BACKGROUND Folate metabolism gene polymorphisms may play an important role in the pathogenesis of autism spectrum disorder(ASD).However,most studies have primarily used single candidate gene typing strategies(such as ...BACKGROUND Folate metabolism gene polymorphisms may play an important role in the pathogenesis of autism spectrum disorder(ASD).However,most studies have primarily used single candidate gene typing strategies(such as targeted polymerase chain reaction technology),and current findings remain inconsistent.AIM To investigate the association of folate metabolism gene polymorphisms with ASD susceptibility and symptom severity among Chinese children.METHODS Whole-exome sequencing(WES)was conducted to systematically screen for coding region variants of key genes in the folate metabolism pathway among children with ASD,focusing on identifying polymorphisms with high mutation frequencies and potential pathogenic effects.A case-control study was then conducted to explore the association of candidate folate metabolism gene polymorphisms with the susceptibility and severity of ASD.RESULTS WES was performed on 70 children with ASD,and the case-control study included 170 children with ASD and 170 healthy controls.WES revealed that 84.3%(59/70)of children with ASD carried potentially pathogenic variants enriched in folate metabolism pathways.MTHFR C677T and MTRR A66G were significantly associated with an increased risk of ASD in both codominant and dominant models(P<0.05).The dominant model of MTRR A66G was also significantly associated with higher scores in the domains of social relations,body and object use,social and adaptive skills,total scores on the Autism Behavior Checklist,as well as emotional reactivity,nonverbal communication,and activity level on the Childhood Autism Rating Scale(P<0.05).CONCLUSION Most children with ASD carry deleterious variants in folate metabolism-related pathways.MTHFR C677T and MTRR A66G mutations are significantly associated with ASD.展开更多
Histopathological examination is still the gold standard for diagnoses of oral-maxillofacial lesions,but it is invasive and time-consuming.Optical coherence tomography(OCT)provides a kind of noninvasive,label-free,rea...Histopathological examination is still the gold standard for diagnoses of oral-maxillofacial lesions,but it is invasive and time-consuming.Optical coherence tomography(OCT)provides a kind of noninvasive,label-free,real-time and high-resolution imaging technology.In this study,in order to assess the feasibility of OCT in oral clinical application,fresh excised tissue specimens from 59 patients undergoing oral-maxillofacial surgery were imaged in detail by using a benchtop sweptsource OCT system.It is shown that different lesions or tissues can be obviously distinguished based on their different microstructural features in OCT images,and the features are similar to those of their corresponding histopathological images.It is proven that OCT has great feasibility and potential as a diagnostic aid for surgeons in oral medicine.展开更多

Two groups of students in first (n=164, mean age=6.65 years at first test) and second grade (n=202, mean age=7.73 years at first test) were followed on orthographic awareness, morphological awareness and rapid automatized naming (RAN) for two years. The children exhibited significant improvement in orthographic awareness, morphological awareness and RAN from grades 1 to 4. More importantly, to the orthographic and morphological awareness, while the children took a leap from grade 1 to 2 and grade 3 to 4, the progress developed at relatively slow rates from grade 2 to 3. In order to assure children's development of orthographic and morphological awareness, evidence-based orthographically and morphologically enhanced instruction is needed for Chinese children in the early elementary years, especially for those at the stage from grade 2 to 3.展开更多

When CTLs co-cultured with the UM cells, most UM cells could be eliminated. Survival UM cells grew slowly and were considered to be survival variants and examined by a microarray analysis. These differential genes were analyzed further with Venn Diagrams and functions related to immune escape. We additionally examined transcriptional changes of manually selected survival variants of UM cells and of clinical UM samples by quantitative real-time polymerase chain reaction (qRT-PCR), and analyzed the correlation of these expressions and patients’ survival.RESULTS: Gene expression analyses revealed a marked up-regulation of SLAMF7 and CCL22 and a significant down-regulation of KRT10, FXYD3 and ABCC2. The expression of these genes in the relapsed UM was significantly greater than those in primary UM. UM patients with overexpression of these genes had a shorter survival period as compared with those of their underexpression.CONCLUSION: Gene expression, in particular of SLAMF7, CCL22, KRT10, FXYD3 and ABCC2, differed between primary UM cells and survival variants of UM cells.展开更多

OBJECTIVE To evaluate the prospective association between cumulative resting heart rate(cumRHR)and rapid renal function decline(RRFD)in a cohort of individuals aged 60 and older.METHODS In the Tianjin Chronic Kidney D...OBJECTIVE To evaluate the prospective association between cumulative resting heart rate(cumRHR)and rapid renal function decline(RRFD)in a cohort of individuals aged 60 and older.METHODS In the Tianjin Chronic Kidney Disease Cohort Study,the individuals who underwent three consecutive physical examinations between 2014 and 2017,with estimated glomerular filtration rate(eGFR)greater than 60 mL/min per 1.73 m2 and aged 60 years or older were enrolled.A total of 27,564 patients were prospectively followed up from January 1,2017 to December 31,2020.The 3-year cumRHR was calculated.The primary outcome was RRFD,defined as an annualized decline in eGFR of 5 mL/min per 1.73 m2 or greater.Logistic and restricted spline regression models and subgroup analysis were used to investigate the association of cumRHR with RRFD after adjusting for all confounders.RESULTS During a median follow-up of 3.2 years,a total of 4,347(15.77%)subjects developed RRFD.In fully-adjusted models,compared with the lowest quartile of cumRHR,the odds ratio(OR)for the highest was 1.44(1.28–1.61),P<0.001.Furthermore,each 1-standard deviation(27.97 beats/min per year)increment in cumRHR was associated with a 17%(P<0.001)increased risk of RRFD,with a linear positive correlation(P for non-linear=0.803).Participants with a 3-year cumRHR≥207(beats/min)*year(equivalent to≥69 beats/min per year in 3 years)were found to be at a higher risk of RRFD.CONCLUSIONS The cumRHR is significantly associated with a higher risk of RRFD among older adults.These results might provide an effective goal for managing and delaying the decline of renal function in the older adults.展开更多

Iconic memory and short-term memory are not only crucial for perception and cognition,but also of great importance to mental health.Here,we first showed that both types of memory could be improved by improving limitin...Iconic memory and short-term memory are not only crucial for perception and cognition,but also of great importance to mental health.Here,we first showed that both types of memory could be improved by improving limiting processes in visual processing through perceptual learning.Normal adults were trained in a contrast detection task for ten days,with their higher-order aberrations(HOA)corrected in real-time.We found that the training improved not only their contrast sensitivity function(CSF),but also their iconic memory and baseline information maintenance for short-term memory,and the relationship between memory and CSF improvements could be well-predicted by an observer model.These results suggest that training the limiting component of a cognitive task with visual perceptual learning could improve visual cognition.They may also provide an empirical foundation for new therapies to treat people with poor sensory memory.展开更多
