摘要
低温是棉花播种出苗期主要的非生物逆境,显著抑制棉花种子萌发;而褪黑素(melatonin,MT)可积极调控植物的抗逆能力。分析MT引发处理对低温胁迫下棉花种子萌发的调控效应,可以为MT在棉花生产中的应用及棉花逆境栽培提供理论依据。以转基因抗虫棉品种冀棉30的种子为试验材料,以未引发的种子作为对照,利用发芽势和发芽率指标,对MT浸种引发处理缓解低温(15℃)胁迫的适宜浓度进行筛选,结果显示,不同浓度MT处理均能够显著提高种子发芽势,但对种子发芽率影响不显著,其中浓度为10μmol/L时种子发芽势最高。之后,采用对比试验设计,以浓度10μmol/L的MT浸种引发处理的种子作为试验组(MT),以未引发的种子作为对照,对15℃低温下种子萌发指标(发芽率、胚根长度),以及萌发过程中抗氧化酶[超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)]活性、渗透调节物质(脯氨酸、可溶性糖)含量以及丙二醛(MDA)含量进行了分析,结果显示,MT处理能够显著促进低温下种子提早萌发和胚根伸长,提高SOD、POD和CAT活性以及脯氨酸和可溶性糖含量,降低MDA含量。用浓度10μmol/L的MT溶液引发处理可有效缓解15℃低温对棉花种子萌发的抑制作用。
Low temperature is the main abiotic stress during cotton sowing and emergence period,significantly inhibiting cotton seed germination.Melatonin(MT)can actively regulate the stress resistance of plants.Analyzing the regulatory effect of MT induced treatment on cotton seed germination under low temperature stress can provide theoretical basis for the application of MT in cotton production and stress cultivation.The seeds of transgenic insect-resistant cotton variety Jimian 30 were used as the experimental materials,and the uninitiated seeds were used as controls.Using germination potential and germination rate indicators,the appropriate concentration for MT soaking treatment to alleviate low temperature(15℃)stress was screened.The results showed that different concentrations of MT treatment significantly increased seed germination potential,but had no significant effect on the seed germination rate,with the highest germination potential observed at a concentration of 10μmol/L.Then a comparative experimental design was adopted,with seeds treated with MT soaking at a concentration of 10μmol/L as the experimental group(MT)and uninitiated seeds as the control.The germination indicators(germination rate and embryonic root length)of seeds at 15℃low temperature,as well as the activities of antioxidant enzymes〔superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)〕,osmoregulatory substances(proline and soluble sugars),and malondialdehyde(MDA)content during germination were analyzed.The results showed that MT treatment could significantly promote early seed germination and embryonic root elongation at low temperatures,increase SOD,POD,and CAT activities,as well as proline and soluble sugar content,and reduce MDA content.The treatment with MT solution with a concentration of 10μmol/L can effectively alleviate the inhibitory effect of 15℃low temperature on cotton seed germination.
