钙调神经磷酸酶(Calcineurin,CN)是唯一的Ca2+/钙调素(Camodulin,CaM)依赖的蛋白磷酸酶,由催化亚基A(CNA, 61 ku)和调节亚基B(CNB, 19 ku) 1:1组成.用 PCR方法构建... 钙调神经磷酸酶(Calcineurin,CN)是唯一的Ca2+/钙调素(Camodulin,CaM)依赖的蛋白磷酸酶,由催化亚基A(CNA, 61 ku)和调节亚基B(CNB, 19 ku) 1:1组成.用 PCR方法构建了仅含CNA催化结构域的编码区cDNA序列.经在E.coli中诱导表达,纯化,制备了CNA催化结构域多肽.以PNPP为底物,研究了其磷酸酶活力.结果表明,在有CaM和CNB时,其活性为CNA活性的20倍.CNA催化结构域多肽的最适反应温度为40℃,其最适反应pH值为8.0.不同离子对其活性的影响为: Mn2+仍为其强激活剂,但 CNA催化结构域多肽不再受 Ca2+的调节.在 6 mmol/LMg2+存在下,加Ca2+或EGTA,催化结构域多肽的活性相同. 展开更多

In the traditional views on developmental biology, the process of a mammal from a zygote to. an adult individual follows continuous changes of space and time environments and is the result of different expressions of target genes. It has long been known that this process is irreversible and the terminal differentiated adult cells, such as cardiac myocytes and neurons, will not divide and differentiate. But recent reports on the two hottest fields - cloning medicine and stem cell biology doubted these concepts. This may lead to a further understanding of the potentiality of mammal development and may provide great chances for commercial and clinical practice. 展开更多
