Fermented food-derived Pichia kudriavzevii A3_KGSM exhibits gastroprotective activity:A cold-restraint stress(CRS)induced mice model study

摘要 This study aimed to explore the probiotic properties and gastroprotective effects of A3_KGSM,isolated from fermented jalebi batter.Identification through 18S rRNA sequencing confirmed the yeast as Pichia kudriavzevii A3_KGSM,which showed strong in vitro probiotic activity,with high viability in acidic conditions(pH 2.0),2%bile salt,and notable auto-aggregation and hydrophobicity.Additionally,the isolate proved sensitive to various antibiotics and exhibited no hemolytic activity.It also producedα-amylase and protease enzymes that aid in breaking down starches and proteins.We then utilised the A3_KGSM strain in a study with CRS-induced mice to evaluate its gastroprotective potential.Adult male mice(27.4±0.46 g)were divided into three groups(n=6):Normal(N),ulcer control(UC),and treatment with P.kudriavzevii A3_KGSM(Y).Daily oral administration of A3_KGSM(108 cells)for 30 days significantly reduced cold-induced gastric ulcers.Macroscopic,histological,and DAPI analyses confirmed the protection of the gastric epithelium.Yeast treatment downregulated proinflammatory markers COX-2(1.18±0.31-fold),iNOS(1.69±0.11-fold),IFN-γ(1.24±0.35-fold),IFN-λ(1.57±0.27-fold),and IL-12(1.44±0.05-fold),while upregulating eNOS(2.97±0.14-fold),catalase(4.36±0.22-fold),Mn-SOD(3.42±0.12-fold),and IL-10(3.06±0.31-fold),indicating notable anti-inflammatory and antioxidant effects.These results suggest P.kudriavzevii A3_KGSM as a promising therapeutic candidate for cold-induced gastric ulcers.
