Scrub typhus is an acute undifferentiated febrile infectious disease transmitted by a chigger(genus Leptotrombidium)bite carrying Orientia(O.)tsutsugamushi, affecting millions of people annually while more than one billion people are susceptible.Endemic areas are expanding to Africa,Europe,Middle East, and South America which is concerning,as despite best efforts, there is no vaccine to combat the bacteria. There are now three species of Orientia and over 20 strains of O.tsutsugamushi.The past attempts to develop a vaccine have been ineffective as they confer homologous strain-specific immunity.Various immunogenic proteins of O.tsutsugamushi have been identified that interact with the extracellular matrix(fibronectin)or vMLL5 receptor and modify the cytoskeleton of non-phagocytic host cells, which aids in host cell adhesion and invasion. These highly conserved proteins involve type specific antigen 56(TSA56),47 kDa,OmpA,and autotransporter proteins(ScaA,ScaB and ScaC).TSA56 is the most immunogenic and contains four types of hypervariable regions.Out of all autotransporter proteins,ScaA provides the homologous strains specific immunity and when coupled with TSA56 it shows better protective immunity against heterologous strains. The review provides detailed insight into the potential immunogenic proteins of Orientia which can be utilized to develop the vaccine.Furthermore, studies focused on highly antigenic proteins will provide more insight into their roles in developing therapeutics and easy-to-handle rapid diagnostic kits. 展开更多

Pesticides have the potential to leave harmful effects on humans, animals, other living organisms, and the environment. Several human metabolic proteins inhibited after exposure to organophosphorus pesticides absorbed through the skin, inhalation, eyes and oral mucosa, are most important targets for this interaction study. The crystal structure of five different proteins, PDBIDs: 3LII, 3NXU, 4GTU, 2XJ1 and 1YXA in Homo sapiens (H. sapiens), interact with organophosphorus pesticides at the molecular level. The 3-D structures were found to be of good quality and validated through PROCHECK, ERRAT and ProSA servers. The results show that the binding energy is maximum -45.21 relative units of cytochrome P450 protein with phosmet pesticide. In terms of H-bonding, methyl parathion and parathion with acetylcholinesterase protein, parathion, methylparathion and phosmet with protein kinase C show the highest interaction. We conclude that these organophosphorus pesticides are more toxic and inhibit enzymatic activity by interrupting the metabolic pathways in H. sapiens. 展开更多

The purpose of this study was to investigate the nuclear magnetic resonance (NMR) assignments of hydrolyzed products extracted from human blood plasma. The correlations between chemical, functional and structural prop- erties of highly toxic pesticides were investigated using the PreADME analysis. We observed that toxic pesticides possessed higher molecular weight and, more hydrogen bond donors and acceptors when compared with less toxic pesticides. The occurrence of functional groups and structural properties was analyzed using ~H-NMR. The ~H- NMR spectra of the phosphomethoxy class of pesticides were characterized by methyl resonances at 3.7-3.9 ppm (8) with the coupling constants of 11-16 Hz (Je-cns). In phosphoethoxy pesticides, the methyl resonance was about 1.4 ppm (8) with the coupling constant of 10 Hz (Je-cH2) and the methylene resonances was 4.2-4.4 ppm (8) with the coupling constant of 0.8 Hz (Jp-cH3), respectively. Our study shows that the values of four parameters such as chemical shift, coupling constant, integration and relaxation time correlated with the concentration of toxic pesticides, and can be used to characterise the proton groups in the molecular structures of toxic pesticides. 展开更多

Groundwater assists farmers to irrigate crops for fulfilling the crop-water requirement.Indian agriculture system is characterized by three cropping seasons known as Kharif(monsoon),Rabi(post-monsoon)and summer(pre-monsoon).In tropical countries like India, monitoring cropping practices using optical remote sensing during Kharif and Rabi seasons is constraint due to the cloud cover, which can be well addressed by microwave remote sensing. In the proposed research, the strength of C-band polarimetric Synthetic Aperture Radar(SAR)time series images were evaluated to classify groundwater irrigated croplands for the Kharif and Rabi cropping seasons of the year 2013.The present study was performed in the Berambadi experimental watershed of Kabini river basin, southern peninsular India.A total of fifteen polarimetric variables were estimated includes four backscattering coefficients(HH,HV,VH,VV)and eleven polarimetric indices for all Radarsat-2 SAR images. The cumulative temporal sum(seasonal and dual-season)of these parameters was supervised classified using Support Vector Machine(SVM)classifier with intensive ground observation samples.Classification results using the best equation(highest accuracy and kappa)shows that the Kharif,Rabi and irrigated double croplands are respectively 9.58 km2(20.6%),16.14 km2(34.7%)and 6.22 km2(13.4%)with a kappa coefficient respectively 0.84,0.74 and 0.94. 展开更多

The curation of genetic variants from biomedical articles is required for various clinical and research purposes. Nowadays, establishment of variant databases that include overall information about variants is becoming quite popular. These databases have immense utility, serving as a user-friendly information storehouse of variants for information seekers. While manual curation is the gold standard method for curation of variants, it can turn out to be time-consuming on a large scale thus necessitating the need for automation. Curation of variants described in biomedical literature may not be straightforward mainly due to various nomenclature and expression issues. Though current trends in paper writing on variants is inclined to the standard nomenclature such that variants can easily be retrieved, we have a massive store of variants in the literature that are present as non-standard names and the online search engines that are pre- dominantly used may not be capable of finding them. For effective curation of variants, knowledge about the overall process of curation, nature and types of difficulties in curation, and ways to tackle the difficulties during the task are crucial. Only by effective curation, can variants be correctly interpreted. This paper presents the process and difficulties of curation of genetic variants with possible solutions and suggestions from our work experience in the field including literature support. The paper also highlights aspects of interpretation of genetic variants and the importance of writing papers on variants following standard and retrievable methods. 展开更多
