血清CD26在糖尿病合并结直肠腺瘤-腺癌序列中的表达及其意义 被引量:2

目的评价血清CD26在糖尿病合并结直肠肿瘤(癌及腺瘤)患者循环中的表达情况及其临床意义。方法收集2012年1月至2013年12月在首都医科大学附属北京世纪坛医院消化内科住院并行电子结肠镜检查的患者214例,其中结直肠腺瘤患者68例,28例合并... 目的评价血清CD26在糖尿病合并结直肠肿瘤(癌及腺瘤)患者循环中的表达情况及其临床意义。方法收集2012年1月至2013年12月在首都医科大学附属北京世纪坛医院消化内科住院并行电子结肠镜检查的患者214例,其中结直肠腺瘤患者68例,28例合并糖尿病;结直肠腺癌患者66例,26例合并糖尿病;内镜检查未见异常者80例,40例合并糖尿病,作为正常对照组。所有入组患者均通过ELISA法检测血清CD26浓度,电化学发光法检测血清CEA浓度,并进行统计学分析。结果从内镜阴性到腺瘤到腺癌,血清CD26逐渐升高,各组间比较,差异有统计学意义(P<0.05)。糖尿病合并结直肠腺癌患者血清CD26、CEA水平明显高于无糖尿病的结直肠腺癌患者,差异有统计学意义(P<0.05)。结论血清CD26水平从内镜阴性到腺瘤到腺癌逐渐升高,提示其可能成为诊断糖尿病合并结直肠癌的生物标志物。 展开更多

目的探讨长链非编码RNA癌症易感基因19(the cancer susceptibility 19,CASC19)在维持巨噬细胞M1型极化状态中的作用及其对结直肠癌细胞生长的影响。方法诱导单核细胞型淋巴瘤细胞Thp1分化为未极化的M0巨噬细胞,进一步诱导M0极化为经典... 目的探讨长链非编码RNA癌症易感基因19(the cancer susceptibility 19,CASC19)在维持巨噬细胞M1型极化状态中的作用及其对结直肠癌细胞生长的影响。方法诱导单核细胞型淋巴瘤细胞Thp1分化为未极化的M0巨噬细胞,进一步诱导M0极化为经典活化的M1巨噬细胞和替代性活化的M2巨噬细胞;采用RT-qPCR检测M1和M2相关分子标记和CASC19的表达,明确巨噬细胞分化状态及CASC19转染效率;使用敲降CASC19的M1型巨噬细胞上清(M1 SI-CM组)及其阴性对照上清(M1 NC-CM组)刺激结直肠癌细胞SW480,采用活细胞实时动态成像、MTS法检测细胞增殖能力,Transwell检测细胞迁移能力。结果RT-qPCR检测结果显示,巨噬细胞诱导极化成功,且CASC19在M1型巨噬细胞中的表达水平高于M0型和M2型(P<0.05);与M1 NC-CM组处理相比,CASC19敲降的M1 SI-CM处理组对结直肠癌细胞SW480增殖和迁移能力的抑制作用更弱(P<0.001);敲降M1型巨噬细胞中的CASC19后M1分子标记CD40、IL-1β的表达均降低,而M2分子标记CD206、IL-10、CD163的表达水平升高(P<0.05)。结论敲降CASC19能减弱M1型巨噬细胞抑制结直肠癌细胞增殖、迁移功能,可能是通过解除CASC19对巨噬细胞M1极化状态的维持作用实现。 展开更多

Dynamic fault tree analysis is widely used for the reliability analysis of the complex system with dynamic failure characteristics. In many circumstances, the exact value of system reliability is difficult to obtain due to absent or insufficient data for failure probabilities or failure rates of components. The traditional fuzzy operation arithmetic based on extension principle or interval theory may lead to fuzzy accumulations. Moreover, the existing fuzzy dynamic fault tree analysis methods are restricted to the case that all system components follow exponential time-to-failure distributions. To overcome these problems, a new fuzzy dynamic fault tree analysis approach based on the weakest n-dimensional t-norm arithmetic and developed sequential binary decision diagrams method is proposed to evaluate system fuzzy reliability. Compared with the existing approach,the proposed method can effectively reduce fuzzy cumulative and be applicable to any time-tofailure distribution type for system components. Finally, a case study is presented to illustrate the application and advantages of the proposed approach. 展开更多

β-elemene is an effective anti-cancer component which has been widely used in clinic.However,it still relies on the extraction from the Chinese medicine plant Curcuma wenyujin,which seriously limits its application.S... β-elemene is an effective anti-cancer component which has been widely used in clinic.However,it still relies on the extraction from the Chinese medicine plant Curcuma wenyujin,which seriously limits its application.Synthetic biology offers a promising approach to satisfy its supply.β-elemene is derived from germacrene A(GA),which is synthesized by germacrene A synthase(GAS),through Cope rearrangement under heat condition instead of enzymatic reaction.In this study,an effective germacrene A synthase(ScGAS)was identified from Solidago canadensis which could produce GA when expressed in E.coli.By introducing the heterogeneous MVA pathway to enrich the FPP pool,the strain yielded 147 mg/L of GA in shake flasks which represented 2.98-fold improvement over the initial one.Moreover,combining molecular docking with phylogeny analysis of ScGAS largely narrowed down the category of its key residues’mutagenesis.The Y376L mutant showed the highest yield of 487 mg/L which was almost 10-fold higher than the initial yield.These results indicate that diverting the metabolism of the host and enzyme mutagenesis based on the combination of molecular docking and phylogeny analysis are of great value to constructing terpenoids chassis. 展开更多
