慢性前列腺炎(Chronic prostatitis,CP)发生与人前列腺组织免疫优势抗原的因果关联,挖掘潜在的CP实验室诊断标志物。方法 本课题组前期的一项研究发现了16种人前列腺组织免疫优势抗原。本研究利用大规模人群全基因组关联研究(Genome-Wide Association Studies,GWAS)数据库,使用孟德尔随机化分析(Mendelian randomization analysis,MR)探索前期研究中热休克蛋白β1(Heat shock protein beta-1,HSP β1)与CP的因果关联。同时,采用MR-Egger回归法、加权中位数法(Weighted Median,WME)、最大似然估计(Maximum likelihood)以及加权模式法(Weighted mode)进行辅助因果推断,以提高结果的可靠性。结果 在open GWAS的数据库中,CP的发生与HSPβ1在外周血的表达量之间呈现显著的正相关性。同样的结果在GWAS Catalog数据库中得到了验证,进一步增加了结果可靠性。结论 本研究结果不仅从生物信息学角度验证了我们先前的研究结果,也为CP的临床诊疗提供了新的靶标和理论依据。 展开更多

目的探讨鸦胆子苦醇对雌性生殖细胞减数分裂启动及早期卵子发生的影响。方法6~8周小鼠雌雄合笼,以雌性小鼠见阴栓记为胚胎0.5d(E0.5)。收集E11.5、E12.5、E13.5、E14.5的雌性胎鼠生殖嵴,分别通过qRT-PCR及Western blotting检测核因子E2(... 目的探讨鸦胆子苦醇对雌性生殖细胞减数分裂启动及早期卵子发生的影响。方法6~8周小鼠雌雄合笼,以雌性小鼠见阴栓记为胚胎0.5d(E0.5)。收集E11.5、E12.5、E13.5、E14.5的雌性胎鼠生殖嵴,分别通过qRT-PCR及Western blotting检测核因子E2(NF-E2)相关因子2(Nrf2)mRNA及蛋白水平。分离E12.5雌性胎鼠的生殖嵴,分别用二甲基亚砜(对照组)、10nmol/L、20nmol/L的Nrf2抑制剂——鸦胆子苦醇、1μmol/L视黄酸或20nmol/L鸦胆子苦醇+1μmol/L视黄酸处理48h后,检测Nrf2通路、减数分裂启动、视黄酸信号通路相关基因的mRNA水平。药物处理48h后继续培养10d,对体外培养的生殖嵴行石蜡切片和HE染色进行原始卵泡计数。结果与对照组相比,10nmol/L、20nmol/L的鸦胆子苦醇显著下调减数分裂启动关键基因Stra8、Sycp3及Dazl的mRNA水平(P<0.05)。与对照组的单位面积卵泡数量(151.1±6.8)相比,10nmol/L、20nmol/L组的单位面积原始卵泡数量显著减少(109.5±12.2,49.1±8.0)(P<0.01)。与对照组相比,20nmol/L鸦胆子苦醇显著抑制视黄酸合成酶(P=0.001)及其受体(P=0.049)的表达。添加视黄酸能部分挽救鸦胆子苦醇对基因Raldh、RARa(P<0.01)及基因Stra8、Sycp3及Dazl的表达抑制作用(P<0.01)。结论鸦胆子苦醇通过抑制Nrf2通路调控减数分裂启动及早期卵子发生。 展开更多
