摘要
为探讨干旱和盐胁迫对种间杂交种子萌发特性的影响,筛选出在萌发期具有优良抗逆性的种间杂交种,以6个白蜡种间杂交F1代种子和2个白蜡新品种为材料,采用不同浓度的PEG-6000和NaCl溶液模拟不同程度的干旱和盐胁迫,以蒸馏水为对照,通过测定种子发芽率、发芽势、发芽指数、活力指数、根长和鲜重等指标,结合主成分分析和隶属函数对8个白蜡种间杂交种子萌发期抗逆性进行综合评价。结果显示:(1)低浓度的干旱胁迫对白蜡种子萌发有一定的促进作用,发芽率、发芽势等指标会上升,随干旱程度的增加,种子萌发受到抑制,发芽率、发芽势、发芽指数等指标下降,另外干旱可以促进根生长,根长变化没有显著差异。(2)随着盐胁迫浓度的增加,种子萌发受到抑制,发芽率、发芽势、活力指数、根长等指标出现显著下降,在100 mmol·L^(-1)的盐浓度下,发芽率、发芽势等各指标与对照差异显著。(3)通过隶属函数对8个白蜡种间杂交种综合评价,可以看到在15%PEG条件下8个白蜡种间杂交种子抗旱性排序为s20>s21>s5>s1>2×8>‘青碧’>‘鲁蜡6号’>s25。(4)在NaCl含量为100 mmol·L^(-1)条件下,白蜡种子的抗盐性由高到低为s5>‘青碧’>‘鲁蜡6号’>s25>s1>2×8>s20>s21。8个品种中综合抗逆性较好的是s5。研究可为白蜡种间杂交种抗盐、抗旱种质的筛选工作提供依据。
In order to explore the effects of drought and salt stress on the germination characteristics of interspecific hybrid seeds and to screen for hybrids with excellent stress resistance during germination,this study used 6 interspecific hybrid F1 generation seeds and two new varieties of Fraxinus as materials.Different concentrations of PEG-6000 and NaCl solutions were applied to simulate varying degrees of drought and salt stress,respectively,with distilled water as the control.By measuring indicators such as seed germination rate,germination potential,germination index,vigor index,root length,and fresh weight,and combining principal component analysis with membership function analysis,the stress resistance of the 8 hybrid seeds during the germination period was comprehensively evaluated.The results indicated that low-concentration drought stress could promote the germination of Fraxinus chinensis seeds,increasing both the germination rate and germination potential.With the increase in drought intensity,seed germination was inhibited,and the germination rate,germination potential,and germination index decreased.In addition,drought promoted root growth,with no significant difference observed in root length.With the increase in salt stress concentration,seed germination was also inhibited,and indicators such as germination rate,germination potential,vigor index,and root length decreased significantly.At a salt concentration of 100 mmol·L^(-1),germination rate,germination potential,and other indicators showed significant differences compared to the control.Based on a comprehensive evaluation of 8 Fraxinus hybrid seeds using membership function analysis,the drought resistance ranking under 15%PEG conditions was as follows:s20>s21>s5>s1>2×8>‘Qingbi’>‘Lula 6’>s25.Under 100 mmol·L^(-1) NaCl conditions,the salt resistance ranking from high to low was:s5>‘Qingbi’>‘Lula 6’>s25>s1>2×8>s20>s21.Among the 8 varieties,s5 demonstrated better comprehensive stress resistance.This study provides a basis for screening salt-resistant and drought-resistant germplasm in Fraxinus chinensis interspecific hybrids.
