Ginseng glucosyl oleanolate exhibits anti-lung cancer activity via regulating cGAS/STING/NF-κB pathway and gut microbiota

摘要 Ginseng is renowned for its medicinal and edible properties,with ginsenosides being a key bioactive component responsible for anti-inflammatory,anti-cancer,and intestinal health maintenance.This research aimed to reveal the anti-lung cancer activities of ginseng glucosyl oleanolate(GGO)transformed from ginsenoside Ro in A549 xenograft nude mice.Through comprehensive analysis,including tumor volume,weight,histopathological staining,and Western blot,we substantiated that GGO significantly inhibited lung cancer(A549 cells)prolif-eration.GGO reduced the volume and weight of the tumors by two-thirds and one-half,respectively.It inhibited the expression of tumor maker Ki67 and promoted apoptosis by increasing the levels of proteins related to apoptosis(Bax and cleaved caspase-3).GGO enhanced anti-tumor immunity by stimulating the cGAS/STING pathway,whilst simultaneously suppressing NF-κB activation to reduce proinflammatory cytokine(TNF-αand IL-6)expression.GGO alleviated colonic damage and modulated gut microbiota associated with cellular processes,including an increase in Enterococcaceae_Enterococcus abundance and a reduction in Bacteroides and Coprobacillus.These findings provide novel insights into the anti-lung cancer effects of GGO and their role in maintaining gut health and microbial homeostasis.
