羟丙基-β-环糊精包合艾纳香挥发油的制备工艺研究

为了制备艾纳香挥发油-羟丙基-β-环糊精(HP-β-CD)包合物,并考察包合物的包合效果及包合后的质量。本研究采用饱和水溶液法制备艾纳香挥发油-HP-β-CD包合物,运用正交试验法对其制备工艺进行优选,分别考察艾纳香挥发油包合物的产率、... 为了制备艾纳香挥发油-羟丙基-β-环糊精(HP-β-CD)包合物,并考察包合物的包合效果及包合后的质量。本研究采用饱和水溶液法制备艾纳香挥发油-HP-β-CD包合物,运用正交试验法对其制备工艺进行优选,分别考察艾纳香挥发油包合物的产率、油利用率、含油率3个指标,并根据上述指标计算其综合评分,优选出最佳工艺条件,最后采用薄层色谱(TLC)和紫外扫描(UV)等方法对包合前后艾纳香挥发油的性质进行考察和分析。结果显示,HP-β-CD将艾纳香挥发油包合的最佳工艺条件为:包合温度50℃、HP-β-CD与艾纳香油的比例8∶1(g∶ml)、包合时间3 h。最佳包合工艺所制备的包合物包合率为83%,综合评分为72.33。TLC和UV等表征结果证明了包合物的生成。本实验研究表明:优选出的艾纳香挥发油-HP-β-CD包合工艺合理、可行,这为艾纳香挥发油进一步的制剂开发提供了数据参考。 展开更多

Peroxiredoxins(Prxs) are a large family of antioxidant enzymes of various types that take part in signal transduction via decomposing reactive oxygen species(ROS). Although extensive efforts have been made over the last decades in understanding the structures and functions of Prxs, type II Prxs in monocots are hardly studied. In this work, a monocot type II Prx gene homologue from Chinese wildrye(Leymus Chinensis), designated as LcTpxII, was isolated and characterized. LcTpxII encoding a 162-amino acid protein contains a thioredoxin domain and a cysteine residue at position 51, suggesting that it is a member of the Prxs family. The LcTpxII is capable of decomposing H2O2 and protecting plasmid DNA from damage caused by ROS. The expression of LcTpxII in Chinese wildrye was induced by 400 mmol/L NaCl and 100 mmol/L Na2CO3 in the experiment. The overexpression of LcTpxII enhances the tolerance of transgenic yeast to 1.6 mol/L NaCl and 10 mmol/L Na2CO3. 展开更多
