摘要
为提高多粘菌素B的发酵产量,降低其生产成本,经紫外诱变,筛选获得一株高产多粘菌素B的多粘芽孢杆菌P01,通过形态特征和16S rDNA序列分析,鉴定菌株P01为多粘芽孢杆菌(Paenibacillus polymyxa),并采用单因素实验和正交实验优化其发酵培养基成分。结果表明,菌株P01在最佳发酵培养基条件下,多粘菌素B的产量是优化前的1.8倍,通过菌株筛选和优化培养基大幅提高了多粘菌素B的产量,为多粘菌素B的工业化生产奠定了基础。
In order to increase the fermentation yield of polymyxin B and reduce its production cost,after UV mutagenesis,a Paenibacillus polymyxa P01 strain with high yield of polymyxin B was obtained by screening.The strain P01 was identified as Paenibacillus polymyxa by morphological characteristics and 16S rDNA sequence analysis.The components of fermentation medium were optimized by single factor test and orthogonal test.The results showed that strain P01 under optimal fermentation medium condition,its yield of polymyxin B was 1.8 times higher than before optimization,and the yield of polymyxin B was significantly increased by strain screening and optimized medium,which laid a foundation for the industrial production of polymyxin B.
