紫外检测器串联离子色谱法同时测定水产品中9种生物胺含量

摘要 目的建立紫外检测器串联离子色谱法(tandem ion chromatography-ultraviolet detection,IC-UV)检测水产品中9种生物胺的方法。方法鱼肉样品经0.2 mmol/L甲烷磺酸(methanesulfonic acid,MSA)溶液超声提取,乙腈沉淀蛋白,过滤膜后进样分析。9种生物胺与样品中共存离子在IonPac CS17(250 mm×4 mm,7μm)阳离子交换色谱柱上可分离。实验过程对淋洗液条件,柱温及检测器波长范围进行优化。结果9种生物胺均具有较宽线性范围,章鱼胺、酪胺、色胺在0.2~20.0 mg/L质量浓度范围,腐胺、尸胺、亚精胺、精胺在0.02~20.00 mg/L质量浓度范围,组胺、苯乙胺在0.10~20.00 mg/L质量浓度范围均具有良好的线性,相关系数(r^(2))≥0.9960,样品加标回收率为83.8%~122.9%。腐胺、尸胺、组胺、苯乙胺、亚精胺、精胺、章鱼胺、酪胺及色胺检出限分别为0.23、0.30、1.63、1.56、0.21、0.26、1.97、2.50和2.88 mg/kg。结论该方法简便、无需复杂衍生前处理、选择性好、灵敏度高,适用于水产品中9种生物胺的检测分析。 Objective To establish a method for determination of 9 kinds of biogenic amines in aquatic products by tandem ion chromatography-ultraviolet detection(IC-UV).Methods Fish samples were ultrasonically extracted with 0.2 mmol/L methanesulfonic acid solution then protein was precipitated with acetonitrile,and analyzed after filtration through a membrane.The 9 kinds of biogenic amines and coexisting ions in the samples were separated on an IonPac CS17 cation exchange chromatographic column(250 mm×4 mm,7μm).The experimental conditions,including eluent condition,column temperature and detector wavelength range,were optimized to achieve optimal performance.Results The 9 kinds of biogenic amines exhibited wide linear ranges:Octopamine,tyramine and tryptamine showed good linearity in the mass concentration range of 0.2–20.0 mg/L,putrescine,cadaverine,spermidine and spermine in the mass concentration range of 0.02–20.00 mg/L;and histamine and phenethylamine in the mass concentration range of 0.10–20.00 mg/L;with linear correlation coefficients(r^(2))≥0.9960 and satisfactory recoveries(83.8%–122.9%).The limits of detection for putrescine,cadaverine,histamine,phenethylamine,spermidine,spermine,octopamine,tyramine and tryptamine were 0.23,0.30,1.63,1.56,0.21,0.26,1.97,2.50 and 2.88 mg/kg,respectively.Conclusion This method is simple,eliminates the complex derivatization pretreatment,and offers excellent selectivity and high sensitivity,making it suitable for the detection and analysis 9 kinds of biogenic amines in aquatic products.
