1株牛病毒性腹泻病毒的分离鉴定及遗传进化分析

Isolation,identification and genetic evolution analysis of a strain of bovine viral diarrhea virus

摘要 从内蒙古自治区呼和浩特市某牛场采集病例的鼻拭子样品,经处理后接种至MDBK细胞,进行病毒分离鉴定并对分离的牛病毒性腹泻病毒(bovine viral diarrhea virus,BVDV)进行5′-UTR和E2基因的克隆和遗传进化分析。结果显示,鼻拭子病料检测为BVDV阳性,接种MDBK细胞后能致细胞出现明显病变;全基因组序列比对结果显示,该分离株与GenBank上公布的BVDV(BJ1308)同源性为98.289%;基因遗传进化分析显示,该分离株为BVDV-1d基因型,将其命名为BVDV-1d-NMG;本研究分离的BVDV毒株与标准毒株的分子生物学特征和理化特性等一致,与标准毒株的抗原性相同,可导致犊牛产生典型临床症状。试验结果为内蒙古地区BVD疫苗毒株的选用、研发及BVD防控提供了参考。 Nasal swab samples from diseased cases were collected from a cattle farm in Hohhot City,Inner Mongolia Autonomous Region.After treatment,the samples were inoculated into Madin-Darby bovine kidney(MDBK)cells for virus isolation and identification.Moreover,the 5'-untranslated region(5'-UTR)and E2 gene of the isolated BVDV were cloned,and a genetic evolutionary analysis was conducted.The results demonstrated that the nasal swab samples from diseased cattle were positive for bovine viral diarrhea virus(BVDV).After inoculation onto MDBK cells,obvious cytopathic effects were observed in the cells.The whole-genome sequence alignment results showed that the homology between the isolated strain and the BVDV strain(BJ1308)published in Gen-Bank was 98.289%.The genetic evolutionary analysis of the genes indicated that the isolated strain belonged to the BVDV-1d genotype,and it was named BVDV-1d-NMG.Thecharacteristics of the BVDV strain isolated in this study are consistent with those of the standard strain.It has the same antigenicity as the standard strain and can induce typical clinical symptoms in calves.The results of this experiment provide a reference for the selection and research and development of BVD vaccine strains,as well as for the prevention and control of BVD in Inner Mongolia region.
