Genotype-specific primer s...A dual real-time quantitative polymerase chain reaction assay (drtqPCR) was established to detect and differentiate between Porcine circovirus-2a (PCV-2a) and Porcine circovirus-2b (PCV-2b). Genotype-specific primer sets and probes were designed by using sequence data published for different PCV-2 strains. Specificity and sensitivity of the drtqPCR were examined by using PCV-2 isolates with known genotype. Among 367 tissue samples, 44.69% (164/367) were PCV-2 positive. From 164 PCV-2 positive samples, 10.98% (18/164), 92.56% (137/164), and 3.31% (9/164) were positive for PCV-2a, PCV-2b, and both genotypes, respectively. These results suggest that the dif-ferential drtqPCR can be used to detect PCV-2 and to differentiate the 2 genotypes from field sam-ples. The PCV-2 infection is quite common in swine of Shanghai area. Furthermore, the PCV-2b infective ratio is far higher than PCV-2a, and PCV-2a/2b mixed infections are also observed but at a lower prevalence in Shanghai area.展开更多
Acupuncture is used worldwide to treat migraine,but its scientific mechanism remains unclear.Here,we report a 1H NMR metabolomics study involving 40 migraine patients and 10 healthy individuals randomly receiving acup...Acupuncture is used worldwide to treat migraine,but its scientific mechanism remains unclear.Here,we report a 1H NMR metabolomics study involving 40 migraine patients and 10 healthy individuals randomly receiving acupuncture or sham acupuncture,followed by machine learning techniques and functional analysis.We found that acupuncture at acupoints particularly enhanced anaerobic glycolysis and modified mitochondrial function by adjusting the levels of plasma pyruvic acid(p=0.012),lactic acid(p=0.031)and citrate(p=0.00079)at a Bonferroni-corrected level of significance compared to the pre-treatment level of these three metabolites in migraine patients.Therefore,acupuncture supplies energy to migraine patients and relieves migraine attacks.In contrast,we observed that sham acupuncture may partially supply energy to migraine patients through lipid metabolism by changing the levels of plasma lipid(p=0.0012),glycerine(p=0.021),and pyruvic acid(p=0.047)at a Bonferroni-corrected level of significance.The functional network analysis further indicates this different way of supplying energy contributes to the different effects of acupuncture and sham acupuncture.Our find-ings reveal novel metabolic evidence for the specific effect of acupuncture in relation to sham acupuncture.This metabolic evidence could enlighten a brand new direction into acupuncture analgesia mechanism,which in turn would pose fresh challenges for future acupuncture research.展开更多
Harnessing the dynamics of complex quantum systems is an area of much interest and a quantum simulator has emerged as a promising platform to probe exotic topological phases.Since the flexibility offered by various co...Harnessing the dynamics of complex quantum systems is an area of much interest and a quantum simulator has emerged as a promising platform to probe exotic topological phases.Since the flexibility offered by various controllable quantum systems has helped gain insight into the quantum simulation of such complicated problems,an analog quantum simulator has recently shown its feasibility to tackle the problems of exploring topological phases.However,digital quantum simulation and the detection of topological phases still remain elusive.Here,we develop and experimentally realize the digital quantum simulation of topological phases with a solid-state quantum simulator at room temperature.Distinct from previous works dealing with static topological phases,the topological phases emulated here are Floquet topological phases.Furthermore,we also illustrate the procedure of digitally simulating a quantum quench and observing the nonequilibrium dynamics of Floquet topological phases.Using a quantum quench,the 0-andπ-energy topological invariants are unambiguously detected through measuring time-averaged spin polarizations.We believe our experiment opens up a new avenue to digitally simulate and detect Floquet topological phases with fast-developed programmable quantum simulators.展开更多
