Paclitaxel(PTX),a valuable natural product derived from Taxus species,exhibits remarkable anti-cancer properties.It penetrates nanopores in microtubule walls,interacting with tubulin on the lumen surface and disrupting microtubule dynamics,thereby inducing cytotoxic effects in cancer cells.PTX and its derivatives have gained approval for treating various diseases due to their low toxicity,high efficiency,and broad-spectrum application.The widespread success and expanding applications of PTX have led to increased demand,raising concerns about accessibility.Consequently,researchers globally have focused on developing alternative production methods and applying nanocarriers in PTX delivery systems to enhance bioavailability.This review examines the challenges and advancements in PTX sourcing,production,physicochemical properties,anti-cancer mechanisms,clinical applications,trials,and chemo-immunotherapy.It aims to provide a comprehensive reference for the rational development and effective utilization of PTX. 展开更多

Application of bone transgenic zebrafish in anti-osteoporosis chemical screening 被引量:9

Background: The zebrafish(Danio rerio) has recently been shown to be an ideal model to study bone disease including osteoporosis. The zebrafish osteoporosis model could be induced by glucocorticoid treatment with chemical staining for reflecting the level of bone mineralization. However, this methodology was unstable.Here, we developed a novel methodology to directly evaluate the bone mass and density.Methods: We generated and used the bone of transgenic zebrafish Tg(ola.sp7:nlsGFP) to evaluate the bone mass and density by measuring the areal extent and the integrated optical density(IOD) of enhanced green fluorescent protein(eGFP).This methodology was further compared with the traditional chemically stained method showing the bone mineralization. Furthermore, genes related to zebrafish osteoporosis were analyzed by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Results: Our results of new methods were consistent with those from chemically stained fish, following glucocorticoid-induction or epimedium flavonoid(FE)-rescue treatments. q RT-PCR analyses on mRNA levels revealed that glucocorticoid induces osteoporosis by downregulating the expression of osteoblast-related factors osterix,osteocalcin, and osteopontin, and upregulating the expression of osteoclast-related factor tartrate-resistant acid phosphatase. In FE-rescued fish, the expression of osteogenic factors osterix, osteocalcin, and osteopontin were increased.Conclusion: Compared to the traditional chemical staining methods, the new osteoporosis model using Tg(ola.sp7:nlsGFP) is more convenient and efficient for studying osteoporosis in vivo, and especially for high-throughput anti-osteoporosis drug screening. 展开更多

超声引导下逆行克氏针内固定治疗大龄儿童锁骨干骨折

目的探讨实时超声引导下逆行克氏针进针法辅助闭合复位内固定治疗大龄儿童锁骨干骨折的临床疗效。方法我院自2020年1月至2023年10月收治儿童锁骨干骨折患儿27例,男19例,女8例。手术方式均采用超声引导下逆行克氏针进针法结合闭合复位克... 目的探讨实时超声引导下逆行克氏针进针法辅助闭合复位内固定治疗大龄儿童锁骨干骨折的临床疗效。方法我院自2020年1月至2023年10月收治儿童锁骨干骨折患儿27例,男19例,女8例。手术方式均采用超声引导下逆行克氏针进针法结合闭合复位克氏针内固定。全程使用超声进行监控,实时超声引导下穿刺逆行进针、闭合复位、顺行进针、固定完成后检查骨折复位质量,最后C臂机透视验证复位效果。结果手术均顺利完成,闭合复位成功率100%,无血管神经损伤、针道感染、克氏针断裂、骨折畸形愈合、骨折不愈合等并发症发生。所有患儿均获得随访,时间7~10个月,平均8.6个月。骨折愈合后即行克氏针取出,愈合时间4~~5周,平均4.3周,患侧肢体达到完全正常活动范围为7~10周,平均8.2周。术后4个月随访患儿QuickDASH评分6.5±1.5分,所有患儿MRC肌力评分均为5分。术后半年随访患儿功能、肌力均正常,未出现再骨折、锁骨短缩畸形等情况。结论超声引导下逆行克氏针进针法行内固定治疗大龄儿童锁骨干骨折具有微创、疗效确切、并发症少、减少放射性损害等优点,操作简单、学习曲线短,便于临床推广。 展开更多

Tim-3 mRNA expression in peripheral blood lymphocytes from asthmatic patients

The study aimed to detect the expression of the Th1-specific cell surface protein T cell Ig and mucin domain-containing molecule-3(Tim-3)mRNA peripheral blood lymphocytes isolated from asthmaticin patients and to examine the correlation among Tim-3 mRNA,interleukin-4(IL-4),interferon-γ(IFN-γ)level,and FEV1/FVC(force expiratory volume in one second/forced vital capacity)to explore the role of Tim-3 in the development and progression of asthmatic inflammation.Tim-3 mRNA expression was detected by reverse transcription polymerase chain reaction(RT-PCR).The IL-4 and IFN-γlevels were determined by using enzyme-linked immunosorbent assay(ELISA).The correlation among Tim-3 mRNA,IL-4,IFN-γlevel,and pulmonary ventilatory capacity was analyzed.The expres-sion of Tim-3 mRNA in patients with acute asthma exacerbation was 0.39�0.06,significantly higher than that in patients at the remission stage and controls(0.18�0.05 and 0.07�0.03,P<0.05).The level of IL-4 in patients with acute asthma exacerbation was 68.42�10.54,significantly higher than that in the patients at the remission stage and controls(41.83�9.37 and 32.75�8.16,P<0.05).The level of IL-4 in patients in remission was significantly higher than that in controls(P<0.05).The level of IFN-γin patients with acute asthma exacerbation was 65.74�7.85,significantly lower than that in patients in remission and the control group(120.84�11.62 and 139.65�13.47,P<0.05).The level of IFN-γin patients in asthma remission was significantly lower than that in controls(P<0.05).Tim-3 mRNA expression was positively correlated with the level of IL-4(r=0.68,P<0.05)and negatively with the level of IFN-γand pulmonary ventilatory capacity(r=–0.85,r=–0.76,both P<0.01).The increased expression of Tim-3 mRNA in peripheral blood lymphocytes might be involved in the development and progression of asthmatic inflammation. 展开更多

This study aims to research the expression of spleen tyrosine kinase(Syk)in non-small cell lung cancer(NSCLC)and the relationship between Syk and clinico-pathologic factors and p53.Immunohistochemistry was applied to detect the expression of Syk and p53 protein in 39 cases of NSCLC(23 cases of lung squamous cell can-cer,16 cases of lung adenocarcinoma)and tumor-sur-rounding normal lung tissues.The positive rate of Syk was 46.15%(18/39)and 100%(39/39)in NSCLC and tumor-surrounding normal lung tissues,respectively.The expres-sion level of Syk in NSCLC was significantly lower than that in tumor-surrounding normal lung tissues(P=0.000).The Syk expression was positively correlated with the p53 expression in NSCLC specimens(P=0.025).There was no significant association between Syk expression and lymph node metastasis,differentiation degree,tumor size and tumor node metastasis(TNM).The present study demonstrated that Syk was aberrantly expressed in the NSCLC and might have a significant impact on tumor growth and progression. 展开更多
