反相高效液相色谱法测定血浆罗哌卡因的浓度

摘要 目的 建立测定罗哌卡因血药浓度的反相高效液相色谱法。方法 血浆样品加入利多卡因作内标 ,经正己烷 二氯甲烷 异丙醇 (10 0∶5 0∶5 )提取后 ,以乙腈 - 0 .0 2 5mol/L磷酸缓冲液 (pH 3.0 ) (体积比 2 0∶80 )为流动相 ,采用Diamonsil钻石C18柱(2 0 0mm× 4 .6mm ,5 μm)分离 ,检测波长 2 2 0nm。 结果 线性范围 0 .0 4 0mg/L~ 7.5 0 0mg/L ,最低检测浓度为 0 .0 2 5mg。应用本法测定了 4只犬静脉注射罗哌卡因 (10 μmoL/kg)后不同时间的血药浓度。 结论 反相高效液相色谱法测定血浆罗哌卡因的浓度方法快速、简便 ,适用于药代动力学研究。 Objective ARP HPLC method for determination of ropivacaine in plasma was developed.Method The sample was treated by the addition of lidocaine as an internal standard,extraction with hexane dichloromethane isopropanol(100∶50∶5 volume ratio),CH 3 CN:0.025 mol/L phosphate buffer solution(pH3.0)(20∶80 volume ratio )as the mobile phase,Diamonsil C 18 (200mn×4.6mm,5μm)as the stationary phase,and the detection wavelength of 220 nm were adopted.Results The linear range was from 0.040 mg/L~7.5 mg/L with γ of 0.9994,The detection limit was 0.020 mg/L.The method described was applied to the determination of ropivacaine in the plasma of dog following mainline of ropivacaine(10μmol/kg).
