P450 enzymes-catalyzed aromatic hydroxylation plays an important role in detoxification,biosynthesis,and potential carcinogenic effect of aromatic compounds.Though it has been explored for decades,the actual process o...P450 enzymes-catalyzed aromatic hydroxylation plays an important role in detoxification,biosynthesis,and potential carcinogenic effect of aromatic compounds.Though it has been explored for decades,the actual process of aromatic hydroxylation and mechanism of regioselectivity catalyzed by cytochrome P450 monooxygenases remained ambiguous.Here,we have resolved these issues.With a stable chiral organofluorine probe,and especially with X-ray data of two isolated arene oxides derivatives,we demonstrate that an arene oxide pathway is definitely involved in P450-catalyzed aromatic hydroxylation.By the capture,isolation,identification and reactivity exploration of the arene 1,2-oxide and arene 2,3-oxide intermediates,together with advanced QM calculations,the mechanism of how two intermediates go to the same product has been elucidated.In addition to the model substrate,we also confirmed that an arene oxide intermediate is involved in the P450-catalyzed hydroxylation pathway of a natural product derivative methyl cinnamate,which indicates that this intermediate appears to be universal in P450-catalyzed aromatic hydroxylation.Our work not only provides the most direct evidence for the arene oxide pathway and new insights into the regioselectivity involved in P450-catalyzed aromatic hydroxylation,but also supplies a new synthetic approach to achieve the dearomatization of aromatic compounds.展开更多
Due to the low transport resistance,mullite hollow fiber(HF)could be regarded as a promising support for preparation of NaA zeolite membranes.However,insufficient coverage and undesired seed penetration often occur in...Due to the low transport resistance,mullite hollow fiber(HF)could be regarded as a promising support for preparation of NaA zeolite membranes.However,insufficient coverage and undesired seed penetration often occur in the seeding step of the mullite support due to the rough surfaces with ultra-large macropores(2-4 μm).This leads to poor intergrowth of the membrane layer.To solve this issue,cationic polyacrylamide(CPAM)was employed to modify the chemical and electrostatic properties of the mullite HF surface,where the electrostatic attraction between the surface and seed crystals could significantly promote the coating effect.In the seeding,the original seeds(3.2 μm)were mixed with the ball-milled seeds(0.3 μm)to improve the seed coverage,i.e.,the original seed was used to block the macropores on the support surface to prevent the penetration of small seeds,while the decked ball-milled seed was used to provide sufficient active sites to induce the intergrowth of the zeolite crystals.The effects of the concentration of CPAM,mixed mass ratio of seeds,and seed concentration on the preparation of NaA zeolite membranes were systematically examined,where the separation performances were characterized by pervaporation(PV)dehydration of 90%(mass)ethanol/water mixtures at 75℃.The results indicated that under a CPAM concentration of 0.7%(mass),an equivalent mass ratio of mixed seeds in a seed concentration of 1.0%(mass),the prepared mullite HF supported NaA zeolite membrane yielded the best PV performance,where the permeation flux and separation factor corresponded to 5.45 kg·m^(-2)·h^(-1)and 10000,respectively,being sufficient for industrial applications.展开更多
The use of amine-based absorbers,such as N-methyldiethanolamine(MDEA),for CO_(2) capture presents a promising strategy for emission reduction as global warming intensifies.However,high energy consumption and limited a...The use of amine-based absorbers,such as N-methyldiethanolamine(MDEA),for CO_(2) capture presents a promising strategy for emission reduction as global warming intensifies.However,high energy consumption and limited absorption/desorption efficiencies constrain its application.To address these challenges,this study developed a composite catalyst derived from fly ash(FA),which is alkali-modified and incorporates nickel(Ni) and iron(Fe) elements,with the aim of enhancing CO_(2) absorption and desorption performance while reducing energy consumption.Experimental results indicated that the Ni/FA-AM catalyst significantly improved adsorption and absorption efficiencies,achieving an 8% increase in capacity,a 63% increase in peak adsorption rate,and a reduction of 6000 s in saturation time.Studies conducted in a wetted-wall column revealed that the absorption process predominantly occurs in the liquid phase.Additionally,the catalyst demonstrated a 19% improvement in desorption performance,a 10% increase in peak desorption rate,and a 24% reduction in energy consumption,while maintaining stability over five consecutive cycles.The alkali-modified and Ni/Fe-enriched fly ash was confirmed to form active acid-base sites,facilitating the formation and disappearance of bicarbonate,thereby enhancing CO_(2) capture efficiency.This was validated through XRD,BET,TPD,PY-IR,TEM,and FTIR with13C NMR characterization.This study highlights the potential of modified fly ash as a low-cost and efficient catalyst for CO_(2) capture.展开更多

目的:对比25G玻璃体切割联合人工晶状体经睫状沟巩膜缝线固定术和无缝线巩膜隧道层间固定术治疗晶状体完全脱位的疗效。方法:回顾性病例对照研究。选取2015-05/2021-09在我院确诊为晶状体完全脱位于玻璃体腔的患者40例40眼,其中行25G玻...目的:对比25G玻璃体切割联合人工晶状体经睫状沟巩膜缝线固定术和无缝线巩膜隧道层间固定术治疗晶状体完全脱位的疗效。方法:回顾性病例对照研究。选取2015-05/2021-09在我院确诊为晶状体完全脱位于玻璃体腔的患者40例40眼,其中行25G玻璃体切割联合人工晶状体经睫状沟巩膜缝线固定术者21眼(缝线组),行25G玻璃体切割联合人工晶状体无缝线巩膜隧道层间固定术者19眼(无缝线组)。随访至术后3mo,观察两组患者手术时间、最佳矫正视力(BCVA)、角膜内皮细胞计数(CECC)、中央角膜厚度(CCT)及术后并发症情况。结果:无缝线组手术用时显著短于缝线组(31.79±6.01min vs 45.38±8.04min,P<0.001)。两组患者术后BCVA均较术前显著改善(均P<0.05),术后1wk无缝线组BCVA(LogMAR)显著优于缝线组(0.32±0.14 vs 0.57±0.25,P<0.001)。术后3mo,两组患者CECC均低于术前(均P<0.01)。缝线组术后1wk CCT大于术前和术后3mo(均P<0.01),无缝线组手术前后CCT无显著变化。随访期间,无缝线组并发症总发生率低于缝线组(26%vs 38%,P>0.05)。结论:25G玻璃体切割联合人工晶状体无缝线巩膜隧道层间固定术治疗晶状体完全脱位手术时间短,术后视力提升快,术后并发症少。展开更多
Mixed-species plantations generally exhibit higher ecosystem multifunctionality than monospecific plantations.However,it is unclear how tree species functional composition influences species mixture effects on ecosyst...Mixed-species plantations generally exhibit higher ecosystem multifunctionality than monospecific plantations.However,it is unclear how tree species functional composition influences species mixture effects on ecosystem multifunctionality.We selected 171 monospecific and mixed-species plantations from nine regions across subtropical China,and quantified 13 key ecosystem functional properties to investigate how species mixture effects on ecosystem multifunctionality are modulated by functional diversity and identity.We found that ecosystem multifunctionality was significantly higher(p<0.05)in mixed tree plantations than in monospecific plantations except the mixed-conifer species plantations.Across all regions,ecosystem multifunctionality was significantly higher(p<0.05)in mixed conifer-broadleaf plantations than in monospecific plantations of the corresponding species,but not different between mixed and monospecific coniferous plantations.The magnitude of species mixture effects on ecosystem multifunctionality varied greatly with tree species compositions.Taking Cunninghamia lanceolata Lamb.as an example,the effects varied from a range of 2.0%–9.6%when mixed with a conifer species to 36%–87%when mixed with a broadleaf species.The functional diversity was the dominate driver shaping ecosystem multifunctionality,while functional identity,as expressed by community-weighted mean of specific leaf area,also had a positive effect on ecosystem multifunctionality through the increased below-ground nitrogen and phosphorus stocks regulated by specific leaf area of the mixing tree species.Our study highlights the important role of functional diversity in shaping ecosystem multifunctionality across region-wide environmental conditions.Mixed conifer-broadleaf tree plantations with distinct functional traits benefit the enhancement of ecosystem multifunctionality,and the magnitude of species mixture effects is modulated by the functional identity of tree species composition;those relationships deserve a special consideration in multifunctional management context of subtropical plantations.展开更多

Low reactivity coke A and high reactivity coke B were chosen and charged into the reaction tube in two methods. The results indicated that the mass loss ratio of high reactivity coke in mixed cokes was more significant than that of single high reactivity coke in the middle stage of reaction. Nevertheless, the mass loss ratio of low reactivity coke in mixed cokes was less than that of single low reactivity coke. It was mainly attributed to gas diffusion and internal reaction of coke. When high and low reactivity cokes were mixed, the practical average mass loss ratio was nearly the same as the weighted average. The microscopic structures of coke indicated that with the increase of reaction time, the external and internal layers of low reactivity coke reacted more uniformly with CO2, whereas the reaction degree of external layer of high reactivity coke was obviously higher.展开更多

From July 2000 to December 2011, 224 patients (mean age: 35.9...We retrospectively evaluated the clinical outcome of penile prosthesis implantation (PPI) in Chinese patients with severe erectile dysfunction (SED). From July 2000 to December 2011, 224 patients (mean age: 35.9±11.8 years, range: 20-75 years) with SED underwent PPI by experienced surgeon according to standard PPI procedure at our centre. A malleable prosthesis (AMS 650) was implanted in 45 cases (20.1%), and a three-piece inflatable prosthesis (AMS 700 CXM or AMS 700 CXR) was implanted in 179 cases (79.9%). Surgical outcomes, including postoperative complications, clinical efficacy and couple satisfaction, were evaluated over than 6 months postoperatively using medical record abstraction, IIEF-5, quality of life (QoL) scores, and the patient/partner sexual satisfaction score proposed by Bhojwani etaL Of the 224 patients eligible for the study, 201 subjects (89.7%) completed follow-up. All of patients could perform sexual intercourse post PPI with the mean postoperative IIEF-5 and QoL scores were 20.02±2.32 and 5.28±0.76, respectively, which were significantly improved compared with the preoperative scores (6.29±1.5 and 2.13±0.84, P〈0.01). Of the 201 men, mechanical malfunction occurred in four cases (2.0%) and three cases were re-implanted new device, and two cases (1.0%) developed a mild curvature of the penis. Scrotal erosion with infection occurred in one case with diabetes mellitus (0.5%) and required complete removal of the implanted AMS 700 CXM. Satisfactory sexual intercourse at least twice per month was reported by 178 men (88.6%), and overall satisfaction with the PPI surgery was reported by 89.0% of men and 82.5% of partners. Patient satisfaction in the three-piece inflatable prosthesis group was higher than in the malleable prosthesis group (P〈0.05). Satisfaction, however, between the types of prostheses, did not differ in the partner survey. PPI is a safe and effective treatment option for Chinese patients with SED and experienced surgeon perform PPI according to standard PPI procedure could reduce the postoperative complications of PPI and could improve patient satisfaction ratio and QoL.展开更多

Methods: According to the requirements of evidence-based medicine(EBM), the literature on acupuncture with distal and local point selection in recent 20 years included in domestic and overseas medical databases was retrieved, and meta-analysis of the included articles was carried out. Results: Thirty-two articles were included finally, totaling 2829 patients. The results of meta-analysis indicated that the curative effect of distal-local point combination was superior to that of simple distal point selection or local point selection during acupuncture–moxibustion treatment for diseases, and there was no obvious difference in the curative effect between simple distal point selection and local point selection. According to the comprehensive analysis of 18 included articles, the difference in total effective rate between distal point selection and local point selection was not statistically significant, i.e. the curative effect of distal point selection and local point selection was equivalent [OR=0.83,95%CI(0.83,1.18), Z=1.04, P0.01]. According to the comprehensive analysis of 20 included articles, the difference in total effective rate between local point selection and distal-local point selection was statistically significant, and the results indicated that the curative effect of distal-local point selection was superior to that of local point selection [OR=0.32, 95%CI(0.23,0.44), Z=6.90, P0.01]. According to the comprehensive analysis of 8 included articles, the difference in total effective rate between distal point selection and distal-local point selection was statistically significant, and the results indicated that the curative effect of distal-local point selection was superior to that of distal point selection [OR=0.20,95%CI(0.10,0.40), Z=4.50, P0.01]. In addition, the analysis of publication bias of this study indicated that publication bias might exist. Since the quality of included articles was generally lower, the above conclusion still needed to be supported by more evidence-based medicine proofs with high quality. Conclusion: The curative effect of distal-local point combination was superior to that of simple distal point selection or local point selection during acupuncture treatment for diseases, and there was no obvious difference in the curative effect between simple distal point selection and local point selection.展开更多

We aimed to investigate the protective effects of C. daurica extracts and phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. We discovered phenylethanoid glycosides from n-butanol extract with large content through extraction and separation. We continue to study the protective effects of phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. INS-1 cells were treated with C. daurica, cell viability assay, RNA-seq technology, superoxide dismutase activity and malonaldehyde content, quantitative real time-PCR and Western blot analysis were used to study the protective effects of C. daurica. Cell viability assay resulted that n-butanol extract and verbascoside, isoacteoside showed protective effects of C. daurica. According to the RNA-seq technology to identify the differentially expressed genes in INS-1 cells, the pathway of gene enrich the protective effect of C. daurica on oxidative stress. SOD activity and the content of MDA indicated that C. daurica could enhance the antioxidant capacity of INS-1 cells. Further investigation indicated C. daurica alleviate oxidative stress by inhibiting INS-1 cell apoptosis. C. daurica may play an anti-diabetic role by inhibiting islet cell apoptosis.展开更多
Since the outbreak of the novel corona virus disease 2019(COVID-19)at the end of 2019,specific antiviral drugs have been lacking.A Chinese patent medicine Toujiequwen granules has been promoted in the treatment of COV...Since the outbreak of the novel corona virus disease 2019(COVID-19)at the end of 2019,specific antiviral drugs have been lacking.A Chinese patent medicine Toujiequwen granules has been promoted in the treatment of COVID-19.The present study was designed to reveal the molecular mechanism of Toujiequwen granules against COVID-19.A network pharmacological method was applied to screen the main active ingredients of Toujiequwen granules.Network analysis of 149 active ingredients and 330 drug targets showed the most active ingredient interacting with many drug targets is quercetin.Drug targets most ffected by the active ingredients were PTGS2,PTGS1,and DPP4.Drug target disease enrichment analysis showed drug targets were significantly enriched in cardiovascular diseases and digestive tract diseases.An"active ingredient-target-disease"network showed that 57 active ingredients from Toujiequwen granules interacted with 15 key targets of COVID-19.There were 53 ingredients that could act on DPP4,suggesting that DPP4 may become a potential new key target for the treatment of COVID-19.GO analysis results showed that key targets were mainly enriched in the cellular response to lipopolysaccharide,cytokine activity and other functions.KEGG analysis showed they were mainly concentrated in viral protein interaction with cytokine and cytokine receptors and endocrine resistance pathway.The evidence suggests that Toujiequwen granules might play an effective role by improving the symptoms of underlying diseases in patients with COVID-19 and multi-target interventions against mutiple signaling pathways related to the pathogenesis of COVID-19.展开更多
BACKGROUND The formation of liver fibrosis is mainly caused by the activation of hepatic stellate cells(HSCs)and the imbalance of extracellular matrix(ECM)production and degradation.The treatment of liver fibrosis mai...BACKGROUND The formation of liver fibrosis is mainly caused by the activation of hepatic stellate cells(HSCs)and the imbalance of extracellular matrix(ECM)production and degradation.The treatment of liver fibrosis mainly includes removing the cause,inhibiting the activation of HSCs,and inhibiting inflammation.NOD-like receptor(NLR)family,caspase activation and recruitment domain(CARD)domain containing 5/NOD27/CLR16.1(NLRC5)is a highly conserved member of the NLR family and is involved in inflammation and immune responses by regulating various signaling pathways such as nuclear factor-κB(NF-κB)signaling.It has been found that NLRC5 plays an important role in liver fibrosis,but its specific effect and possible mechanism remain to be fully elucidated.AIM To investigate the role of NLRC5 in the activation and reversion of HSCs induced with transforming growth factor-β(TGF-β)and MDI,and to explore its relationship with liver fibrosis.METHODS A total of 24 male C57BL/6 mice were randomly divided into three groups,including normal,fibrosis,and recovery groups.Twenty-four hours after a liver fibrosis and spontaneous reversion model was established,the mice were sacrificed and pathological examination of liver tissue was performed to observe the degree of liver fibrosis in each group.LX-2 cells were cultured in vitro and treated with TGF-β1 and MDI.Real-time quantitative PCR(qPCR)and Western blot were used to analyze the expression levels of NLRC5,α-smooth muscle actin(α-SMA),and collagen type I alpha1(Col1a1)in each group.The activity of NF-κB in each group of cells transfected with NLRC5-siRNA was detected.RESULTS Compared with the normal mice,the expression level of NLRC5 increased significantly(P<0.01)in the fibrosis group,but decreased significantly in the recovery group(P<0.01).In in vitro experiments,the content of NLRC5 was enhanced after TGF-β1 stimulation and decreased to a lower level when treated with MDI(P<0.01).The expression ofα-SMA and Col1a1 proteins and mRNAs in TGF-β1-mediated cells was suppressed by transfection with NLRC5-siRNA(P<0.01).Western blot analysis showed that the expression of NF-κB p65 protein and phosphorylated IκBα(p-IκBα)was increased in the liver of mice in the fibrosis group but decreased in the recovery group(P<0.01),and the protein level of nuclear p65 and p-IκBαwas significantly increased after treatment with NLRC5-siRNA(P<0.01).CONCLUSION NLRC5 may play a key role in the development and reversal of hepatic fibrosis through the NF-κB signaling pathway,and it is expected to be one of the clinical therapeutic targets.展开更多
This study sought to investigate the clinical efficacy and safety of combined oral therapy with sildenafil and doxazosin GITS compared to sildenafil monotherapy in treating Chinese patients with erectile dysfunction ...This study sought to investigate the clinical efficacy and safety of combined oral therapy with sildenafil and doxazosin GITS compared to sildenafil monotherapy in treating Chinese patients with erectile dysfunction (ED) and lower urinary tract symptoms secondary to benign prostatic hyperplasia (BPH/LUTS). The trial was conducted in hospitals in Beijing, Shanghai, Changsha, Wuhan and Guangzhou, five major cities in China. A total of 250 patients diagnosed with ED and BPH/LUTS aged 50-75 years, and who had International Index of Erection Function-5 (IIEF-5) scores ≤ 21 and International Prostate Symptom Score (IPSS) ≥ 10 points, were enrolled and randomly divided into Group A (168 cases; doxazosin GITS 4 mg once daily plus sildenafi125-100 mg on demand) and Group B (82 cases; sildenafi125-100 mg on demand). Efficacies were evaluated by IIEF-5 and IPSS scores and a quality of life (QoL) questionnaire, and adverse effects were evaluated during the treatment period. There were no statistically significant differences in mean age, and IIEF-5, IPSS and QoL scores pre-treatment between the two groups. After treatment, IIEF-5, IPSS and QoL scores were significantly improved in Group A, while only IIEF-5 scores were significantly improved in Group B compared with pre-treatment. There were no significant differences in side effects between the two groups. The results indicated that combined therapy with sildenafil and doxazosin GITS for the treatment of ED and BPH/LUTS is safe and effective compared to sildenafil monotherapy.展开更多

DHCR24 overexpression confers neuroprotection against apoptosis caused by amyloid β deposition. The present study aimed to construct two recombinant adenoviruses driving DHCR24 expression specifically in neurons. Two SYN1 promoter DNA fragments were obtained from human (h) and rat (r). Recombinant Ad-r(h)SYN1-DHCR24 was transfected into AD-293, N2A (mouse neuroblastoma), and MIN6 (mouse pancreatic carcinoma) cells. Western blot analysis showed DHCR24 was specially expressed in 293 and N2A cells, but no specific band was found in MIN6 cells. This demonstrates that the recombinant adenoviruses successfully express DHCR24, and no expression is observed in non-neuronal cells. TUNEL assay results showed apoptosis was inhibited in adenovirus-transfected neurons. Detecting reactive oxygen species by immunoflu- orescence, we found that adenovirus transfection inhibits apoptosis through scavenging excess reactive oxygen species. Our findings show that the recombinant DHCR24 adenoviruses induce neuron-specific DHCR24 expression, and thereby lay the foundation for further studies on DHCR24 gene therapy for Alzheimer's disease.展开更多

Numerical analysis was used to develop solutions for these challenges. Based on an optimized furnace structure and operating conditions from numerical analysis, experiments were performed to grow monocrystalline silicon using the single-seed casting technique. The results revealed that this technique is highly superior to the popular high-performance multicrystalline and multiseed casting mono-like techniques.展开更多
Potentilla sericea is a heavy metal hyperaccumulator landscaping plant.MYB transcription factors play an important role in regulating plant stress response to adversity.However,there are few studies on MYB transcripti...Potentilla sericea is a heavy metal hyperaccumulator landscaping plant.MYB transcription factors play an important role in regulating plant stress response to adversity.However,there are few studies on MYB transcription factors in stress tolerance in Potentilla sericea.In this study,the PsMYB62 gene was successfully cloned from Potentilla sericea.Methods:Bioinformatic analysis and real-time quantitative PCR(qPCR)methods were used to evaluate this gene.The transgenic A.thaliana were obtained by flower dipping and the gene function was identified by determining physiological indicators under cadmium stress.Results:The open reading frame of PsMYB62 is 942 bp,which encodes 313 amino acids(aa)and belongs to the R2R3 MYB transcription factor.The plant overexpression vector PBI121-PsMYB62-GFP was constructed and successfully transferred into A.thaliana.The relative expression level of PsMYB62 was significantly increased by CdCl_(2),NaCl,ABA,and mannitol treatments.The germination rate of transgenic seeds was higher than those of wild type(WT)and empty vector(EV)under different concentrations of cadmium treatment.Upon treatment with 100μmol·L^(−1)of CdCl_(2)·2.5H_(2)O,the activities of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)in the transgenic plants were significantly higher than those in the WT and EV.The contents of H_(2)O_(2),O_(2)·−and malondialdehyde(MDA)in transgenic lines were increased,but lower than those in WT and EV.The expression levels of AtGSH,AtPCS,and AtNAS4 that were related to the regulation of cadmium were increased,but the expression levels of transgenic lines were higher than those of WT and EV.Conclusion:The above results showed that PsMYB62 could be induced by cadmium and could improve the cadmium resistance of plants.展开更多

文摘P450 enzymes-catalyzed aromatic hydroxylation plays an important role in detoxification,biosynthesis,and potential carcinogenic effect of aromatic compounds.Though it has been explored for decades,the actual process of aromatic hydroxylation and mechanism of regioselectivity catalyzed by cytochrome P450 monooxygenases remained ambiguous.Here,we have resolved these issues.With a stable chiral organofluorine probe,and especially with X-ray data of two isolated arene oxides derivatives,we demonstrate that an arene oxide pathway is definitely involved in P450-catalyzed aromatic hydroxylation.By the capture,isolation,identification and reactivity exploration of the arene 1,2-oxide and arene 2,3-oxide intermediates,together with advanced QM calculations,the mechanism of how two intermediates go to the same product has been elucidated.In addition to the model substrate,we also confirmed that an arene oxide intermediate is involved in the P450-catalyzed hydroxylation pathway of a natural product derivative methyl cinnamate,which indicates that this intermediate appears to be universal in P450-catalyzed aromatic hydroxylation.Our work not only provides the most direct evidence for the arene oxide pathway and new insights into the regioselectivity involved in P450-catalyzed aromatic hydroxylation,but also supplies a new synthetic approach to achieve the dearomatization of aromatic compounds.

文摘Due to the low transport resistance,mullite hollow fiber(HF)could be regarded as a promising support for preparation of NaA zeolite membranes.However,insufficient coverage and undesired seed penetration often occur in the seeding step of the mullite support due to the rough surfaces with ultra-large macropores(2-4 μm).This leads to poor intergrowth of the membrane layer.To solve this issue,cationic polyacrylamide(CPAM)was employed to modify the chemical and electrostatic properties of the mullite HF surface,where the electrostatic attraction between the surface and seed crystals could significantly promote the coating effect.In the seeding,the original seeds(3.2 μm)were mixed with the ball-milled seeds(0.3 μm)to improve the seed coverage,i.e.,the original seed was used to block the macropores on the support surface to prevent the penetration of small seeds,while the decked ball-milled seed was used to provide sufficient active sites to induce the intergrowth of the zeolite crystals.The effects of the concentration of CPAM,mixed mass ratio of seeds,and seed concentration on the preparation of NaA zeolite membranes were systematically examined,where the separation performances were characterized by pervaporation(PV)dehydration of 90%(mass)ethanol/water mixtures at 75℃.The results indicated that under a CPAM concentration of 0.7%(mass),an equivalent mass ratio of mixed seeds in a seed concentration of 1.0%(mass),the prepared mullite HF supported NaA zeolite membrane yielded the best PV performance,where the permeation flux and separation factor corresponded to 5.45 kg·m^(-2)·h^(-1)and 10000,respectively,being sufficient for industrial applications.

文摘The use of amine-based absorbers,such as N-methyldiethanolamine(MDEA),for CO_(2) capture presents a promising strategy for emission reduction as global warming intensifies.However,high energy consumption and limited absorption/desorption efficiencies constrain its application.To address these challenges,this study developed a composite catalyst derived from fly ash(FA),which is alkali-modified and incorporates nickel(Ni) and iron(Fe) elements,with the aim of enhancing CO_(2) absorption and desorption performance while reducing energy consumption.Experimental results indicated that the Ni/FA-AM catalyst significantly improved adsorption and absorption efficiencies,achieving an 8% increase in capacity,a 63% increase in peak adsorption rate,and a reduction of 6000 s in saturation time.Studies conducted in a wetted-wall column revealed that the absorption process predominantly occurs in the liquid phase.Additionally,the catalyst demonstrated a 19% improvement in desorption performance,a 10% increase in peak desorption rate,and a 24% reduction in energy consumption,while maintaining stability over five consecutive cycles.The alkali-modified and Ni/Fe-enriched fly ash was confirmed to form active acid-base sites,facilitating the formation and disappearance of bicarbonate,thereby enhancing CO_(2) capture efficiency.This was validated through XRD,BET,TPD,PY-IR,TEM,and FTIR with13C NMR characterization.This study highlights the potential of modified fly ash as a low-cost and efficient catalyst for CO_(2) capture.

In the receiver, these two hyperchaotic systems are adopted as response systems at the same time. The digital signals are recovered via comparing the discrete signals of the two error systems. Numerical simulations show the effectiveness of the method.
文摘目的:对比25G玻璃体切割联合人工晶状体经睫状沟巩膜缝线固定术和无缝线巩膜隧道层间固定术治疗晶状体完全脱位的疗效。方法:回顾性病例对照研究。选取2015-05/2021-09在我院确诊为晶状体完全脱位于玻璃体腔的患者40例40眼,其中行25G玻璃体切割联合人工晶状体经睫状沟巩膜缝线固定术者21眼(缝线组),行25G玻璃体切割联合人工晶状体无缝线巩膜隧道层间固定术者19眼(无缝线组)。随访至术后3mo,观察两组患者手术时间、最佳矫正视力(BCVA)、角膜内皮细胞计数(CECC)、中央角膜厚度(CCT)及术后并发症情况。结果:无缝线组手术用时显著短于缝线组(31.79±6.01min vs 45.38±8.04min,P<0.001)。两组患者术后BCVA均较术前显著改善(均P<0.05),术后1wk无缝线组BCVA(LogMAR)显著优于缝线组(0.32±0.14 vs 0.57±0.25,P<0.001)。术后3mo,两组患者CECC均低于术前(均P<0.01)。缝线组术后1wk CCT大于术前和术后3mo(均P<0.01),无缝线组手术前后CCT无显著变化。随访期间,无缝线组并发症总发生率低于缝线组(26%vs 38%,P>0.05)。结论:25G玻璃体切割联合人工晶状体无缝线巩膜隧道层间固定术治疗晶状体完全脱位手术时间短,术后视力提升快,术后并发症少。

文摘Mixed-species plantations generally exhibit higher ecosystem multifunctionality than monospecific plantations.However,it is unclear how tree species functional composition influences species mixture effects on ecosystem multifunctionality.We selected 171 monospecific and mixed-species plantations from nine regions across subtropical China,and quantified 13 key ecosystem functional properties to investigate how species mixture effects on ecosystem multifunctionality are modulated by functional diversity and identity.We found that ecosystem multifunctionality was significantly higher(p<0.05)in mixed tree plantations than in monospecific plantations except the mixed-conifer species plantations.Across all regions,ecosystem multifunctionality was significantly higher(p<0.05)in mixed conifer-broadleaf plantations than in monospecific plantations of the corresponding species,but not different between mixed and monospecific coniferous plantations.The magnitude of species mixture effects on ecosystem multifunctionality varied greatly with tree species compositions.Taking Cunninghamia lanceolata Lamb.as an example,the effects varied from a range of 2.0%–9.6%when mixed with a conifer species to 36%–87%when mixed with a broadleaf species.The functional diversity was the dominate driver shaping ecosystem multifunctionality,while functional identity,as expressed by community-weighted mean of specific leaf area,also had a positive effect on ecosystem multifunctionality through the increased below-ground nitrogen and phosphorus stocks regulated by specific leaf area of the mixing tree species.Our study highlights the important role of functional diversity in shaping ecosystem multifunctionality across region-wide environmental conditions.Mixed conifer-broadleaf tree plantations with distinct functional traits benefit the enhancement of ecosystem multifunctionality,and the magnitude of species mixture effects is modulated by the functional identity of tree species composition;those relationships deserve a special consideration in multifunctional management context of subtropical plantations.

文摘In order to effectively utilize the high reactivity coke, the gasification characteristics of high and low reactivity cokes were investigated at 1100 ℃. Low reactivity coke A and high reactivity coke B were chosen and charged into the reaction tube in two methods. The results indicated that the mass loss ratio of high reactivity coke in mixed cokes was more significant than that of single high reactivity coke in the middle stage of reaction. Nevertheless, the mass loss ratio of low reactivity coke in mixed cokes was less than that of single low reactivity coke. It was mainly attributed to gas diffusion and internal reaction of coke. When high and low reactivity cokes were mixed, the practical average mass loss ratio was nearly the same as the weighted average. The microscopic structures of coke indicated that with the increase of reaction time, the external and internal layers of low reactivity coke reacted more uniformly with CO2, whereas the reaction degree of external layer of high reactivity coke was obviously higher.

文摘We retrospectively evaluated the clinical outcome of penile prosthesis implantation (PPI) in Chinese patients with severe erectile dysfunction (SED). From July 2000 to December 2011, 224 patients (mean age: 35.9±11.8 years, range: 20-75 years) with SED underwent PPI by experienced surgeon according to standard PPI procedure at our centre. A malleable prosthesis (AMS 650) was implanted in 45 cases (20.1%), and a three-piece inflatable prosthesis (AMS 700 CXM or AMS 700 CXR) was implanted in 179 cases (79.9%). Surgical outcomes, including postoperative complications, clinical efficacy and couple satisfaction, were evaluated over than 6 months postoperatively using medical record abstraction, IIEF-5, quality of life (QoL) scores, and the patient/partner sexual satisfaction score proposed by Bhojwani etaL Of the 224 patients eligible for the study, 201 subjects (89.7%) completed follow-up. All of patients could perform sexual intercourse post PPI with the mean postoperative IIEF-5 and QoL scores were 20.02±2.32 and 5.28±0.76, respectively, which were significantly improved compared with the preoperative scores (6.29±1.5 and 2.13±0.84, P〈0.01). Of the 201 men, mechanical malfunction occurred in four cases (2.0%) and three cases were re-implanted new device, and two cases (1.0%) developed a mild curvature of the penis. Scrotal erosion with infection occurred in one case with diabetes mellitus (0.5%) and required complete removal of the implanted AMS 700 CXM. Satisfactory sexual intercourse at least twice per month was reported by 178 men (88.6%), and overall satisfaction with the PPI surgery was reported by 89.0% of men and 82.5% of partners. Patient satisfaction in the three-piece inflatable prosthesis group was higher than in the malleable prosthesis group (P〈0.05). Satisfaction, however, between the types of prostheses, did not differ in the partner survey. PPI is a safe and effective treatment option for Chinese patients with SED and experienced surgeon perform PPI according to standard PPI procedure could reduce the postoperative complications of PPI and could improve patient satisfaction ratio and QoL.
文摘To evaluate the effect of distal point selection, local point selection, and distal-local point selection on acupuncture efficacy. Methods: According to the requirements of evidence-based medicine(EBM), the literature on acupuncture with distal and local point selection in recent 20 years included in domestic and overseas medical databases was retrieved, and meta-analysis of the included articles was carried out. Results: Thirty-two articles were included finally, totaling 2829 patients. The results of meta-analysis indicated that the curative effect of distal-local point combination was superior to that of simple distal point selection or local point selection during acupuncture–moxibustion treatment for diseases, and there was no obvious difference in the curative effect between simple distal point selection and local point selection. According to the comprehensive analysis of 18 included articles, the difference in total effective rate between distal point selection and local point selection was not statistically significant, i.e. the curative effect of distal point selection and local point selection was equivalent [OR=0.83,95%CI(0.83,1.18), Z=1.04, P0.01]. According to the comprehensive analysis of 20 included articles, the difference in total effective rate between local point selection and distal-local point selection was statistically significant, and the results indicated that the curative effect of distal-local point selection was superior to that of local point selection [OR=0.32, 95%CI(0.23,0.44), Z=6.90, P0.01]. According to the comprehensive analysis of 8 included articles, the difference in total effective rate between distal point selection and distal-local point selection was statistically significant, and the results indicated that the curative effect of distal-local point selection was superior to that of distal point selection [OR=0.20,95%CI(0.10,0.40), Z=4.50, P0.01]. In addition, the analysis of publication bias of this study indicated that publication bias might exist. Since the quality of included articles was generally lower, the above conclusion still needed to be supported by more evidence-based medicine proofs with high quality. Conclusion: The curative effect of distal-local point combination was superior to that of simple distal point selection or local point selection during acupuncture treatment for diseases, and there was no obvious difference in the curative effect between simple distal point selection and local point selection.

文摘Cymbaria daurica L. is a well-known traditional Mongolian medicine, which has been used to treat diabetesrelated conditions characterized by persistent thirst and hunger, copious urination, and weight loss. We aimed to investigate the protective effects of C. daurica extracts and phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. We discovered phenylethanoid glycosides from n-butanol extract with large content through extraction and separation. We continue to study the protective effects of phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. INS-1 cells were treated with C. daurica, cell viability assay, RNA-seq technology, superoxide dismutase activity and malonaldehyde content, quantitative real time-PCR and Western blot analysis were used to study the protective effects of C. daurica. Cell viability assay resulted that n-butanol extract and verbascoside, isoacteoside showed protective effects of C. daurica. According to the RNA-seq technology to identify the differentially expressed genes in INS-1 cells, the pathway of gene enrich the protective effect of C. daurica on oxidative stress. SOD activity and the content of MDA indicated that C. daurica could enhance the antioxidant capacity of INS-1 cells. Further investigation indicated C. daurica alleviate oxidative stress by inhibiting INS-1 cell apoptosis. C. daurica may play an anti-diabetic role by inhibiting islet cell apoptosis.

文摘Since the outbreak of the novel corona virus disease 2019(COVID-19)at the end of 2019,specific antiviral drugs have been lacking.A Chinese patent medicine Toujiequwen granules has been promoted in the treatment of COVID-19.The present study was designed to reveal the molecular mechanism of Toujiequwen granules against COVID-19.A network pharmacological method was applied to screen the main active ingredients of Toujiequwen granules.Network analysis of 149 active ingredients and 330 drug targets showed the most active ingredient interacting with many drug targets is quercetin.Drug targets most ffected by the active ingredients were PTGS2,PTGS1,and DPP4.Drug target disease enrichment analysis showed drug targets were significantly enriched in cardiovascular diseases and digestive tract diseases.An"active ingredient-target-disease"network showed that 57 active ingredients from Toujiequwen granules interacted with 15 key targets of COVID-19.There were 53 ingredients that could act on DPP4,suggesting that DPP4 may become a potential new key target for the treatment of COVID-19.GO analysis results showed that key targets were mainly enriched in the cellular response to lipopolysaccharide,cytokine activity and other functions.KEGG analysis showed they were mainly concentrated in viral protein interaction with cytokine and cytokine receptors and endocrine resistance pathway.The evidence suggests that Toujiequwen granules might play an effective role by improving the symptoms of underlying diseases in patients with COVID-19 and multi-target interventions against mutiple signaling pathways related to the pathogenesis of COVID-19.
文摘BACKGROUND The formation of liver fibrosis is mainly caused by the activation of hepatic stellate cells(HSCs)and the imbalance of extracellular matrix(ECM)production and degradation.The treatment of liver fibrosis mainly includes removing the cause,inhibiting the activation of HSCs,and inhibiting inflammation.NOD-like receptor(NLR)family,caspase activation and recruitment domain(CARD)domain containing 5/NOD27/CLR16.1(NLRC5)is a highly conserved member of the NLR family and is involved in inflammation and immune responses by regulating various signaling pathways such as nuclear factor-κB(NF-κB)signaling.It has been found that NLRC5 plays an important role in liver fibrosis,but its specific effect and possible mechanism remain to be fully elucidated.AIM To investigate the role of NLRC5 in the activation and reversion of HSCs induced with transforming growth factor-β(TGF-β)and MDI,and to explore its relationship with liver fibrosis.METHODS A total of 24 male C57BL/6 mice were randomly divided into three groups,including normal,fibrosis,and recovery groups.Twenty-four hours after a liver fibrosis and spontaneous reversion model was established,the mice were sacrificed and pathological examination of liver tissue was performed to observe the degree of liver fibrosis in each group.LX-2 cells were cultured in vitro and treated with TGF-β1 and MDI.Real-time quantitative PCR(qPCR)and Western blot were used to analyze the expression levels of NLRC5,α-smooth muscle actin(α-SMA),and collagen type I alpha1(Col1a1)in each group.The activity of NF-κB in each group of cells transfected with NLRC5-siRNA was detected.RESULTS Compared with the normal mice,the expression level of NLRC5 increased significantly(P<0.01)in the fibrosis group,but decreased significantly in the recovery group(P<0.01).In in vitro experiments,the content of NLRC5 was enhanced after TGF-β1 stimulation and decreased to a lower level when treated with MDI(P<0.01).The expression ofα-SMA and Col1a1 proteins and mRNAs in TGF-β1-mediated cells was suppressed by transfection with NLRC5-siRNA(P<0.01).Western blot analysis showed that the expression of NF-κB p65 protein and phosphorylated IκBα(p-IκBα)was increased in the liver of mice in the fibrosis group but decreased in the recovery group(P<0.01),and the protein level of nuclear p65 and p-IκBαwas significantly increased after treatment with NLRC5-siRNA(P<0.01).CONCLUSION NLRC5 may play a key role in the development and reversal of hepatic fibrosis through the NF-κB signaling pathway,and it is expected to be one of the clinical therapeutic targets.
文摘This study sought to investigate the clinical efficacy and safety of combined oral therapy with sildenafil and doxazosin GITS compared to sildenafil monotherapy in treating Chinese patients with erectile dysfunction (ED) and lower urinary tract symptoms secondary to benign prostatic hyperplasia (BPH/LUTS). The trial was conducted in hospitals in Beijing, Shanghai, Changsha, Wuhan and Guangzhou, five major cities in China. A total of 250 patients diagnosed with ED and BPH/LUTS aged 50-75 years, and who had International Index of Erection Function-5 (IIEF-5) scores ≤ 21 and International Prostate Symptom Score (IPSS) ≥ 10 points, were enrolled and randomly divided into Group A (168 cases; doxazosin GITS 4 mg once daily plus sildenafi125-100 mg on demand) and Group B (82 cases; sildenafi125-100 mg on demand). Efficacies were evaluated by IIEF-5 and IPSS scores and a quality of life (QoL) questionnaire, and adverse effects were evaluated during the treatment period. There were no statistically significant differences in mean age, and IIEF-5, IPSS and QoL scores pre-treatment between the two groups. After treatment, IIEF-5, IPSS and QoL scores were significantly improved in Group A, while only IIEF-5 scores were significantly improved in Group B compared with pre-treatment. There were no significant differences in side effects between the two groups. The results indicated that combined therapy with sildenafil and doxazosin GITS for the treatment of ED and BPH/LUTS is safe and effective compared to sildenafil monotherapy.

DHCR24 overexpression confers neuroprotection against apoptosis caused by amyloid β deposition. The present study aimed to construct two recombinant adenoviruses driving DHCR24 expression specifically in neurons. Two SYN1 promoter DNA fragments were obtained from human (h) and rat (r). Recombinant Ad-r(h)SYN1-DHCR24 was transfected into AD-293, N2A (mouse neuroblastoma), and MIN6 (mouse pancreatic carcinoma) cells. Western blot analysis showed DHCR24 was specially expressed in 293 and N2A cells, but no specific band was found in MIN6 cells. This demonstrates that the recombinant adenoviruses successfully express DHCR24, and no expression is observed in non-neuronal cells. TUNEL assay results showed apoptosis was inhibited in adenovirus-transfected neurons. Detecting reactive oxygen species by immunoflu- orescence, we found that adenovirus transfection inhibits apoptosis through scavenging excess reactive oxygen species. Our findings show that the recombinant DHCR24 adenoviruses induce neuron-specific DHCR24 expression, and thereby lay the foundation for further studies on DHCR24 gene therapy for Alzheimer's disease.

文摘To grow high-quality and large-size monocrystalline silicon at low cost, we proposed a single-seed casting technique. To realize this technique, two challenges—polycrystalline nucleation on the crucible wall and dislocation multiplication inside the crystal—needed to be addressed. Numerical analysis was used to develop solutions for these challenges. Based on an optimized furnace structure and operating conditions from numerical analysis, experiments were performed to grow monocrystalline silicon using the single-seed casting technique. The results revealed that this technique is highly superior to the popular high-performance multicrystalline and multiseed casting mono-like techniques.

文摘Potentilla sericea is a heavy metal hyperaccumulator landscaping plant.MYB transcription factors play an important role in regulating plant stress response to adversity.However,there are few studies on MYB transcription factors in stress tolerance in Potentilla sericea.In this study,the PsMYB62 gene was successfully cloned from Potentilla sericea.Methods:Bioinformatic analysis and real-time quantitative PCR(qPCR)methods were used to evaluate this gene.The transgenic A.thaliana were obtained by flower dipping and the gene function was identified by determining physiological indicators under cadmium stress.Results:The open reading frame of PsMYB62 is 942 bp,which encodes 313 amino acids(aa)and belongs to the R2R3 MYB transcription factor.The plant overexpression vector PBI121-PsMYB62-GFP was constructed and successfully transferred into A.thaliana.The relative expression level of PsMYB62 was significantly increased by CdCl_(2),NaCl,ABA,and mannitol treatments.The germination rate of transgenic seeds was higher than those of wild type(WT)and empty vector(EV)under different concentrations of cadmium treatment.Upon treatment with 100μmol·L^(−1)of CdCl_(2)·2.5H_(2)O,the activities of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)in the transgenic plants were significantly higher than those in the WT and EV.The contents of H_(2)O_(2),O_(2)·−and malondialdehyde(MDA)in transgenic lines were increased,but lower than those in WT and EV.The expression levels of AtGSH,AtPCS,and AtNAS4 that were related to the regulation of cadmium were increased,but the expression levels of transgenic lines were higher than those of WT and EV.Conclusion:The above results showed that PsMYB62 could be induced by cadmium and could improve the cadmium resistance of plants.
文摘Based on the database of Chinese industrial industries,a model is constructed to empirically analyze the interaction between knowledge spillovers and R&D in manufacturing industries;the mean productivity values of county and city regions have a significant positive effect on firms'R&D,which gradually decreases;an interaction term between the number of neighboring firms and the average total factor productivity of industries in different regional scopes is added,and the greater the number of neighboring firms in the neighborhood,the greater the spillover effect on research and development.In order to increase the innovation input of companies,they need to be given the space to folly exchange ideas.
文摘During the 14th Five Year Plan period,the main task of talent team construction in China’s asset appraisal industry was to develop innovative talent training models.Therefore,this article focuses on the talent cultivation model of integrating industry and education in asset evaluation in universities,systematically summarizes the theoretical and practical significance of research on asset evaluation talent cultivation models in universities,and explores the construction measures of asset evaluation talent cultivation models based on the integration of industry and education that meet social needs and the development of the times[1].At the same time,the strategy of constructing a deep integration talent training system was explored,guided by the integration of industry and education,to cultivate asset evaluation composite talents with strong practical skills.The aim is to provide a reference for improving the quality of asset evaluation professionals in China and promoting the development of asset evaluation talent training models.

文摘The charge transport and accumulation in oil-paper can cause the insulation degradation.So far,the most widely used model to simulate space charge transport and accumulation is the bipolar charge transport(BCT)model,which can well simulate the space charge dynamics.However,there are two shortcomings in the algorithms for solving the BCT model.One is that there is almost no use of vectorisation technology,which may increase the complexity of the algorithm,the other is the usage of fixed step size which might bring extra computation cost.In view of this,an adaptive time‐stepping transient up-stream finite element method(FEM)is developed to solve the BCT model considering trapping/detrapping,as well as the recombination phenomenon under DC condition in this article.Then,a vectorisation technology is used in the method to optimise the al-gorithm.Moreover,the adaptive time‐stepping method is introduced in simulation to reduce computation time and calculation amount.Simulation results are obtained by programing and later presented,which are basically consistent with the corresponding experimental results.Therefore,the proposed method is expected to promote the opti-mization design of the oil-paper insulation system.
