Transcriptomic and metabolomic analyses insight into the synergistic effects of nitric oxide and hydrogen sulfide fumigation on enhancing postharvest antioxidant defense and phenylpropane metabolism in‘crystal’grape

‘Crystal’grapes are thin and juicy skin and brittle pericarp tissues,which are extremely perishable with a short shelf life after harvest,and in order to examine the influence of NO and H_(2)S fumigation on the phys... ‘Crystal’grapes are thin and juicy skin and brittle pericarp tissues,which are extremely perishable with a short shelf life after harvest,and in order to examine the influence of NO and H_(2)S fumigation on the physiological metabolism of‘Crystal’grape during postharvest storage.In this study,transcriptomic and metabolomic analyses revealed the impact of nitric oxide(NO)and hydrogen sulfide(H_(2)S)fumigation on the physiological metabolism,disease resistance in‘Crystal’grape.The results demonstrated that the combined treatment of NO and H_(2)S preserved the storage quality of‘Crystal’grape.This positive preservation effect was attributed to enhanced antioxidant capacity and the stimulation of phenolic secondary metabolite biosynthesis.The analysis were mainly enriched in phenylpropanoid and flavonoid biosynthesis pathways.NO+H_(2)S up-regulated the expression of key genes in phenylpropanoid metabolism,including VvPAL,Vv4CL,VvFLS,VvCCR,and VvPOD,and increased the enzymatic activities of phenylalamine ammonia lyase(PAL),cinnamate 4-Hydroxylase(C4H),peroxidase(POD),and beta-glucosidase(BGLU).Furthermore,with the positive regulation of the phenylpropanoid metabolic,NO+H_(2)S also activated the flavonoid biosynthesis,promoting the accumulation of secondary metabolites such as quercetin,kaempferol,myricetin,and(+)-catechin.These changes enhanced postharvest disease resistance and antioxidant capacity in‘Crystal’grape.Additionally,NO+H_(2)S treatment reduced malondialdehyde(MDA)and hydrogen peroxide(H_(2)O2)accumulation by upregulating antioxidant enzymes,including ascorbate peroxidase(APX),catalase(CAT),peroxidase(POD)and glutathione reductase(GR),thereby mitigating oxidative stress.This process helps maintain redox homeostasis,reduces membrane lipid peroxidation,and delays postharvest quality deterioration. 展开更多

This study aimed to prepare an adenosine triphosphate(ATP)liposome by reverse evaporation to improve the stability of exogenous ATP and regulate its sustained release.Its microstructure and physicochemical indexes were systematically characterized to obtain the optimal drug-lipid ratio.The results showed that egg yolk lecithin(EYL)/ATP-5 exhibited minimal vesicle size with good encapsulation efficiency(63.26%±0.20%).The storage stability and thermogravimetric(TG)results showed that EYL/ATP-5 exhibited better dispersion prop-erties during storage;however,it did not significantly affect the thermal stability of liposomes.Transmission electron microscopy results further corroborated that the liposomes at this ratio exhibited a homogeneous and unaggregated vesicular structure,which was favorable for the maintenance of liposome stability.EYL/ATP-5 treatment was used for the postharvest preservation of white mushrooms.The results showed that EYL/ATP-5 treatment maintained the firmness of white mushroom substrate after harvesting,delayed browning,inhibited its respiration,and postponed the quality deterioration of white mushrooms.In conclusion,the ATP liposomes obtained in this study provided a new idea for storing and preserving harvested edible fungi. 展开更多

Recent advances in nanocarriers for bioactives have focused on finding more sustained-release,natural materials.In this study,genipin-crosslinked soyβ-conglycinin(β-CG)-curcumin nanoparticles(G-SCP)significantly enhanced the stability of curcumin and released curcumin controllably in the gastric environment.The optimal intra-particle crosslinkage for G-SCP was obtained under the genipin concentration less than 0.2 mg/mL and stirring at 37℃ for 24 h,which was confirmed by ninhydrin assay,amino acid analysis,dynamic light scattering(DLS)and flourier transform infrared spectroscopy(FT-IR)analysis.Transmission electron microscopy(TEM),DLS and release of trichloroacetic acid(TCA)-soluble nitrogen were used to systematically evaluate the sustained-release behavior and mechanism of curcumin from the G-SCP.The anti-cancer activity of oral-intake curcumin was investigated by collecting the G-SCP digestion supernatant and analyzing its cellular uptake and proliferation in HepG2 cells.The results revealed that the bioaccessible curcumin released from G-SCP had stronger anti-proliferation for HepG2 cells than free curcumin.On normal liver cells L02,interestingly,bioaccessible curcumin had considerably lower cytotoxicity than it did on HepG2 cells.These findings suggested that the G-SCP was a protein-based carrier that could effectively transfer insoluble bioactives in the gastric environment. 展开更多
