研制了一种新型冠状病毒(coronavirus, SARS-CoV-2)体外转录RNA标准物质,用于SARS-CoV-2病毒RNA检测的量值溯源.目标基因范围涵盖已公布的SARS-CoV-2病毒检测的3个主要靶标基因:核壳蛋白N基因全长(基因组坐标:28274-29533, GenBank:MT02... 研制了一种新型冠状病毒(coronavirus, SARS-CoV-2)体外转录RNA标准物质,用于SARS-CoV-2病毒RNA检测的量值溯源.目标基因范围涵盖已公布的SARS-CoV-2病毒检测的3个主要靶标基因:核壳蛋白N基因全长(基因组坐标:28274-29533, GenBank:MT027064.1)、包膜蛋白E基因全长(基因组坐标:26245-26472, GenBank:MT027064.1)、开放阅读框1ab(ORF1ab)基因片段(基因组坐标:13321-15540, GenBank:MT027064.1).通过采用数字聚合酶链式反应(digital PCR, dPCR)方法进行多实验室联合定值,确定了该标准物质的量值为N、E和ORF1ab基因的拷贝数浓度.研究结果表明,该RNA标准物质量值可靠,均匀性良好,稳定性可满足运输要求.进一步地,通过在临床检验、多家不同原理的病毒检测试剂盒生产企业、方法开发实验室等试用,验证了RNA标准物质的普适性.这一RNA标准物质的研制为新型冠状病毒检测试剂盒的质量控制提供了计量学依据,为防控流行性疾病提供了便利. 展开更多

Ratiometric DNA biosensors,which utilize DNA as the recognition element and integrate dual signals from diverse sensing platforms including fluorescence,electrochemistry,and surface-enhanced Raman scattering(SERS),have demonstrated remarkable proficiency in detecting a wide range of targets.In this review,we showcase the significant progress achieved by ratiometric biosensors.Firstly,ratiometric biosensors have made notable advancements in analyzing real samples.These include the analysis of pH values near cancer cells,quantification of miRNA in human cell lysates,detection of human telomerase RNA in cell extracts,and performing DNA logicgated in situ bioimaging on cell membranes.Secondly,excellent sensitivity has been attained through the utilization of effective amplification methods such as RCA,HCR,and CHA,among others.Thirdly,the construction of stable reference signals has resulted in significantly improved precision for ratiometric biosensors.This breakthrough has overcome matrix effects,enabling reliable detection in real samples with high selectivity.This review provides a comprehensive overview of recent advancements in strategies employed by ratiometric DNA biosensors.We present three types of biosensors based on distinct sensing platforms:fluorescent,electrochemical,and SERS biosensors.Additionally,we discuss future directions and primary challenges in the development of ratiometric DNA biosensors. 展开更多
