METHODS All clinical data of ...AIM To investigate the long-term prognosis in peptic ulcer patients continuing taking antithrombotics after ulcer bleeding, and to determine the risk factors that influence the prognosis. METHODS All clinical data of peptic ulcer patients treated from January 1, 2009 to January 1, 2014 were retrospectively collected and analyzed. Patients were divided into either a continuing group to continue taking antithrombotic drugs after ulcer bleeding or a discontinuing group to discontinue antithrombotic drugs. The primary outcome of follow-up in peptic ulcer bleeding patients was recurrent bleeding, and secondary outcome was death or acute cardiovascular disease occurrence. The final date of follow-up was December 31, 2014. Basic demographic data, complications, and disease classifications were analyzed and compared by t- or χ2-test. The number of patients that achieved various outcomes was counted and analyzed statistically. A survival curve was drawn using the Kaplan-Meier method, and the differencewas compared using the log-rank test. COX regression multivariate analysis was applied to analyze risk factors for the prognosis of peptic ulcer patients. RESULTS A total of 167 patients were enrolled into this study. As for the baseline information, differences in age, smoking, alcohol abuse, and acute cardiovascular diseases were statistically significant between the continuing and discontinuing groups(70.8 ± 11.4 vs 62.4 ± 12.0, P < 0.001; 8(8.2%) vs 15(21.7%), P < 0.05; 65(66.3%) vs 13(18.8%), P < 0.001). At the end of the study, 18 patients had recurrent bleeding and three patients died or had acute cardiovascular disease in the continuing group, while four patients had recurrent bleeding and 15 patients died or had acute cardiovascular disease in the discontinuing group. The differences in these results were statistically significant(P = 0.022, P = 0.000). The Kaplan-Meier survival curve indicated that the incidence of recurrent bleeding was higher in patients in the continuing group, and the risk of death and developing acute cardiovascular disease was higher in patients in the discontinuing group(log-rank test, P = 0.000 for both). Furthermore, COX regression multivariate analysis revealed that the hazard ratio(HR) for recurrent bleeding was 2.986(95%CI: 067-8.356, P = 0.015) in the continuing group, while HR for death or acute cardiovascular disease was 5.216(95%CI: 1.035-26.278, P = 0.028).CONCLUSION After the occurrence of peptic ulcer bleeding, continuing antithrombotics increases the risk of recurrent bleeding events, while discontinuing antithrombotics would increase the risk of death and developing cardiovascular disease. This suggests that clinicians should comprehensively consider the use of antithrombotics after peptic ulcer bleeding.展开更多

Objective:To explore the effects of aerobic exercise on the levels of inflammatory factors in patients with type 2 diabetes.Methods:Randomized controlled studies(RCTs)on the effects of aerobic exercise on inflammatory...Objective:To explore the effects of aerobic exercise on the levels of inflammatory factors in patients with type 2 diabetes.Methods:Randomized controlled studies(RCTs)on the effects of aerobic exercise on inflammatory factors in patients with type 2 diabetes were searched from electronic databases.Two researchers independently screened the literatures for data extraction and quality assessment.Meta-analysis was performed using RevMan 5.4 software.Results:A total of 17 literatures were included,including 1186 patients.The results of Meta-analysis showed that aerobic exercise significantly reduced the plasma levels of CRP,TNF-α,IL-6,and leptin in T2DM patients,but did not significantly reduce the plasma concentration of adiponectin.However,the left and right asymmetry of the"inverted funnel"plot indicated that there was a certain publication bias in the included studies.Conclusion:Existing evidence shows that aerobic exercise has a good effect on the improvement of chronic inflammatory state in patients with type 2 diabetes mellitus,suggesting that lifestyle changes can play a good role in adjunctive therapy.展开更多
BACKGROUND Traumatic injuries,such as falling,car accidents,and crushing mostly cause spinal fractures in young and middle-aged people,and>50%of them are thoracolumbar fractures.This kind of fracture is easily comb...BACKGROUND Traumatic injuries,such as falling,car accidents,and crushing mostly cause spinal fractures in young and middle-aged people,and>50%of them are thoracolumbar fractures.This kind of fracture is easily combined with serious injuries to peripheral nerves and soft tissues,which causes paralysis of the lower limbs if there is no timely rehabilitation treatment.Young patients with thoracolumbar fractures find it difficult to recover after the operation,and they are prone to depression,low self-esteem,and other negative emotions.AIM To investigate the association between anxiety,depression,and social stress in young patients with thoracolumbar spine fractures and the effect on rehabilitation outcomes.METHODS This study retrospectively analyzed 100 patients admitted to the orthopedic department of Honghui Hospital,Xi’an Jiaotong University who underwent thoracolumbar spine fracture surgery from January 2022 to June 2023.The general data of the patients were assessed with the Hamilton anxiety scale(HAMA),Hamilton depression scale(HAMD),life events scale,and social support rating scale(SSRS)to identify the correlation between anxiety,depression scores,and social stress and social support.The Japanese Orthopedic Association(JOA)was utilized to evaluate the rehabilitation outcomes of the patients and to analyze the effects of anxiety and depression scores on rehabilitation.RESULTS According to the scores of HAMD and HAMA in all patients,the prevalence of depression in patients was 39%(39/100),and the prevalence of anxiety was 49%(49/100).Patients were categorized into non-depression(n=61)and depression(n=39),non-anxiety(n=51),and anxiety(n=49)groups.Statistically significant differences in gender,occupation,Pittsburgh Sleep Quality Index(PSQI)score,and monthly family income were observed between the non-depression and depression groups(P<0.05).A significant difference in occupation and PSQI score was found between the non-anxiety and anxiety groups.Both depression(r=0.207,P=0.038)and anxiety scores(r=0.473,P<0.001)were significantly and positively correlated with negative life events.The difference in negative life event scores as well as SSRS total and item scores was statist-ically significant between patients in the non-depression and depression groups(P<0.05).The difference between the non-anxiety and anxiety groups was statistically significant(P<0.05)in the negative life event scores as well as the total SSRS scores.Additionally,JOA scores were significantly lower in both anxious and depressed patients.CONCLUSION Young patients with thoracolumbar fractures are prone to anxiety and depression.Patients’anxiety and depression are closely associated with social pressure,which reduces the life pressure of young patients with thoracolumbar fractures,enhances social support,and improves the psychology of anxiety and depression.,which affects patients’recovery.展开更多
Epidemiological studies have indicated that branched-chain amino acids(BCAAs)increased and gut microbiota disordered in type 2 diabetes mellitus(T2DM).This study aimed to investigate the mechanism of Lactiplantibacill...Epidemiological studies have indicated that branched-chain amino acids(BCAAs)increased and gut microbiota disordered in type 2 diabetes mellitus(T2DM).This study aimed to investigate the mechanism of Lactiplantibacillus plantarum strain 84-3(Lp84-3)combined with Staphylococcus aureus bacteriophage on ameliorating T2DM.Here we perform a case-control study and identify that Staphylococcus_phage was inversely correlated with fasting blood glucose(FBG).It revealed that Lp84-3 could inhibit the growth of S.aureus,and Lp84-3 contains BCAAs degradation enzymes in its genome.Furthermore,Lp84-3 alone or combined with S.aureus bacteriophage interventions can improve blood glucose,insulin resistance,triglycerides,interleukin-1β,tumor necrosis factor-α(TNF-α),BCAAs,and acetyllactate synthase(ALS)in db/db mice.Lp84-3 and S.aureus bacteriophage decreased S.aureus,Malacoplasma iowae,and Oscillibacter sp.,and increased some beneficial such as L.plantarum and Muribaculaceae bacterium.Transcriptomic analyses revealed that Lp84-3 and S.aureus bacteriophage activated the PI3K/AKT/GLUT4 signaling pathway and upregulated key genes of Il22,Hgf,Col6a1,Gh,Itga10,Fgf23,and Prl involved in glucose metabolism in hypothalamus.Collectively,Lp84-3 and S.aureus bacteriophage alleviate T2DM by modulating gut microbiota and enhancing glucose metabolism in hypothalamus,supporting its potential use as a promising functional compound microecological agent for alleviating T2DM.展开更多
Autophagy directly regulates the amyloid beta-peptide(Aβ)clearance,and its dysfunction occurs in the early pathogenesis of Alzheimer's disease(AD).We previously reported that docosahexaenoic acid-acylated astaxan...Autophagy directly regulates the amyloid beta-peptide(Aβ)clearance,and its dysfunction occurs in the early pathogenesis of Alzheimer's disease(AD).We previously reported that docosahexaenoic acid-acylated astaxanthin monoester(AST-DHA)showed neuroprotection against AD pathology.However,its in-depth mechanism and autophagic responses in AD brains are poorly understood.Herein,SH-SY5Y cells overexpressing the Swedish mutation(K595N/M596L)of APP gene were established to preliminarily evaluate the actions of AST-DHA on reducing Aβ_(1-42)levels and regulating autophagy.In microglial BV2 cells,AST-DHA and free astaxanthin(F-AST)recovered p62 and LC3Ⅱ/Ⅰlevels,and restored autophagy flux by rescuing the late phase of microglial autophagy.Notably,autophagic inhibitor bafilomycin A1 blunted the abilities of AST-DHA to reduce Aβ_(1-42)and fibral Aβ,suggesting that AST-DHA probably promoted Aβclearance in a microglial autophagy-dependent manner.Further studies in APP/PS1 mice verified that dietary AST-DHA and F-AST promoted Aβphagocytosis via microglial autophagy.Significant decreases of Iba1 and p62 were observed around Aβplaque in the hippocampus and cortex using triple fluorescence staining.Furthermore,AST-DHA exhibited superior performance over F-AST in restoring autophagic dysfunction,ameliorating Aβburden and cognitive deficit.Our findings suggest a possible mechanism of AST-DHA in improving AD by which it restores microglial autophagy to facilitate cerebral Aβclearance.It supports the future application of AST-DHA as an autophagic regulator in maintaining brain function.展开更多
Biological load-bearing materials,like the nacre in shells,have a unique staggered structure that supports their superior mechanical properties.Engineers have been encouraged to imitate it to create load-bearing bio-i...Biological load-bearing materials,like the nacre in shells,have a unique staggered structure that supports their superior mechanical properties.Engineers have been encouraged to imitate it to create load-bearing bio-inspired materials which have excellent properties not present in conventional composites.To create such materials with desirable mechanical properties,the optimum structural parameters combination must be selected.Moreover,the optimal design of bio-inspired composites needs to take into account the trade-offs between various mechanical properties.In this paper,multi-objective optimization models were developed using structural parameters as design variables and mechanical properties as optimization objectives,including stiffness,strength,toughness,and dynamic damping.Using the NSGA-II optimization algorithm,a set of optimal solutions were solved.Additionally,three different structures in natural nacre were introduced in order to utilize the better structure when design bio-inspired materials.The range of optimal solutions that obtained using results from previous research were examined and explained why this collection of optimal solution ranges is better.Also,optimal solutions were compared with the structural features and mechanical properties of real nacre and artificial biomimetic composites to validate our models.Finally,the optimum design strategies can be obtained for nacre-like composites.Our research methodically proposes an optimization method for achieving load-bearing bio-inspired materials with excellent properties and creates a set of optimal solutions from which designers can select the one that best suits their preferences,allowing the fabricated materials to demonstrate preferred performance.展开更多

METHODS: Hematological changes and changes of lymphocyte subpopulations were detected in Smad3"/- mice using cell counter and flow cytometry, respectively, and compared to their littermate controls. RESULTS: The numbers of neutrophils and lymphocytes in peripheral blood were significantly increased in Smad3^-/- mice compared to littermate controls. CD19^+ expressing cells in blood and spleen, and CD8^+ T cells in thymus were all markedly decreased in Smad3^-/- mice. More important, Smad3^-/- mice had an increased population of CD4^+CD25^+ T cells in peripheral lymphoid tissues, including thymus, spleen, and lymph nodes. CONCLUSION: These observations suggest that the changes of lymphocyte subpopulations might play a role in susceptibility to inflammation of Smad3^-/- mice.展开更多

In this study, we applied structured illumination microscopy(SIM) to capture the distribution of Cy5-Dextran at different incubation periods in living cells. The results showed that Cy5-Dextran could be absorbed by He La cells. In addition, we clarified that Cy5-Dextran exhibited differential organelle distribution(lysosomal or mitochondrial) in a time-dependent manner. Moreover,lysosomal Cy5-Dextran localization was found to be independent of the autophagy process, while Cy5-Dextran localized to the mitochondria triggered a pro-apoptotic event, upregulating the levels of reactive oxygen species(ROS) to accelerate mitochondrial fragmentation. This work uses a visualized strategy to reveal the anti-tumor bioactivity of dextran, which was achieved by regulating apoptosis and autophagy.展开更多

Measured by single crystal X-ray crystallography, complexes Col and Co3 adopted distorted tetrahedral structures around the cobalt center. Upon activation by ethylaluminium sesquichloride (EASC), these cobalt complexes exhibited high catalytic activity and cis-l,4-selectivity towards 1,3-butadiene polymerization. The effects of ligand environment, polymerization temperature, and cocatalyst types on the polymerization were investigated in detail. Interestingly, the binuclear Co(Ⅱ) complexes exhibited high thermal stability, and the polymer yields were up to 97.2% even at a high temperature of 70 ℃.展开更多
目的探讨感染性休克患者血清肽酰基精氨酸脱亚胺酶2(peptidylarginine deiminase type 2,PAD2)表达水平与急性生理学和慢性健康状况评价Ⅱ(acute physiology and chronic health evaluationⅡ,APACHEⅡ)评分的相关性。方法选取内江市第...目的探讨感染性休克患者血清肽酰基精氨酸脱亚胺酶2(peptidylarginine deiminase type 2,PAD2)表达水平与急性生理学和慢性健康状况评价Ⅱ(acute physiology and chronic health evaluationⅡ,APACHEⅡ)评分的相关性。方法选取内江市第一人民医院2020年6月~2022年6月收治的103例感染性休克患者作为研究组,采用APACHEⅡ评分根据患者病情严重程度将其分为轻度组(n=9)、中度组(n=51)和重度组(n=13),另外选取103例同期在该院体检且一般资料与研究组患者相匹配的健康者作为对照组。采用酶联免疫吸附法测定感染性休克患者血清PAD2表达水平;采用Spearman法分析感染性休克患者血清PAD2表达水平与APACHEⅡ评分的相关性;采用Logistic回归分析影响感染性休克患者病情严重程度的相关因素;采用受试者工作特征(receiver operating characteristic,ROC)曲线分析血清PAD2对中重度感染性休克的诊断价值。结果研究组与对照组血肌酐(137.52±9.01μmol/L vs 112.22±8.67μmol/L)水平及血小板计数(74.58±5.19 vs 86.02±5.34)×109/L比较,差异具有统计学意义(t=20.535,15.591,均P<0.05);研究组患者血清PAD2表达水平(42.47±6.22 ng/ml)高于对照组(38.59±5.31 ng/ml),差异具有统计学意义(t=4.815,P<0.05);感染性休克患者血清PAD2表达水平和APACHEⅡ评分均随病情严重程度的增加而逐渐升高(F=3.777,176.582,均P<0.05);感染性休克患者血清PAD2表达水平与APACHEⅡ评分呈正相关(r=0.859,P<0.05);血肌酐(OR=1.927)、PAD2(OR=1.803)及APACHEⅡ评分(OR=1.657)均为发生中重度感染性休克的危险因素(均P<0.05),血小板计数(OR=0.781)则是发生中重度感染性休克的保护因素(P<0.05)。血清PAD2诊断中重度感染性休克的曲线下面积(area under the curve,AUC)为0.880,敏感度、特异度分别为75.73%(95%CI:0.701~0.826)和90.29%(95%CI:0.851~0.935),对中重度感染性休克具有较高的诊断价值。结论血清PAD2表达水平与APACHEⅡ评分呈正相关,且对中重度感染性休克具有较好诊断价值。展开更多
Objective:This trial is aimed at evaluating the efficacy and safety of acupuncture on treating FC.Methods:We describe the protocol for a randomized,patient-assessor-blinded,sham controlled trial.Seventy-two eligible p...Objective:This trial is aimed at evaluating the efficacy and safety of acupuncture on treating FC.Methods:We describe the protocol for a randomized,patient-assessor-blinded,sham controlled trial.Seventy-two eligible patients will be randomly assigned to the intervention group(acupuncture)or the control group(sham acupuncture).All treatment will be given 26 sessions of acupuncture or sham acupuncture over 8 weeks(5 times per week in the first 2 weeks,3 times per week during weeks 3-6,and 2 times per week during weeks 7 and 8).Each treatment will last for 20 min.The primary outcome is the change in mean complete spontaneous bowel movements(CSBMs)per week.The secondary out?comes are patient assessment of constipation quality of life questionnaire(PAC-QOL),self-rating anxiety scale(SAS),and the dosage of the medication.All adverse events will be recorded in detail and managed by corresponding researchers as quickly as possible.Outcomes will be evaluated at baseline(1 week before treatment),2 weeks after intervention begins,6 weeks after intervention begins,8 weeks after intervention begins,4 weeks follow-up,and 12 weeks follow-up.Discussion:The results of this study will provide the evidence of the efficacy and safety of acupuncture as a traditional treatment methods for functional constipation.Trial registration:Chinese Clinical Trial Registry,ChiCTR-INR-17011472.Registered on 23 May 2017.展开更多
The motivation for this study is that the quality of deep fakes is constantly improving,which leads to the need to develop new methods for their detection.The proposed Customized Convolutional Neural Network method in...The motivation for this study is that the quality of deep fakes is constantly improving,which leads to the need to develop new methods for their detection.The proposed Customized Convolutional Neural Network method involves extracting structured data from video frames using facial landmark detection,which is then used as input to the CNN.The customized Convolutional Neural Network method is the date augmented-based CNN model to generate‘fake data’or‘fake images’.This study was carried out using Python and its libraries.We used 242 films from the dataset gathered by the Deep Fake Detection Challenge,of which 199 were made up and the remaining 53 were real.Ten seconds were allotted for each video.There were 318 videos used in all,199 of which were fake and 119 of which were real.Our proposedmethod achieved a testing accuracy of 91.47%,loss of 0.342,and AUC score of 0.92,outperforming two alternative approaches,CNN and MLP-CNN.Furthermore,our method succeeded in greater accuracy than contemporary models such as XceptionNet,Meso-4,EfficientNet-BO,MesoInception-4,VGG-16,and DST-Net.The novelty of this investigation is the development of a new Convolutional Neural Network(CNN)learning model that can accurately detect deep fake face photos.展开更多

文摘This paper employs the theories and methods of music anthropology to conduct a deep analysis of the newly composed Guangdong Han Opera“Tianfeng Haiyu Meihuadu.”By exploring its musical forms,performance rituals,and the cultural connotations of the Hakka people,it reveals the unique cultural memory embedded in the musical aspects of the opera.The study finds that“Tianfeng Haiyu Meihuadu”is not only a representative work of the newly composed Guangdong Han Opera but also an important carrier of Hakka cultural inheritance and dissemination.Its music and performance are closely linked to the history,emotions,and social life of the Hakka people,providing a vivid example for understanding the characteristics of Hakka culture and the functions of musical culture.

文摘Guangdong Han Opera,known as the“Peony of the South,”is a treasured art form with a history of over 300 years.As one of the three major opera genres in Guangdong,it primarily uses Pi and Huang tuning,incorporating Kunqu,Gaoqiang,Chuiqiang,and minor tunes,with Zhongzhou rhyme as its stage language.The roles are divided into seven categories:Sheng,Dan,Chou,Gong,Po,and Jing(including Hongjing and Wujing).Its accompaniment instruments,particularly the Touxian,Dasuluo,and Haotou,are distinctive,making it an excellent Pi and Huang opera genre in southern China.In 2008,Guangdong Han Opera was listed in the national intangible cultural heritage,recognizing its artistic value and entrusting it with the mission of inheriting and developing this opera genre.This paper focuses on the Guangdong Han Opera“Wang Zhaojun”and employs methods such as literature review and case analysis to explore its achievements in secondary creation and artistic innovation.The opera has achieved innovations in script,character,music,and stagecraft,and made breakthroughs in performance form,narrative technique,and cultural connotation,promoting the inheritance and development of Guangdong Han Opera and providing insights into contemporary drama innovation.

文摘AIM To investigate the long-term prognosis in peptic ulcer patients continuing taking antithrombotics after ulcer bleeding, and to determine the risk factors that influence the prognosis. METHODS All clinical data of peptic ulcer patients treated from January 1, 2009 to January 1, 2014 were retrospectively collected and analyzed. Patients were divided into either a continuing group to continue taking antithrombotic drugs after ulcer bleeding or a discontinuing group to discontinue antithrombotic drugs. The primary outcome of follow-up in peptic ulcer bleeding patients was recurrent bleeding, and secondary outcome was death or acute cardiovascular disease occurrence. The final date of follow-up was December 31, 2014. Basic demographic data, complications, and disease classifications were analyzed and compared by t- or χ2-test. The number of patients that achieved various outcomes was counted and analyzed statistically. A survival curve was drawn using the Kaplan-Meier method, and the differencewas compared using the log-rank test. COX regression multivariate analysis was applied to analyze risk factors for the prognosis of peptic ulcer patients. RESULTS A total of 167 patients were enrolled into this study. As for the baseline information, differences in age, smoking, alcohol abuse, and acute cardiovascular diseases were statistically significant between the continuing and discontinuing groups(70.8 ± 11.4 vs 62.4 ± 12.0, P < 0.001; 8(8.2%) vs 15(21.7%), P < 0.05; 65(66.3%) vs 13(18.8%), P < 0.001). At the end of the study, 18 patients had recurrent bleeding and three patients died or had acute cardiovascular disease in the continuing group, while four patients had recurrent bleeding and 15 patients died or had acute cardiovascular disease in the discontinuing group. The differences in these results were statistically significant(P = 0.022, P = 0.000). The Kaplan-Meier survival curve indicated that the incidence of recurrent bleeding was higher in patients in the continuing group, and the risk of death and developing acute cardiovascular disease was higher in patients in the discontinuing group(log-rank test, P = 0.000 for both). Furthermore, COX regression multivariate analysis revealed that the hazard ratio(HR) for recurrent bleeding was 2.986(95%CI: 067-8.356, P = 0.015) in the continuing group, while HR for death or acute cardiovascular disease was 5.216(95%CI: 1.035-26.278, P = 0.028).CONCLUSION After the occurrence of peptic ulcer bleeding, continuing antithrombotics increases the risk of recurrent bleeding events, while discontinuing antithrombotics would increase the risk of death and developing cardiovascular disease. This suggests that clinicians should comprehensively consider the use of antithrombotics after peptic ulcer bleeding.

文摘The demand for low-carbon MgO–C refractories is ever growing to meet the development of advanced steelmaking technologies and efficient energy conservation.Meanwhile,to improve the oxidation resistance and inhibit the weakness of low-carbon MgO–C refractories,antioxidants are necessary.The application of ternary carbides that focused on improving the oxidation performance of MgO–C refractories has been explored,and the ternary carbides including Al_(4)O_(4)C,Al8B_(4)C_(7),Al_(4)SiC_(4),Ti_(2)AlC,Ti_(3)AlC_(2),and Ti_(3)SiC_(2)have been proved effective.The crystal structure,physical properties,oxidation behavior,and synthesis of these ternary carbides were summarized,and their oxidation mechanism in assisting antioxidation of MgO–C refractories was discussed.In addition,the potential aspects related to the usage and development of ternary carbides in low-carbon MgO–C refractories were proposed.

文摘Objective:To explore the effects of aerobic exercise on the levels of inflammatory factors in patients with type 2 diabetes.Methods:Randomized controlled studies(RCTs)on the effects of aerobic exercise on inflammatory factors in patients with type 2 diabetes were searched from electronic databases.Two researchers independently screened the literatures for data extraction and quality assessment.Meta-analysis was performed using RevMan 5.4 software.Results:A total of 17 literatures were included,including 1186 patients.The results of Meta-analysis showed that aerobic exercise significantly reduced the plasma levels of CRP,TNF-α,IL-6,and leptin in T2DM patients,but did not significantly reduce the plasma concentration of adiponectin.However,the left and right asymmetry of the"inverted funnel"plot indicated that there was a certain publication bias in the included studies.Conclusion:Existing evidence shows that aerobic exercise has a good effect on the improvement of chronic inflammatory state in patients with type 2 diabetes mellitus,suggesting that lifestyle changes can play a good role in adjunctive therapy.
文摘BACKGROUND Traumatic injuries,such as falling,car accidents,and crushing mostly cause spinal fractures in young and middle-aged people,and>50%of them are thoracolumbar fractures.This kind of fracture is easily combined with serious injuries to peripheral nerves and soft tissues,which causes paralysis of the lower limbs if there is no timely rehabilitation treatment.Young patients with thoracolumbar fractures find it difficult to recover after the operation,and they are prone to depression,low self-esteem,and other negative emotions.AIM To investigate the association between anxiety,depression,and social stress in young patients with thoracolumbar spine fractures and the effect on rehabilitation outcomes.METHODS This study retrospectively analyzed 100 patients admitted to the orthopedic department of Honghui Hospital,Xi’an Jiaotong University who underwent thoracolumbar spine fracture surgery from January 2022 to June 2023.The general data of the patients were assessed with the Hamilton anxiety scale(HAMA),Hamilton depression scale(HAMD),life events scale,and social support rating scale(SSRS)to identify the correlation between anxiety,depression scores,and social stress and social support.The Japanese Orthopedic Association(JOA)was utilized to evaluate the rehabilitation outcomes of the patients and to analyze the effects of anxiety and depression scores on rehabilitation.RESULTS According to the scores of HAMD and HAMA in all patients,the prevalence of depression in patients was 39%(39/100),and the prevalence of anxiety was 49%(49/100).Patients were categorized into non-depression(n=61)and depression(n=39),non-anxiety(n=51),and anxiety(n=49)groups.Statistically significant differences in gender,occupation,Pittsburgh Sleep Quality Index(PSQI)score,and monthly family income were observed between the non-depression and depression groups(P<0.05).A significant difference in occupation and PSQI score was found between the non-anxiety and anxiety groups.Both depression(r=0.207,P=0.038)and anxiety scores(r=0.473,P<0.001)were significantly and positively correlated with negative life events.The difference in negative life event scores as well as SSRS total and item scores was statist-ically significant between patients in the non-depression and depression groups(P<0.05).The difference between the non-anxiety and anxiety groups was statistically significant(P<0.05)in the negative life event scores as well as the total SSRS scores.Additionally,JOA scores were significantly lower in both anxious and depressed patients.CONCLUSION Young patients with thoracolumbar fractures are prone to anxiety and depression.Patients’anxiety and depression are closely associated with social pressure,which reduces the life pressure of young patients with thoracolumbar fractures,enhances social support,and improves the psychology of anxiety and depression.,which affects patients’recovery.

文摘Epidemiological studies have indicated that branched-chain amino acids(BCAAs)increased and gut microbiota disordered in type 2 diabetes mellitus(T2DM).This study aimed to investigate the mechanism of Lactiplantibacillus plantarum strain 84-3(Lp84-3)combined with Staphylococcus aureus bacteriophage on ameliorating T2DM.Here we perform a case-control study and identify that Staphylococcus_phage was inversely correlated with fasting blood glucose(FBG).It revealed that Lp84-3 could inhibit the growth of S.aureus,and Lp84-3 contains BCAAs degradation enzymes in its genome.Furthermore,Lp84-3 alone or combined with S.aureus bacteriophage interventions can improve blood glucose,insulin resistance,triglycerides,interleukin-1β,tumor necrosis factor-α(TNF-α),BCAAs,and acetyllactate synthase(ALS)in db/db mice.Lp84-3 and S.aureus bacteriophage decreased S.aureus,Malacoplasma iowae,and Oscillibacter sp.,and increased some beneficial such as L.plantarum and Muribaculaceae bacterium.Transcriptomic analyses revealed that Lp84-3 and S.aureus bacteriophage activated the PI3K/AKT/GLUT4 signaling pathway and upregulated key genes of Il22,Hgf,Col6a1,Gh,Itga10,Fgf23,and Prl involved in glucose metabolism in hypothalamus.Collectively,Lp84-3 and S.aureus bacteriophage alleviate T2DM by modulating gut microbiota and enhancing glucose metabolism in hypothalamus,supporting its potential use as a promising functional compound microecological agent for alleviating T2DM.

文摘Autophagy directly regulates the amyloid beta-peptide(Aβ)clearance,and its dysfunction occurs in the early pathogenesis of Alzheimer's disease(AD).We previously reported that docosahexaenoic acid-acylated astaxanthin monoester(AST-DHA)showed neuroprotection against AD pathology.However,its in-depth mechanism and autophagic responses in AD brains are poorly understood.Herein,SH-SY5Y cells overexpressing the Swedish mutation(K595N/M596L)of APP gene were established to preliminarily evaluate the actions of AST-DHA on reducing Aβ_(1-42)levels and regulating autophagy.In microglial BV2 cells,AST-DHA and free astaxanthin(F-AST)recovered p62 and LC3Ⅱ/Ⅰlevels,and restored autophagy flux by rescuing the late phase of microglial autophagy.Notably,autophagic inhibitor bafilomycin A1 blunted the abilities of AST-DHA to reduce Aβ_(1-42)and fibral Aβ,suggesting that AST-DHA probably promoted Aβclearance in a microglial autophagy-dependent manner.Further studies in APP/PS1 mice verified that dietary AST-DHA and F-AST promoted Aβphagocytosis via microglial autophagy.Significant decreases of Iba1 and p62 were observed around Aβplaque in the hippocampus and cortex using triple fluorescence staining.Furthermore,AST-DHA exhibited superior performance over F-AST in restoring autophagic dysfunction,ameliorating Aβburden and cognitive deficit.Our findings suggest a possible mechanism of AST-DHA in improving AD by which it restores microglial autophagy to facilitate cerebral Aβclearance.It supports the future application of AST-DHA as an autophagic regulator in maintaining brain function.

文摘Biological load-bearing materials,like the nacre in shells,have a unique staggered structure that supports their superior mechanical properties.Engineers have been encouraged to imitate it to create load-bearing bio-inspired materials which have excellent properties not present in conventional composites.To create such materials with desirable mechanical properties,the optimum structural parameters combination must be selected.Moreover,the optimal design of bio-inspired composites needs to take into account the trade-offs between various mechanical properties.In this paper,multi-objective optimization models were developed using structural parameters as design variables and mechanical properties as optimization objectives,including stiffness,strength,toughness,and dynamic damping.Using the NSGA-II optimization algorithm,a set of optimal solutions were solved.Additionally,three different structures in natural nacre were introduced in order to utilize the better structure when design bio-inspired materials.The range of optimal solutions that obtained using results from previous research were examined and explained why this collection of optimal solution ranges is better.Also,optimal solutions were compared with the structural features and mechanical properties of real nacre and artificial biomimetic composites to validate our models.Finally,the optimum design strategies can be obtained for nacre-like composites.Our research methodically proposes an optimization method for achieving load-bearing bio-inspired materials with excellent properties and creates a set of optimal solutions from which designers can select the one that best suits their preferences,allowing the fabricated materials to demonstrate preferred performance.
文摘脱水应答元件结合蛋白(Dehydration-responsive element binding proteins,DREBs)是一类重要的植物耐逆相关转录因子。蒙古沙冬青Ammopiptanthus mongolicus是中国西北荒漠区特有的强耐逆常绿阔叶灌木。为探明其AmDREB1F基因在耐受非生物逆境中的功能和作用机理,文中对该基因编码蛋白的亚细胞定位、表达模式和转基因拟南芥的耐逆性进行了分析。结果表明:AmDREB1F编码的蛋白质定位于细胞核内;在室内培养幼苗中,该基因在正常条件下不表达,在低温和干旱胁迫下有较明显表达,在高盐和高温胁迫下仅有微弱表达,而在脱落酸(Abscisicacid,ABA)处理下不表达;在野外生长植株的叶片中,其表达量在秋末、冬季和早春远高于其他季节,而不同器官相比,其在根和未成熟果荚中的表达量远高于其他器官;将AmDREB1F在拟南芥中组成型表达可提高多个受DREBs调控的胁迫响应基因的转录水平,增强转基因株系对干旱、高盐和低温以及氧化胁迫的耐性,同时导致其生长发育延滞,外施赤霉素3可消除生长延滞现象;将该基因进行胁迫诱导表达也可提高转基因拟南芥对上述非生物胁迫的耐受性,而不影响其生长发育。这些结果说明AmDREB1F可能通过ABA非依赖的信号途径在响应和耐受逆境胁迫中起正调节作用。

文摘AIM: To investigate the changes of lymphocyte subpopulations, especially CD4^+CD25^ T regulatory cells in Smad3^-/- mice. METHODS: Hematological changes and changes of lymphocyte subpopulations were detected in Smad3"/- mice using cell counter and flow cytometry, respectively, and compared to their littermate controls. RESULTS: The numbers of neutrophils and lymphocytes in peripheral blood were significantly increased in Smad3^-/- mice compared to littermate controls. CD19^+ expressing cells in blood and spleen, and CD8^+ T cells in thymus were all markedly decreased in Smad3^-/- mice. More important, Smad3^-/- mice had an increased population of CD4^+CD25^+ T cells in peripheral lymphoid tissues, including thymus, spleen, and lymph nodes. CONCLUSION: These observations suggest that the changes of lymphocyte subpopulations might play a role in susceptibility to inflammation of Smad3^-/- mice.

文摘Theranostic visualization of dextran at the nanoscale is beneficial for understanding the bioregulatory mechanisms of this molecule. In this study, we applied structured illumination microscopy(SIM) to capture the distribution of Cy5-Dextran at different incubation periods in living cells. The results showed that Cy5-Dextran could be absorbed by He La cells. In addition, we clarified that Cy5-Dextran exhibited differential organelle distribution(lysosomal or mitochondrial) in a time-dependent manner. Moreover,lysosomal Cy5-Dextran localization was found to be independent of the autophagy process, while Cy5-Dextran localized to the mitochondria triggered a pro-apoptotic event, upregulating the levels of reactive oxygen species(ROS) to accelerate mitochondrial fragmentation. This work uses a visualized strategy to reveal the anti-tumor bioactivity of dextran, which was achieved by regulating apoptosis and autophagy.

文摘A series of mono-and binuclear Co(Ⅱ) complexes (Col-Co7) supported by quinoline-2-imidate ligands were synthesized and thoroughly characterized. Measured by single crystal X-ray crystallography, complexes Col and Co3 adopted distorted tetrahedral structures around the cobalt center. Upon activation by ethylaluminium sesquichloride (EASC), these cobalt complexes exhibited high catalytic activity and cis-l,4-selectivity towards 1,3-butadiene polymerization. The effects of ligand environment, polymerization temperature, and cocatalyst types on the polymerization were investigated in detail. Interestingly, the binuclear Co(Ⅱ) complexes exhibited high thermal stability, and the polymer yields were up to 97.2% even at a high temperature of 70 ℃.
文摘目的探讨感染性休克患者血清肽酰基精氨酸脱亚胺酶2(peptidylarginine deiminase type 2,PAD2)表达水平与急性生理学和慢性健康状况评价Ⅱ(acute physiology and chronic health evaluationⅡ,APACHEⅡ)评分的相关性。方法选取内江市第一人民医院2020年6月~2022年6月收治的103例感染性休克患者作为研究组,采用APACHEⅡ评分根据患者病情严重程度将其分为轻度组(n=9)、中度组(n=51)和重度组(n=13),另外选取103例同期在该院体检且一般资料与研究组患者相匹配的健康者作为对照组。采用酶联免疫吸附法测定感染性休克患者血清PAD2表达水平;采用Spearman法分析感染性休克患者血清PAD2表达水平与APACHEⅡ评分的相关性;采用Logistic回归分析影响感染性休克患者病情严重程度的相关因素;采用受试者工作特征(receiver operating characteristic,ROC)曲线分析血清PAD2对中重度感染性休克的诊断价值。结果研究组与对照组血肌酐(137.52±9.01μmol/L vs 112.22±8.67μmol/L)水平及血小板计数(74.58±5.19 vs 86.02±5.34)×109/L比较,差异具有统计学意义(t=20.535,15.591,均P<0.05);研究组患者血清PAD2表达水平(42.47±6.22 ng/ml)高于对照组(38.59±5.31 ng/ml),差异具有统计学意义(t=4.815,P<0.05);感染性休克患者血清PAD2表达水平和APACHEⅡ评分均随病情严重程度的增加而逐渐升高(F=3.777,176.582,均P<0.05);感染性休克患者血清PAD2表达水平与APACHEⅡ评分呈正相关(r=0.859,P<0.05);血肌酐(OR=1.927)、PAD2(OR=1.803)及APACHEⅡ评分(OR=1.657)均为发生中重度感染性休克的危险因素(均P<0.05),血小板计数(OR=0.781)则是发生中重度感染性休克的保护因素(P<0.05)。血清PAD2诊断中重度感染性休克的曲线下面积(area under the curve,AUC)为0.880,敏感度、特异度分别为75.73%(95%CI:0.701~0.826)和90.29%(95%CI:0.851~0.935),对中重度感染性休克具有较高的诊断价值。结论血清PAD2表达水平与APACHEⅡ评分呈正相关,且对中重度感染性休克具有较好诊断价值。

文摘Objective:This trial is aimed at evaluating the efficacy and safety of acupuncture on treating FC.Methods:We describe the protocol for a randomized,patient-assessor-blinded,sham controlled trial.Seventy-two eligible patients will be randomly assigned to the intervention group(acupuncture)or the control group(sham acupuncture).All treatment will be given 26 sessions of acupuncture or sham acupuncture over 8 weeks(5 times per week in the first 2 weeks,3 times per week during weeks 3-6,and 2 times per week during weeks 7 and 8).Each treatment will last for 20 min.The primary outcome is the change in mean complete spontaneous bowel movements(CSBMs)per week.The secondary out?comes are patient assessment of constipation quality of life questionnaire(PAC-QOL),self-rating anxiety scale(SAS),and the dosage of the medication.All adverse events will be recorded in detail and managed by corresponding researchers as quickly as possible.Outcomes will be evaluated at baseline(1 week before treatment),2 weeks after intervention begins,6 weeks after intervention begins,8 weeks after intervention begins,4 weeks follow-up,and 12 weeks follow-up.Discussion:The results of this study will provide the evidence of the efficacy and safety of acupuncture as a traditional treatment methods for functional constipation.Trial registration:Chinese Clinical Trial Registry,ChiCTR-INR-17011472.Registered on 23 May 2017.

文摘The motivation for this study is that the quality of deep fakes is constantly improving,which leads to the need to develop new methods for their detection.The proposed Customized Convolutional Neural Network method involves extracting structured data from video frames using facial landmark detection,which is then used as input to the CNN.The customized Convolutional Neural Network method is the date augmented-based CNN model to generate‘fake data’or‘fake images’.This study was carried out using Python and its libraries.We used 242 films from the dataset gathered by the Deep Fake Detection Challenge,of which 199 were made up and the remaining 53 were real.Ten seconds were allotted for each video.There were 318 videos used in all,199 of which were fake and 119 of which were real.Our proposedmethod achieved a testing accuracy of 91.47%,loss of 0.342,and AUC score of 0.92,outperforming two alternative approaches,CNN and MLP-CNN.Furthermore,our method succeeded in greater accuracy than contemporary models such as XceptionNet,Meso-4,EfficientNet-BO,MesoInception-4,VGG-16,and DST-Net.The novelty of this investigation is the development of a new Convolutional Neural Network(CNN)learning model that can accurately detect deep fake face photos.
