Synergistic Effect of Silicone Macromolecular Charring Agent and Ammonium Polyphosphate on Improving Flame Retardancy and Mechanical Properties of Ethylene-Butyl Acrylate Copolymer Composites

Power cables are important pieces of equipment for energy transmission,but achieving a good balance between flame retardancy and mechanical properties of cable sheaths remains a challenge.In this work,a novel intumescent flame retardant(IFR)system containing silicone-containing macromolecular charring agent(Si-MCA)and ammonium polyphosphate(APP)was designed to synergistically improve the flame retardancy and mechanical properties of ethylene-butyl acrylate copolymer(EBA)composites.The optimal mass ratio of APP/Si-MCA was 3/1 in EBA composites(EBA/APP-Si-31),corresponding to the best flame retardancy with 31.2% of limited oxygen index(LOI),V-0 rating in UL-94 vertical burning test,and 76.4%reduction on the peak of heat release rate(PHRR)in cone calorimeter test.The enhancement mechanism was attributed to the synergistic effect of APP/Si-MCA during combustion,including the radical-trapping effect,the dilution effect of non-flammable gases,and the barrier effect of the intumescent char layer.Meanwhile,the tensile results indicated that EBA/APP-Si-31 also exhibited good mechanical properties with the addition of maleic anhydride-grafted polyethylene(PE-g-MA)as the compatibilizer.Thus,the APP/Si-MCA combination is an effective IFRs system for preparing high-performance EBA composites,and it will promote their applications as cable sheath materials. 展开更多

Biaxial compression mechanical properties of NPR anchor solid under different crack dip angles

With the rapid development of deep resource extraction and underground space construction,the design of anchored support systems for jointed rock masses in complex stress environments faces significant challenges.This study investigates the influence of prefabricated crack dip angles on the mechanical properties of anchored rock masses in deep soft rock roadways.By constructing similarity models of NPR(Negative Poisson’s Ratio)and PR(Positive Poisson’s Ratio)anchored solids,biaxial compression experiments under varying crack dip angles were conducted.Strain gauges,3D Digital Image Correlation(3D DIC),and acoustic emission monitoring were employed to systematically analyze the strength characteristics,deformation-damage evolution,and energy dissipation mechanisms of the two types of anchor systems.The results show that:(1)The stress-strain curves of anchored solids with prefabricated cracks exhibit a distinct bimodal characteristic.Compared to PR anchors,NPR anchors show 20%and 23%improvements in peak strength and elastic modulus,respectively,with residual strength enhanced by up to 34%.(2)Owing to high pre-tightening force and large deformation capacity,NPR anchors maintain superior integrity under increasing crack dip angles,demonstrating more uniform free-surface displacement and localized shear-tensile composite crack patterns.(3)Acoustic emission analysis reveals that NPR anchors exhibit higher cumulative energy absorption(300%improvement over PR anchors)and lack low-rate energy development phases,indicating enhanced ductility and impact resistance at high crack dip angles.(4)Crack dip angle critically governs failure mechanisms by modulating the connectivity between shear cracks and prefabricated fissures:bimodal effects dominate at low angles,while vertical tensile crack propagation replaces bimodal behavior at high angles.The study proposes prioritizing NPR anchor cables in deep engineering applications and optimizing support parameters based on crack dip angles to mitigate stress concentration and ensure the long-term stability of surrounding rock. 展开更多

The growth and yield of peanut are negatively affected by continuous cropping.Arbuscular mycorrhizal fungi(AMF)and calcium ions(Ca^(2+))have been used to improve stress resistance in other plants,but little is known about their roles in peanut seedling growth under continuous cropping.This study investigated the possible roles of the AMF Glomus mosseae combined with exogenous Ca^(2+)in improving the physiological responses of peanut seedlings under continuous cropping.G.mosseae combined with exogenous Ca^(2+)can enhance plant biomass,Ca^(2+)level,and total chlorophyll content.Under exogenous Ca^(2+)application,the F_v/F_m in arbuscular mycorrhizal(AM)plant leaves was higher than that in the control plants when they were exposed to high irradiance levels.The peroxidase,superoxide dismutase,and catalase activities in AM plant leaves also reached their maximums,and accordingly,the malondialdehyde content was the lowest compared to other treatments.Additionally,root activity,and content of total phenolics and flavonoids were significantly increased in AM plant roots treated by Ca^(2+)compared to either G.mosseae inoculation or Ca^(2+)treatment alone.Transcription levels of AhCaM,AhCDPK,AhRAM1,and AhRAM2 were significantly improved in AM plant roots under exogenous Ca^(2+)treatment.This implied that exogenous Ca^(2+)might be involved in the regulation of G.mosseae colonization of peanut plants,and in turn,AM symbiosis might activate the Ca^(2+)signal transduction pathway.The combination of AMF and Ca^(2+)benefitted plant growth and development under continuous cropping,suggesting that it is a promising method to cope with the stress caused by continuous cropping. 展开更多

AIM: To investigate the inflammatory amplification effect of high-mobility group box 1(HMGB1) in Aspergillus fumigatus(A. fumigatus) keratitis and the relationship between lectin-like oxidized low-density lipoprotein receptor 1(LOX-1) and HMGB1 in keratitis immune responses.METHODS: Phosphate buffer saline(PBS), and Boxb were injected into BALB/c mice subconjunctivally before the corneas were infected with A. fumigatus. RAW264.7 macrophages and neutrophils were pretreated with PBS and Boxb to determine the HMGB1 inflammatory amplification effects. Abdominal cavity extracted macrophages were pretreated with Boxb and Poly(I)(a LOX-1 inhibitor) before A. fumigatus hyphae stimulation to prove the the relationship between the two molecules. LOX-1, interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), macrophage inflammatory protein-2(MIP-2) and IL-10 were assessed by polymerase chain reaction and Western blot.RESULTS: Pretreatment with Boxb exacerbated corneal inflammation. In macrophages and neutrophils, A. fumigatus induced LOX-1, IL-1β, TNF-α and MIP-2 expression in Boxb group was higher than those in PBS group. Poly(I) treatments before infection alleviated the proinflammatory effects of Boxb in abdominal cavity extracted macrophages. Pretreatment with Boxb did not influence Dectin-1 mRNA levels in macrophages and neutrophils.CONCLUSION: In fungal keratitis, HMGB1 is a proinflammatory factor in the first line of immune response. HMGB1 mainly stimulates neutrophils and macrophages to produce inflammatory cytokines and chemokines during the immune response. LOX-1 participates in HMGB1 induced inflammatory exacerbation in A. fumigatus keratitis. 展开更多

A field study and theoretical calculations were performed to clarify the levels, profiles, and distributions of polybrominated dibenzo-p-dioxins and dibenzofurans(PBDD/Fs) in a cement kiln co-processing solid waste, with a focus on the PBDF formation mechanism.The raw materials contributed greatly to input of PBDD/Fs into the cement kiln. The PBDD/F concentrations in the raw materials were much higher than those in particle samples from different process stages in the cement kiln. The PBDD/F concentrations in the clinkers were1.40% of the concentrations in the raw materials, which indicated that the high destruction efficiencies for PBDD/Fs by cement kiln. PBDD/F distribution patterns in particle samples collected from different process stages indicated the cement kiln backend was a major site for PBDD/F formation. PBDFs with high levels of halogenation, such as heptabrominated furans(Hp BDF), were the dominant contributors to the total PBDD/F concentrations and accounted for 42%–73% of the total PBDD/F concentrations in the particle samples. Our results showed that co-processing of municipal solid waste in a cement kiln may influence the congener profile of PBDD/Fs, especially for the higher halogenated PBDD fraction. In addition, there were significant correlations between the decabromodiphenyl ether and heptabrominated furan concentrations, which is an indicator of transformation from polybrominated diphenyl ethers to PBDD/Fs. Theoretical calculations were performed and demonstrated that elimination of HBr and Br_2 from polybrominated diphenyl ethers were the dominant formation pathways for PBDD/Fs. These pathways differed from that for polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs). 展开更多

Deformation characteristics and residual curvature during LP plate leveling process were investigated based on curva ture integral by elastic plastic differences. It was found that entry intermesh section is reduced with increasing thick- ness and entry intermesh should be from 0.7 to 1.1 mm for LP plate with 42/54 mm in thickness. In order to get satisfactory flatness along longitudinal direction, small entry intermesh should be selected and exit intermesh should be 0 ram. In addition, thickness section for I.P plate which can be leveled by 11 roller leveler is from 24 to 60 mm. Finally, there is an intermesh schedule that could level LP plate with initial curvature less than elastic limit curvature. 展开更多

AIM:To elucidate the effect of rapamycin on regulating the production of interleukin(IL)-1β in Aspergillus fumigatus(A.fumigatus)-induced keratitis and to verify whether the expression of IL-1β in A.fumigatus keratitis is associated with the mammalian target of rapamycin(mT OR)/Toll-like receptor 4(TLR4) signaling pathway.METHODS:Fungal keratitis mouse models of susceptible C57 BL/6 mice were established using A.fumigatus.The mice were subsequently treated with rapamycin.The protein levels of p-mT OR,TLR4,and IL-1β in normal and infected corneal tissue were measured by Western blot.The TLR4 and IL-1β m RNA levels were determined by real-time polymerase chain reaction(PCR).RESULTS:In C57 BL/6 mice,rapamycin treatment decreased the clinical scores and production of the pro-inflammatory cytokine,IL-1β.The expression of TLR4,stimulated by A.fumigatus,was reduced as well when the mT OR signaling pathway was suppressed by rapamycin.CONCLUSION:Rapamycin is beneficial for the outcome of fungal keratitis and has an inhibitory effect expression of the inflammatory cytokine IL-1β.The inhibitory effect on IL-1β expression can be associated with the mT OR/TLR4 signaling pathway in A.fumigatus infection in mice. 展开更多

The expression of dectin-1 mRNA was measured by real-time qPCR at the stimulation of 0,4,8 and 24h separately. Dectin-1 protein was detected by immunocytochemistry at 0 and 24h separately. ·RESULTS: Dectin-1 expressed in corneal epithelium of normal persons and FK patients. Vitro cellular experiment showed that the expression of dectin-1 mRNA in HCE cells began to increase after stimulation of AF antigens at 4h,and dectin-1 protein expression increased after stimulation at 24h. · CONCLUSION: Dectin-1 expressed in corneal epithelium of normal persons. Innate immunity,particularly macrophages,is critical for intestinal homeostasis.Sulforaphane,a dietary isothiocyanate from cruciferous vegetables,has been reported to protect against intestinal inflammation.However,the role of macrophages in sulforaphane mediated intestinal inflammation and the underlying molecular mechanisms have not been studied yet.In this study,sulforaphane effectively attenuated dextran sodium sulphate(DSS)induced intestinal inflammation in murine model.Of note,sulforaphane skewed the switching from classically(M1)to alternatively(M2)activated phenotype both in intestinal and bone marrow-derived macrophages(BMDMs).The expression levels of M1 associated maker genes induced by DSS or lipopolysaccharide(LPS)plus interferon gamma-γ(IFN-γ)were suppressed by sulforaphane while M2 marker gene expression levels were improved.This resulted in alteration of inflammatory mediators,particularly interleukin-10(IL-10),both in colon tissues and culture medium of BMDMs.Subsequently,IL-10 was found to mediate the sulforaphane induced M2 phenotype switching of BMDMs through the activation of STAT3 signaling.This was confirmed by immunofluorescence analysis with increased number of p-STAT3-positive cells in the colon sections.Moreover,anti-IL-10 neutralizing antibody significantly interfered M2 phenotyping of BMDMs induced by sulforaphane with reduced STAT3 phosphorylation.Findings here introduced a potential utilization of sulforaphane for intestinal inflammation treatment with macrophages as the therapeutic targets. 展开更多

AIM: To investigate the expression of interleukin(IL)-33 in the cornea and human corneal epithelial cells(HCECs) exposed to Aspergillus fumigatus(A. fumigatus), and to determine the function of IL-33/ST2/p38 signaling pathway in the immune response of corneal epithelial cells to A. fumigatus infection.METHODS: The mRNA and protein expression of IL-33 in HCECs and mice corneas were examined by quantitative real-time reverse transcription polymerase chain reaction(q RT-PCR) and Western blot analysis, respectively. IL-33 expression was also detected in cornea samples from healthy donors and patients with fungal keratitis with immunohistochemistry. The cultured HCECs were treated with inactive A. fumigatus hyphae at various concentrations with or without recombinant human IL-33 protein, soluble recombinant ST2 protein, specific ST2 neutralizing antibody, or the mitogen-activated protein kinase(MAPK) p38 inhibitor SB203580 for evaluation of the expression and activation of IL-33/ST2/p38 signaling in the regulation of proinflammatory cytokines. The production levels of IL-6 and IL-1β were determined by qR T-PCR and enzymelinked immunosorbent assay(ELISA). The proliferation of HCECs was determined by a Cell Counting Kit-8(CCK8) assay and cell count.RESULTS: IL-33 expression levels increased in the corneal tissues of patients with fungal keratitis and in mice corneas of experimental A. fumigatus infection,as well as in HCECs with infection of A. fumigatus. A. fumigatus strongly stimulated HCECs-generated proinflammatory cytokine(IL-6 and IL-1β) production at both the mRNA and protein levels. This production of proinflammatory mediators stimulated by A. fumigatus was further stimulated by IL-33 and was prevented by soluble ST2 protein or ST2 neutralizing antibody. Moreover, IL-33 naturally promoted the p38 phosphorylation induced by A. fumigatus, which was suppressed by soluble ST2 protein. The MAPK p38 inhibitor SB203580 also inhibited the A. fumigatus-induced proinflammatory cytokine production. IL-33 administration for 48 h and 72 h promoted the proliferation of HCECs, which was attenuated by treatment with soluble recombinant human ST2 protein.CONCLUSION: A. fumigatus elevates IL-33 expression in human and mice corneas and HCECs. Thus, IL-33/ST2/p38 signaling may play an important role in amplifying the immune response of corneal epithelial cells to A. fumigatus infection. Besides, IL-33 promotes the cell proliferation of HCECs via its receptor ST2. These findings suggest a novel autocrine mechanism of amplification of the fungalinduced inflammatory response in the corneal epithelium, highlighting a potential therapeutic target for fungal keratitis. 展开更多

Effects of four different drying methods on the colour, texture, sensory quality, microstructure, bacterial viability and storage stability of probiotic-enriched apple snacks were assessed. The drying methods were air drying (AD), freeze drying (FD), freeze drying followed by microwave vacuum drying (FD+MVD) and air drying followed by explosion puffing drying (AD+EPD). Overall, FD+MVD can be used as a suitable drying method for the development of probiotic enriched apple snacks in consideration of colour, texture, sensory quality, bacterial viability and storage stability. Probiotic bacteria in FD+MVD-dried samples remained above 1×106 CFU g 1 for 120 days at 25℃C. Interestingly, bacterial viability in FD+MVD-dried samples turned out to be significantly higher than FD-dried samples during storage for 120 days. 展开更多

AIM:To investigate the expression of dendritic cell-associated C-type lectin-1 (dectin-1) at the early period of Aspergillus fumigatus infection in rat’s corneal epithelium. ·METHODS:A total of 72 Wistar rats were randomly divided into three groups:A, B and C. The right eyes were chosen as experimental eyes. Group A was control group. Rats in group B were not inoculated with Aspergillus fumigatus. Group C was taken as Aspergillus fumigatus keratitis model. Rats in group B and C (six from each group) were executed randomly at 4, 8, 16 and 24 hours after experimental model being established to assess the expression of dectin-1 mRNA through real-time PCR. Another six rats in group B and C were executed randomly at 24 hours to assess the expression of dectin-1 protein through immunohistochemistry. ·RESULTS:The results of real-time PCR indicated that dectin-1 mRNA expression was low in corneal epithelium of normal rats’. There was no significantly difference of dectin-1 mRNA expression in group A and B (P 】0.05). The expression of Aspergillus fumigatus infected corneal epithelium increased gradually after 8 hours in group C. The synchronous expression of group A and C had significant difference (P 【0.01). Immunohistochemisty discovered that dectin-1 receptor existed in normal rat’s corneal epithelium . Dectin-1 protein increased after 24 hours in group C. There was a significant difference of synchronous expression in group B and C(P【0.01). · CONCLUSION:Dectin-1 exists in rat’s cornealepithelium and its expression significantly increases at the early period of Aspergillus fumigatus infection. Dectin-1 is a pattern recognition receptor that expresses in corneal epithelium and involves in immune response to Aspergillus fungal keratitis. 展开更多

化学防治是“预防为主,综合防治”植保方针的重要组成部分。经过几十年的发展,我国农药使用技术从原始粗放发展到现代智能,经历了手动工具、背负手动、背负机动、自走机动、精准智能的“四级跳”转变,实现了“人背机器”向“机器背人”... 化学防治是“预防为主,综合防治”植保方针的重要组成部分。经过几十年的发展,我国农药使用技术从原始粗放发展到现代智能,经历了手动工具、背负手动、背负机动、自走机动、精准智能的“四级跳”转变,实现了“人背机器”向“机器背人”“人机分离”的演变。近十年来,我国植保无人飞机低容量喷雾技术发展迅速,植保无人飞机保有量从695架增至25.1万架,年作业面积由9万hm^(2)次提升至1.78亿hm^(2)次,直接推动智慧植保的创新发展,无人化智慧植保施药技术体系正逐渐替代传统农药使用方式。智慧植保施药技术以“感知决策执行”为核心框架,融合卫星遥感、无人机近地感知及物联网传感等多源信息,结合边缘计算设备实现高效监管处理;基于决策支持系统、强化学习与知识图谱生成精准方案;依托变量施药处方图技术、机器视觉对靶喷雾及雾滴飘移控制技术,实现农药减量的同时保障病虫害防治效果。未来,人工智能大模型将整合文本、图像与传感数据为植保施药提供智能决策;多智能体协同系统集合无人机集群作业实现植保全流程自主化,并最终构建“感知决策执行优化”闭环管理,为农药减量化与粮油作物单产提升提供可持续技术支撑。 展开更多

AIM:To determine the roles of high-mobility group box1(HMGB1)in pro-inflammation,host immune response and its potential target for treatment in Aspergillus fumigatus(A.fumigatus)keratitis.METHODS:Expression of HMGB1 was tested in C57 BL/6 normal and infected corneas.Dual immunostaining tested coexpression of HMGB1 with TLR4 or LOX-1.C57 BL/6 mice were pretreated with Box A or PBS and then infected.Clinical scores,polymerase chain reaction,ELISA,and MPO assay were used to assess the disease response.Flow cytometry were used to test the effect of Box A on reactive oxygen species(ROS)expression after A.fumigatus stimulation in polymorphonuclear neutrophilic leukocytes(PMN).C57 BL/6 peritoneal macrophages were pretreated with Box B before A.fumigatus stimulation,and MIP-2,IL-1β,TNF-α,HMGB1 and LOX-1 were measured.Macrophages were pretreated with Box B or Box B combined with Poly(I)(an inhibitor of LOX-1)before stimulating with A.fumigatus,and MIP-2,IL-1β,TNF-α,LOX-1,p38-MAPK,p-p38-MAPK were measured.RESULTS:HMGB1 levels were elevated in C57 BL/6 mice after infection.HMGB1 co-expressed with TLR4,and LOX-1 in infiltrated cells.Box A vs PBS treated C57 BL/6 mice had lower clinical scores and down-regulated corneal HMGB1,MIP-2,IL-1βexpression and neutrophil influx.Box B treatment amplified expression of MIP-2,IL-1β,TNF-α,HMGB1 and LOX-1 that induced by A.fumigatus in macrophage.Compared to the treatment of Box B only,the protein expression of IL-1β,TNF-αshowed inhibition of Box B combined with Poly(I),which also reduced the A.fumigatusevoked protein level of LOX-1 and phosphorylation level of p38-MAPK.The production of A.fumigatus-stimulated ROS was significantly declined after Box A pretreatment in PMN.CONCLUSION:Blocking HMGB1 reduces the disease response in C57 BL/6 mice.HMGB1 can amplify the host immune response through p38-MAPK,and is a target for treatment of A.fumigatus keratitis. 展开更多

The effects of fermenting, boiling, and steaming processes on the levels of free amino acids, organic acids, antioxidant properties (FRAP and DPPH), phenolic compounds, and angiotensine converting enzyme (ACE) inhibition of sweet potato tips were investigated. Seventeen free amino acids were found in the tips. Fermenting, boiling, and steaming processes significantly reduced total free amino acid contents and steaming processes caused smaller losses than fermenting and boiling processes. Five organic acids were investigated in the tips. In comparison with the total organic acid content of unprocessed sweet potato tips, steaming and boiling processes significantly reduced it, while fermenting process increased it. Chlorogenic acid, rutin, and quercetin were investigated in the tips. The total phenolic compounds contents of sweet potato tips significantly increased by steaming and boiling processes, but a substantial loss was caused by fermenting process. A similar result was shown in FRAP value. The result of ACE inhibitory activity indicated that boiling, steaming, and fermenting processes significantly increased ACE inhibitory activity. 展开更多

The effects of the plasticizer poly(ethylene glycol)(PEG)on crystallization properties of equimolar poly(L-lactide)(PLLA)/poly(D-lactide)(PDLA)blends were investigated.Forma-tion of the stereocomplex-type poly(lactide acid)(sc-PLA)crystallites was confirmed by Wide-angle X-ray diffraction(WAXD)and differential scanning calorimetry(DSC)analyses.Sc-PLA crystallites without any homochiral poly(lactide acid)(hc-PLA)formed,as the result of the incorporation of the plasticizer PEG(more than or equal to 10%(wt))at a processing temperature(240℃).More-over,when the Mw of PEG reached 1000 g·mol^(-1),the crystal-lizability of stereocomplex crystallites was the best.Isothermal crystallization kinetics further revealed that PEG could accelerate the crystallization rate of sc-PLA,with the optimum crystallization kinetic parameters being obtained at 10%(wt)PEG.Several crys-tallization kinetics equations were applied to describe the effect of PEG on the crystallization behavior of sc-PLA.The influence of PEG on the spherocrystal morphologies of sc-PLA was also inves-tigated using polarized optical microscopy. 展开更多

METHODS: The corneas of C57BL/6 mice were pretreated with LOX-1 inhibitor Poly(I) or Dectin-1 siRNA separately before the infection of A. fumigatus. Polymerase chain reaction (PCR) and Western blot were used to detect the expression of IL-10. RESULTS: The mRNA and protein expressions of IL-10 were significantly increased in mice with A. fumigatus keratitis. Compared with the group pretreated with sterile water before infection, Poly(I) pretreatment suppressed IL-10 expression significantly. Compared with the group pretreated with scrambled siRNA before infection, Dectin-1 siRNA pretreatment significantly reduced IL-10 expression in response to A. fumigatus infection. CONCLUSION: LOX-1 and Dectin-1 regulate IL-10 production in mouse A. fumigatus keratitis. 展开更多

AIM: To investigate whether high-mobility group box 1(HMGB1) Boxb exacerbates BALB/c mice corneal immune responses and inflammatory through the Toll-like receptor 4(TLR4)/myeloid differentiation primary response 88(My D88)-dependent signaling pathway in Aspergillus fumigatus(A. fumigatus) keratitis.METHODS: The mice corneas were pretreated with phosphate buffer saline(PBS), Boxb before A. fumigatus infection. The abdominal cavity extracted macrophages were pretreated with PBS, Boxb, TLR4 inhibitor(CLI-095), Dimethyl sulfoxide(DMSO) separately before A. fumigatus hyphae stimulation. HMGB1 was detected in normal and infected mice corneas and macrophages by real-time reverse transcriptase polymerase chain reaction(RT-PCR), the TLR4, My D88, interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) were detected by Western blot and PCR.RESULTS: In BALB/c mice corneas, the expressions of TLR4, HMGB1, IL-1β, TNF-α were increased after A. fumigatus infection. While pretreatment with Boxb significantly increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α compared with PBS control after infection. In BALB/c mice abdominal cavity extracted macrophages, pretreatment with Boxb increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α, while pretreatment with CLI-095 and Boxb significantly decreased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α. CONCLUSION: In A. fumigatus keratitis, Boxb play a proinflammatory role in corneal anti-fungi immune response through the HMGB1-TLR4-My D88 signal pathway. 展开更多

AIM: To explore whether nicotinamide adenine dinucleotide phosphate(NADPH) oxidase 2(NOX2) is expressed in fungal keratitis in mice and investigate its role in this disease.METHODS: NOX2 expression was detected in C57 BL/6 mice. After testing the inhibitory effect of diphenyleneiodonium chloride(DPI) on NOX2, its impact on clinical performance, myeloperoxidase levels, the number of colonies forming units, the level of H3, the generation of reactive oxygen species(ROS) and the release of cytokines [NF-κB, interleukin-17 A(IL-17 A), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α), Nrf2, IL-10, and TGF-β] were compared. A one-way ANOVA and an unpaired, two-tailed Student’s t-test was used to determine the statistical significance.RESULTS: NOX2 expression was significantly increased after Aspergillus fumigatus injection in corneas and that this increase could be reduced by treatment with DPI. DPI treatment produced more severe inflammation and resulted in higher clinical scores, more neutrophils infiltration, a weakened ability to clear fungi, the release of fewer ROS and the formation of neutrophil extracellular traps. Treatment with DPI increased the expression of the proinflammatory cytokines NF-κB, IL-17 A, IL-6, and TNF-α and decreased the expression of the anti-inflammatory cytokines Nrf2, IL-10 and TGF-β compared to their expression levels without DPI treatment.CONCLUSION: NOX2 plays an important role against Aspergillus fumigatus in the mouse cornea through killing fungi and limiting the degree of inflammation. 展开更多
