[Objectives] To evaluate the effects of Huanglian Jiedu Decoration( HLJD) on the treatment of rat gingivitis with excess internal heat through injecting lipopolysaccharide( LPS) in tooth-supporting tissues. [Methods]Gingival inflammation was established by LPS injection. Male SD rats were randomly divided into 7 groups: normal control group( no treatment group),solvent control group,model group( LPS injection),HLJD low dose group( 0. 25 g/kg),HLJD medium dose group( 0. 50 g/kg),HLJD high dose group( 1. 00 g/kg),and metronidazole and fenbufen capsule( MFC) group. Water intake and urine volume were observed through metabolic cage feeding; gingival tissue protein expressions were detected by LC-ESI-MS/MS method; oxidative stress-related molecules such as total superoxide dismutase( T-SOD) and hydrogen peroxidase catalase( CAT) were detected by the biochemical kit combined with a microplate reader; expressions of TSC related with energy metabolism-related signaling pathway were detected by western blotting. [Results]When rat gingivitis was injected by LPS,both water intake and urine volume were decreased,the levels of T-SOD and CAT were decreased,and the expressions of fructose-bisphosphate aldolase C,triosephosphate isomerase 1 and TSC2 were increased. Huanglian Jiedu Decoration can increase water intake and urine volume,increase the levels of T-SOD and CAT,reduce the expressions of fructose-bisphosphate aldolase C and triosephosphate isomerase 1,and inhibit the expression of energy metabolism-related pathway molecule TSC2. [Conclusions] Huanglian Jiedu Decoration can treat rat gingivitis through inhibiting energy metabolism and oxidative stress such as TSC signal pathway and glycolysis related molecules. 展开更多

A novel photocatalyst of mesoporous graphitic carbon nitride(g-C_3N_4) co-doped with Co and Mo(Co/Mo-MCN) has been one-pot synthesized via a simple template-free method; cobalt chloride and molybdenum disulfide were used as the Co and Mo sources, respectively. The characterization results evidently indicate that molybdenum disulfide functions as Mo sources to incorporate Mo atoms in the framework of g-C_3N_4 and as a catalyst for promoting the decomposition of g-C_3N_4, resulting in the creation of mesopores. The obtained Co/Mo-MCN exhibited a significant enhancement of the photocatalytic activity in H_2 evolution(8.6 times) and Rhodamine B degradation(10.1 times) under visible light irradiation compared to pristine g-C_3N_4. Furthermore, density functional theory calculations were applied to further understand the photocatalytic enhancement mechanism of the optical absorption properties at the atomic level after Co-or Mo-doping. Finite-di erence time-domain simulations were performed to evaluate the e ect of the mesopore structures on the light absorption capability. The results revealed that both the bimetal doping and the mesoporous architectures resulted in an enhanced optical absorption; this phenomenon was considered to have played a critical role in the improvement in the photocatalytic performance of Co/Mo-MCN. 展开更多

Goose meat is rich in amino acids with unique taste and aroma traits that are targeted by housing and feeding systems.This study aimed to investigate the impacts of in-house feeding(IHF)and artificial pasture grazing(AGF)systems on nutrient composition,flavor,and taste of thigh muscles of goose.The proximate analysis showed that,compared to IHF group,AGF group contained significantly higher contents of moisture and crude protein than ash and intramuscular fat.The findings of transcriptome analysis revealed that 139 differentially expressed genes(DEGs)displayed differential expression(102 genes downregulated and 37 genes upregulated),and only 13 DEGs were involved in fatty acid metabolic process,protein binding,and calcium ion binding.Together,79 and 16 significantly differential metabolites(SDMs)were identified through LC-MS/MS and GC-TOF-MS,respectively.Furthermore,we performed KEGG enrichment analysis of DEGs and related them with enriched signaling pathways attained from SDMs.Collectively,we detected 4 common KEGG pathways involved in the development of flavor and taste in AGF geese,namely sphingolipid metabolism,arginine and proline metabolism,alanine,aspartate and glutamate metabolism,and valine,leucine and isoleucine degradation.In these pathways,the upregulated uridine diphosphate(UDP)-glycosyltransferase 8(UGT8),glycine amidino-transferase(GATM),and aldehyde dehydrogenase(ALDH)and downregulated ceramide synthase 6(CERS6)genes may regulate the synthesis of metabolites including L-serine,phosphatidylethanolamine,L-arginine,L-glutamate,lysyl-proline,D-ornithine,putrescine,and alpha-ketoisocaproic acid 2 in pasture-fed group.These genes and metabolites may play a key role in regulating protein accumulation,IMF deposition,meat flavor,and taste development in pasture-fed group. 展开更多

Here we developed a saccharic colorimetric method based on the combination of chemoselective ligation and enzyme-specific catalysis using aminooxy/ hydrazine-functionalized gold nanoparticles (AO/AuNPs or H/AuNPs). In the detection of galactose (Gal), galactohexodialdose (GHDA), the galactose oxidase (GalOx)-catalyzed product, has an aldehyde group, which allows it to chemoselectively react with an aminooxy or hydrazine group at the outer layer of AO/AuNPs or H/AuNPs by oxime/hydrazone click chemistry to form oxime or hydrozone. Consequent134 through the specific recognition of 1,4-phenylenediboronic acid (PDBA) on cis-diols, GHDA, which contains two pairs of hydroxyls in the cis form, can bind not only with AO/AuNPs or H/AuNPs, but also with PDBA to form boronate diester, thereby triggering the aggregation of AuNPs and causing the corresponding color change. As GalOx catalyzed specific substrates, the amount of Gal correlated with the production of GHDA and the extent of AuNPs aggregation, thus allowing a simple and easily operatable colorimetric method for Gal detection to be developed. Under the optimized experimental conditions, the ratios of absorbance at a wavelength of 617 nm to that at 536 nm vary linearly with the logarithmic values of Gal concentrations within a wide range of 500 nM to 5 mM. Moreover, this colorimetric method shows anti-interference capability and high sensitivity with a detection limit of 21 nM. Thus, a universal platform for accurate and specific colorimetric analysis can be established through the integration of chemoselective ligation with enzyme specific catalysis. 展开更多

Zika virus (ZIKV) is a previously-neglected, but newly-emerging zoonotic pathogen that has swept most of the western hemisphere. The unexpected attention aroused worldwide lies in the explosive spread of ZIKV to Brazil and USA in 2015 and its rare correlation to micro- cephaly in newborns, raising a serious challenge to public health. We show the origin and fast dissemination of these epidemic ZIKA isolates. Phylogenetic analyses revealed that all the newly-emerging ZIKV isolates belong to the Asian-American Lineage. Structural analyses of the ZIKV E surface protein suggested that it might adopt a similar entry mechanism to that of other Flavi-viruses like Dengue Virus and West Nile Virus. 展开更多
