Exercise training benefits pancreatic islet by modulating the insulinlike growth factor 1/phosphatidylinositol 3-kinase/protein kinase B pathway

BACKGROUND Diabetes is characterized by insulin resistance as well as impaired insulin production,withβ-cell dysfunction playing a critical role in disease progression.Exercise is known to improve insulin sensitivity,but its effects on pancreatic islet quality and function remain poorly understood.This work hypothesized that swimming training enhances glycemic control and insulin secretion by upregulating the insulin-like growth factor 1(IGF-1)/phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)pathway in streptozotocin(STZ)-induced diabetic rats.AIM To investigate the effects of swimming on pancreatic islet quality and function in STZ-induced diabetic rats via the IGF-1/PI3K/AKT pathway.METHODS Twenty-six Sprague-Dawley rats were grouped into diabetic and control groups,with each group further split into exercise and sedentary subgroups.Diabetic rats were induced with STZ.The exercise groups underwent swimming training for 60 minutes/day,5 days/week,for 8 weeks.Body weight,food intake,blood glucose,insulin,lipids,and muscle glycogen were measured.Pancreatic islet morphology and the protein expression levels of IGF-1,PI3K,and AKT were analyzed.Data were analyzed using two-way repeated-measure ANOVA,followed by Tukey’s post-hoc test.RESULTS Exercise training significantly improved body weight[diabetic exercise group(D-Ex):390.66±50.14 g vs diabetic sedentary group(D-Sed):315.89±50.12 g,P<0.05],reduced blood glucose(D-Ex:12.21±4.43 mmol/L vs D-Sed:17.79±2.05 mmol/L,P<0.05),and increased insulin levels(D-Ex:53.50±15.31 pmol/L vs D-Sed:25.31±10.23 pmol/L,P<0.05)in diabetic rats.It also enhanced islet morphology,increased IGF-1 expression,and activated the PI3K/AKT pathway(P<0.05).In-vitro experiments confirmed that IGF-1 positively regulated insulin expression and inhibitedβ-cell apoptosis via the PI3K/AKT pathway.CONCLUSION Exercise training improves pancreatic islet quality and function in diabetic rats by modulating the IGF-1/PI3K/AKT pathway,highlighting its therapeutic potential for diabetes management. 展开更多

Electron beam selective melting (EBSM) is a promising additive manufacturing (AM) technology. The EBSM process consists of three major procedures:(1) spreading a powder layer, (2) preheating to slightly sinter the powder, and (3) selectively melting the powder bed. The highly transient multi-physics phenomena involved in these procedures pose a significant challenge for in situ experimental observation and measurement. To advance the understanding of the physical mechanisms in each procedure, we leverage high- fidelity modeling and post-process experiments. The models resemble the actual fabrication procedures, including (1) a powder-spreading model using the discrete element method (DEM), (2) a phase field (PF) model of powder sintering (solid-state sintering), and (3) a powder-melting (liquid-state sintering) model using the finite volume method (FVM). Comprehensive insights into all the major procedures are provided, which have rarely been reported. Preliminary simulation results (including powder particle packing within the powder bed, sintering neck formation between particles, and single-track defects) agree qualitatively with experiments, demonstrating the ability to understand the mechanisms and to guide the design and optimization of the experimental setup and manufacturing process. 展开更多

Manipulation of light-matter interaction is critical in modern physics, especially in the strong coupling regime, where the generated half-light, half-matter bosonic quasiparticles as polaritons are important for fundamental quantum science and applications of optoelectronics and nonlinear optics. Two-dimensional transition metal dichalcogenides (TMDs) are ideal platforms to investigate the strong coupling because of their huge exciton binding energy and large absorption coefficients. Further studies on strong exciton-plasmon coupling by combining TMDs with metallic nanostructures have generated broad interests in recent years. However, because of the huge plasmon radiative damping, the observation of strong coupling is significantly limited at room temperature. Here, we demonstrate that a large Rabi splitting (~300 meV) can be achieved at ambient conditions in the strong coupling regime by embedding Ag-WS2 heterostructure in an optical microcavity. The generated quasiparticle with part-plasmon, part-exciton and part-light is analyzed with Hopfield coefficients that are calculated by using three-coupled oscillator model. The resulted plasmon-exciton polaritonic hybrid states can efficiently enlarge the obtained Rabi splitting, which paves the way for the practical applications of polaritonic devices based on ultrathin materials. 展开更多

Laser scanning confocal endomicroscope(LSCEM)has emerged as an imaging modality which provides noninvasive,in vivo imaging of biological tissue on a microscopic scale.Scientific visualizations for LSCEM datasets captured by current imaging systems require these datasets to be fully acquired and brought to a separate rendering machine.To extend the features and capabilities of this modality,we propose a system which is capable of performing realtime visualization of LSCEM datasets.Using field-programmable gate arrays,our system performs three tasks in parallel:(1)automated control of dataset acquisition;(2)imaging-rendering system synchronization;and(3)realtime volume rendering of dynamic datasets.Through fusion of LSCEM imaging and volume rendering processes,acquired datasets can be visualized in realtime to provide an immediate perception of the image quality and biological conditions of the subject,further assisting in realtime cancer diagnosis.Subsequently,the imaging procedure can be improved for more accurate diagnosis and reduce the need for repeating the process due to unsatisfactory datasets. 展开更多

The hierarchical structure of the composite cathodes brings in significant chemical complexity related to the interfaces,such as cathode electrolyte interphase.These interfaces account for only a small fraction of the volume and mass,they could,however,have profound impacts on the cell-level electrochemistry.As the investigation of these interfaces becomes a crucial topic in the battery research,there is a need to properly study the surface chemistry,particularly to eliminate the biased,incomplete characterization provided by techniques that assume the homogeneous surface chemistry.Herein,we utilize nano-resolution spatially-resolved x-ray spectroscopic tools to probe the heterogeneity of the surface chemistry on LiNi0.8Mn0.1Co0.1O2 layered cathode secondary particles.Informed by the nano-resolution mapping of the Ni valance state,which serves as a measurement of the local surface chemistry,we construct a conceptual model to elucidate the electrochemical consequence of the inhomogeneous local impedance over the particle surface.Going beyond the implication in battery science,our work highlights a balance between the high-resolution probing the local chemistry and the statistical representativeness,which is particularly vital in the study of the highly complex material systems. 展开更多

Retrospective analysis of clinical characteristics and prognostic factors for pathologic fracture in 44 patients with extremity osteosarcoma

Objective: The aim of this study was to identify the clinical features and prognostic factors associated with extremity osteosarcoma with pathologic fracture. Methods: The clinical records of 271 patients with extremity osteosarcomas were retrospectively reviewed. The data obtained covered the period from October 2003 to May 2012, and included sex, age, tumor site etc. The mean follow-up time was 25.2 months(ranged from 1 to 117). Chi-square method and Kaplan-Meier method were used to compare clinical differences and overall survival between patients with or without pathologic fracture, respectively. The univariate analysis was used to determine the prognostic factors related with survival rate by log-rank test. The multivariate analysis of prognosis was performed by COX proportional hazards regression model. Results: The proportions of patients having a tumor's diameter of 10 cm or more(P = 0.038), locating upper limbs(P = 0.004) and receiving amputation surgery(P = 0.02) were significantly higher with pathological fracture group than without pathological fracture group. The local recurrence rate(P = 0.000) was also significantly higher in the pathological fracture group. The median survival time of patients with or without pathological fracture was 16(95% confidence interval: 14.6–17.4) months and 22(95% confidence interval: 19.8–24.1) months(P = 0.002). The Log-rank univariate analysis indicated that the tumor size, Enneking's surgical staging, Karnofsky performance status(KPS) score, cycles of adjuvant chemotherapy, local recurrence and metastasis were significantly related to overall survival. Multivariate Cox regression analysis revealed KPS score, cycles of adjuvant chemotherapy and metastasis were independent prognostic factors(P < 0.05). Conclusion: Compared with the patients without pathological fracture, a higher proportion of patients receiving amputation surgery or having larger tumor size, humeral osteosarcoma or local recurrence was observed in patients with pathological fracture, and the prognosis of these patients was poor. The independent prognostic factors of extremity osteosarcoma with pathologic fracture were the KPS score, cycles of adjuvant chemotherapy and metastasis. 展开更多

Objective:The aim of this study was to evaluate the efficacy,adverse reaction and survival of MAID protocol as first-line treatment on 137 metastatic soft tissue sarcomas patients.Methods:One hundredand thirty-seven patients with metastatic soft tissue sarcoma were given MAID protocol chemotherapy which consists of ifosfamide at a total dose of 8-10 g/m2 in 4-5 days,adriamycin at a dose of 60 mg/m2 on first day and dacarbazine at a dose of 200 mg/m2/d in 4-5 days.Twenty one days were regarded as a cycle and 2-6 cycles were completed.The median cycle number was 4.Results:Four cases had complete remission(CR)(2.9%),thirteen cases achieved partial remission(PR)(9.5%),eighty-eight cases maintained a stable disease(SD)(64.2%),and thirty-two cases had progressive disease(PD)(23.4%).The overall response rate(RR) was 12.4%,disease control rate(DCR) was 76.6%.The progression-free survival(PFS) was 2-14 months and median PFS was 5.00 ± 1.12 months.The overall survival(OS) was 3-19 months and median OS was 8.00 ± 1.32 months.Three months PFS rate was 56.9%,six months PFS rate was 27.2% and one year PFS rate was 7.8%.Three months OS rate was 100.0%,six months OS rate was 41.5% and one year OS rate was 10.7%.The main adverse reactions were bone marrow supression,nausea/vomiting and alopecia.Other adverse reactions were seldom observed.Conclusion:The effect of MAID protocol as first-line treatment on metastatic soft tissue sarcomas patients was exact,the treatment can control disease progression effectively and the side effects can be tolerable.It was obviously very worthy in clinical use as one of the first-line chemotherapy protocols on advanced soft tissue sarcomas. 展开更多

背景 产前抑郁对产妇健康和妊娠结局具有重要影响。既往研究表明,产妇产前抑郁与社会支持相关、社会支持与分娩恐惧相关,但关于产妇产前抑郁、社会支持、分娩恐惧三者之间关系的研究较少,且未查见关于社会支持与分娩恐惧对初产妇产前抑... 背景 产前抑郁对产妇健康和妊娠结局具有重要影响。既往研究表明,产妇产前抑郁与社会支持相关、社会支持与分娩恐惧相关,但关于产妇产前抑郁、社会支持、分娩恐惧三者之间关系的研究较少,且未查见关于社会支持与分娩恐惧对初产妇产前抑郁影响的研究。目的 了解初产妇产前抑郁现状,分析其产前抑郁与社会支持和分娩恐惧的相关性,并进一步探讨社会支持和分娩恐惧对初产妇产前抑郁的影响,为改善该群体的心理健康状况提供参考。方法 选取2022年12月—2023年9月成都市温江区人民医院住院部收治的380名初产妇为研究对象。采用自制调查表、爱丁堡产后抑郁量表(EPDS)、社会支持评定量表(SSRS)以及分娩态度量表(CAQ)进行调查。采用Pearson相关分析检验初产妇各量表评分之间的相关性。采用多元线性回归分析检验产前抑郁的影响因素。结果 共发放问卷380份,回收有效问卷372份(97.89%)。检出222名(59.68%)初产妇存在产前抑郁。初产妇EPDS评分与SSRS评分呈负相关(r=-0.283,P<0.01),与CAQ评分呈正相关(r=0.341,P<0.01)。多元线性回归分析结果 显示,社会支持(β=-0.166,P<0.01)和分娩恐惧水平(β=0.269,P<0.01)是初产妇产前抑郁的影响因素。结论 初产妇产前抑郁情况不容乐观,其产前抑郁水平与社会支持和分娩恐惧密切相关。 展开更多

长江经济带发展战略实施以来,在“生态优先、绿色发展”“共抓大保护、不搞大开发”理念指引下,长江干线船舶水污染物已从直接排放、达标排放逐步向“入江零排放”转变。按照致公党中央关于长江生态环境保护民主监督工作的部署要求,课... 长江经济带发展战略实施以来,在“生态优先、绿色发展”“共抓大保护、不搞大开发”理念指引下,长江干线船舶水污染物已从直接排放、达标排放逐步向“入江零排放”转变。按照致公党中央关于长江生态环境保护民主监督工作的部署要求,课题组于2024年6月-7月前往合肥、芜湖、铜陵、池州,就“安徽省长江流域船舶和港口污染防控”课题开展实地调研。调研发现:安徽长江流域船舶和港口污染防控已取得显著成效,但还存在监管体制协调性不足、信息系统数据共享有待提升、重建设轻应用、污染物接收配套经费精细化程度不高、船舶供需协调性与危化品船员培训监管力度有待提升等问题。建议:贯彻落实中共二十届三中全会精神,全面推进安徽港航生态环境治理责任体系、监管体系、市场体系、法律法规政策体系建设。通过统一执法、多方监管,整合资源、注重激励,科学研判、加强船舶生命周期管理,重视人员培训和预防,建立跨地方跨部门协商机制等措施,进一步提升安徽省港航污染防控效果,促进长江流域航运业高质量发展。 展开更多

金属卤化物钙钛矿因具有优异的光电特性,被广泛应用于光电探测器。然而,溶液法制备钙钛矿多晶薄膜的工艺复杂,且薄膜的连续性和致密度不高,进而影响其光电性能。使用溶液处理离心铸造法快速制备了连续且致密的CsPbBr_(3)多晶薄膜,采用... 金属卤化物钙钛矿因具有优异的光电特性,被广泛应用于光电探测器。然而,溶液法制备钙钛矿多晶薄膜的工艺复杂,且薄膜的连续性和致密度不高,进而影响其光电性能。使用溶液处理离心铸造法快速制备了连续且致密的CsPbBr_(3)多晶薄膜,采用叉指金电极构建了金属-半导体-金属结构的CsPbBr_(3)微米晶薄膜光电探测器。利用离心力增强CsPbBr_(3)微米晶粒之间的接触,形成连续且致密的高质量薄膜,最终有效地提升了CsPbBr_(3)多晶薄膜光电探测器的性能。所制备的光电探测器表现出出色的弱光电探测能力,在520 nm波长光源照射下,器件的光响应度和比探测率分别达到525 mA/W和2.8×10^(12)Jones。研究结果表明,通过离心铸造法能够有效地提高溶液处理的钙钛矿多晶薄膜的质量,进而提升其光电器件性能。 展开更多

地基云的自动观测和识别对分析大气运动趋势和天气预测具有指导意义。针对目前地基云图分类算法准确率不高、在嵌入式终端部署困难的问题,提出了一种基于残差网络结构的地基云图分类网络模型GBcNet及基于ZYNQ的硬件实现架构,PS端用于加... 地基云的自动观测和识别对分析大气运动趋势和天气预测具有指导意义。针对目前地基云图分类算法准确率不高、在嵌入式终端部署困难的问题,提出了一种基于残差网络结构的地基云图分类网络模型GBcNet及基于ZYNQ的硬件实现架构,PS端用于加载模型的权重参数和云图数据,PL端实现DDR3读写控制和GBcNet的硬件加速。设计了滑窗、卷积层、池化层、批量归一化层和全连接层等模块的加速IP核。实验在CCSN数据集上进行,结果表明,提出的模型在PC端的准确率达到96.02%。采用现场可编程门阵列(FPGA)硬件加速后,准确率仍然保持在94.5%。与PC端模型的识别率相比,各云类的识别精度损失均不超过3%,整体精度损失小于1.5%;FPGA的最大资源占用不超过48%,单张地基云图推理时间为0.13 s。相较于现有地基云的识别方法,识别准确率高且推理时间较短。提出的识别模型和硬件加速方法为便携式地基云观测设备的研制提供了一种参考方案。 展开更多

研究制备周期有序的高纵横比GaN纳米阵列,有效改善光电器件的光吸收和提取。利用聚苯乙烯胶体球(PS球)和介质掩模刻蚀,四甲基氢氧化铵(TMAH)腐蚀辅助的方法,制备高纵横比的GaN纳米柱阵列。在制备过程中,掩膜层的刻蚀选择比决定纳米柱的... 研究制备周期有序的高纵横比GaN纳米阵列,有效改善光电器件的光吸收和提取。利用聚苯乙烯胶体球(PS球)和介质掩模刻蚀,四甲基氢氧化铵(TMAH)腐蚀辅助的方法,制备高纵横比的GaN纳米柱阵列。在制备过程中,掩膜层的刻蚀选择比决定纳米柱的高度和形貌,感应耦合等离子体刻蚀(ICP)参数和TMAH腐蚀时间影响纳米柱的形貌。研究了不同掩膜层的选择比、ICP气体流量和种类、TMAH腐蚀时间对纳米柱形貌的影响,优化调整了工艺。扫描电子显微镜(SEM)表征纳米柱形貌,分析表明:(1)PS球、100 nm氮化硅和20 nm镍组合掩膜的选择比最高,制备出直径高度比大于1∶5的高纵横比纳米柱;(2)ICP刻蚀的最佳参数为64 sccm氯气,8 sccm三氯化硼,ICP功率100 W,BIAS功率70 W,腔压0.6 Pa,刻蚀速率约90 nm/min,侧壁倾斜角度约80°;(3)纳米柱的侧壁会随着TMAH的腐蚀时间变得垂直光滑,腐蚀时间不够侧壁呈阶梯状台阶,腐蚀时间过长纳米柱从底部脱落,最佳腐蚀时间为30 min,并从原子结构解释了湿化学腐蚀行为。 展开更多

目的:分析评价八段锦对抑郁症状改善的临床效果,为八段锦功法的运用提供证据支持。方法:检索并收集在国内外数据库公开发表的关于八段锦对抑郁症状改善的临床随机对照试验。经筛选后,使用Cochrane与NoteExpress 3.2来评估文献质量以及... 目的:分析评价八段锦对抑郁症状改善的临床效果,为八段锦功法的运用提供证据支持。方法:检索并收集在国内外数据库公开发表的关于八段锦对抑郁症状改善的临床随机对照试验。经筛选后,使用Cochrane与NoteExpress 3.2来评估文献质量以及进行文献管理。应用RevMan5.4.1软件进行统计分析,根据Q检验即P值的大小,评估文献有无异质性及其大小,从而使用固定或随机效应模型,然后计算OR值,在此基础上并绘制森林图。结果:共纳入8项RCTs。结果显示,与对照组相比,试验组行八段锦功法治疗能降低SDS、SAS与HAMD评分水平。结论:现有证据表明,八段锦功法对抑郁症状的改善效果明显,可有效减轻患者的抑郁程度,改善焦虑症状,为抑郁症患者提供更广泛的选择可能。 展开更多

目的:探究选择性血清素再摄取抑制剂(SSRIs)联合作业疗法(OT)对卒中后抑郁(PSD)患者神经递质、心理水平及日常生活能力改善情况。方法:纳入90例PSD患者作为研究对象,按照治疗方式不同分为对照组(n=45)和试验组(n=45)。对照组采用SSRIs治... 目的:探究选择性血清素再摄取抑制剂(SSRIs)联合作业疗法(OT)对卒中后抑郁(PSD)患者神经递质、心理水平及日常生活能力改善情况。方法:纳入90例PSD患者作为研究对象,按照治疗方式不同分为对照组(n=45)和试验组(n=45)。对照组采用SSRIs治疗;试验组采用SSRIs结合OT治疗。比较两组患者治疗后总有效率、去甲肾上腺素(NE)、5-羟色胺(5-HT)、脑源性神经营养因子(BDNF)、胰岛素样生长因子-1(IGF-1)水平、汉密尔顿抑郁量表(HAMD)评分、日常生活活动(ADL)评分、匹兹堡睡眠质量指数量表(PSQI)评分及不良反应发生情况。结果:两组患者治疗总有效率试验组95.56%高于对照组的80.00%(P<0.05);治疗后,两组患者血清NE、IGF-1、5-HT、BDNF水平均改善(P<0.05),且试验组改善优于对照组(P<0.05);两组患者ADL评分均升高(P<0.05),PSQI评分均降低(P<0.05),且试验组患者ADL、PSQI评分均较对照组改善更佳(P<0.05);试验组患者不良反应总发生率为2.22%,低于对照组的17.78%(P<0.05)。结论:PSD患者使用SSRIs联合OT治疗,能改善心理水平、睡眠质量,调控神经递质,进而提高日常生活能力,增加治疗安全性,优势显著。 展开更多

随着抗生素在医疗、农业以及畜牧业等行业的广泛应用,地表水中抗生素污染问题已经对人类健康和生态系统的稳定构成了严重威胁。我国西南地区与南亚、东南亚多个国家接壤,因其独特的地理位置和复杂的地形地貌,地表水环境中抗生素的分布... 随着抗生素在医疗、农业以及畜牧业等行业的广泛应用,地表水中抗生素污染问题已经对人类健康和生态系统的稳定构成了严重威胁。我国西南地区与南亚、东南亚多个国家接壤,因其独特的地理位置和复杂的地形地貌,地表水环境中抗生素的分布特征及环境效应具有一定特殊性。目前,已有研究结果显示在我国西南地区地表水中检测到多种抗生素,且发现抗生素具有典型的分布特征。通过综述近年来西南地区地表水中抗生素的污染现状及其分布特征,并探讨抗生素对水生生态系统、微生物群落以及人类健康的潜在影响,期望为该地区地表水环境中抗生素污染的控制与科学管理提供一定参考依据。 展开更多
