Hair Growth Promoting Effect of Trichoxidil<up>TM</up>: A New Natural Compound for Hair Loss

Thus, the present study aimed to investigate the effect of Trichoxidil<sup>TM</sup>, a phytocomplex obtained from a blend of essential oils, in the treatment of hair loss caused by AGA. <strong>Methods:</strong> The CCD1072Sk cells were cultured for the 24-hour cell viability assessment and cytotoxicity of Trichoxidil<sup>TM</sup>. The expression of mRNA levels from KGF, IGF-1, and VEGF in fibroblasts was evaluated by RT-qPCR. Thirty-three volunteers, diagnosed with AGA, men and women, aged between 25 and 50 years, were divided into Control group, without treatment (n = 5);Trichosol<sup>TM</sup> vehicle group, without active (n = 5);Hydroalcoholic vehicle group, without active (n = 4);Trichosol<sup>TM</sup> vehicle group, with 2.5% minoxidil (n = 5);Hydroalcoholic vehicle group, with 2.5% minoxidil (n = 5);Trichosol<sup>TM</sup> group with 2.5% Trichoxidil<sup>TM</sup> (n = 5) and Hydroalcoholic vehicle group with 2.5% Trichoxidil<sup>TM</sup> (n = 4) to dermoscopic and histologic. <strong>Results:</strong> Fibroblasts exhibited higher proliferation when treated with higher concentrations of Trichoxidil<sup>TM</sup>. Trichoxidil<sup>TM</sup> significantly increased the expression of KGF, IGF-1, and VEGF mRNA in fibroblasts cells. Analysis of the capillary density showed that Trichoxidil<sup>TM</sup> associated with Trichosol<sup>TM</sup> vehicle, was the most effective association. In addition, it was observed an increased more effectively the percentage of anagen phase and reduction of the telogen when compared to other formulations. <strong>Conclusion:</strong> Trichoxidil<sup>TM</sup> promoted proliferative effects and positively modulated the expression of growth factors IGF-1, VEGF, and KGF, being a promising candidate for the treatment of hair loss caused by AGA. 展开更多

Background: Most anxiety complaints are treated with pharmacological measures involving barbiturates and benzodiazepines, in which they may end up causing tolerance and pharmacological dependence. Integrative approaches such as aromatherapy are used in addition to medications to improve sleep quality and reduce anxiety. Thus, PinetoninTM, a phytocomplex obtained from a blend of essential oils aims to aid in the symptoms of stress and anxiety. Methods: The cytotoxicity of PinetoninTM was evaluated MTT assay using fibroblasts and astrocytes showed reduction in the cell viability only at high concentrations. Evaluation of intracellular calcium and determination of residual glutamate in the supernatant of astrocyte cultures showed agonist action of dihydroxyphenylglycine (DHPG) increasing linearly the concentration of intracellular calcium and the glutamate levels in the supernatants of the cultures. On the other hand, cultures of astrocytes treated with PinetoninTM showed residual glutamate levels in the supernatants reducing proportionally, as well as, intracellular calcium reduction. The determination of salivary cortisol showed a significant reduction in salivary cortisol levels in the group that received PinetoninTM. The evaluation of the electroencephalogram in patients treated with PinetoninTM had a significant increase (P Results: A reduction in dose-dependent cell viability was observed when compared to cultures of PinetoninTM treated fibroblasts with control culture. When PinetoninTM and linole are administered in astrocytic cells, there was a reduction of the intracellular concentration of Ca2+ against a control group treated with DHPG agonist. The evaluation of salivary cortisol demonstrated a reduction when the patient group was treated with PinetoninTM by purchasing the results against the group of patients treated with saline. Reinforcing the relaxed state of that group, alpha waves have been increased and reductions in beta waves. Conclusion: The results obtained after intranasal administration of PinetoninTM suggest that this phytocomplex can help reduce the symptoms of stress and the better quality of life. 展开更多

The human hair follicle, a mini-organ formed with neuroectodermal-meso-dermal interaction, is a complex structure, in the active steady state (anagen) the dermal papilla can be considered as a ball of extracellular matrix, surrounding specialized fibroblasts. The cross-talk of dermal papilla with neighbouring matrix cells results in the maintenance of hair fibre production. This study aimed to investigate the proliferative potential of the compound TrichotechTM, a phytocomplex obtained from a mixture of essential oils, on cultured human fibroblasts and its ability to modulate the gene expression of FGF-7 and FGF-10. TrichotechTM was shown to enhance fibroblasts proliferation in concentrations of 0.5% to 2.0%, and also increase the percentage of cells in the S/G2/M phases of the cell cycle. TrichotechTM at both 1.0% and 2.0% induced a statistically significant effect on wound healing assay compared to the untreated control. We examined the interaction between cell survival (PI3K/Akt) and mitogenic (Ras/MAPK) signal transduction pathways after TrichotechTM treatment (1.0% and 2.0%) on the fibroblast cell line. TrichotechTM caused phosphorylation of ERK1/2, as well as greater phosphorylation of MEK in comparison with both the untreated control and ERK1/2. PI3K and AKT, however, were not shown to be significantly more phosphorylated following TrichotechTM exposure. To verify the relative expression of mRNA for FGF-7 and FGF-10 genes, a real-time polymerase chain reaction (qPCR) protocol was used. Results show the increase in mRNA expression by fibroblasts after treatment with TrichotechTM. In both concentrations tested, TrichotechTM was found to increase the expression of FGF-7 and FGF-10. Sirius red staining allows for rapid assessment of collagen content, it showed a significant increase in collagen content in treated fibroblasts. Further investigation concerning TrichotechTM could be helpful towards the development of new bioactive phytocomplexes for dermatological and trichological use. 展开更多
