Elucidating interaction mechanisms of catechin,proanthocyanidin,and tea polyphenol with porcine serum albumin:Insights into functional properties and complex formation

摘要 This study aimed to investigate the interaction between porcine serum albumin(PSA)and three polyphenols,namely catechin(CA),proanthocyanidin(PC),and tea polyphenol(TP).The polyphenol binding equivalents of CA,PC,and TP were 50.8367±0.4584,15.0732±0.3594,and 45.2216±0.0321 mg/g,respectively.In comparison to PSA,the DPPH radical scavenging activity of CA-PSA,PC-PSA,and TP-PSA was augmented by 32.32%,33.78%,and 39.85%,respectively.And the ABTS radical scavenging activity of CA-PSA,PC-PSA,and TP-PSA was increased 75.81%,34.6%,and 67.84%,respectively.The turbidity of the complexes increased significantly,while the surface hydrophobicity decreased significantly(P<0.05).Meanwhile,the structural properties were analyzed.The result of UV-vis spectroscopy suggested that the maximum absorption peaks of the three complexes were observed a red shift.In descending order,the peaks of CA-PSA,PC-PSA,and TP-PSA were discovered to be 304 nm,296 nm,and 292 nm.The fluorescence spectroscopy showed that polyphenols exhibited a notable fluorescence quenching effect on PSA.The intensity of the fluorescence peak ranked as follows:CA-PSA<TP-PSA<PC-PSA.Based on the results of FT-IR,theα-helix and random coil content of three complexes increased,while theβ-sheet content decreased significantly(P<0.05).In addition,the SEM analysis confirmed that polyphenols changed the PSA protein structure.The molecular docking analysis revealed that CA,PC,and TP formed hydrogen bonds and hydrophobic interactions with PSA.The binding energies of the complexes,presented in descending order,were as follows:CA-PSA(-23.90 kJ/mol)<TP-PSA(-21.64 kJ/mol)<PC-PSA(-13.77 kJ/mol).The findings of this investigation provide invaluable insights into the utilization of livestock by-products.
