目的探讨白头翁皂苷B4和常春藤皂苷C(HSC,又名白头翁皂苷B5)质量比为4∶1的组合物对帕金森病和阿尔茨海默病模型小鼠的保护作用及机制。方法取60只健康雄性C57BL/6小鼠,随机抽取10只作为对照组,剩余50只小鼠ip1-甲基-4-苯基-1,2,3,6-四... 目的探讨白头翁皂苷B4和常春藤皂苷C(HSC,又名白头翁皂苷B5)质量比为4∶1的组合物对帕金森病和阿尔茨海默病模型小鼠的保护作用及机制。方法取60只健康雄性C57BL/6小鼠,随机抽取10只作为对照组,剩余50只小鼠ip1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)制备帕金森病(PD)模型,模型构建成功后随机分为模型组、左旋多巴胺片(阳性对照,30 mg·kg^(-1))组和白头翁皂苷B4+HSC高、中、低剂量(20、10、5 mg·kg^(-1))组,每组10只,ig给药14 d,每天1次,对照、模型组小鼠ig等量0.9%的氯化钠溶液。每2天观察动物震颤行为出现与消失的时间,于给药第7、14天展开行为学实验并记录评分;HE染色法观察脑组织黑质病理学变化;免疫组化检测脑组织α-突触核蛋白表达;实时荧光定量PCR(qRTPCR)检测小鼠脑组织多巴胺转运体(DAT)、酪氨酸羟化酶(TH)mRNA表达。将50只APP/PS1转基因小鼠随机分为模型组、盐酸多奈哌齐(阳性药,1.5 mg·kg^(-1))组和白头翁皂苷B4+HSC高、中、低剂量(20、10、5 mg·kg^(-1))组,取10只同背景APP/PS1转基因阴性小鼠作为对照组,每天1次,连续ig给药28 d,模型组和对照组ig等量0.9%的氯化钠溶液。给药结束后进行行为学实验并记录评分;ELISA法检测小鼠血清中炎症因子白细胞介素(IL-6、IL-1β)、肿瘤坏死因子-α(TNF-α)水平、环氧化酶2(COX-2)和氧化应激因子活性氧(ROS),试剂盒检测超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)水平。结果在PD模型小鼠中,与模型组比较,白头翁皂苷B4+HSC显著降低震颤麻痹评分(P<0.01、0.001),显著降低旷场实验评分(P<0.01、0.001),显著降低爬杆实验评分(P<0.01、0.001),显著降低悬挂实验评分(P<0.01、0.001),显著降低游泳实验评分(P<0.01、0.001),显著降低行为学实验综合评分(P<0.001),明显改善黑质体结构损伤,明显降低α-突触核蛋白表达,显著升高小鼠脑组织DAT和TH mRNA水平(P<0.05、0.01、0.001)。在AD模型小鼠中,与模型组比较,白头翁皂苷B4+HSC显著缩短水迷宫定位航行实验逃避潜伏期(P<0.001),明显延长在目标象限的累计停留时间(P<0.05、0.01、0.001),显著增加准确穿越平台所在位置的次数(P<0.05、0.01),显著降低血清炎症因子水平(P<0.05、0.01、0.001),显著降低ROS水平、升高SOD和GSH-Px的水平(P<0.05、0.01、0.001)。结论白头翁皂苷+HSC对PD和AD小鼠具有保护作用,可能通过发挥抗炎、抗氧化,改善脑组织和神经元损伤等来实现。 展开更多

软相尺寸对反贝壳复合材料压缩性能的影响

文章采用一步球磨结合真空热压工艺制备了由Al_(2)O_(3)/Al纳米复合材料(硬相)和纯铝(软相)构成的反贝壳结构复合材料,研究了软相尺寸(从微米级到亚微米级)对复合材料压缩性能的影响,并对其变形失效机制进行了分析。结果表明:在保持高... 文章采用一步球磨结合真空热压工艺制备了由Al_(2)O_(3)/Al纳米复合材料(硬相)和纯铝(软相)构成的反贝壳结构复合材料,研究了软相尺寸(从微米级到亚微米级)对复合材料压缩性能的影响,并对其变形失效机制进行了分析。结果表明:在保持高失效应变的前提下,抗压强度随软相尺寸的减小而逐渐提高,尤其是从微米级减小到亚微米级时,抗压强度达到494 MPa,比微米级时至少提高60%;当软相尺寸为微米级时,复合材料中的裂纹萌生于软硬相界面处,并沿着界面-硬相-软相扩展,最终形成沿着与加载方向呈45°的宏观主裂纹。同时,软相可有效阻挡二次裂纹的扩展,当软相尺寸减小到亚微米级别时,裂纹萌生于软相变形带附近并沿着变形带-硬相扩展,逐渐演变成与加载方向呈±45°的均匀分散于整个复合材料的微裂纹,即软相尺寸从微米级减小到亚微米级时,增韧机制由软相对裂纹的阻挡转变为微裂纹的增殖和均匀分散。该研究结果为制备高强韧铝基复合材料提供重要参考价值。 展开更多

Some natural soils and cultivated soils on sloping land in southern Jiangsu Province, China were chosen to study soil degradation associated with erosion. Soil erosion intensity was investigated using the 137Cs tracer method. Soil particle-size distribution, soil organic matter (OM), total nitrogen (TN) and total phosphorus (TP) were measured, and the effects of erosion on soil physical and chemical properties were analyzed statistically using SYSTAT8.0. Results indicated that erosion intensity of cultivated soils was greater than that of the natural soils, suggesting that cultivation increased soil loss. Erosion also led to an increase of coarser soil particle proportion, especially in natural soils. In addition, silt was the primary soil particle lost due to erosion. However, in cultivated fields, coarser soil particles over time were attributed not only to soil erosion but also to mechanical eluviation as a result of farming activities. Moreover, erosion caused a decrease in soil OM, TN and TP as well as thinning of the soil layer. 展开更多

In this paper, removal of NOx(namely DeNOx) from flue gas by radical injection combined with NaOH solution(26% by weight of NaOH in water) scrubbing was investigated. The experimental results showed that the steady streamer corona occurs through adjusting the flow rate of the oxygen fed into the nozzles electrode. The vapor in the oxygen has influence on the V I characteristics of corona discharge. Both HNO 2 and HNO 3 come into being in the plasma reactor and the DeNOx efficiency in the plasma reactor is more than 60%. The overall DeNOx efficiency of the whole system reaches 81 7% when the NaOH solution scrubbing is collaborated. 展开更多

Manipulation of mouse genome has merged as one of the most important approaches for studying genefunction and establishing the disease model because of the high homology between human genome and mouse genome. In this study, the chemical mutagen ethylnitrosourea (ENU) was employed for inducing germ cell mutations in maleC57BL/6J mice. The first generation (G1) of the backcrossof these mutated mice, totally 3172, was screened for abnor-mal phenotypes on gross morphology, behavior, learning and memory, auditory brainstem response (ABR), electrocardio-gram (ECG), electroretinogram (ERG), flash-visual evoked potential (F-VEP), bone mineral density, and blood sugarlevel. 595 mice have been identified with specific dominantabnormalities. Fur color changes, eye defects and hearing loss occurred at the highest frequency. Abnormalities related to metabolism alteration are least frequent. Interestingly, eye defects displayed significant left-right asymmetry and sexpreference. Sex preference is also observed in mice with ab-normal bone mineral density. Among 104 G1 generation mutant mice examined for inheritability, 14 of them have been confirmed for passing abnormal phenotypes to their progenies. However, we did not observe behavior abnormali-ties of G1 mice to be inheritable, suggesting multi-gene con-trol for these complicated functions in mice. In conclusion, the generation of these mutants paves the way for under-standing molecular and cellular mechanisms of these ab-normal phenotypes, and accelerates the cloning of disease-related genes. 展开更多

Removal of NOx ( DeNOx, NOx is the total of NO and NO2) from flue gas by radical injection has been investigated . The discharge characteristics were examined and the steady streamer corona was acquired by adjusting the nozzle gases properly. It was found that an increase in the voltage resulted in a decrease in the NO concentration and the concentration of the NO2 increased at low voltages but decreased as the voltage rose to a certain level. The DeNOx efficiency increased as the applied voltage rose and reached a maximum of 70% when the voltage approached the break-down voltage. The hypothetical mechanism of NOx removal suggested that the radicals formed in the discharge process converted the NO and NO2 into acidic species. The Monte Carlo method was used to calculate the rate coefficients and the productivity of the radicals, and then the concentrations of both NO and NO2 and the DeNOx efficiencies were calcu-lated with chemical kinetics. The calculated DeNOx efficien-cies were comparable with the experimental DeNOx efficien-cies at low voltages, but were lower at high voltages. 展开更多
