The study area Caofeidian 18-1/2 structure is located in the Shadongnan structural belt at the southeast subduction end of the Shaleitian salient in the western Bohai Sea. The characteristics of reservoirs and fluid inclusions from 13 core samples near the buried hills in the study area are studied,and regional geology and conditions for reservoir formation are analyzed to reveal the characteristics and the processes of reservoir formation. Phase I oil and gas inclusions are mainly developed,and the abundance of oil and gas inclusions in this period is high( GOI is about 15%). The homogenization temperature of the hydrocarbon-containing brine inclusions accompanying them is mainly 90-120 ℃ . The simulation results of burial history and thermal history show that the main charging period of oil and gas is the present Himalayan tectonic movement period since 8 Ma,and mainly through unconformities,faults,and drainage systems,they are migrated and accumulated into fault anticline traps of Dongying Formation mudstone( E_d). 展开更多

Effects of Kaixin Powder on Expression of 5-HT Receptor in Hippocampus of Depressed Rats Induced by CUMS 被引量:4

Objective To observe the influence of Kaixin Powder on ethology,content of 5-HT in the hippocampus, expression of m RNA, and protein in 5-HT1 A and 5-HT2 A receptors in the hippocampus of depressed rats induced by chronic unpredictable mild stress(CUMS). Methods Twenty-four male Wistar rats were randomly divided into blank,model, Trazodone, and Kaixin Powder groups, six rats in each group. In addition to the blank control group, other groups were established the depression model induced by CUMS combined with isolated feeding. At the same time, Trazodone group and Kaixin Powder group were treated with corresponding drugs for 3 weeks. After 3 weeks of administration, the rats were sacrificed, and a series of indexes were measured such as the contents of 5-HT, m RNA expression levels of 5-HT1 A and 5-HT2 A receptors, protein expression levels of 5-HT1 A and 5-HT2 A receptors, and so on. Results A series of indexes in the model group were decreased significantly such as the body weight growth, the sugar water intake, the score of Open Field Test, the content of5-HT in the hippocampus, expression of m RNA, and protein in 5-HT1 A receptor, while the expression of m RNA and protein in 5-HT2 A receptor were increased significantly.Compared with the model group, the indexes were ameliorated in Trazodone and Kaixin Powder groups. Kaixin Powder is better than Trazodone in decreasing the level of protein in 5-HT2 A receptor. Conclusion The result indicated that the depression performance of depressed rats induced by CUMS can be ameliorated by Kaixin Powder,and the mechanism maybe concerned with increasing the contents of 5-HT, exciting 5-HT1 A receptor, and antagonising 5- HT2 A receptor. 展开更多

Circ_0087199 depletion alleviates lps-induced oxidative injury and cell cycle arrest in human periodontal ligament cells via the mir-527/tlr4 axis

Background Circular RNAs(circRNAs)participate in the development of periodontitis.The present work aims to reveal the role and mechanism of circ_0087199 in human periodontal ligament cell(PDLC)injury during periodontitis.Methods PDLCs were treated with lipopolysaccharides(LPS)to establish a periodontitis cell model.Quantitative real-time polymerase chain reaction was used to detect the expression of circ_0087199,miR-527,toll-like receptor 4(TLR4).Western blotting assay was performed to assess protein expression.Cell viability,proliferation,apoptosis and inflammation were investigated by cell counting kit-8,EdU assay,flow cytometry and enzyme-linked immunosorbent assay,respectively.Oxidative stress was evaluated by malondialdehyde assay kit and superoxide dismutase activity assay kit.The interaction between miR-527 and circ_0087199 or TLR4 was confirmed by a dual-luciferase reporter assay.Results Circ_0087199 and TLR4 expression levels were significantly increased,while miR-527 was decreased in the periodontal ligament tissues of periodontitis patients and LPS-stimulated PDLCs when compared with controls.LPS treatment inhibited cell viability and proliferation but induced cell apoptosis,inflammation and oxidative stress,whereas these effects were attenuated after circ_0087199 knockdown.Circ_0087199 bound to miR-527 and regulated LPS-caused PDLC damage by targeting miR-527.Additionally,the overexpression of TLR4,a target gene of miR-527,rescued miR-527 mimic-mediated effects on LPS-treated PDLCs.Further,the regulation of circ_0087199 toward TLR4 involved miR-527.Conclusion Circ_0087199 knockdown attenuated LPS-induced apoptosis,inflammation and oxidative stress of PDLCs by regulating the miR-527/TLR4 pathway. 展开更多
