To separate and redefine the ambiguous Holosticha-complex, a confusing group of hypotrichous ciliates, six strains belonging to five morphospecies of three genera, Holosticha heterofoissneri, Anteholosticha sp. popl, ...To separate and redefine the ambiguous Holosticha-complex, a confusing group of hypotrichous ciliates, six strains belonging to five morphospecies of three genera, Holosticha heterofoissneri, Anteholosticha sp. popl, Anteholosticha sp. pop2, A. manca, A. gracilis and Nothoholostichafasciola, were analyzed using 12 restriction enzymes on the basis of amplified ribosomal DNA restriction analysis. Nine of the 12 enzymes could digest the DNA products, four (HinfⅠ, Hind Ⅲ, Msp Ⅰ, Taq Ⅰ) yielded species-specific restriction patterns, and Hind Ⅲ and Taq Ⅰ produced different pattems for two Anteholosticha sp. populations. Distinctly different restriction digestion haplotypes and similarity indices can be used to separate the species. The secondary structures of the five species were predicted based on the ITS2 transcripts and there were several minor differences among species, while two Anteholosticha sp. populations were identical. In addition, phylogenies based on the SSrRNA gene sequences were reconstructed using multiple algorithms, which grouped them generally into four clades, and exhibited that the genus Anteholosticha should be a convergent assemblage. The fact that Holosticha species clustered with the oligotrichs and choreotrichs, though with very low support values, indicated that the topology may be very divergent and unreliable when the number of sequence data used in the analyses is too low.展开更多
Variations in physical-chemical factors, species composition, abundance and biomass of nano-and micro-phytoplankton assemblages, as well as their responses to environmental factors, were investigated over a complete c...Variations in physical-chemical factors, species composition, abundance and biomass of nano-and micro-phytoplankton assemblages, as well as their responses to environmental factors, were investigated over a complete cycle (6 months) in a semi-enclosed shrimp-farming pond near Qingdao, northern China. The aim was to establish the temporal patterns of phytoplankton communities and to evaluate protists as suitable bioindicators to water quality in mariculture systems. A total of 34 taxa with nine dominant species were identified, belonging to six taxonomic groups (dinoflagellates, diatoms, cryptophyceans, chlorophyceans, euglenophyceans and chrysophyceans). A single peak of protist abundance occurred in October, mainly due to chlorophyceans, diatoms and chrysophyceans. Two biomass peaks in July and October were primarily due to dinoflagellates and diatoms. Temporal patterns of the phytoplankton communities significantly correlated with the changes in nutrients, temperature and pH, especially phosphate, either alone or in combination with NO3-N and NH3-N. Species diversity, evenness and richness indices were clearly correlated with water temperature and/or salinity, whereas the biomass/abundance ratio showed a significant correlation with NO3-N. The results suggest that phytoplankton are potentially useful bioindicators to water quality in semi-enclosed mariculture systems.展开更多
The ciliate genus Protocruzia belongs to one of the most ambiguous taxa considering its systematic position,possibly as a member of the classes Heterotrichea,Spirotrichea or Karyorelictea,which is tentatively placed i...The ciliate genus Protocruzia belongs to one of the most ambiguous taxa considering its systematic position,possibly as a member of the classes Heterotrichea,Spirotrichea or Karyorelictea,which is tentatively placed into Spirotrichea in Lynn’s 2008 system.To test these hypotheses,multigene trees(Bayesian inference,evolutionary distance,maximum parsimony,and maximum likelihood) were constructed using the small subunit rRNA(SSU rRNA) gene,internal transcribed spacer 2(ITS2) and a protein coding gene(histone H4).All analyses agree that:(1) four morphotypes of Protocruzia from different geographical origins group together and form a monophyletic clade,which cannot be assigned to any of the eleven described ciliate classes;(2) it is invariably positioned on an isolated branch separated from the class Spirotrichea suggesting that this clade should be clearly removed from Spirotrichea;(3) this leads us to hypothesize that this taxon may indeed represent a lineage on a class rank.Based on the fact that it is,both morphologically and in molecular features,closely related to the heterotrichs,Colpodea and Oligohymenophorea,Protocruziida might be an ancestral form for the subphylum Intramacronucleata in the evolutionary line from the class Heterotrichea(subphylum Postciliodesmatophora) to higher taxa.展开更多
