Objective:To systematically evaluate the clinical efficacy of Kuntai Capsules combined with Diane-35 in the treatment of polycystic ovarian syndrome(PCOS).Methods:The randomized controlled trials(RCTs)of Kuntai Capsul...Objective:To systematically evaluate the clinical efficacy of Kuntai Capsules combined with Diane-35 in the treatment of polycystic ovarian syndrome(PCOS).Methods:The randomized controlled trials(RCTs)of Kuntai Capsules combined with Diane-35 in the treatment of PCOS from databases establishment to April 2020 were searched in Chinese and English databases,and then meta-analysis was performed using RecMan 5.3 software.Results:A total of 132 articles were collected and 14 studies were included.The total sample size was 1254 cases,including 627 cases in the experimental group and 627 cases in the control group.Meta-analysis showed that the combined group was significantly superior to the single group in terms of overall response rate(OR=5.53,95%CI[3.25,9.41],P<0.01),ovulation rate(OR=3.54,95%CI[2.13,5.88],P<0.01),and pregnancy rate(OR=3.21,95%CI[2.00,5.14],P<0.01).In terms of serum sex hormone levels,the combined group was significantly better than the singer group for the regulation of follicle-stimulating hormone(FSH)(MD=-2.09,95%CI[-2.16,-2.01],P<0.01),luteinizing hormone(LH)(MD=-2.15,95%CI[-2.35,-1.95],P<0.01),estrogen(E2)(MD=36.47,95%CI[34.97,37.96],P<0.01),and testosterone(T)(MD=-1.95,95%CI[-2.77,-1.14],P<0.01).In addition,the combined group also showed some advantages over the single group in improving the level of insulin resistance(HOMA-IR)and pancreatic beta cells function(HOMA-β).Conclusion:Kuntai Capsules combined with Diane-35 had certain efficacy advantage in the treatment of PCOS compared with control group.However,the data indicate that the methodological quality of the studies are low,there was potential publication bias,no define conclusion could be drawn,and more high-quality RCTs studies are needed for further validation.展开更多

Cyclooxygenase-2(COX-2) plays a critical role in OC development. This study was designed to evaluate the effects of COX-2 on migration and cisplatin(cis-dichloro diammine platinum, CDDP) resistance of OC cells and explore its related mechanisms. Methods: Cell counting kit-8(CCK-8) assay was used to detect the cytotoxicity effects of celecoxib(CXB) and CDDP on SKOV3 and ES2 cells. The effect of COX-2 on migration was evaluated via the healing test. Western blot and real-time quantitative polymerase chain reaction(q PCR) were used to analyze E-cadherin, vimentin, Snail, and Slug levels. Results: COX-2 promoted drug-resistance and cell migration. CXB inhibited these effects. The combination of CDDP and CXB increased tumor cell sensitivity, reduced the amount of CDDP required, and shortened treatment administration time. COX-2 upregulation increased the expression of Snail and Slug, resulting in E-cadherin expression downregulation and vimentin upregulation. Conclusions: COX-2 promotes cancer cell migration and CDDP resistance and may serve as a potential target for curing OC.展开更多

Background:The molecular mechanism underlying the involvement of the Transferrin receptor(TFRC)in cervical cancer remains poorly understood.This study aims to elucidate the role of TFRC in cervical cancer by analyzing...Background:The molecular mechanism underlying the involvement of the Transferrin receptor(TFRC)in cervical cancer remains poorly understood.This study aims to elucidate the role of TFRC in cervical cancer by analyzing data from The Cancer Genome Atlas(TCGA)and Genotype-Tissue Expression(GTEx)databases.Methods:TFRC protein expression was obtained from Human Protein Altas(HPA).All datas were collected from TCGA and GTEx.In this study,we analyzed the expression of TFRC in cervical cancer and its clinical significance.Through Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene set enrichment analyses(GSEA),investigated the related molecular pathways of TFRC.The relationship between TFRC and immune infiltration was then examined.The prognosis of different immune cell subsets was then analyzed after dividing cervical cancer patients into high and low expression of TFRC groups.Results:TFRC is highly expressed in various tumor tissues compared to control normal tissues,including cervical cancer.An increased expression of TFRC was associated with higher Tumor(T)and Node(N)stage,as well as a higher clinical stage.Kaplan–Meier(KM)survival analysis investigated that higher TFRC expression patients have a poor overall survival(OS),disease specific survival(DSS)and progress free interval(PFI).Both KEGG and GSEA enriched signaling pathway by high TFRC and low TFRC groups.There was a significant negative linear correlation between TFRC expression and immune infiltration.TFRC affects the prognosis of cervical cancer patients through immune pathway.Conclusions:Cervical cancer patients with TFRC expression may have a worse prognosis.展开更多
Reactive oxygen species(ROS)are highly prevalent in skin-related impairments and accelerate chronic ulcer progression.The routine subcutaneous administration approaches combining drug delivery with microenvironment in...Reactive oxygen species(ROS)are highly prevalent in skin-related impairments and accelerate chronic ulcer progression.The routine subcutaneous administration approaches combining drug delivery with microenvironment intervention are widely developed for skin-related treatment but lack effective outcomes.Herein,we present a cuttlefish ink-derived nanoparticles(CNPs)-integrated microneedles patch,silk fibroin and cuttlefish ink-derived melanin nanoparticles(SC-MNs),that can easily be inserted into the skin and alleviate ROS.The microneedle tips,formed from silk fibroin and treated with methanol vapor annealing,turn to increasedβ-sheet and enhanced mechanical strength.Meanwhile,the tips can rapidly detach from SC-MNs in mildly acidic conditions due to the introduction of NaHCO_(3).SC-MNs also exhibited a unique ROS obliteration capacity.Furthermore,under near-infrared irradiation,SC-MNs triggered photothermal performance,which elicited reliable tumor cell-killing effects.Collectively,these SC-MN patches described here can provide a promising platform for combined ROS-scavenging and photothermal therapy,which makes them a potential candidate in skin-related disease management.展开更多

The aim of this study was to compare the percentage of T cells, natural killer (NK) cells and natural killer T (NKT) cells within decidual parietalis of normal pregnant controls (NP) and patients with intraheptic cholestasis of pregnancy (ICP), and to investigate the production of interleukin-4 (IL-4), interferon-γ (IFN-γ) in the culture supernatant of decidual parietalis mononuclear cells (DPMCs). Compared with controls, the decidua parietalis from ICP were characterized with significant increased percentages of CD3^+CD56^+ cells, CD3^+CD56^+ cells, CD56^+CD16+ cells, CD56^+CD16^+ cells, CD56^+NKG2D+ ceils, and the significant decreased percentages of CD3^+ cells, CD3^+CD4^+ cells. There were no differences found for the percentage of CD3^+CD8^+ cells, CD56^+NKG2A^+ cells between control and study group. In addition, the enhanced concentration of IFN-γ was presented in culture supernatant of DPMCs from ICP. It was suggested that the increased NK cells, NKT cells and the decreased T cells in the decidual parietalis and over-secretion of IFN-γ could be correlated with the pathophysiology of ICP patients. In this study, to investigate whether NK cells and NKT cells, instead of T cells, are the dominant populations of peripheral blood in early pregnancy, flow cytometry was used to detect the percentage and intracellular cytokine expressions of T cells, NK cells, NKT cdls in peripheral blood of non-pregnant women and early pregnant women. In our result, the percentages of NK calls and NKT calls were significantly increased in pregnancy compared to non-pregnancy. However, the percentage of T cells was not changed. We did not detect the Th2-dominance of total lymphocytes or T cells in peripheral blood of early pregnant women and there were also no significant changes of type 1 and type 2 cytokines in T cells, but IFN-γ production in both NK and NKT cells was decreased in early pregnancy. These results suggest that the innate immune system including NK cells and NKT cells should play a pivotal role in pregnancy progression. Type 1/type 2 shift mechanisms in innate immune system during the human early pregnancy should be paid more attention.展开更多

However, CF is vulnerable to shilling attackers who inject fake user profi...Collaborative filtering (CF) is a technique commonly used for personalized recommendation and Web service quality-of-service (QoS) prediction. However, CF is vulnerable to shilling attackers who inject fake user profiles into the system. In this paper, we first present the shilling attack problem on CF-based QoS recommender systems for Web services. Then, a robust CF recommendation approach is proposed from a user similarity perspective to enhance the resistance of the recommender systems to the shilling attack. In the approach, the generally used similarity measures are analyzed, and the DegSim (the degree of similarities with top k neighbors) with those measures is selected for grouping and weighting the users. Then, the weights are used to calculate the service similarities/differences and predictions. We analyzed and evaluated our algorithms using WS-DREAM and Movielens datasets. The experimental results demonstrate that shilling attacks influence the prediction of QoS values, and our proposed features and algorithms achieve a higher degree of robustness against shilling attacks than the typical CF algorithms.展开更多
