AIM:To investigate the anti-proliferation and apoptosisinducing effects of sodium aescinate(SA)on retinoblastoma Y79 cells and its mechanism.METHODS:Y79 cells were cultured at different drug concentrations for different periods of time(24,48,and 72 h).The inhibitory effect of SA on proliferation of Y79 cells was detected by the cell counting kit-8(CCK-8)assay,and the morphology of Y79 cells in each group was observed under an inverted microscope.An IC50 of 48 h was selected for subsequent experiments.After pretreatment with SA for 24 and 48 h,cellular DNA distribution and apoptosis were detected by flow cytometry.Real-time qunatitative polymerase chain reaction(RT-qPCR)and Western blot were used to assess changes in related genes(CDK1,CyclinB1,Bax,Bcl-2,caspase-9,caspase-8,and caspase-3).RESULTS:SA inhibited proliferation and induced apoptosis of Y79 cells in a time-dependent and concentrationdependent manner.Following its intervention in the cell cycle pathway,SA can inhibit the expression of CDK1 and Cyclin B1 at the mRNA and protein levels,and block cells in the G2/M phase.In caspase-related apoptotic pathways,up-regulation of Bax and down-regulation of Bcl-2 caused caspase-9 to self-cleave and further activate caspase-3.What’s more,the caspase-8-mediated extrinsic apoptosis pathway was activated,and the activated caspase-8 was released into the cytoplasm to activate caspase-3,which as a member of the downstream apoptotic effect group,initiates a caspase-cascade reaction that induces cell apoptosis.CONCLUSION:SA inhibits the proliferation of Y79 cells by arresting the cell cycle at the G2/M phase,and induces apoptosis via the caspase-related apoptosis pathway,indicating that SA may have promising potential as a chemotherapeutic drug. 展开更多

AIM:To explore the effect of the Andrographis paniculata(A.paniculata)polysaccharide on the proliferation and apoptosis of human retinoblastoma(RB)Y79 cells and its mechanism.METHODS:The refined A.paniculata polysaccharide was obtained using techniques such as water extraction,ethanol precipitation,and decompression concentration.The inhibition effect of the A.paniculata polysaccharide on the proliferation of Y79 cells was detected by cell proliferation assay.Flow cytometr y was used for the detection of cell apoptosis rate and cycle change.Realtime qunatitative polymerase chain reaction(RT q PCR)and Western blotting were used to detect the expression of cell apoptosis signal pathway-related factors(caspase-3,caspase-8,and caspase-9)and cell cycle signal pathwayrelated factors(CDK1 and cyclin B1)at the transcriptional and translational levels.RESULTS:Infrared and ultraviolet spectrum scanning showed that the extracted drug was a polysaccharide withhigh purity.After being treated with different concentrations of A.paniculata polysaccharide for different periods of time,the Y79 cells showed different degrees of proliferation inhibition.Flow cytometric observations showed that the cell apoptosis rate and the proportion of cells blocked in the G2/M phase were significantly increased after A.paniculata polysaccharide treatment.Further analysis revealed that the m RNA and protein expression of caspase-3,caspase-8,and caspase-9 in the A.paniculata polysaccharide treatment groups increased significantly compared with that in the control groups,while the expression of CDK1 and cyclin B1 decreased significantly.CONCLUSION:The A.paniculata polysaccharide could inhibit the proliferation and induce apoptosis of Y79 cells.Its possible mechanism is via the upregulation of caspase-3,caspase-8,and caspase-9 expression in the cell apoptotic signaling pathway and the downregulation of CDK1 and cyclin B1 expression in the cell cycle signaling pathway. 展开更多

Retinoblastoma(RB)is a highly aggressive ocular tumor,and due to socioeconomic and medical constraints,many children receive treatment only in the metaphase and advanced clinical stages,resulting in high rates of blindness and disability.Although several approaches exist in the treatment of RB,some children with the disease do not have satisfactory results because of various factors.Plantderived natural products have shown definite therapeutic effects in the treatment of various tumors and are also widely used in the study of RB.We review plant-derived natural products used in the study of anti-RB to provide ideas for the clinical application of these drugs and the development of new therapeutic drugs. 展开更多

In this paper,the numerical simulation method is used to study the flow resistance law of u-shaped channels under rotating conditions based on similarity theory.The study compares three geometric models:real model,completely similar model and incompletely similar model for cooling typical U-shaped channels inside turbine rotating blades.The completely similar model is geometrically 4.8 times magnification of the real model,and the rotation radius ratio of the real model is 5.4 times that of the incomplete similar model.It is found that the friction factor of the completely similar model increases with the rotation number,and the difference varies from 6%to 38%.The friction factor of the model after incomplete similarity amplification decreases with the increase of rotation number,and the difference varies from-2%to-30%.The friction factor of the laboratory imperfectly similar amplification model combined the effects of the above two laws,and the predicted difference was within 12%.This study provides a theoretical basis for subsequent experiments related to flow resistance. 展开更多
