目的通过观察接种乙型肝炎病毒(hepatitis B virus,HBV)后新生树鼩体内细胞因子TLR-4和IL-6的变化,初步探讨HBV感染过程中宿主体内的免疫反应。方法将31只树鼩幼崽分为实验组(n=25)和对照组(n=6)。实验组接种人HBV,接种后第8周至第24周...目的通过观察接种乙型肝炎病毒(hepatitis B virus,HBV)后新生树鼩体内细胞因子TLR-4和IL-6的变化,初步探讨HBV感染过程中宿主体内的免疫反应。方法将31只树鼩幼崽分为实验组(n=25)和对照组(n=6)。实验组接种人HBV,接种后第8周至第24周定期检测树鼩外周血中乙肝表面抗原(HBsAg)和HBV-DNA,根据检测结果将实验组分为慢性感染组7只(血清HBsAg或HBV-DNA持续阳性时间达到24周)和疑似慢性感染组5只(血清HBsAg或HBV-DNA不连续出现2次或2次以上阳性),其余13只Hbs Ag和HBV-DNA均阴性,不纳入研究;对照组不做任何处理。采用双抗体夹心法检测树鼩外周血TLR-4、IL-6蛋白表达水平,RT-PCR法检测树鼩肝组织中TLR-4、IL-6 mRNA的表达水平。结果观察期间内,慢性感染组和疑似慢性感染组外周血TLR-4与IL-6蛋白的表达均高于对照组(P<0.05);肝组织中IL-6的mRNA表达水平高于对照组(P<0.05);慢性感染组TLR-4 mRNA表达水平高于对照组(P<0.05)。结论 TLR-4、IL-6在树鼩HBV感染模型中表达升高,可能在HBV感染过程中起重要作用。展开更多

Along with the aimed product quer...With an attempt to synthesize high-value isoquercitrin(quercetin-3-O-β-D-glucopyranoside), we carried out the biotransformation of quercetin(1) by Gliocladium deliquescens NRRL 1086. Along with the aimed product quercetin 3-O-β-D-glycoside(2), three additional metabolites, 2-protocatechuoyl-phlorogucinol carboxylic acid(3), 2,4,6-trihydroxybenzoic acid(4), and protocatechuic acid(5), were also isolated. The time-course experiments revealed that there were two metabolic routes, regio-selectivity glycosylation and quercetin 2,3-dioxygenation, co-existing in the culture. Both glycosylation and oxidative cleavage rapidly took place after quercetin feeding; about 98% quercetin were consumed within the initial 8 h and the oxdized product(2-protocatechuoyl-phlorogucinol carboxylic acid) was hydrolyzed into two phenolic compounds(2,4,6-trihydroxybenzoic acid and protocatechuic acid). We also investigated the impact of glucose content and metal ions on the two reactions and found that high concentrations of glucose significantly inhibited the oxidative cleavage and improved the yield of isoquercitrin and that Ca^(2+), Fe^(2+), Mn^(2+), Mg^(2+), and Zn^(2+) inhibited glycosylation. To test the promiscuity of this culture, we selected other four flavonols as substrates; the results demonstrated its high regio-selectivity glycosylation ability towards flavonols at C-3 hydroxyl. In conclusion, our findings indicated that the versatile microbe of G. deliquescens NRRL 1086 maitained abundant enzymes, deserving further research.展开更多
Effectively controlling bacterial infections and reducing oxidative stress and inflammatory reactions are important steps in wound healing.However,owing to the improper use of antibiotics and inadequate control of inf...Effectively controlling bacterial infections and reducing oxidative stress and inflammatory reactions are important steps in wound healing.However,owing to the improper use of antibiotics and inadequate control of infections in recent years,the emergence of many broadspectrum drug-resistant strains has exacerbated the threat of infected wounds to human health.Recent studies have shown that bimetallic nanozymes may become an effective means of treating drug-resistant bacterial infections because of their unique physical properties and excellent antibacterial properties.In this study,silver iron bimetallic nanozymes with multiple enzyme activities(peroxidase,glutathione peroxidase,and superoxide dismutase)were successfully synthesized for the treatment of skin wounds.Notably,the prepared V_(o)-AgFeO_(2−x) exhibited different enzyme activities under different pH conditions.In acidic environments,V_(o)-AgFeO_(2−x) can catalyze H2O2 to generate reactive oxygen species(ROS),deplete glutathione(GSH),and kill bacteria.In a neutral environment,V_(o)-AgFeO_(2−x) can eliminate free radicals,control inflammatory reactions,and accelerate wound healing.In vivo experiments have shown that V_(o)-AgFeO_(2−x) can promote the healing of infected wounds and has good biological safety.These findings suggest that it can be used as a safe and efficient antibacterial drug to achieve effective treatment of bacterial infection-induced wounds.展开更多
文摘目的探讨载脂蛋白F(apolipoprotein F,APOF)和人胰岛素样生长因子酸不稳定亚基(the insulin-like grouth factor binding protein acid labile subunit,IGFALS)在原发性肝癌组织中的表达及临床意义。方法选取2014年1月1日至2015年12月30日于广西医科大学附属肿瘤医院经手术切除及病理确诊为原发性肝癌88例,实时荧光定量PCR(qRT-PCR)法检测88例肝癌组织及其相应癌旁组织和6例正常肝组织中APOF基因和IGFALS基因mRNA的相对表达量,并分析两者表达与肝癌患者临床病理特征的关系。结果肝癌组织中APOF mRNA相对表达量低于癌旁组织和正常肝组织(0.156±0.019 vs 0.971±0.010,P<0.001;0.156±0.019 vs 81.140±0.092,P<0.001);肝癌组织中IGFALS mRNA相对表达量亦低于癌旁组织及正常肝组织(0.111±0.016 vs 1.090±0.054,P<0.001;0.111±0.016 vs 1.101±0.211,P<0.001)。APOF mRNA表达与门静脉侵犯相关(P=0.004);IGFALS mRNA表达与肿瘤分化程度、门静脉侵犯相关(P<0.05)。APOF mRNA表达水平与IGFALS mRNA表达水平呈正相关(r=0.332,P<0.01)。结论 APOF和IGFALS基因在肝癌组织中表达下调,二者表达呈正相关,且与门静脉侵犯有关,可能参与了肝癌的发生发展。
