Bacterial and cyanobacterial diversities determined by T-RFLP analyses in the Jiaozhou Bay 被引量:4

The methods of DAPI staining epifluorescence microscopy and T-RFLP analysis were used to analyze the microbial abundance and diversity in surface seawater sampled from 12 stations inside and outside of the Jiaozhou Bay during a survey on 12 and 13 September 2004. The abundance of total microbes is in the range of 10^6~ 10^7 cells/cm^3, similar to those of most semi-enclosed bays in the temperate areas in the world. The highest microbial densities occur in the northeastern part of the Jiaozhou Bay, around the mouths of Loushan and Licun Rivers and the Hongdao aquacultural farming areas, suggesting that the degree and characteristics of pollutions, along with geographical and hydrological effects, may be important determinants affecting the abundance and distribution of bacteria in the Jiaozhou Bay. Bacterial communities inside and outside of the Jiaozhou Bay can be grouped into three classes based on T-RFLP patterns and cluster analyses. Stations at the water channel of the bay mouth and outside, such as D1, D3, D5, D6 and D7, are grouped together to stand for the outside bacterial community interacting with the environment outside of the Jiaozhou Bay. Stations of the innermost side of the Jiaozhou Bay, such as A3, A5, B2 and Y1, are grouped together to stand for the residential bacteria community. Stations C1, C3 and CA are grouped together and may stand for the transitional bacterial assemblage between the residential community and the outside community. However, there is no such a defined relationship for the case of cyanobacterial diversity, indicating the fact that cyanobacteria are more flexible and adaptable to all kinds of conditions. 展开更多

目的 对比肌肉骨骼超声与磁共振成像(MRI)诊断类风湿性膝关节炎及评估其活动度的价值。方法 对类风湿性关节炎累及膝关节患者122例行肌肉骨骼超声及MRI检查,以关节镜检查结果为“金标准”,比较肌肉骨骼超声及MRI对类风湿性膝关节炎的诊... 目的 对比肌肉骨骼超声与磁共振成像(MRI)诊断类风湿性膝关节炎及评估其活动度的价值。方法 对类风湿性关节炎累及膝关节患者122例行肌肉骨骼超声及MRI检查,以关节镜检查结果为“金标准”,比较肌肉骨骼超声及MRI对类风湿性膝关节炎的诊断价值,并对所有患者进行疾病活动度分级,观察类风湿性膝关节炎疾病活动度与滑膜厚度、关节积液及血流信号分级的相关性。结果 122例患者244个膝关节,经关节镜明确受累关节为152个(62.30%),滑膜增厚138个(90.79%),均伴有关节腔积液,关节骨质改变113个(74.34%),骨侵蚀95个(62.50%),血管翳形成113个(74.34%)。肌肉骨骼超声正确检出受累关节136个,准确率为89.47%(136/152),MRI正确检出受累关节135个,准确率为88.82%(135/152),2种检查方式对滑膜增生、骨质改变、骨侵蚀及血管翳检出率比较,差异无统计学意义(P>0.05),但肌肉骨骼超声关节积液检出率高于MRI(P<0.05)。肌肉骨骼超声及MRI诊断类风湿性膝关节炎的特异度、敏感度、诊断符合率、Kappa值均较高。类风湿性膝关节炎患者DAS28评分与血流分级、滑膜血流分级、髌上囊积液厚度均呈正相关(P<0.05)。结论 肌肉骨骼超声及MRI对类风湿性膝关节炎均具有较高的诊断价值,但与MRI相比,肌肉骨骼超声对关节积液的诊断价值较高,二者均可通过对滑膜厚度、髌上囊积液厚度及血流分级的检测来判断膝关节疾病活动度,对临床诊疗及病情监测具有一定的借鉴意义。 展开更多

为了解猪链球菌2型菌株对昆明小鼠的致病性,并建立其感染昆明小鼠模型,本试验分别取2头疑似患链球菌病的病死猪脑组织、血液分别涂布于含5%胎牛血清的TSA固体培养基中进行细菌分离;并对分离获得的细菌进行形态观察、PCR鉴定、药物敏感... 为了解猪链球菌2型菌株对昆明小鼠的致病性,并建立其感染昆明小鼠模型,本试验分别取2头疑似患链球菌病的病死猪脑组织、血液分别涂布于含5%胎牛血清的TSA固体培养基中进行细菌分离;并对分离获得的细菌进行形态观察、PCR鉴定、药物敏感性试验、小鼠致病性试验及组织病理切片观察。结果显示,分离菌为革兰氏阳性短链球菌,经PCR鉴定为猪链球菌2型,命名为LY株;药敏试验证实LY株对青霉素、红霉素、氯霉素、苯唑西林、环丙沙星、诺氟沙星敏感,对四环素、多粘菌素B、克林霉素不敏感;LY株对小鼠致死剂量为2.2×10^(9) cfu/只;组织病理切片观察发现小鼠肺脏、脾脏及心肌均出现不同程度的病变。本研究明确了山东省猪链球2型毒株的毒力,为疫苗候选菌株的筛选提供了理论依据。 展开更多

The influencing factors of extracellular polysac-charide(EPS)produced from a strain of lactic acid bacteria(LAB L15)were studied by using the phenol-H2SO4 method.It was demonstrated that the strain produced EPS at the most amount when it was incubated for 40-48 h and when the pH value was 4 under 30℃.Glucose was the most suitable carbon source for LAB-producing EPS.The rough EPS was obtained from L15 culture after centrifugation,dialysis,deprotein,decoloration,and ethanol-precipitation.The sample was at least composed of two polysaccharides that were completely different in molecular weight and the amount.The purified EPS was passed through the SephadexG-200 column and it showed that it was a sample purified by thin layer chromatography. 展开更多
