BACKGROUND At present,the incidence of diabetic nephropathy is increasing year by year,and there are many studies on the pathogenesis of diabetic nephropathy,but it is still not completely clear.The final pathological...BACKGROUND At present,the incidence of diabetic nephropathy is increasing year by year,and there are many studies on the pathogenesis of diabetic nephropathy,but it is still not completely clear.The final pathological result of diabetic nephropathy is mainly glomerular cell fibrosis,and the roles of micro-RNA(miRNA)-29 and DNA methyl transferase(DNMTs)in cell fibrosis have been confirmed in other studies,but there is a lack of relevant research in the kidney at present.AIM To study the potential involvement of miRNA-29a-3p in fibrosis related to diabetic kidney disease(DKD).METHODS The expression of miR-29a-3p,DNMT3A/3B,fibrosis-related molecules,Wnt3a,β-catenin,Janus kinase 2,and signal transducer and activator of transcription 3 was assessed in SV40MES13 cells and diabetic mice using quantitative real-time PCR and western blotting.Furthermore,the expression changes of fibrosis-related molecules were further analyzed using immunofluorescence and immunohistochemical blotting.The renal pathological changes of DKD in each group were also studied using hematoxylin-eosin and periodate-Schiff reaction staining.RESULTS In both the in vivo and in vitro experiments,it was observed that high glucose induction significantly decreased miR-29a-3p expression.As a result of this downregulation,DKD-related fibrosis was found to be promoted,as confirmed by elevated expression levels ofα-smooth muscle actin,collagen type I,and fibronectin.MiR-29a-3p targets the 3’non-coding regions of DNMT3A and DNMT3B and inhibits their expression.Inhibition of DNMT3A and DNMT3B can reverse the effect of miR-29a-3p downregulation on DKD-related fibrosis.CONCLUSION MiR-29a-3p can regulate Wnt/β-catenin and Janus kinase/signal transducer and activator of transcription signal pathways by regulating and inhibiting the expression of DNMT3A/3B and thus participate in the inhibition of DKD-related fibrosis.展开更多
This article presents an interpretation of the seismic source mechanisms for 905 earthquakes with MS>2.0,occurring in the central and southern sections of the Tan-Lu Fault Zone and its adjacent areas from 1970 to 2...This article presents an interpretation of the seismic source mechanisms for 905 earthquakes with MS>2.0,occurring in the central and southern sections of the Tan-Lu Fault Zone and its adjacent areas from 1970 to 2023.Utilizing the damped stress tensor method,we have inverted the spatial characteristics of the stress field variations in this study area,at a resolution of 1.0°×1.0°.The results indicate that the maximum principal stress direction within the central and southern sections of the Tan-Lu Fault Zone exhibits a spatially continuous change,rotating counterclockwise from East-West(EW)to Northeast-East(NEE),albeit with local variations.When dividing along the Tan-Lu Fault Zone,it is observed that on its western side—the North China block—a near EW stress field predominates.In contrast,on its eastern side—the Ludong-Huanghai Block—stress fields primarily exhibit NEE and Northeast-East(NE)orientations,underscoring the role of the Tan-Lu Fault Zone as a significant boundary between tectonic blocks.In regions located between latitudes 30°-34°N and longitudes 113°-115°E on the western side of these fault zone sections,maximum principal stress follows a radial distribution pattern indicative of a complex stress field.Conversely,on the eastern side of this fault zone,maximum principal stress direction remains relatively consistent,primarily displaying NEE and NE distributions.This reflects the fact that this area is situated under a tectonic background characterized by near NEE-NE direction for the Ludong-Huanghai Block.However,within an area bounded by latitudes 30°-32°N and longitudes 120°-122°E on the eastern flank of the Tan-Lu Fault Zone,maximum principal stress direction appears more intricate,with radial distribution patterns suggesting influences not only from near EW-NEE movements associated with the North China Block,but also from westward subduction processes related to Philippine Sea plate dynamics.Through our analysis of historical earthquakes in this region,we conclude that the moderate to strong seismic activity within this area is significantly related to the tectonic stress environment,with regions of complex tectonic stress often being the most seismically active.展开更多
Hard carbon stands out as the most promising candidate for anodes in sodium-ion battery.Nevertheless,addressing the challenges of low initial Coulombic efficiency and rate performance is crucial for practical applicat...Hard carbon stands out as the most promising candidate for anodes in sodium-ion battery.Nevertheless,addressing the challenges of low initial Coulombic efficiency and rate performance is crucial for practical applications.In this study,we employed a dimensionally designed approach,using six different biomass precursors,to preserve their inherent fine hierarchical morphological structures and appearances during the synthesis of selfsupporting carbon materials.Benefiting from its low-tortuosity structure that facilitates electron and ion transport,as well as its surface-enriched C=O functional groups and significant closed micropore areas,the obtained carbon material exhibits excellent electrochemical performance in sodium-ion storage,demonstrated by finite element simulation.Notably,the carbonized basswood exhibited a remarkable initial Coulombic efficiency of up to 92.4%and demonstrated outstanding rate performance,achieving a capacity of 223.3 mAh·g^(-1)at a high current density of2 A·g^(-1).In addition,thorough investigation was conducted on the influence of microstructure on the sodium storage behavior of hard carbon.Ex situ X-ray diffraction(XRD)was used to confirm that the capacity in the plateau region originates from interlayer insertion and closed-pore filling,which is consistent with the results obtained from smallangle X-ray scattering.These findings underscore the immense potential of leveraging surface functionalization and structural design to bolster the performance of hard carbon,paving the way for promising future advancements in this field.展开更多

METHODS: CD73 and HIF-1α expressions wer...AIM: To investigate the expression of CD73 and hypoxia-inducible factor-1α (HIF-1α) in human gastric carcinoma, and explore their clinical significance and prognostic value. METHODS: CD73 and HIF-1α expressions were detected by immunohistochemistry in consecutive sections of tissue samples from 68 gastric carcinoma patients. The peritumor tissues 2 cm away from the tumor were obtained and served as controls. The presence of CD73 and HIF-1α was analyzed by immunohis-tochemistry using the Envision technique. RESULTS: CD73 and HIF-1α expressions in gastric carcinoma were significantly higher than those in gastric mucosal tissues as control (P < 0.001) and showed a close correlation (Spearman r = 0.390, P = 0.001). Overexpression of CD73 was positively correlated with differentiation of tumor (P = 0.000), histopathology (P = 0.041), depth of invasion (P < 0.001), nodal status (P = 0.003), metastasis (P = 0.013), and the American Joint Committee on Cancer (AJCC) stage (P < 0.001). High expression of HIF-1α was positively correlated with tumor diameter (P = 0.031), depth of invasion (P = 0.022), and AJCC stage (P = 0.035). The overall survival rate was low in the patients with high expression of CD73 (P < 0.001). Moreover, CD73+/HIF-1α+ patients had the worst prognosis (P < 0.001). CD73 expression was proven to be an independent predictor for patients with gastric carcinoma by both multivariate Cox regression analysis (P = 0.021) and receiver operating characteristic curves (P = 0.001).CONCLUSION: CD73 expression correlates closely with HIF-1α expression in gastric carcinoma. CD73 could be an independent prognostic indicator for gastric carcinoma.展开更多
Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.Methods: A total of 210 sows(parities 3–6) wer...Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.Methods: A total of 210 sows(parities 3–6) were randomly allocated into six diets(n = 35) throughout gestation to feed graded levels of dietary fiber(DF), including a corn-soybean meal-based control diet with no wheat bran inclusion(CON, 12.5% DF), a wheat bran-rich diet(DF1, 17.4% DF), and another 4 diets(DF2, 17.7% DF;DF3, 18.1%DF;DF4, 18.4% DF;DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX(guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout gestation.Results: DF treatment during gestation resulted in normal fecal score(1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet(P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment(P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation(P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group(P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. Fecal acetate, propionate, and butyrate of sows were markedly increased in late gestation, and butyrate contents in feces of gestating sows were significantly affected by DF levels(P < 0.01).Serum concentrations of pro-inflammatory TNF-α were decreased and anti-inflammatory IL-10 was increased on day30 of gestation by DF levels(P < 0.05).Conclusions: In summary, increasing dietary fiber levels by guar gum and cellulose during gestation improved the reproductive performance of sows, which might be related to changes in immunity and gut microbiota of sows.展开更多

Increasing number of findings of toxin-producing bacteria in the cells of dinoflagellate such as Alexandriumtamarense supports the hypothesis of the bacterial origin of PSP toxins. Evidence that there are specific symbiosis bacterial taxa associated with the phytoplankton indicates the presence of specific selective mechanisms between them, and implies that the symbiosis bacteria have some vital function to the benefit of the dinoflagellates. Studies on the role of toxin-producing symbiosis bacteria in the marine ecosystem are considered to be becoming more important. Although toxigenic bacteria could be isolated from toxic dinoflagellates, it was not clearly proven whether the isolated bacterial strains based on culture-dependent manner and the corresponding intracellular bacteria were the same because of microbial unculturability. This paper aims to demonstrate the biodiversity of the symbiotic bacteria associated with toxic dinoflagellate A. tamarense using the culture-indepen- dent high-throughput pyrosequencing method, as well as the phylogenetic analysis based on 16S rDNA sequences of the symbiotic cultivable bacteria strains isolated from toxic Alexander tamarense.展开更多

Therefore, the present study focuses on the investi- gation of the protective effect of oat bran extract by enzymatic hydrolysates on human dermal fibroblast injury induced by hydrogen peroxide (H2O2). Kjeldahl determination, phenol-sulfuric acid method, and high-performance liquid chromatography (HPLC) analysis indicated that the enzymatic products of oat bran contain a protein amount of 71.93%, of which 97.43% are peptides with a molecular range from 438.56 to 1301.01 Da. Assays for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity indicate that oat peptide-dch extract has a direct and concentration-dependent antioxidant activity. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) colorimetric assay and the TdT-mediated digoxigenin-dUTP nick-end labeling (TUNEL) assay for apoptosis showed that administration of H2O2 in human dermal fibroblasts caused cell damage and apoptosis. Pre-incubation of human dermal fibroblasts with the Oatp for 24 h markedly inhibited human dermal fibroblast injury induced by H2O2, but ap- plication oat peptides with H2O2 at same time did not. Pre-treatment of human dermal fibroblasts with Oatp significantly reversed the H2O2-induced decrease of superoxide dismutase (SOD) and the inhibition of malondialdehyde (MDA). The results demonstrate that oat peptides possess antioxidant activity and are effective against H2O2-induced human dermal fibroblast injury by the enhanced activity of SOD and decrease in MDA level. Our results suggest that oat bran will have the potential to be further explored as an antioxidant functional food in the prevention of aging-related skin injury.展开更多
Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both p...Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both pelagic and benthic Microcystis and their characteristics in Lake Caohai,we conducted a 1-year investigation from December 2015to December 2016 to gain a seasonal outlook on the distribution and dynamics of cell abundance,colony size and intracellular microcystins(MCs)of Microcystis.The results indicated that the Microcystis bloom occupied primarily the northeastern region and then moved gradually from lakeshore to lake center.The perennial southwesterly winds and the water inflow from northeast to southwest in Lake Caohai determined the spatiotemporal distribution of pelagic Microcystis.Benthic Microcystis was mainly distributed in the northeastern region in summer,occupied the lake center in autumn and then occupied the southeastern region in winter,determined by the sedimentation of pelagic Microcystis and the death of benthic Microcystis.Small colonies(20-60μm)overwintered more easily in both water column and sediment.The concentrations of intracellular toxin of benthic Microcystis were observed to be significantly higher than those of pelagic Microcystis.This might be because Microcystis synthesized large amount of MCs to acclimate to an unfavorable benthic environment.This knowledge on the dynamics of Microcystis expands our understanding of mechanisms underpinning the formation of Microcystis blooms.展开更多
BACKGROUND As human placenta-derived mesenchymal stem cells(hP-MSCs)exist in a physiologically hypoxic microenvironment,various studies have focused on the influence of hypoxia.However,the underlying mechanisms remain...BACKGROUND As human placenta-derived mesenchymal stem cells(hP-MSCs)exist in a physiologically hypoxic microenvironment,various studies have focused on the influence of hypoxia.However,the underlying mechanisms remain to be further explored.AIM The aim was to reveal the possible mechanisms by which hypoxia enhances the proliferation of hP-MSCs.METHODS A hypoxic cell incubator(2.5%O2)was used to mimic a hypoxic microenvironment.Cell counting kit-8 and 5-ethynyl-20-deoxyuridine incorporation assays were used to assay the proliferation of hP-MSCs.The cell cycle was profiled by flow cytometry.Transcriptome profiling of hP-MSCs under hypoxia was performed by RNA sequencing.CD99 mRNA expression was assayed by reverse transcription-polymerase chain reaction.Small interfering RNA-mediated hypoxia-inducible factor 1α(HIF-1α)or CD99 knockdown of hP-MSCs,luciferase reporter assays,and the ERK1/2 signaling inhibitor PD98059 were used in the mechanistic analysis.Protein expression was assayed by western blotting;immunofluorescence assays were conducted to evaluate changes in expression levels.RESULTS Hypoxia enhanced hP-MSC proliferation,increased the expression of cyclin E1,cyclin-dependent kinase 2,and cyclin A2,and decreased the expression of p21.Under hypoxia,CD99 expression was increased by HIF-1α.CD99-specific small interfering RNA or the ERK1/2 signaling inhibitor PD98059 abrogated the hypoxia-induced increase in cell proliferation.CONCLUSION Hypoxia promoted hP-MSCs proliferation in a manner dependent on CD99 regulation of the MAPK/ERK signaling pathway in vitro.展开更多

[Method] With Yunyan 87 as tested flue-cured tobacco varieties, three treatments (chemical fertilizer (control), chemical fertilizer+solid amino acid, chemical fertilizer+liquid amino acid) were set up to study the effects of amino acid fertilizer on flue-cured tobacco fertilizer reducing and efficiency increasing by means of contrast in the same field. [Result] The flue-cured tobacco applied solid amino acid fertilizer had the best performances of overall agronomic traits and the highest yield, while liquid amino acid fertilizer was conductive to the top leaf opening, making the raw tobacco appearance best, and could increase the proportion of middle and high quality tobacco, gaining good economic benefits. The flue-cured tobacco treated by liquid amino acid fertilizer had the optimal ratio of total sugar to nicotine and disaccharide differences in tobacco leaves, and the best coordination of chemical composition. Compared with the contrel, tobacco treated by amino acid fertilizer had less irritating and more comfortable taste, and the tobacco treated by liquid amino acid had light miscellaneous gas, and the best overall performance of sensory quality. [Conclusion] Chemical fertilizer+liquid amino acids is conductive to promote the growth and development of flue-cured tobacco and improve the quality of tobacco leaves.展开更多
BACKGROUND The effect of hypoxia on mesenchymal stem cells(MSCs)is an emerging topic in MSC biology.Although long non-coding RNAs(lncRNAs)and messenger RNAs(mRNAs)are reported to play a critical role in regulating the...BACKGROUND The effect of hypoxia on mesenchymal stem cells(MSCs)is an emerging topic in MSC biology.Although long non-coding RNAs(lncRNAs)and messenger RNAs(mRNAs)are reported to play a critical role in regulating the biological characteristics of MSCs,their specific expression and co-expression profiles in human placenta-derived MSCs(hP-MSCs)under hypoxia and the underlying mechanisms of lncRNAs in hP-MSC biology are unknown.AIM To reveal the specific expression profiles of lncRNAs in hP-MSCs under hypoxia and initially explored the possible mechanism of lncRNAs on hP-MSC biology.METHODS Here,we used a multigas incubator(92.5%N_(2),5%CO_(2),and 2.5%O_(2))to mimic the potential of hP-MSCs.RNA sequencing technology was applied to identify the exact expression profiles of lncRNAs and mRNAs under hypoxia.RESULTS We identified 289 differentially expressed lncRNAs and 240 differentially expressed mRNAs between the hypoxia and normoxia groups.Among them,the lncRNA SNHG16 was upregulated under hypoxia,which was also validated by reverse transcription-polymerase chain reaction.SNHG16 was confirmed to affect hP-MSC proliferation rates using a SNHG16 knockdown model.SNHG16 overexpression could significantly enhance the proliferation capacity of hP-MSCs,activate the PI3K/AKT pathway,and upregulate the expression of cell cycle-related proteins.CONCLUSION Our results revealed the specific expression characteristics of lncRNAs and mRNAs in hypoxiacultured hP-MSCs and that lncRNA SNHG16 can promote hP-MSC proliferation through the PI3K/AKT pathway.展开更多

文摘This article presents an interpretation of the seismic source mechanisms for 905 earthquakes with MS>2.0,occurring in the central and southern sections of the Tan-Lu Fault Zone and its adjacent areas from 1970 to 2023.Utilizing the damped stress tensor method,we have inverted the spatial characteristics of the stress field variations in this study area,at a resolution of 1.0°×1.0°.The results indicate that the maximum principal stress direction within the central and southern sections of the Tan-Lu Fault Zone exhibits a spatially continuous change,rotating counterclockwise from East-West(EW)to Northeast-East(NEE),albeit with local variations.When dividing along the Tan-Lu Fault Zone,it is observed that on its western side—the North China block—a near EW stress field predominates.In contrast,on its eastern side—the Ludong-Huanghai Block—stress fields primarily exhibit NEE and Northeast-East(NE)orientations,underscoring the role of the Tan-Lu Fault Zone as a significant boundary between tectonic blocks.In regions located between latitudes 30°-34°N and longitudes 113°-115°E on the western side of these fault zone sections,maximum principal stress follows a radial distribution pattern indicative of a complex stress field.Conversely,on the eastern side of this fault zone,maximum principal stress direction remains relatively consistent,primarily displaying NEE and NE distributions.This reflects the fact that this area is situated under a tectonic background characterized by near NEE-NE direction for the Ludong-Huanghai Block.However,within an area bounded by latitudes 30°-32°N and longitudes 120°-122°E on the eastern flank of the Tan-Lu Fault Zone,maximum principal stress direction appears more intricate,with radial distribution patterns suggesting influences not only from near EW-NEE movements associated with the North China Block,but also from westward subduction processes related to Philippine Sea plate dynamics.Through our analysis of historical earthquakes in this region,we conclude that the moderate to strong seismic activity within this area is significantly related to the tectonic stress environment,with regions of complex tectonic stress often being the most seismically active.

文摘Hard carbon stands out as the most promising candidate for anodes in sodium-ion battery.Nevertheless,addressing the challenges of low initial Coulombic efficiency and rate performance is crucial for practical applications.In this study,we employed a dimensionally designed approach,using six different biomass precursors,to preserve their inherent fine hierarchical morphological structures and appearances during the synthesis of selfsupporting carbon materials.Benefiting from its low-tortuosity structure that facilitates electron and ion transport,as well as its surface-enriched C=O functional groups and significant closed micropore areas,the obtained carbon material exhibits excellent electrochemical performance in sodium-ion storage,demonstrated by finite element simulation.Notably,the carbonized basswood exhibited a remarkable initial Coulombic efficiency of up to 92.4%and demonstrated outstanding rate performance,achieving a capacity of 223.3 mAh·g^(-1)at a high current density of2 A·g^(-1).In addition,thorough investigation was conducted on the influence of microstructure on the sodium storage behavior of hard carbon.Ex situ X-ray diffraction(XRD)was used to confirm that the capacity in the plateau region originates from interlayer insertion and closed-pore filling,which is consistent with the results obtained from smallangle X-ray scattering.These findings underscore the immense potential of leveraging surface functionalization and structural design to bolster the performance of hard carbon,paving the way for promising future advancements in this field.

文摘AIM: To investigate the expression of CD73 and hypoxia-inducible factor-1α (HIF-1α) in human gastric carcinoma, and explore their clinical significance and prognostic value. METHODS: CD73 and HIF-1α expressions were detected by immunohistochemistry in consecutive sections of tissue samples from 68 gastric carcinoma patients. The peritumor tissues 2 cm away from the tumor were obtained and served as controls. The presence of CD73 and HIF-1α was analyzed by immunohis-tochemistry using the Envision technique. RESULTS: CD73 and HIF-1α expressions in gastric carcinoma were significantly higher than those in gastric mucosal tissues as control (P < 0.001) and showed a close correlation (Spearman r = 0.390, P = 0.001). Overexpression of CD73 was positively correlated with differentiation of tumor (P = 0.000), histopathology (P = 0.041), depth of invasion (P < 0.001), nodal status (P = 0.003), metastasis (P = 0.013), and the American Joint Committee on Cancer (AJCC) stage (P < 0.001). High expression of HIF-1α was positively correlated with tumor diameter (P = 0.031), depth of invasion (P = 0.022), and AJCC stage (P = 0.035). The overall survival rate was low in the patients with high expression of CD73 (P < 0.001). Moreover, CD73+/HIF-1α+ patients had the worst prognosis (P < 0.001). CD73 expression was proven to be an independent predictor for patients with gastric carcinoma by both multivariate Cox regression analysis (P = 0.021) and receiver operating characteristic curves (P = 0.001).CONCLUSION: CD73 expression correlates closely with HIF-1α expression in gastric carcinoma. CD73 could be an independent prognostic indicator for gastric carcinoma.

文摘Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.Methods: A total of 210 sows(parities 3–6) were randomly allocated into six diets(n = 35) throughout gestation to feed graded levels of dietary fiber(DF), including a corn-soybean meal-based control diet with no wheat bran inclusion(CON, 12.5% DF), a wheat bran-rich diet(DF1, 17.4% DF), and another 4 diets(DF2, 17.7% DF;DF3, 18.1%DF;DF4, 18.4% DF;DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX(guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout gestation.Results: DF treatment during gestation resulted in normal fecal score(1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet(P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment(P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation(P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group(P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. Fecal acetate, propionate, and butyrate of sows were markedly increased in late gestation, and butyrate contents in feces of gestating sows were significantly affected by DF levels(P < 0.01).Serum concentrations of pro-inflammatory TNF-α were decreased and anti-inflammatory IL-10 was increased on day30 of gestation by DF levels(P < 0.05).Conclusions: In summary, increasing dietary fiber levels by guar gum and cellulose during gestation improved the reproductive performance of sows, which might be related to changes in immunity and gut microbiota of sows.

文摘Hepatocellular carcinoma(HCC)is the most common primary malignant liver tumor in China.Preoperative diagnosis of HCC is challenging because of atypical imaging manifestations and the diversity of focal liver lesions.Artificial intelligence(AI),such as machine learning(ML)and deep learning,has recently gained attention for its capability to reveal quantitative information on images.Currently,AI is used throughout the entire radiomics process and plays a critical role in multiple fields of medicine.This review summarizes the applications of AI in various aspects of preoperative imaging of HCC,including segmentation,differential diagnosis,prediction of histo-pathology,early detection of recurrence after curative treatment,and evaluation of treatment response.We also review the limitations of previous studies and discuss future directions for diagnostic imaging of HCC.
文摘Paralytic shellfish poisoning (PSP) toxins are potent neurotoxins mainly produced by dinoflagellates and being concentrated in bivalves through food web transfer. Increasing number of findings of toxin-producing bacteria in the cells of dinoflagellate such as Alexandriumtamarense supports the hypothesis of the bacterial origin of PSP toxins. Evidence that there are specific symbiosis bacterial taxa associated with the phytoplankton indicates the presence of specific selective mechanisms between them, and implies that the symbiosis bacteria have some vital function to the benefit of the dinoflagellates. Studies on the role of toxin-producing symbiosis bacteria in the marine ecosystem are considered to be becoming more important. Although toxigenic bacteria could be isolated from toxic dinoflagellates, it was not clearly proven whether the isolated bacterial strains based on culture-dependent manner and the corresponding intracellular bacteria were the same because of microbial unculturability. This paper aims to demonstrate the biodiversity of the symbiotic bacteria associated with toxic dinoflagellate A. tamarense using the culture-indepen- dent high-throughput pyrosequencing method, as well as the phylogenetic analysis based on 16S rDNA sequences of the symbiotic cultivable bacteria strains isolated from toxic Alexander tamarense.
文摘Oat contains different components that possess antioxidant properties; no study to date has addressed the antioxidant effect of the extract of oat bran on the cellular level. Therefore, the present study focuses on the investi- gation of the protective effect of oat bran extract by enzymatic hydrolysates on human dermal fibroblast injury induced by hydrogen peroxide (H2O2). Kjeldahl determination, phenol-sulfuric acid method, and high-performance liquid chromatography (HPLC) analysis indicated that the enzymatic products of oat bran contain a protein amount of 71.93%, of which 97.43% are peptides with a molecular range from 438.56 to 1301.01 Da. Assays for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity indicate that oat peptide-dch extract has a direct and concentration-dependent antioxidant activity. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) colorimetric assay and the TdT-mediated digoxigenin-dUTP nick-end labeling (TUNEL) assay for apoptosis showed that administration of H2O2 in human dermal fibroblasts caused cell damage and apoptosis. Pre-incubation of human dermal fibroblasts with the Oatp for 24 h markedly inhibited human dermal fibroblast injury induced by H2O2, but ap- plication oat peptides with H2O2 at same time did not. Pre-treatment of human dermal fibroblasts with Oatp significantly reversed the H2O2-induced decrease of superoxide dismutase (SOD) and the inhibition of malondialdehyde (MDA). The results demonstrate that oat peptides possess antioxidant activity and are effective against H2O2-induced human dermal fibroblast injury by the enhanced activity of SOD and decrease in MDA level. Our results suggest that oat bran will have the potential to be further explored as an antioxidant functional food in the prevention of aging-related skin injury.

文摘Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both pelagic and benthic Microcystis and their characteristics in Lake Caohai,we conducted a 1-year investigation from December 2015to December 2016 to gain a seasonal outlook on the distribution and dynamics of cell abundance,colony size and intracellular microcystins(MCs)of Microcystis.The results indicated that the Microcystis bloom occupied primarily the northeastern region and then moved gradually from lakeshore to lake center.The perennial southwesterly winds and the water inflow from northeast to southwest in Lake Caohai determined the spatiotemporal distribution of pelagic Microcystis.Benthic Microcystis was mainly distributed in the northeastern region in summer,occupied the lake center in autumn and then occupied the southeastern region in winter,determined by the sedimentation of pelagic Microcystis and the death of benthic Microcystis.Small colonies(20-60μm)overwintered more easily in both water column and sediment.The concentrations of intracellular toxin of benthic Microcystis were observed to be significantly higher than those of pelagic Microcystis.This might be because Microcystis synthesized large amount of MCs to acclimate to an unfavorable benthic environment.This knowledge on the dynamics of Microcystis expands our understanding of mechanisms underpinning the formation of Microcystis blooms.

文摘BACKGROUND As human placenta-derived mesenchymal stem cells(hP-MSCs)exist in a physiologically hypoxic microenvironment,various studies have focused on the influence of hypoxia.However,the underlying mechanisms remain to be further explored.AIM The aim was to reveal the possible mechanisms by which hypoxia enhances the proliferation of hP-MSCs.METHODS A hypoxic cell incubator(2.5%O2)was used to mimic a hypoxic microenvironment.Cell counting kit-8 and 5-ethynyl-20-deoxyuridine incorporation assays were used to assay the proliferation of hP-MSCs.The cell cycle was profiled by flow cytometry.Transcriptome profiling of hP-MSCs under hypoxia was performed by RNA sequencing.CD99 mRNA expression was assayed by reverse transcription-polymerase chain reaction.Small interfering RNA-mediated hypoxia-inducible factor 1α(HIF-1α)or CD99 knockdown of hP-MSCs,luciferase reporter assays,and the ERK1/2 signaling inhibitor PD98059 were used in the mechanistic analysis.Protein expression was assayed by western blotting;immunofluorescence assays were conducted to evaluate changes in expression levels.RESULTS Hypoxia enhanced hP-MSC proliferation,increased the expression of cyclin E1,cyclin-dependent kinase 2,and cyclin A2,and decreased the expression of p21.Under hypoxia,CD99 expression was increased by HIF-1α.CD99-specific small interfering RNA or the ERK1/2 signaling inhibitor PD98059 abrogated the hypoxia-induced increase in cell proliferation.CONCLUSION Hypoxia promoted hP-MSCs proliferation in a manner dependent on CD99 regulation of the MAPK/ERK signaling pathway in vitro.

文摘The post-hepatectomy recurrence rate of hepatocellular carcinoma(HCC)is persistently high,affecting the prognosis of patients.An effective therapeutic option is crucial for achieving long-term survival in patients with postoperative recurrences.Local ablative therapy has been established as a treatment option for resectable and unresectable HCCs,and it is also a feasible approach for recurrent HCC(RHCC)due to less trauma,shorter operation times,fewer complications,and faster recovery.This review focused on ablation techniques,description of potential candidates,and therapeutic and prognostic implications of ablation for guiding its application in treating intrahepatic RHCC.

文摘[Objective] The aim was to explore the effect of amino acid fertilizer on the growth, development and quality of flue-cured tobacco. [Method] With Yunyan 87 as tested flue-cured tobacco varieties, three treatments (chemical fertilizer (control), chemical fertilizer+solid amino acid, chemical fertilizer+liquid amino acid) were set up to study the effects of amino acid fertilizer on flue-cured tobacco fertilizer reducing and efficiency increasing by means of contrast in the same field. [Result] The flue-cured tobacco applied solid amino acid fertilizer had the best performances of overall agronomic traits and the highest yield, while liquid amino acid fertilizer was conductive to the top leaf opening, making the raw tobacco appearance best, and could increase the proportion of middle and high quality tobacco, gaining good economic benefits. The flue-cured tobacco treated by liquid amino acid fertilizer had the optimal ratio of total sugar to nicotine and disaccharide differences in tobacco leaves, and the best coordination of chemical composition. Compared with the contrel, tobacco treated by amino acid fertilizer had less irritating and more comfortable taste, and the tobacco treated by liquid amino acid had light miscellaneous gas, and the best overall performance of sensory quality. [Conclusion] Chemical fertilizer+liquid amino acids is conductive to promote the growth and development of flue-cured tobacco and improve the quality of tobacco leaves.

文摘BACKGROUND The effect of hypoxia on mesenchymal stem cells(MSCs)is an emerging topic in MSC biology.Although long non-coding RNAs(lncRNAs)and messenger RNAs(mRNAs)are reported to play a critical role in regulating the biological characteristics of MSCs,their specific expression and co-expression profiles in human placenta-derived MSCs(hP-MSCs)under hypoxia and the underlying mechanisms of lncRNAs in hP-MSC biology are unknown.AIM To reveal the specific expression profiles of lncRNAs in hP-MSCs under hypoxia and initially explored the possible mechanism of lncRNAs on hP-MSC biology.METHODS Here,we used a multigas incubator(92.5%N_(2),5%CO_(2),and 2.5%O_(2))to mimic the potential of hP-MSCs.RNA sequencing technology was applied to identify the exact expression profiles of lncRNAs and mRNAs under hypoxia.RESULTS We identified 289 differentially expressed lncRNAs and 240 differentially expressed mRNAs between the hypoxia and normoxia groups.Among them,the lncRNA SNHG16 was upregulated under hypoxia,which was also validated by reverse transcription-polymerase chain reaction.SNHG16 was confirmed to affect hP-MSC proliferation rates using a SNHG16 knockdown model.SNHG16 overexpression could significantly enhance the proliferation capacity of hP-MSCs,activate the PI3K/AKT pathway,and upregulate the expression of cell cycle-related proteins.CONCLUSION Our results revealed the specific expression characteristics of lncRNAs and mRNAs in hypoxiacultured hP-MSCs and that lncRNA SNHG16 can promote hP-MSC proliferation through the PI3K/AKT pathway.
