摘要
Dendrobium is a traditional Chinese plant homologous to medicine and food.It is distributed throughout China,but the differences in total flavonoid content and some biological activities of Dendrobium from different regions and growing environments have received less attention.In this study,the traditional organic solvent extraction method was used to optimize the extraction process.The stabilizing radicals 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2′-azino-bis-3-ethylbenzothiazole-6-sulfonic acid(ABTS)have been used to estimate antioxidant activity.The inhibitory effect on advanced glycation end products(AGEs)was determined by constructing an in vitro anti-glycation system of bovine serum albumin(BSA)-glucose/fructose,BSA-methylglyoxal(MGO),and BSA-glyoxal(GO).A high-performance liquid chromatography-mass spectrometry(HPLC-MS)technique was established to characterize the flavonoids and phenolic acids in Dendrobium officinale extracts.The results showed that the optimal extraction process involved dissolving the sample in 70%acetone(1:30 g/mL)and extracting at 80℃ for 30 min.Using antioxidant capacity as an indicator,and considering the total flavonoid content in Dendrobium across different environments and varieties,we selected Dendrobium officinale cultivated in greenhouses in Anhui Province(AO.3)(DPPH:IC_(50)=321.7μg/mL,ABTS:IC_(50)=95.91μg/mL)for antiglycation research.The maximum inhibition of AGEs by AO.3 total flavonoid extract was 62.16%,56.57%,and 40.05%in the three systems,respectively.These findings contribute to an in-depth understanding of the medicinal potential of Dendrobium and promote its further development and utilization in pharmaceuticals,nutraceuticals,and food additives.
