BACKGROUND Rotavirus(RV),a primary cause of diarrhea-related mortality in 2021,has been shown to damage intestinal epithelial cells while upregulating intestinal stem cells(ISCs)activities.ISCs within the crypt niche ...BACKGROUND Rotavirus(RV),a primary cause of diarrhea-related mortality in 2021,has been shown to damage intestinal epithelial cells while upregulating intestinal stem cells(ISCs)activities.ISCs within the crypt niche drive the continuous self-renewal of intestinal epithelium,preserving its barrier functions.Paneth cells secrete antimicrobial peptide and signaling molecules within the intestine crypt,thereby playing a crucial role in intestinal immune defense and providing ISCs functional support.However,the regulatory function of Paneth cells under pathological conditions,such as RV infection,remains unclear.AIM To determine the impact of RV infection on Paneth cells and how Paneth cells regulate ISCs during intestinal injury repair.METHODS We constructed a reference genome for the RV enteric cytopathogenic human orphan virus strain and reanalyzed published single-cell RNA sequencing data to investigate Paneth cell responses to RV-induced intestinal injury.We derived Paneth-ISC communication networks using CellChat,tracked ISC differentiation with pseudotime analysis,and validated our findings in leucine-rich repeat-containing G protein-coupled receptor 5-enhanced green fluorescent protein-internal ribosomal entry site-Cre recombinase estrogen receptor variant 2 mice and organoids via immunofluorescence,flow cytometry,and reverse transcription quantitative polymerase chain reaction.RESULTS We found that RV directly infects Paneth cells,leading to a reduction in mature Paneth cells and an increase in kallikrein 1-high immature Paneth cells.Paneth-ISC communication was significantly enhanced.In particular,the bone morphogenic protein 7(BMP7)-activin A receptor type 2B/BMP receptor type 1A-Smad pathway was upregulated post-infection,suggesting that Paneth cells suppress excessive ISC proliferation.Functional validation confirmed activation of this pathway.CONCLUSION Paneth cells regulate ISC proliferation during RV infection by activating BMP7 signaling,limiting excessive stem cell expansion and preserving crypt homeostasis for effective epithelial repair.展开更多

The discharge and plasma characteristics of Ag magnetron sputtering discharge operated near the electron series resonance(ESR)oscillation,which was excited using the driving frequency of 27.12 MHz,was investigated.The...The discharge and plasma characteristics of Ag magnetron sputtering discharge operated near the electron series resonance(ESR)oscillation,which was excited using the driving frequency of 27.12 MHz,was investigated.The imaginary part of impedance was found to undergo a transition from capacitive to inductive on varying radio-frequency(RF)power,and the conditions for the ESR excitation were satisfied.The current–voltage(I–V)characteristic of discharge showed that the lower discharge voltage with higher current was an important feature of RF magnetron sputtering operated near the ESR oscillation,which was caused by the small impedance Z originated from the mutual compensation between the sheath capacitive reactance and the plasma inductive reactance.The higher electron temperature,ion flux density and ion energy as well as the moderate electron density were obtained.The interaction of higher energy ions on substrate surface improved the crystallographic quality of Ag films.Therefore,the 27.12 MHz magnetron sputtering operated near the ESR oscillation has better deposition characteristics than that of commercial 13.56 MHz RF magnetron sputtering.展开更多
Silks have various advantages compared with synthetic polymer fibers,such as sustainability,mechanical properties,luster,as well as air and humidity permeability.However,the functionalization of silks has not yet been...Silks have various advantages compared with synthetic polymer fibers,such as sustainability,mechanical properties,luster,as well as air and humidity permeability.However,the functionalization of silks has not yet been fully developed.Functionalization techniques that retain or even improve the sustainability of silk production are required.To this end,a low-cost,effective,and scalable strategy to produce TCSs by integrating yarn-spinning and continuous dip coating technique is developed herein.TCSs with extremely long length(>10 km),high mechanical performance(strength of 443.1 MPa,toughness of 56.0 MJ m−3,comparable with natural cocoon silk),and good interfacial bonding were developed.TCSs can be automatically woven into arbitrary fabrics,which feature super-hydrophobicity as well as rapid and programmable thermochromic responses with good cyclic performance:the response speed reached to one second and remained stable after hundreds of tests.Finally,applications of TCS fabrics in temperature management and dynamic textile displays are demonstrated,confirming their application potential in smart textiles,wearable devices,flexible displays,and human–machine interfaces.Moreover,combination of the fabrication and the demonstrated applications is expected to bridge the gap between lab research and industry and accelerate the commercialization of TCSs.展开更多
Sympathetic activation and the kidney play critical roles in hypertension and chronic heart failure.The role of the kidney in sympathetic activation is still not well known.In this study,we revealed an excitatory rena...Sympathetic activation and the kidney play critical roles in hypertension and chronic heart failure.The role of the kidney in sympathetic activation is still not well known.In this study,we revealed an excitatory renal reflex(ERR)in rats induced by chemical stimulation of the kidney that regulated sympathetic activity and blood pressure.The ERR was induced by renal infusion of capsaicin,and evaluated by the changes in renal sympathetic outflow,blood pressure,and heart rate.Renal infusion of capsaicin dose-dependently increased the contralateral renal sympathetic nerve activity,mean arterial pressure,and heart rate.Capsaicin in the corticomedullary border had greater effects than in the cortex or medulla.Intravenous infusion of capsaicin had no significant effects.The effects of renal infusion of capsaicin were abolished by ipsilateral renal denervation,but were not affected by bilateral sinoaortic denervation.Renal infusion of capsaicin increased the ipsilateral renal afferent activity.The ERR was also induced by renal infusion of bradykinin,adenosine,and angiotensin II,but not by ATP.Renal infusion of capsaicin increased c-Fos expression in the paraventricular nucleus(PVN)of hypothalamus.Lesion of neurons in the PVN with kainic acid abolished the capsaicin-induced ERR.These findings indicate that chemical stimulation of kidney causes an excitatory reflex,leading to sympathetic activation,pressor response,and accelerated heart rate.The PVN is an important central nucleus in the pathway of the ERR.展开更多

Energy harvesting textiles(EHTs)have attracted much attention in wearable electronics and the internet-of-things for real-time mechanical energy harvesting associated with human activities.However,to satisfy practical...Energy harvesting textiles(EHTs)have attracted much attention in wearable electronics and the internet-of-things for real-time mechanical energy harvesting associated with human activities.However,to satisfy practical application requirements,especially the demand for long-term use,it is challenging to construct an energy harvesting textile with elegant trade-off between mechanical and triboelectric performance.In this study,an energy harvesting textile was constructed using natural silk inspired hierarchical structural designs combined with rational material screening;this design strategy provides multiscale opportunities to optimize the mechanical and triboelectric performance of the final textile system.The resulting EHTs with traditional advantages of textiles showed good mechanical properties(tensile strength of 237±13 MPa and toughness of 4.5±0.4 MJ m−3 for single yarns),high power output(3.5 mW m−2),and excellent structural stability(99%conductivity maintained after 2.3 million multi-type cyclic deformations without severe change in appearance),exhibiting broad application prospects in integrated intelligent clothing,energy harvesting,and human-interactive interfaces.展开更多

In addition, three therm...The free thermal vibration of functionally graded material(FGM) cylindrical shells containing porosities is investigated. Both even distribution and uneven distribution are taken into account. In addition, three thermal load types, i.e., uniform temperature rise(UTR), nonlinear temperature rise(NLTR), and linear temperature rise(LTR), are researched to explore their effects on the vibration characteristics of porous FGM cylindrical shells. A modified power-law formulation is used to describe the material properties of FGM shells in the thickness direction. Love’s shell theory is used to formulate the straindisplacement equations, and the Rayleigh-Ritz method is utilized to calculate the natural frequencies of the system. The results show that the natural frequencies are affected by the porosity volume fraction, constituent volume fraction, and thermal load. Moreover,the natural frequencies obtained from the LTR have insignificant differences compared with those from the NLTR. Due to the calculation complexity of the NLTR, we propose that it is reasonable to replace it by its linear counterpart for the analysis of thin porous FGM cylindrical shells. The present results are verified in comparison with the published ones in the literature.展开更多
This study proposes a rational strategy for the design,fabrication and system integration of the humanoid intelligent display platform(HIDP)to meet the requirements of highly humanized mechanical properties and intell...This study proposes a rational strategy for the design,fabrication and system integration of the humanoid intelligent display platform(HIDP)to meet the requirements of highly humanized mechanical properties and intelligence for human-machine interfaces.The platform’s sandwich structure comprises a middle lightemitting layer and surface electrodes,which consists of silicon elastomer embedded with phosphor and silk fibroin ionoelastomer,respectively.Both materials are highly stretchable and resilient,endowing the HIDP with skin-like mechanical properties and applicability in various extreme environments and complex mechanical stimulations.Furthermore,by establishing the numerical correlation between the amplitude change of animal sounds and the brightness variation,the HIDP realizes audiovisual interaction and successful identification of animal species with the aid of Internet of Things(IoT)and machine learning techniques.The accuracy of species identification reaches about 100%for 200 rounds of random testing.Additionally,the HIDP can recognize animal species and their corresponding frequencies by analyzing sound characteristics,displaying real-time results with an accuracy of approximately 99%and 93%,respectively.In sum,this study offers a rational route to designing intelligent display devices for audiovisual interaction,which can expedite the application of smart display devices in human-machine interaction,soft robotics,wearable sound-vision system and medical devices for hearing-impaired patients.展开更多
AIM: To investigate the mechanism of interleukin (IL)-6 secretion through blocking the IL-17A/IL-17A recepto (IL-17RA) signaling pathway with a short hairpin RNA (shRNA) in hepatic stellate cells (HSCs) in vitro . MET...AIM: To investigate the mechanism of interleukin (IL)-6 secretion through blocking the IL-17A/IL-17A recepto (IL-17RA) signaling pathway with a short hairpin RNA (shRNA) in hepatic stellate cells (HSCs) in vitro . METHODS: HSCs were derived from the livers of adul male Sprague-Dawley rats. IL-6 expression was evalu ated using real-time quantitative polymerase chain reaction and enzyme linked immunosorbent assay. The phosphorylation activity of p38 mitogen activated pro tein kinases (MAPK) and extracellular regulated pro tein kinases (ERK) 1/2 upon induction by IL-17A and suppression by IL-17RA shRNA were examined using Western blotting.RESULTS: IL-6 expression induced by IL-17A was significantly increased compared to control in HSCs (P < 0.01 in a dose-dependent manner). Suppression of IL17RA using lentiviral-mediated shRNA inhibited IL-6 expression induced by IL-17A compared to group with only IL-17A treatment (1.44 ± 0.17 vs 4.07 ± 0.43, P < 0.01). IL-17A induced rapid phosphorylation of p38 MAPK and ERK1/2 after 5 min exposure, and showed the strongest levels of phosphorylation of p38 MAPK and ERK1/2 at 15 min in IL-17A-treated HSCs. IL-6 mRNA expression induced by IL-17A (100 ng/mL) for 3 h exposure was inhibited by preincubation with specific inhibitors of p38 MAPK (SB-203580) and ERK1/2 (PD-98059) compared to groups without inhibitors preincubation (1.67 ± 0.24, 2.01 ± 0.10 vs 4.08 ± 0.59, P < 0.01). Moreover, lentiviral-mediated IL-17RA shRNA 1 inhibited IL-17A-induced IL-6 mRNA expression compared to random shRNA in HSCs (1.44 ± 0.17 vs 3.98 ± 0.68, P < 0.01). Lentiviral-mediated IL17RA shRNA 1 inhibited phosphorylation of p38 MAPK and ERK1/2 induced by 15 min IL-17A (100 ng/mL) exposure. CONCLUSION: Down-regulation of the IL-17RA receptor by shRNA decreased IL-6 expression induced by IL-17A via p38 MAPK and ERK1/2 phosphorylation in HSCs. Suppression of IL-17RA expression may be a strategy to reduce the inflammatory response induced by IL-17A in the liver.展开更多
Orchidaceae are one of the largest families of angiosperms in terms of species richness.In the last decade,numerous studies have delved into reconstructing the phylogenetic framework of Orchidaceae,leveraging data fro...Orchidaceae are one of the largest families of angiosperms in terms of species richness.In the last decade,numerous studies have delved into reconstructing the phylogenetic framework of Orchidaceae,leveraging data from plastid,mitochondrial and nuclear sources.These studies have provided new insights into the systematics,diversification and biogeography of Orchidaceae,establishing a robust foundation for future research.Nevertheless,pronounced controversies persist regarding the precise placement of certain lineages within these phylogenetic frameworks.To address these discrepancies and deepen our understanding of the phylogenetic structure of Orchidaceae,we provide a comprehensive overview and analysis of phylogenetic studies focusing on contentious groups within Orchidaceae since 2015,delving into discussions on the underlying reasons for observed topological conflicts.We also provide a novel phylogenetic framework at the subtribal level.Furthermore,we examine the tempo and mode underlying orchid species diversity from the perspective of historical biogeography,highlighting factors contributing to extensive speciation.Ultimately,we delineate avenues for future research aimed at enhancing our understanding of Orchidaceae phylogeny and diversity.展开更多

文摘This study proposes a rational strategy for the design,fabrication and system integration of the humanoid intelligent display platform(HIDP)to meet the requirements of highly humanized mechanical properties and intelligence for human-machine interfaces.The platform’s sandwich structure comprises a middle lightemitting layer and surface electrodes,which consists of silicon elastomer embedded with phosphor and silk fibroin ionoelastomer,respectively.Both materials are highly stretchable and resilient,endowing the HIDP with skin-like mechanical properties and applicability in various extreme environments and complex mechanical stimulations.Furthermore,by establishing the numerical correlation between the amplitude change of animal sounds and the brightness variation,the HIDP realizes audiovisual interaction and successful identification of animal species with the aid of Internet of Things(IoT)and machine learning techniques.The accuracy of species identification reaches about 100%for 200 rounds of random testing.Additionally,the HIDP can recognize animal species and their corresponding frequencies by analyzing sound characteristics,displaying real-time results with an accuracy of approximately 99%and 93%,respectively.In sum,this study offers a rational route to designing intelligent display devices for audiovisual interaction,which can expedite the application of smart display devices in human-machine interaction,soft robotics,wearable sound-vision system and medical devices for hearing-impaired patients.

文摘AIM: To investigate the mechanism of interleukin (IL)-6 secretion through blocking the IL-17A/IL-17A recepto (IL-17RA) signaling pathway with a short hairpin RNA (shRNA) in hepatic stellate cells (HSCs) in vitro . METHODS: HSCs were derived from the livers of adul male Sprague-Dawley rats. IL-6 expression was evalu ated using real-time quantitative polymerase chain reaction and enzyme linked immunosorbent assay. The phosphorylation activity of p38 mitogen activated pro tein kinases (MAPK) and extracellular regulated pro tein kinases (ERK) 1/2 upon induction by IL-17A and suppression by IL-17RA shRNA were examined using Western blotting.RESULTS: IL-6 expression induced by IL-17A was significantly increased compared to control in HSCs (P < 0.01 in a dose-dependent manner). Suppression of IL17RA using lentiviral-mediated shRNA inhibited IL-6 expression induced by IL-17A compared to group with only IL-17A treatment (1.44 ± 0.17 vs 4.07 ± 0.43, P < 0.01). IL-17A induced rapid phosphorylation of p38 MAPK and ERK1/2 after 5 min exposure, and showed the strongest levels of phosphorylation of p38 MAPK and ERK1/2 at 15 min in IL-17A-treated HSCs. IL-6 mRNA expression induced by IL-17A (100 ng/mL) for 3 h exposure was inhibited by preincubation with specific inhibitors of p38 MAPK (SB-203580) and ERK1/2 (PD-98059) compared to groups without inhibitors preincubation (1.67 ± 0.24, 2.01 ± 0.10 vs 4.08 ± 0.59, P < 0.01). Moreover, lentiviral-mediated IL-17RA shRNA 1 inhibited IL-17A-induced IL-6 mRNA expression compared to random shRNA in HSCs (1.44 ± 0.17 vs 3.98 ± 0.68, P < 0.01). Lentiviral-mediated IL17RA shRNA 1 inhibited phosphorylation of p38 MAPK and ERK1/2 induced by 15 min IL-17A (100 ng/mL) exposure. CONCLUSION: Down-regulation of the IL-17RA receptor by shRNA decreased IL-6 expression induced by IL-17A via p38 MAPK and ERK1/2 phosphorylation in HSCs. Suppression of IL-17RA expression may be a strategy to reduce the inflammatory response induced by IL-17A in the liver.

文摘Orchidaceae are one of the largest families of angiosperms in terms of species richness.In the last decade,numerous studies have delved into reconstructing the phylogenetic framework of Orchidaceae,leveraging data from plastid,mitochondrial and nuclear sources.These studies have provided new insights into the systematics,diversification and biogeography of Orchidaceae,establishing a robust foundation for future research.Nevertheless,pronounced controversies persist regarding the precise placement of certain lineages within these phylogenetic frameworks.To address these discrepancies and deepen our understanding of the phylogenetic structure of Orchidaceae,we provide a comprehensive overview and analysis of phylogenetic studies focusing on contentious groups within Orchidaceae since 2015,delving into discussions on the underlying reasons for observed topological conflicts.We also provide a novel phylogenetic framework at the subtribal level.Furthermore,we examine the tempo and mode underlying orchid species diversity from the perspective of historical biogeography,highlighting factors contributing to extensive speciation.Ultimately,we delineate avenues for future research aimed at enhancing our understanding of Orchidaceae phylogeny and diversity.

文摘In order to improve the quality of low-dose computational tomography (CT)images, the paper proposes an improved image denoising approach based on WGAN-gpwith Wasserstein distance. For improving the training and the convergence efficiency, thegiven method introduces the gradient penalty term to WGAN network. The novelperceptual loss is introduced to make the texture information of the low-dose imagessensitive to the diagnostician eye. The experimental results show that compared with thestate-of-art methods, the time complexity is reduced, and the visual quality of low-doseCT images is significantly improved.
